Bile-acid-mediated decrease in endoplasmic reticulum stress: a potential contributor to the metabolic benefits of ileal interposition surgery in UCD-T2DM rats.

Post-operative increases in circulating bile acids have been suggested to contribute to the metabolic benefits of bariatric surgery; however, their mechanistic contributions remain undefined. We have previously reported that ileal interposition (IT) surgery delays the onset of type 2 diabetes in UCD-T2DM rats and increases circulating bile acids, independently of effects on energy intake or body weight. Therefore, we investigated potential mechanisms by which post-operative increases in circulating bile acids improve glucose homeostasis after IT surgery. IT, sham or no surgery was performed on

A primary dysregulation in the immunoregulatory role of the intestinal mucosal epithelial cell in inflammatory bowel disease pathogenesis? Biology of inflammatory response as tissue pattern entities in Crohn's versus ulcerative colitis.

Within a framework of dual involvement of mucosa and submucosa on the one hand, and of the muscularis propria of the bowel wall on the other, it might be valid to consider involvement of the vascular supply as the essential means in itself of not only causing the morphologic lesions in inflammatory bowel disease, but also especially in accounting for persisting patterns of inflammatory response both in ulcerative colitis and in Crohn's disease. Inflammatory bowel disease as a group constitutes a spectrum of biologic and pathobiologic manifestations in terms not only of inflammatory involvement

Emerging infectious diseases: vulnerabilities, contributing factors and approaches.

We live in an ever more connected global village linked through international travel, politics, economics, culture and human-human and human-animal interactions. The realization that the concept of globalization includes global exposure to disease-causing agents that were formerly confined to small, remote areas and that infectious disease outbreaks can have political, economic and social roots and effects is becoming more apparent. Novel infectious disease microbes continue to be discovered because they are new or newly recognized, have expanded their geographic range, have been shown to caus

Microbiome varies by body site in the yellow mud turtle, Kinosternon flavescens, from the Chihuahuan Desert.

Microbiomes are important to the ecology and evolution of their associated animal hosts. These important functions range broadly from individual animal health to population-level adaptations. Turtle-associated microbiomes are under increasing investigation, given turtles' conservation status and the unique natural history of the shell as an evolutionary-developmental novelty. Many components of the turtle-microbiome interaction remain understudied, including how microbial communities assemble based on host and environmental factors. Here, we hypothesized that age, habitats, and body sites woul

Paired genomic and proteomic analysis of Acinetobacter towneri from waste sludge reveals an extensive repertoire of enzymes for breakdown of hydrocarbons.

Crude oil, primarily consisting of alkanes, and plastic derivatives are integral to modern society, yet their widespread use has led to persistent pollution that devastates ecosystems worldwide. Enzymatic and microbial biodegradation are highly desirable to become alternatives to conventional methods for the control of these pollutants in terrestrial and aquatic environments. Consequently, investigating the enzymatic machinery of specialized microbes that thrive in contaminated areas and possess the catabolic potential to degrade alkanes and plastics is of considerable interest. In this study,

The role of gut microbiota in osteoporosis: underlying mechanisms, clinical associations, and emerging biomaterials.

Osteoporosis is a prevalent metabolic skeletal disorder characterized by reduced bone mass, deteriorated trabecular microarchitecture, and increased fragility fracture risk, imposing substantial global medical, social and economic burdens. Current first-line antiresorptive and anabolic therapeutics are severely constrained by long-term adverse reactions, insufficient patient adherence, and compromised bone microenvironment remodeling capacity, leaving a large unmet clinical demand for multitargeted and translational interventions. The gut-bone axis has been recognized as a core interorgan regu

Assessing the effects of ex vivo hormonal exposure on oxidative responses in equine leukocytes: A preliminary study.

Breed differences exist between horses and ponies in circulating concentrations of several hormones, notably ACTH and insulin. These hormones regulate stress and metabolic responses, but in other species, they also impact leukocyte oxidant responses. The effects of these hormones on equine leukocytes have not been evaluated to date. If equine leukocytes are similarly regulated, breed differences in increased plasma hormone concentrations or altered sensitivity to them at the leukocyte level could result in breed-related differences in oxidant responses or oxidative status. The objective of thi

Engineered Nanotechnology: An Effective Therapeutic Platform for the Chronic Cutaneous Wound.

The healing of chronic wound infections, especially cutaneous wounds, involves a complex cascade of events demanding mutual interaction between immunity and other natural host processes. Wound infections are caused by the consortia of microbial species that keep on proliferating and produce various types of virulence factors that cause the development of chronic infections. The mono- or polymicrobial nature of surface wound infections is best characterized by its ability to form biofilm that renders antimicrobial resistance to commonly administered drugs due to poor biofilm matrix permeability

Genomic Insights into Adaptations of Trimethylamine-Utilizing Methanogens to Diverse Habitats, Including the Human Gut.

Archaea of the order Methanomassiliicoccales use methylated amines such as trimethylamine as the substrates for methanogenesis. They form two large phylogenetic clades and reside in diverse environments, from soil to the human gut. Two genera, one from each clade, inhabit the human gut: Methanomassiliicoccus, which has one cultured representative, and "Candidatus Methanomethylophilus," which has none. Questions remain regarding their distribution across biomes and human populations, their association with other taxa in the gut, and whether host genetics correlate with their abundance. To gain

Lactoferrin and Its Derived Peptides: An Alternative for Combating Virulence Mechanisms Developed by Pathogens.

