Neutrophils in glioblastoma: orchestrators of the tumor microenvironment and immune evasion.

Glioblastoma (GBM) is the most aggressive primary brain tumor in adults and remains refractory to current therapies. Beyond profound immunosuppression, GBM is characterized by a complex tumor microenvironment (TME) in which neutrophils have emerged as critical yet understudied regulators of tumor progression and immune evasion. Tumor-associated neutrophils (TANs) display marked functional plasticity, acquiring pro-tumor or anti-tumor phenotypes depending on microenvironmental cues. GBM recruits and reprograms infiltrating neutrophils through chemokine-driven trafficking, hypoxia, and tumor-der

Mitogen-Activated Protein Kinases: Therapeutic Signaling Catalysts in Viral Immune Evasion.

The mitogen-activated protein kinase (MAPK) pathways, ERK, JNK, and p38, are key regulators of immune responses during viral infections. These signaling cascades control cytokine production, T cell activity, and antigen presentation. However, many viruses can hijack MAPK pathways to avoid immune detection, promote their replication, and establish chronic infection. In this review, we discuss how different viruses, including HSV-1, HBV, HCMV, and SARS-CoV-2, manipulate MAPK signaling to alter host cell functions. A particular focus is given to the CD1d-iNKT cell axis, which plays a critical rol

Transcriptome Analysis Reveals Root Endophyte Serendipita indica-Mediated Growth Promotion in Wheat Is Associated with Enhanced Photosynthesis.

Serendipita indica (S. indica), a root endophytic fungus of the Sebacinaceae family, promotes growth and increases biomass accumulation in a wide range of plant species. However, the mechanism underlying S. indica-mediated growth promotion in wheat (Triticum aestivum L.), particularly its effects on chlorophyll accumulation, remains poorly understood. In this study, colonization by S. indica significantly enhanced shoot growth, biomass accumulation, plant height, fresh weight, dry weight and chlorophyll content in wheat seedlings. Transcriptome analysis revealed extensive transcriptional repro

The FaAP2-11 Transcription Factor Putatively Regulates Cell Wall Remodeling and Stress-Acclimation Responses Under Drought and Salinity Stress.

APETALA2/ethylene-responsive factor (AP2/ERF) transcription factors are key regula-tors of plant responses to abiotic stress, integrating environmental and hormonal signals into coordinated transcriptional programs. RNA-seq analysis of strawberry (Fragaria × ananassa) leaves and roots subjected to drought and salinity identified 58 differentially expressed FaAP2/ERF genes. Promoter analysis revealed cis-regulatory elements associated with abscisic acid, jasmonate, ethylene, salicylic acid, and drought-responsive signaling. Among these genes, FaAP2-11 was selected for functional characterizatio

Disease-Stage Synchronized Nanozyme Therapy for Polymicrobial Sepsis through Adaptive Catalytic and Immune Reprogramming.

Sepsis remains a leading cause of mortality because current therapies fail to address its dynamically evolving pathophysiology, in which infection, oxidative stress, and immune dysfunction emerge sequentially and interdependently. Here, we present a pH-adaptive nanozyme platform (MICP@HG) that orchestrates stage-specific antibacterial and immunomodulatory activities throughout sepsis progression. The platform integrates near-infrared imaging, catalytic therapy, and immune regulation into a single construct. In acidic infectious microenvironments, the Cu-piceatannol shell exhibits peroxidase-mi

Glutaminolysis Blockade-Empowered Bimodal Nanodepot for Spatially Complementary Sono-Thermal Ablation via PANoptosis and STING Activation Against Large Tumors.

Microwave ablation (MWA) for large hepatocellular carcinoma (HCC) is frequently compromised by non-uniform thermal distribution and immunosuppressive niche formation. To address these challenges, we engineered an injectable bimodal nanodepot (TCPD) via catalytic crosslinking of dopamine-modified hyaluronic acid by microwave thermal converter cesium-doped Prussian blue, enabling co-delivery of glutaminase 1 inhibitor Telaglenastat and sonosensitizer Chlorin e6. Under combined MWA and ultrasound irradiation, TCPD nanodepot disrupts cellular antioxidant defenses and bioenergetic pathways through

Repurposing metformin as a dual-function agent to combat E. coli-induced mastitis: Mechanistic insights into biofilm dispersion and AMPK/SIRT1-mediated NF-κB inhibition.

