Multiple cerebral cavernous malformations associated with extracranial mesenchymal anomalies.

Cerebral cavernous malformations (CCM) are common hamartous dysplasias characterized by abnormally dilated vascular channels. CCM mostly occur sporadically, and multiple occurrence of CCM is highly suggestive of a genetic origin of the disorder. Typical clinical symptoms are seizures, hemorrhages, focal neurological deficits, and headaches. Three genes have so far been described that are responsible for most cases of familial CCM and more than half of the sporadic cases with multiple CCM (CCM1-3). The coincidence of CCM and other vascular anomalies has been described before. The present review

Limbic neurogenesis/plasticity in the R6/2 mouse model of Huntington's disease.

Huntington's disease is an inherited neurodegenerative condition characterized by movement disorders, and mood and cognitive disturbance. Mammalian neurogenesis persists into adulthood in the subventricular zone and dentate gyrus of the hippocampus. Neurogenesis is abnormal in the dentate gyrus in the R6/2 transgenic mouse model of Huntington's disease. We have now found that the number of immature neurons (doublecortin-positive cells) is markedly reduced in the piriform and insular cortex but not in the temporal germinal layer or caudal subventricular zone of R6/2 mice. Furthermore, numbers o

Hypoparathyroidism-retardation-dysmorphism syndrome in a girl: A new variant not caused by a TBCE mutation--clinical report and review.

Hypoparathyroidism-retardation-dysmorphism (HRD) or Sanjad-Sakati syndrome (SSS) (OMIM 241410) is a rare autosomal recessive (AR) inherited condition, characterized by congenital hypoparathyroidism (hypoPTH), retardation, seizures, and a typical facial dysmorphism, consisting of prominent forehead, deep-set eyes, and abnormal external ears. This disorder has been mapped to the long arm of chromosome 1 (1q42-q43) and mutations in the gene coding for tubulin-specific chaperone E (TBCE) have been identified as the cause of the disease. Mutations in the same gene were also reported in patients wit

Bardet-Biedl syndrome type 4 (BBS4)-null mice implicate Bbs4 in flagella formation but not global cilia assembly.

The functions of the proteins encoded by the Bardet-Biedl syndrome (BBS) genes are unknown. Mutations in these genes lead to the pleiotropic human disorder BBS, which is characterized by obesity, retinopathy, polydactyly, renal and cardiac malformations, learning disabilities, and hypogenitalism. Secondary features include diabetes mellitus and hypertension. Recently, it has been suggested that the BBS phenotypes are the result of a lack of cilia formation or function. In this study, we show that mice lacking the Bbs4 protein have major components of the human phenotype, including obesity and

[Expression of survivin in different stages of carcinogenesis and progression of breast cancer].

BACKGROUND & OBJECTIVE: Survivin is one of the newly identified apoptosis inhibitor; it can block the cell apoptosis by inhibiting the function of the enzyme caspase-3 and caspase-7. Present studies indicate that survivin is overexpressed in malignant tumor. This current study was designed to investigate the effects of survivin in tumorigenesis and progression of breast carcinoma through observing the expression of survivin in the tissue of normal mammary, cystic hyperplasia mammary, atypical hyperplasia mammary and breast carcinoma. METHODS: The expression of survivin in the tissue of normal

Cloning and characterization of an inversion breakpoint at 6q23.3 suggests a role for Map7 in sacral dysgenesis.

Here we report on a male patient with sacral dysgenesis (SD) and constitutional pericentric inversion of chromosome 6 (p11.2;q23.3). SD is a heterogeneous group of congenital anomalies with complex genetic etiology. Previously, a patient with sacral abnormalities and an interstitial deletion of 6q23-->q25 region has been described. We speculated that a susceptibility gene for SD lies in 6q23.3 region (disrupted in both patients), and therefore, cloning of the breakpoint in our patient would lead to the identification of the disrupted gene. We performed FISH analysis followed by Southern blot a

Developmentally regulated expression of Survivin in human pancreatic islets.

Islet cell apoptosis plays a role in both normal development of the endocrine pancreas and in the pathogenesis of Type I and Type II diabetes. The molecular mechanisms regulating islet cell death and survival in both normal and pathological situations are still not completely elucidated. The inhibitor of apoptosis protein (IAP) Survivin has an anti-apoptotic function mediated by several mechanisms; these include inhibiting caspase 3 and caspase 7. Survivin expression has been reported in human fetal islets and it may play a role in pancreatic remodeling and islet homeostasis. However, there ar

BBS4 is a minor contributor to Bardet-Biedl syndrome and may also participate in triallelic inheritance.

Bardet-Biedl syndrome (BBS) is an uncommon multisystemic disorder characterized primarily by retinal dystrophy, obesity, polydactyly, and renal dysfunction. BBS has been modeled historically as an autosomal recessive trait, under which premise six independent BBS loci (BBS1-BBS6) have been mapped in the human genome. However, extended mutational analyses of BBS2 and BBS6, the first two BBS genes cloned, suggest that BBS exhibits a more complex pattern of inheritance, in which three mutations at two loci simultaneously are necessary and sufficient in some families to manifest the phenotype. We

Functions of Vrp1p in cytokinesis and actin patches are distinct and neither requires a WH2/V domain.

