An immunometabolic signature of major depressive disorder in systemic lupus erythematosus.

OBJECTIVES: Systemic lupus erythematosus (SLE) frequently affects the central nervous system, leading to neuropsychiatric SLE (NPSLE). Major depressive disorder in SLE (SLEMDD) is the most frequent manifestation of NPSLE and is believed to arise from an immune-mediated process. However, biomarkers for SLEMDD remain lacking. The aim of this study was to identify candidate immunometabolic biomarkers associated with SLEMDD. METHODS: We analysed deep flow cytometry immune phenotyping, gut microbiota profiling, and targeted mass spectrometry-based metabolomics from 99 patients from the LUPIL-2 stu

Harnessing necroptosis via RIPK1 degradation to potentiate radio-immunotherapy in cervical cancer.

Immune checkpoint inhibitors (ICIs) are first-line therapy for cervical cancer (CC), yet their efficacy is limited to a subset of patients owing to low tumor immunogenicity. Single-cell RNA sequencing revealed that CC patients exhibiting robust immune responses following radiotherapy (RT) showed upregulation of necroptosis along with lower baseline RIPK1 expression. In vitro and in vivo experiments further illustrated that RT alone upregulates RIPK1 expression. Inspired by these findings, we developed a hafnium (Hf)-based nanoscale metal-organic framework loaded with LD4172, a RIPK1 degrader (

Self-sustaining ROS-responsive hydrogel enabled delivery of PROTAC/Ce6 nanoparticles via PDT-Epigenetic protein degradation to amplify pyroptosis for enhanced cancer immunotherapy.

Cancer immunotherapy exhibits limited efficacy in immunologically "cold" tumors due to insufficient T-cell infiltration and immunosuppressive tumor microenvironment. The efficacy of photodynamic therapy (PDT) is restricted by inadequate tumor-specific aggregation of photosensitizers and therapeutic resistance regulated by epigenetics. Herein, we engineered a light-driven, ROS-responsive hydrogel (PACL@Gel) for loading peptide-lipid scaffold nanoparticles encapsulating proteolysis targeting chimera (PROTAC) molecule and photosensitizer to enhance cancer immunotherapy. The system utilizes both e

Single-Cell RNA Sequencing of Baseline Immune Profiles After Third Vaccination Associated with Subsequent SARS-CoV-2 Infection in Naïve Individuals.

Even though vaccines protected many from infection, not all were protected, and vaccinated individuals displayed a wide range of clinical outcomes, from complete protection against infection to multiple breakthrough infections. This study aimed to identify baseline differences following identical ChAdOx1/ChAdOx1/BNT162b2 in infection-free and breakthrough-infected individuals to find molecular signatures linked to enhanced SARS-CoV-2 protection. Samples from a previous longitudinal study were analyzed, classifying subjects as 'Protected' or 'Infected' based on infection status over two years.

A Dual Adjuvant System for Intranasal Boosting of Local and Systemic Immunity for Influenza Vaccination.

Systemically vaccinated individuals against COVID-19 and influenza may continue to support viral replication and shedding in the upper airways, contributing to the spread of infections. Thus, a vaccine regimen that enhances mucosal immunity in the respiratory mucosa is needed to prevent a pandemic. Intranasal/pulmonary (IN) vaccines can promote mucosal immunity by promoting IgA secretion at the infection site. Here, we demonstrate that an intramuscular (IM) priming-IN boosting regimen with an inactivated influenza A virus adjuvanted with the liposomal dual TLR4/7 adjuvant (Fos47) enhances syst

T and B Lymphocyte Transcriptional States Differentiate between Sensitized and Unsensitized Individuals in Alpha-Gal Syndrome.

The mechanisms of pathogenesis driving alpha-gal syndrome (AGS) are not fully understood. Differences in immune gene expression between AGS individuals and non-allergic controls may illuminate molecular pathways and targets critical for AGS development. We performed immune expression profiling with RNA from the peripheral blood mononuclear cells (PBMCs) of seven controls, 15 AGS participants, and two participants sensitized but not allergic to alpha-gal using the NanoString nCounter PanCancer immune profiling panel, which includes 770 genes from 14 different cell types. The top differentially

Nonpathologic Infection of Macaques by an Attenuated Mycobacterial Vaccine Is Not Reactivated in the Setting of HIV Co-Infection.