Escherichia coli-induced bovine mastitis represents a major challenge in dairy production due to the prevalence of multidrug-resistant strains. This study repurposes metformin as a dual-function agent that simultaneously targets bacterial virulence and host inflammation. Epidemiological surveillance identified phylogroup B1 as the most prevalent (52.5%) and resistant E. coli lineage. Against a representative B1 strain, metformin potently inhibited and dispersed bacterial biofilms, and synergized with conventional β-lactam antibiotics. Bacterial transcriptomics revealed metformin downregulated

Palmitoylethanolamide (PEA) regulates cell cycle progression and promotes an anti-inflammatory transcriptomic signature in C2C12 skeletal muscle cells.

Palmitoylethanolamide (PEA) is an endogenous lipid mediator with immunomodulatory actions, yet its effects in skeletal muscle remain poorly defined. We examined whether PEA influences myogenesis and profiled the acute transcriptomic response of differentiated C2C12 myotubes to 10 μM PEA. PEA decreased myotube number (90.3 ± 10.6 vs. 112.6 ± 10.1 control) while increasing nuclear fusion index (37.8 ± 5.7% vs. 30.7 ± 3.2%); myotube area was unchanged. In myoblasts, 24 h PEA increased G0/G1 (48.2 ± 1.2% vs. 42.3 ± 1.9%) and reduced S-phase (21.7 ± 1.2% vs. 25.5 ± 1.2%), consistent with G1 arrest.

Countering the Faltering Ability of Warming Oceans to Absorb CO2 by Adding Silica and Other Nanoparticles.

Diatoms play a central role in marine CO2-fixation and the marine food web, yet their survival is increasingly threatened by intensifying marine heatwaves. Here, we demonstrate that engineered nanoparticles (ENPs) can significantly enhance diatom thermotolerance. The diatom Skeletonema costatum pre-exposed to 0.1 mg/L SiO2 NPs exhibited significantly higher survival rates (from 43 to 64-71%), increased biomass, enhanced photosynthetic pigment content, and a bolstered antioxidant system compared to untreated, heat-stressed controls. Multiomics analyses revealed that SiO2 NPs precondition the ce

Relationship between DNA methylation and ethylene in plants: A review.

DNA methylation, as a central mechanism of epigenetic regulation, plays a key role in coordinating plant growth and stress responses. Simultaneously, the gaseous phytohormone ethylene is also a crucial regulator in these processes. Both form a metabolic competition through S-adenosyl-L-methionine (SAM) and mutually regulate each other at the level of gene expression. Studies demonstrate that DNA methylation functions to either enhance or block ethylene synthesis and ethylene signal transduction, thereby regulating a spectrum of developmental processes including seed development, root growth, f

Closing the CO2-to-glycolic acid loop through efficient methanol bioconversion enabled by cofactor-neutral metabolic rewiring.

Chemo-biological valorization of CO2 into value-added chemicals, such as the biodegradable plastic monomer glycolic acid (GA), represents a promising frontier in sustainable manufacturing. However, the realization of this synergistic loop is hindered by the low carbon efficiency of C1 bioconversion. Herein, we address this challenge by engineering an efficient methanol bioconversion platform that selectively upgrades CO2-derived methanol into GA. To address metabolic redox constraints inherent to methanol assimilation that severely limit carbon yield, we reprogrammed Komagataella phaffii by co

Targeting mitochondrial dysfunction to quell neuroinflammation in central nervous system (CNS): a new strategy for treating CNS disease with nanomedicines.

Neuroinflammation has emerged as an important pathogenic factor in central nervous system (CNS) disease, including various neurodegenerative diseases, brain injuries, and autoimmune conditions, affecting approximately 3 billion people worldwide. Mitochondrial dysfunction within neurovascular unit (NVU) cells not only initiates neuronal damage and blood-brain barrier (BBB) disruption but also critically reprograms immunometabolism, shifting microglia and infiltrating immune cells toward a pro-inflammatory, glycolysis-dominant state while impairing anti-inflammatory, oxidative phosphorylation-de

Placental programming in maternal malnutrition as a crucial determinant of offspring health: A critical-interpretative narrative review.

Maternal malnutrition during pregnancy is a critical environmental factor that can reprogram placental development and, consequently, influence fetal growth and offspring health across the lifespan. According to the Developmental Origins of Health and Disease (DOHaD) theory, adverse intrauterine conditions - such as nutrient deficiencies or excesses - may induce morphological and functional alterations in the placenta, thereby increasing the risk of chronic noncommunicable diseases (NCDs) in adulthood. This critical-interpretative narrative review compiles evidence on how maternal nutritional

From ribosomopathies to therapeutic targets: ribosomal alterations in pediatric leukemogenesis and tumorigenesis.

