Two unique biological response-modifier glucans beneficially regulating gut microbiota and faecal metabolome in a non-alcoholic steatohepatitis animal model, with potential applications in human health and disease.

OBJECTIVE: The gut microbiome and its metabolites are influenced by age and stress and reflect the metabolism and health of the immune system. We assessed the gut microbiota and faecal metabolome in a static animal model of non-alcoholic steatohepatitis (NASH). DESIGN: This model was subjected to the following treatments: reverse osmosis water, AFO-202, N-163, AFO-202+N-163 and telmisartan treatment. Faecal samples were collected at 6 and 9 weeks of age. The gut microbiome was analysed using 16S ribosomal RNA sequences acquired by next-generation sequencing, and the faecal metabolome was anal

High tolerance to instrument drifts by differential chemical isotope labeling LC-MS: A case study of the effect of LC leak in long-term sample runs on quantitative metabolome analysis.

Metabolomics study of a biological system often involves the analysis of many comparative samples over a period of several days or weeks. This process of long-term sample runs can encounter unexpected instrument drifts such as small leaks in liquid chromatography-mass spectrometry (LC-MS), degradation of column performance, and MS signal intensity change. A robust analytical method should ideally tolerate these instrumental drifts as much as possible. In this work, we report a case study to demonstrate the high tolerance of differential chemical isotope labeling (CIL) LC-MS method for quantita

Systemic Metabolomic Profiling of Acute Myeloid Leukemia Patients before and During Disease-Stabilizing Treatment Based on All-Trans Retinoic Acid, Valproic Acid, and Low-Dose Chemotherapy.

Acute myeloid leukemia (AML) is an aggressive malignancy, and many elderly/unfit patients cannot receive intensive and potentially curative therapy. These patients receive low-toxicity disease-stabilizing treatment. The combination of all-trans retinoic acid (ATRA) and the histone deacetylase inhibitor valproic acid can stabilize the disease for a subset of such patients. We performed untargeted serum metabolomic profiling for 44 AML patients receiving treatment based on ATRA and valproic acid combined with low-dose cytotoxic drugs (cytarabine, hydroxyurea, 6-mercaptopurin) which identified 88

Elevated CO2 ameliorates O3-induced stress and enhances growth, metabolomic, and yield attributes of two wheat (Triticum aestivum L.) cultivars under fully open-air field conditions.

Two wheat cultivars (PBW-154, WH-1105), grown in the free-air CO2 and O3 enrichment (FAOCE) facility, were exposed to alone or combined elevated CO2 (ECO2) and/or elevated O3 (EO3). EO3 exposure (+ 20 ppb above ambient) caused a reduction in growth and yield in both wheat cultivars. ECO2 exposure (550 ppm) enhanced plant growth, photosynthetic rate (A), and yield. Total phenol, flavonoid, gallic, ferulic, caffeic acid, and kaempferol content showed significant induction in leaves of O3-exposed cultivars at the flowering phase. Significant increases in glucose, sucrose, d-galactose, and malic a

Circulating Amino Acid Profiles in Adults with Abnormal Body Mass Index: Associations with Triglycerides, HDL Cholesterol, LDL Cholesterol, and Total Cholesterol in an Exploratory Cross-Sectional Metabolomic Pilot Study.

BACKGROUND/OBJECTIVES: Circulating amino acids are not only markers of nutritional status; in experimental and interventional models they have been linked to hepatic lipogenesis, lipoprotein assembly, mitochondrial fatty acid oxidation, and bile acid conjugation, which makes them plausible candidate correlates of obesity-related lipid dysregulation. Despite this, most metabolomic studies of excess adiposity have focused either on a single lipid parameter-typically triglycerides-or only on branched-chain amino acids (BCAAs). This pilot study was designed to generate hypotheses about these assoc
