ERP evidence for semantic facilitation of tool-grasping by tool-using actions.

Research has shown that the human visual system recognizes manipulated objects more efficiently than faces, numbers, or text, largely due to their manipulability. According to the two-action system theory, manipulative actions can be categorized into grasping actions and using actions, providing a framework for understanding how these two types of actions are represented. For a long time, researchers assumed that correctly grasping an object ensured its appropriate use. However, the concept of 'second-order planning' challenges this view, suggesting that using actions can influence the selecti

Time Course of the Development and Loss of Delta-9-tetrahydrocannabinol Tolerance: Effects on Hypothermia and Spontaneous Locomotor Activity in Mice.

Deregulation of cannabis use is gradually expanding in Europe and the United States. However, the biological processes driving tolerance to delta-9-tetrahydrocannabinol (Δ9-THC), the main psychoactive component of cannabis, remain unclear. Thus, this study aimed to investigate the mechanisms and time course of tolerance development and loss to Δ9-THC in mice. Male ICR mice (7 weeks old) were administered Δ9-THC once daily for 3 days and then divided into three groups according to the washout period (3-, 10-, and 17-day washout groups). After each washout, changes in body temperature and locomo

Partial TG6 loss of function causes motor deficits in male mice.

Mutations in Transglutaminase 6 (TG6) have been linked to a genetic form of spinocerebellar ataxia, namely SCA35. In recent years, several mutations associated with this disease have been identified. While some of them did not alter TG6 enzymatic activity, others induced a dominant-negative loss-of-function and altered subcellular localization. We previously observed that mutations identified in patients, which showed detrimental effects on neuronal viability in vitro, including mislocalization and activation of the unfolded protein response, were consistently characterized by a loss of TG6 en

Urine protein/osmolality and creatinine/osmolality ratios for assessing proteinuria and motor function in children with spinal muscular atrophy.

BACKGROUND: Spinal muscular atrophy (SMA) is characterized by the degeneration of anterior horn motor neurons. While nusinersen has altered the natural course of SMA, it may accumulate in the kidneys, leading to proteinuria. Therefore, routine pre-dose urine monitoring for proteinuria is recommended. To address this issue, we aimed to evaluate the urinary protein-to-osmolality ratio (UPr/Osm). We also aimed to evaluate the relation of urinary creatinine-to-osmolality ratio (UCr/Osm) to motor function scores. METHODS: A retrospective study was performed on 42 SMA patients. UPr/Cr, UPr/Osm, and

Synergistic effect of carboxymethyl chitosan and phloroglucinol against rotenone induced Parkinson's disease in zebrafish model.

Parkinson's disease (PD) is a prevalent neurodegenerative disorder that impairs motor function in humans. This research explores the combined neuroprotective effects of carboxymethyl chitosan (CMC) and phloroglucinol (PGL) in a zebrafish (ZF) model of PD induced by rotenone (ROT). The structural features of the CMC-PGL conjugate were analysed using UV-visible, Fourier Transform Infrared (FT-IR), and fluorescence spectroscopy. The phenolic content of conjugated sample was estimated and confirmed the conjugation of CMC with PGL. The LD50 of CMC-PGL was determined as 8 μg/mL in ZF embryo (ZFE). I

Use of Hidden Markov Models to Identify Behavioral Patterns in Accelerometry Data of Subterranean Rodents in Field Enclosures.

Activity rhythms of laboratory rodents are usually measured by running wheels, and although wheel running activity-or-rest data enable straightforward rhythmic analyses, it provides limited behavioral information. In subterranean rodents (tuco-tucos), we used bio-loggers (accelerometers) to measure activity rhythms in both lab and field conditions, detecting diverse movements that compose activity. However, understanding these different accelerometer-detected activity components requires more complex analytical tools. Here we used supervised hidden Markov models (HMMs) as a machine learning an

Comparison of CO-OP and goal-directed training on occupational performance and functional status in children with cerebral palsy: Three-armed randomised trial.

INTRODUCTION: Cerebral palsy (CP) is a neurological disorder that impacts motor skills and daily functioning in children. While conventional occupational therapy aims to improve these areas, newer approaches like 'Cognitive Orientation to Daily Occupational Performance' (CO-OP) and 'Goal-Directed Training' (GDT) show promise. However, their comparative effectiveness in enhancing occupational performance and functional status in children with CP remains underexplored. This study aimed to investigate and compare the effects of the CO-OP and GDT on the occupational performance and functional stat

Curcumin treatment reduces motor impairments and protects against rotenone-induced neurodegeneration in a rat model of Parkinson disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by degeneration of dopaminergic neurons, leading to motor impairments (bradykinesia, rigidity, tremor) and non-motor symptoms (cognitive decline, psychiatric disturbances). This study investigated the neuroprotective effects of curcumin in a rotenone-induced PD rat model. Male albino rats received daily intraperitoneal injections of rotenone (2 mg/kg) and/or curcumin (200 mg/kg) for 9 weeks. Behavioral assessments revealed that curcumin treatment significantly improved motor function, reducing catalepsy duration

Multi-scale self-attention approach for analysing motor imagery signals in brain-computer interfaces.