Failure to replace Bacille Calmette-Guerin vaccines with efficacious anti-tuberculosis (TB) vaccines have prompted outside-the-box thinking, including pulmonary vaccination to elicit local immunity. Inhalational MtbΔsigH, a stress-response-attenuated strain, protected against lethal TB in macaques. While live mycobacterial vaccines show promising efficacy, HIV co-infection and the resulting immunodeficiency prompts safety concerns about their use. We assessed the persistence and safety of MtbΔsigH, delivered directly to the lungs, in the setting of HIV co-infection. Macaques were aerosol-vacci

Anti-PD-1/PD-L1 therapy for infectious diseases: learning from the cancer paradigm.

OBJECTIVES: Immune checkpoint pathways regulate optimal host immune responses against transformed cells, induce immunological memory, and limit tissue pathology. Conversely, aberrant immune checkpoint activity signifies a poor prognosis in cancer and infectious diseases. Host-directed therapy (HDT) via immune checkpoint blockade has revolutionized cancer treatment with therapeutic implications for chronic infections, thus laying the foundation for this review. METHODS: Online literature searches were performed via PubMed, PubMed Central, and Google using the keywords "immune checkpoint inhibi

Innate DNA sensing is impaired in HIV patients and IFI16 expression correlates with chronic immune activation.

The innate immune system has been recognized to play a role in the pathogenesis of HIV infection, both by stimulating protective activities and through a contribution to chronic immune activation, the development of immunodeficiency and progression to AIDS. A role for DNA sensors in HIV recognition has been suggested recently, and the aim of the present study was to describe the influence of HIV infection on expression and function of intracellular DNA sensing. Here we demonstrate impaired expression of interferon-stimulated genes in responses to DNA in peripheral blood monuclear cells from HI

Determinants of vaccine-induced resistance in animal models of pulmonary tuberculosis.

A more effective vaccine will be essential if the global tuberculosis (TB) pandemic is ever to be controlled. A large number of new tuberculosis vaccines have been developed, representing the whole range of modern strategies for vaccine formulation and delivery. There is currently no alternative to testing these new vaccines in experimental animals challenged with virulent Mycobacteriurn tuberculosis in order to assess their protective efficacy. Although such testing is being carried out in several animal species (mice, guinea pigs, rabbits), all rational models include pulmonary challenge wit

Boosting long-term immunity with bioengineered nanovaccines.

The development of vaccine platforms capable of inducing durable immunity remains a crucial frontier. While conventional vaccines offer proven efficacy, their often limited durability poses challenges for rapidly evolving pathogens and cancer immunotherapy. Recent advances in biomaterials and nanotechnology have enabled the rational design of bioengineered nanovaccines. These systems leverage engineered polymeric, lipid-based, and hybrid materials to achieve controlled antigen release, targeted lymph node delivery, and enhanced cross-presentation. Their "bioengineered" functionality allows the

Cognitive impairment in the HTLV-1 infection: a comparative study associated with functional performance.

Human T cell leukemia virus type-I (HTLV-1) infection courses with a myelopathy, the tropical spastic paraparesis (HAM/TSP). In a case-control study, we compared the neuropsychological profile and functional characteristics in two case HTLV-1-infected groups (asymptomatic and with HAM/TSP) with a control group negative for HTLV-1. Subjects were paired for age, sex, and educational features. The case group differed from control group in neuropsychological measures such as in episodic memory recall, executive functions, and fine motor dexterity measure. Individuals with HAM/TSP have more depress

Immunotoxicology of beryllium lung disease.

Beryllium induces non-caseating granulomatous inflammation in humans exposed to the metal dust or fumes in both occupational and non-occupational settings. The resulting condition, chronic beryllium disease (CBD), affects principally the lungs, lymphatics, and skin and continues to plague modern industry. Beryllium exerts several important immunotoxic effects, including induction of a beryllium-antigen specific adaptive immune response and the triggering of inflammatory and innate immune responses. Genetic susceptibility plays a role in CBD adaptive immune responses, mainly mediated through si

Potential Mechanism of Paraquat-Induced Blood-Brain Barrier Disruption: IL-17A as a Risk Accelerator for Disrupting bEnd.3 Cells Tight Junction.