Vrp1 (verprolin, End5) is a Saccharomyces cerevisiae actin-associated protein and is related to mammalian Wiskott-Aldrich syndrome protein (WASP)-interacting protein (WIP). Vrp1-deficient (vrp1 Delta) cells are inviable at high temperature, have partially depolarized cortical actin patches and have defects in both actomyosin ring-dependent and Hof1 (Cyk2)-dependent pathways of cytokinesis. We demonstrate here that N-Vrp1(1-364) and C-Vrp1(364-817) are each sufficient to restore viability, actomyosin ring constriction and Hof1 localization at 37 degrees C to vrp1 Delta. C-Vrp1, like Vrp1, parti

Ubiquitin-positive degenerating neurites in the brainstem in Parkinson's disease.

Characteristic ubiquitin-positive, tau-negative, degenerating neurites were present in brainstem regions known to be involved in idiopathic Parkinson's disease. Corresponding changes were entirely absent from controls and from the brainstems obtained from patients who had died with Alzheimer's disease, motor neuron disease and multiple system atrophy. In Parkinson's disease cases degenerating neurites were particularly striking in the dorsal motor nucleus of the vagus. In this nucleus the density of degenerating neurites was inversely related to the duration of Parkinson's disease symptoms. So

Hypoxia Reprograms a Ciliary Scaffold into a Metabolic-Epigenetic Trafficking Hub via a Succinate-Driven DNMT3L-ASXL2 Axis.

Environmental hypoxia is a major yet mechanistically unresolved driver of male infertility. While the canonical hypoxia-inducible factor (HIF) and reactive oxygen species (ROS) pathways dominate current models, how chronic oxygen deprivation persistently rewires spermatogenic epigenetics and flagellar architecture remains unknown. Here, we identify CEP162-traditionally defined as a static ciliary transition zone scaffold-as a dynamic oxygen-sensitive signaling hub that orchestrates a pathogenic metabolic-epigenetic-trafficking axis. In human asthenozoospermia and murine hypoxia models, CEP162

Integrated transcriptomic and metabolomic ecotoxicity assessment of earthworms exposed to ZnO and Mn-doped ZnO nanomaterials.

INTRODUCTION: Zinc oxide (ZnO)-containing nanomaterials may elicit toxic responses in environmental organisms such as earthworms. In this study, we investigate the time-dependent gene expression and metabolomic response in earthworms exposed to ZnO and Mn-doped ZnO (ZnO:Mn) nanomaterials (both at 850 mg/kg soil) and MnCl2. METHODS: Earthworms were exposed to ZnO nanomaterial (850 mg/kg soil) for 2, 3, 4, or 7 days, to ZnO:Mn nanomaterial (850 mg/kg soil) for 2, 4, 7, or 14 days, and to MnCl2 (45 mg/kg soil) for 14 days. SELENOT, a Key Membrane-Bound Selenoprotein, Mediates Selenium's Protection Against Lead-Induced Renal Aging in Chickens by Modulating Inflammation and Autophagy.

Lead (Pb) pollution is a global public health issue, yet the mechanisms underlying Pb-induced kidney aging remain poorly understood. Selenium (Se), an essential trace element critical for kidney health, primarily exerts its physiological roles via selenoproteins; among them, membrane-bound selenoproteins strategically localized at the endoplasmic reticulum and plasma membrane, have emerged as potential molecular links in Se-mediated protection. In this study, Hyline chicken models (treated with 350.00 mg/L Pb or/and 1.00 mg/kg Se) and HK-2 cell models (treated with 200 μM Pb or/and 2.5 μM Se)

Pretreatment Chromosomal Instability Correlates With Radiation Sensitivity in Squamous Cell Cancers.

PURPOSE: Continuous chromosome missegregation over successive mitotic divisions, known as chromosomal instability (CIN), is common in cancer. Though it has been associated with treatment resistance and poor prognosis, increasing CIN above a maximally tolerated threshold leads to cell death because of loss of essential chromosomes. Because radiation causes CIN, we hypothesize that pre-existing CIN sensitizes tumor cells to radiation therapy. METHODS AND MATERIALS: We induced mitotic defects that lead to CIN in FaDu (head and neck cancer, HNC) and HeLa (cervical) cells by knocking down or overe

Real-world pharmacovigilance signals of antibody-drug conjugates: reporting patterns by payload, linker, target, and sex in FAERS.

Antibody-drug conjugates (ADCs) have transformed cancer therapy by delivering potent cytotoxic agents directly to tumor cells, yet their complex structure creates unique safety challenges. This retrospective pharmacovigilance study utilized the FDA Adverse Event Reporting System (FAERS) database from 2004Q1 to 2025Q4 to characterize adverse-event reporting patterns of 15 approved ADCs across 100,536 ADC-PT-report records derived from 34,041 unique FAERS reports. Using disproportionality algorithms, we identified heterogeneous adverse-event reporting patterns across ADCs grouped by payload, lin