Due to the emergence of multidrug-resistant pathogens, it is necessary to develop options to fight infections caused by these agents. Lactoferrin (Lf) is a cationic nonheme multifunctional glycoprotein of the innate immune system of mammals that provides numerous benefits. Lf is bacteriostatic and/or bactericidal, can stimulate cell proliferation and differentiation, facilitate iron absorption, improve neural development and cognition, promote bone growth, prevent cancer and exert anti-inflammatory and immunoregulatory effects. Lactoferrin is present in colostrum and milk and is also produced

An exhaustive cell-based screen coupled with an intracellular-induced lux-based reporter identified bioactive molecules that inhibit host cell infection by intracellular pathogens.

INTRODUCTION: Antibiotic resistance poses a critical and escalating global health crisis, leading to higher morbidity and mortality associated with infectious diseases. This problem is significantly exacerbated by intracellular bacterial pathogens, which are often shielded from conventional antibiotics and foster the emergence of persister populations. Recently, host-directed therapy (HDT) has been emerging as a promising strategy that aims to modulate host cellular processes or immune responses to enhance bacterial clearance. Nonetheless, the inherent complexity of host biology makes identify

Switching cell-penetrating peptides from "deliverer" to "enforcer" by hydrophobic amino acid end-tagging to combat intracellular bacterial infections.

The presence of recalcitrant intracellular bacteria (ICB) and antimicrobial resistance poses a significant challenge to global public health. Antimicrobial peptides (AMPs) and cell-penetrating peptides (CPPs) play irreplaceable roles in combating multidrug-resistant (MDR) bacteria and drug delivery, respectively, but how to combine the advantages of both to design peptide-based biomaterials to fight ICB is still a formidable challenge. Here we constructed a miniature peptide library containing thirty AMPs with dual functions of antimicrobial and cell-penetrating properties based on CPP transac

African Swine Fever Virus Envelope Glycoprotein CD2v Interacts with Host CSF2RA to Regulate the JAK2-STAT3 Pathway and Inhibit Apoptosis to Facilitate Virus Replication.

African swine fever (ASF) is a highly infectious disease caused by the African swine fever virus (ASFV) in swine. It is characterized by the death of cells in infected tissues. However, the molecular mechanism of ASFV-induced cell death in porcine alveolar macrophages (PAMs) remains largely unknown. In this study, transcriptome sequencing of ASFV-infected PAMs found that ASFV activated the JAK2-STAT3 pathway in the early stages and apoptosis in the late stages of infection. Meanwhile, the JAK2-STAT3 pathway was confirmed to be essential for ASFV replication. AG490 and andrographolide (AND) inh

Suppression of DC-SIGN and gH Reveals Complex, Subset-Specific Mechanisms for KSHV Entry in Primary B Lymphocytes.

Kaposi sarcoma-associated herpesvirus (KSHV) is the causative agent of multiple cancers in immunocompromised patients including two lymphoproliferative disorders associated with KSHV infection of B lymphocytes. Despite many years of research into the pathogenesis of KSHV associated diseases, basic questions related to KSHV molecular virology remain unresolved. One such unresolved question is the cellular receptors and viral glycoproteins needed for KSHV entry into primary B lymphocytes. In this study, we assess the contributions of KSHV glycoprotein H (gH) and the cellular receptor DC-SIGN to

Temporal shifts in antibiotic resistance elements govern phage-pathogen conflicts.

Bacteriophage predation selects for diverse antiphage systems that frequently cluster on mobilizable defense islands in bacterial genomes. However, molecular insight into the reciprocal dynamics of phage-bacterial adaptations in nature is lacking, particularly in clinical contexts where there is need to inform phage therapy efforts and to understand how phages drive pathogen evolution. Using time-shift experiments, we uncovered fluctuations in Vibrio cholerae's resistance to phages in clinical samples. We mapped phage resistance determinants to SXT integrative and conjugative elements (ICEs),

Intestinal inflammation alters mucosal carbohydrate foraging and monosaccharide incorporation into microbial glycans.

Endogenous carbohydrates released from the intestinal mucus represent a constant source of nutrients to the intestinal microbiota. Mucus-derived carbohydrates can also be used as building blocks in the biosynthesis of bacterial cell wall components, thereby influencing host mucosal immunity. To assess the uptake of endogenous carbohydrates by gut microbes in healthy mice and during intestinal inflammation, we applied azido-monosaccharides that can be tracked on bacterial cell walls after conjugation with fluorophores. In interleukin-10 deficient mice, changes in the gut microbiota were accompa

Bronchial Epithelial Cells on the Front Line to Fight Lung Infection-Causing Aspergillus fumigatus.

Aspergillus fumigatus is an environmental filamentous fungus that can be pathogenic for humans, wherein it is responsible for a large variety of clinical forms ranging from allergic diseases to life-threatening disseminated infections. The contamination occurs by inhalation of conidia present in the air, and the first encounter of this fungus in the human host is most likely with the bronchial epithelial cells. Although alveolar macrophages have been widely studied in the Aspergillus-lung interaction, increasing evidence suggests that bronchial epithelium plays a key role in responding to the