Paraquat (PQ) is a globally and widely used high-residue herbicide, also regarded as a potential environmental risk factor for neurodegenerative diseases (NDs). However, how PQ exerts neurotoxicity remains a mystery. The blood-brain barrier (BBB) plays a crucial role in communicating between the central nervous system (CNS) and the peripheral blood. In this article, the core genes, which were mainly based on the potential targets for PQ, MPTP, and Diquat chemicals from the CTD database, intersected with the potential targets related to the symptoms of NDs from databases GeneCards and OMIM. The

Comprehensive analysis of ASB3 as a prognostic biomarker in hepatocellular carcinoma.

BACKGROUND: Some reports have indicated a high expression level of ASB3 in various cancers, but its role in hepatocellular carcinoma (HCC) remains elusive. METHODS: ASB3 levels and clinical features were obtained from the TCGA database. Meanwhile, the expression levels of ASB3 in tumor and paraneoplastic tissues were further verified by qRT-PCR and Imunohistochemistry (IHC). ASB3-related downstream molecular analysis was carried out with Gene Ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG). Pathways linked to ASB3 expression were identified by means of gene set enrichment

Immune Responses to Broad-Spectrum Antibiotic Treatment and Fecal Microbiota Transplantation in Mice.

Compelling evidence demonstrates the pivotal role of the commensal intestinal microbiota in host physiology and the detrimental effects of its perturbations following antibiotic treatment. Aim of this study was to investigate the impact of antibiotics induced depletion and subsequent restoration of the intestinal microbiota composition on the murine mucosal and systemic immunity. To address this, conventional C57BL/6j mice were subjected to broad-spectrum antibiotic treatment for 8 weeks. Restoration of the intestinal microbiota by peroral fecal microbiota transplantation (FMT) led to reestabl

Cytometry by time-of-flight immunophenotyping identifies a blood Sjögren's signature correlating with disease activity and glandular inflammation.

BACKGROUND: Mass cytometry has recently emerged as a promising tool for clinical research. However, few studies have demonstrated its benefit for patient stratification and biomarker identification. Primary Sjögren's syndrome (pSS) is a prototype of chronic autoimmune disease, the pathogenesis of which remains unclear and for which treatment does not exist. OBJECTIVE: This observational case-control study was designed to discover new cellular biomarkers and therapeutic targets in patients with pSS. METHODS: Forty-nine patients with pSS and 45 control subjects were enrolled for clinical evalu

Pathogenicity and immunogenicity of a vaccine strain of Listeria monocytogenes that relies on a suicide plasmid to supply an essential gene product.

Listeria monocytogenes is a bacterial pathogen that elicits a strong cellular immune response and thus has potential use as a vaccine vector. An attenuated strain, L. monocytogenes dal dat, produced by deletion of two genes (dal and dat) used for d-alanine synthesis, induces cytotoxic T lymphocytes and protective immunity in mice following infection in the presence of d-alanine. In order to obviate the dependence of L. monocytogenes dal dat on supplemental d-alanine yet retain its attenuation and immunogenicity, we explored mechanisms to allow transient endogenous synthesis of the amino acid.

Targeting fibroblast activation protein in solid tumors via LNP-mediated CAR-mRNA delivery promotes durable regression in murine models.

The therapeutic potential of chimeric antigen receptor (CAR) T-cell therapy in treating solid tumors is highly recognized, yet the complex and immunosuppressive nature of the tumor microenvironment, poor accessibility, and the instability of target antigens pose substantial challenges. Here, we present an mRNA-LNP-based therapeutic strategy that delivers mRNA encoding a fibroblast activation protein (FAP)-specific CAR to reprogram host immune cells in vivo and target cancer-associated fibroblasts within the tumor stroma. In multiple solid tumor mouse models, this approach, combined with chemot