BACKGROUND: Motor imagery-based electroencephalogram (EEG) brain-computer interface (BCI) technology has seen tremendous advancements in the past several years. Deep learning has outperformed more traditional approaches, such next-gen neuro-technologies, in terms of productivity. It is still challenging to develop and train an end-to-end network that can sufficiently extract the possible characteristics from EEG data used in motor imaging. Brain-computer interface research is largely reliant on the fundamental problem of accurately classifying EEG data. There are still many challenges in the f

The role of the sensory input intervention in recovery of the motor function in hypoxic ischemic encephalopathy rat model.

Motor disturbances predominantly characterize hypoxic-ischemic encephalopathy (HIE). Among its intervention methods, environmental enrichment (EE) is strictly considered a form of sensory intervention. However, limited research uses EE as a single sensory input intervention to validate outcomes postintervention. A Sprague-Dawley rat model subjected to left common carotid artery ligation and exposure to oxygen-hypoxic conditions is used in this study. EE was achieved by enhancing the recreational and stress-relief items within the cage, increasing the duration of sunlight, colorful items exposu

RANBP17 Overexpression Restores Nucleocytoplasmic Transport and Ameliorates Neurodevelopment in Induced DYT1 Dystonia Motor Neurons.

DYT1 dystonia is a debilitating neurological movement disorder, and it represents the most frequent and severe form of hereditary primary dystonia. There is currently no cure for this disease due to its unclear pathogenesis. In our previous study utilizing patient-specific motor neurons (MNs), we identified distinct cellular deficits associated with the disease, including a deformed nucleus, disrupted neurodevelopment, and compromised nucleocytoplasmic transport (NCT) functions. However, the precise molecular mechanisms underlying these cellular impairments have remained elusive. In this study

A Detailed Re-Examination of the Period Gene Rescue Experiments Shows That Four to Six Cryptochrome-Positive Posterior Dorsal Clock Neurons (DN1p) of Drosophila melanogaster Can Control Morning and Evening Activity.

Animal circadian clocks play a crucial role in regulating behavioral adaptations to daily environmental changes. The fruit fly Drosophila melanogaster exhibits 2 prominent peaks of activity in the morning and evening, known as morning (M) and evening (E) peaks. These peaks are controlled by 2 distinct circadian oscillators located in separate groups of clock neurons in the brain. To investigate the clock neurons responsible for the M and E peaks, a cell-specific gene expression system, the GAL4-UAS system, has been commonly employed. In this study, we re-examined the two-oscillator model for t

Orexin neurons mediate temptation-resistant voluntary exercise.

Despite the well-known health benefits of physical activity, many people underexercise; what drives the prioritization of exercise over alternative options is unclear. We developed a task that enabled us to study how mice freely and rapidly alternate between wheel running and other voluntary activities, such as eating palatable food. When multiple alternatives were available, mice chose to spend a substantial amount of time wheel running without any extrinsic reward and maintained this behavior even when palatable food was added as an option. Causal manipulations and correlative analyses of ap

Brachial Plexus Root Avulsion Injury-Induced Endothelin-Converting Enzyme-Like 1 Overexpression Is Associated with Injured Motor Neurons Survival.

Brachial plexus root avulsion (BPRA) injury arises from challenging delivery during childbirth, sports-related incidents, or car accidents, leading to extensive loss of motor neurons (MNs) and subsequent paralysis, including both motor and sensory impairment. Surgical nerve re-implantation cannot effectively restore motor function, and the survival of injured MNs is vital for axon regeneration and re-innervating the target muscles. Therefore, identifying novel molecular targets to improve injured MNs survival is of great significance in the treatment of BPRA injuries. Endothelin-converting enz

Implementation of automated behavior metrics to evaluate voluntary wheel running effects on inflammatory-erosive arthritis and interstitial lung disease in TNF-Tg mice.

BACKGROUND: Although treatment options and algorithms for rheumatoid arthritis (RA) have improved remarkably in recent decades, there continues to be no definitive cure or pharmacologic intervention with reliable long-term efficacy. For this reason, the combination of medications and healthy lifestyle modifications are essential for controlling joint disease, and extra-articular manifestations of RA, such as interstitial lung disease (ILD) and other lung pathologies, which greatly impact morbidity and mortality. Generally, exercise has been deemed beneficial in RA patients, and both patients a