Ribosomes are central to cellular growth and proteome maintenance, and cancer cells frequently depend on increased ribosome biogenesis and altered translation to sustain proliferation, stress tolerance, and metabolic rewiring. Paradoxically, inherited "ribosomopathies" caused by germline defects in ribosomal proteins or ribosome biogenesis factors present with tissue hypoplasia and bone marrow failure early in life, yet confer substantially increased lifetime risk of myelodysplastic syndrome (MDS), acute myeloid leukemia (AML), and selected solid tumors. In parallel, somatic alterations affect

Calcific Aortic Valve Disease: Mechanism and Future Therapeutic Strategies.

Calcific aortic valve disease (CAVD) is an active pathological process driven by complex cellular and molecular mechanisms rather than passive aging. The disease is characterized by endothelial dysfunction, lipid infiltration, inflammation, extracellular matrix remodeling, and osteogenic differentiation of valvular interstitial cells, ultimately leading to hydroxyapatite deposition and progressive valve calcification. Key signaling pathways, including Notch, Wnt/β-catenin, BMP2, and TGF-β, play critical roles in osteogenic reprogramming, while inflammatory cytokines such as IL-6, IL-1β, and TN

A human cytokine response atlas to reconstruct underlying gene regulatory networks.

Circulating cytokines encode immune state, yet their pleiotropy and cell-type specificity make constructing a unified atlas of immune cell responses to them challenging. Here, I transformed a single-cell atlas of approximately 10 million human peripheral blood mononuclear cells from 12 donors exposed to 90 cytokines into a multiscale model of cytokine response. A GPU-accelerated implementation of dimension-scalable single-cell perturbation integration network (D-SPIN) allowed for the creation of a signed, directed model of 9.6 million cells, 1,634 immune regulatory genes, and 40 cellular progr

Epstein-Barr Virus-Mediated Tumorigenesis in Nasopharyngeal Carcinoma.

This chapter focuses on Epstein-Barr virus (EBV)-mediated tumorigenesis in nasopharyngeal carcinoma (NPC). EBV-mediated epithelial cell transformation is a crucial early event in the development of NPC. NPC and other EBV-associated lymphoepithelioma-like carcinoma have latency II program and a genomic landscape characterized by NF-κB pathway activating mutations, implicating the presence of viral latency program-specific oncogenesis. After the establishment of persistent EBV infection in nasopharyngeal epithelial cells, latency II viral genes (such as EBNA1, LMP1) are expressed to drive malign

Phosphate binders suppress glutaminase activity for treatment of cirrhosis and hepatic encephalopathy murine models.

Hyperactivated glutaminase1 (GLS1) promotes the progression of cirrhosis via the reprogramming of hepatic stellate cells (HSCs). Hepatic encephalopathy (HE), the main complication of cirrhosis characterized by abnormal ammonia metabolism, is also associated with increased glutaminase activation in intestinal epithelial cells (IECs). The enzymatic activity of glutaminase depends on inorganic phosphate (Pi). In this study, a retrospective study of serum Pi levels was performed in 185 cirrhosis-HE patients. The pharmacology and pharmacodynamics of Pi binders (sevelamer and lanthanum carbonate) we

The Rab7 C-terminal intrinsically disordered region is a key determinant of development and pathogenicity in Aspergillus flavus.

Rab7 GTPases coordinate late endosome maturation, vesicle trafficking, and vacuole biogenesis across eukaryotes, yet the functional contribution of the Rab7 C-terminal tail to organismal physiology remains incompletely defined. Here we present the crystal structure of Aspergillus flavus Rab7 (AflRab7), which reveals a well-ordered GTPase core, whereas its C-terminal tail is conserved across Aspergillus species yet lacks interpretable electron density, consistent with an intrinsically disordered region (IDR). Biochemical assays show that deletion of this IDR does not alter basal GTP hydrolysis,

A Pilot Proteomics Analysis of Mycobacterium tuberculosis Lacking a Putative Short-Chain Dehydrogenase Rv0148.

Mycobacterium tuberculosis (Mtb) is an intracellular pathogen that survives in host cells by resisting hostile antimicrobial defenses. However, the molecules and mechanisms that contribute to Mtb's intracellular survival are not fully understood. We have previously reported that Rv0148, a putative short-chain dehydrogenase/reductase, plays a significant role in Mtb stress response and virulence in in vitro and in vivo models. To further understand the role of Rv0148 in regulating global functions of Mtb, we performed comparative pilot proteomic analysis between Δrv0148 mutant, and wild-type (W
