A small viral protein suppresses immune amplification by two distinct mechanisms.

Diverse viral suppressors of RNA interference (RNAi) and RNA silencing (VSRs) interact directly with core protein and/or RNA components of the host RNAi pathway. However, the specific counter-defense function of any VSR biochemical activity is fully validated only when it is shown as essential for viral infection in the wild-type but not mutant hosts defective in antiviral RNAi. Here, we investigated the role of VSR activities for direct binding to small-interfering RNA duplexes (siRNA), long double-stranded RNA (dsRNA), or RNA-dependent RNA polymerase 1 (RDR1) during plant infection by wild-t

Metabolic adaptability and nutrient scavenging in Toxoplasma gondii: insights from ingestion pathway-deficient mutants.

UNLABELLED: The obligate intracellular parasite Toxoplasma gondii replicates within a specialized compartment called the parasitophorous vacuole (PV). Recent work showed that despite living within a PV, Toxoplasma endocytoses proteins from the cytosol of infected host cells via a so-called ingestion pathway. The ingestion pathway is initiated by dense granule protein GRA14, which binds host endosomal sorting complex required for transport (ESCRT) machinery to bud vesicles into the lumen of the PV. The protein-containing vesicles are internalized by the parasite and trafficked to the plant vacu

ACBD3 Is an Essential Pan-enterovirus Host Factor That Mediates the Interaction between Viral 3A Protein and Cellular Protein PI4KB.

The enterovirus genus of the picornavirus family includes a large number of important human pathogens such as poliovirus, coxsackievirus, enterovirus A71, and rhinoviruses. Like all other positive-strand RNA viruses, genome replication of enteroviruses occurs on rearranged membranous structures called replication organelles (ROs). Phosphatidylinositol 4-kinase IIIβ (PI4KB) is required by all enteroviruses for RO formation. The enteroviral 3A protein recruits PI4KB to ROs, but the exact mechanism remains elusive. Here, we investigated the role of acyl-coenzyme A binding domain containing 3 (ACB

Interaction of lipocalin 2, transferrin, and siderophores determines the replicative niche of Klebsiella pneumoniae during pneumonia.

UNLABELLED: Pathogenic bacteria require iron for replication within their host. Klebsiella pneumoniae and other Gram-negative pathogens produce the prototypical siderophore enterobactin (Ent) to scavenge iron in vivo. In response, mucosal surfaces secrete lipocalin 2 (Lcn2), an innate immune protein that binds Ent to disrupt bacterial iron acquisition and promote acute inflammation during colonization. A subset of K. pneumoniae isolates attempt to evade Lcn2 by producing glycosylated Ent (Gly-Ent, salmochelin) or the alternative siderophore yersiniabactin (Ybt). However, these siderophores are

Lysine benzoylation of fatty acid β-oxidation core enzyme FoxA regulates the aflatoxin biosynthesis by benzoyltransferase EsaA in the pathogenic fungus Aspergillus flavus.

Lysine benzoylation (Kbz) is a newly identified post-translational modification, which participates in the regulation of a variety of cellular processes. However, the function of Kbz in pathogenic fungi remains unclear. Here, we first identified that FoxA is a benzoylated protein with two benzoylated sites at lysines 425 and 433. Mutations of Kbz sites in FoxA significantly reduced long-chain fatty acid (LCFA) utilization activity and exhibited a phenotype similar to that of foxA gene deletion mutant, including decreased conidiation and aflatoxin production, reduced seed colonization, and incr

Interdomain Flexibility of Chikungunya Virus nsP2 Helicase-Protease Differentially Influences Viral RNA Replication and Infectivity.

Chikungunya virus (CHIKV) is a mosquito-borne alphavirus responsible for chikungunya fever. Nonstructural protein 2 (nsP2), a multifunctional protein essential for viral replication, has an N-terminal helicase region (nsP2h), which has both nucleotide triphosphatase and RNA triphosphatase activities, as well as a C-terminal cysteine protease region (nsP2p), which is responsible for nonstructural polyprotein processing. The two functional units are connected through a linker of 14 residues. Although crystal structures of the helicase and protease regions of CHIKV nsP2 have been solved separatel

PgFur participates differentially in expression of virulence factors in more virulent A7436 and less virulent ATCC 33277 Porphyromonas gingivalis strains.

BACKGROUND: Porphyromonas gingivalis is considered a keystone pathogen responsible for chronic periodontitis. Although several virulence factors produced by this bacterium are quite well characterized, very little is known about regulatory mechanisms that allow different strains of P. gingivalis to efficiently survive in the hostile environment of the oral cavity, a typical habitat characterized by low iron and heme concentrations. The aim of this study was to characterize P. gingivalis Fur homolog (PgFur) in terms of its role in production of virulence factors in more (A7436) and less (ATCC 3

Zebrafish Models for the Mechanosensory Hair Cell Dysfunction in Usher Syndrome 3 Reveal That Clarin-1 Is an Essential Hair Bundle Protein.

UNLABELLED: Usher syndrome type III (USH3) is characterized by progressive loss of hearing and vision, and varying degrees of vestibular dysfunction. It is caused by mutations that affect the human clarin-1 protein (hCLRN1), a member of the tetraspanin protein family. The missense mutation CLRN1(N48K), which affects a conserved N-glycosylation site in hCLRN1, is a common causative USH3 mutation among Ashkenazi Jews. The affected individuals hear at birth but lose that function over time. Here, we developed an animal model system using zebrafish transgenesis and gene targeting to provide an exp

[Spongiform encephalitis].

The authors present a personal interpretation of the pathogeny of prionic processes, showing the role played by heredity. They also present the very surprising fact that a protein codified by a mutant gene can act as transmissible agent in a serious illness opening new chapters to the pathology of a series of important diseases. This is a completely new perspective in the biology.

Medulloblastoma-associated mutations in the RNA helicase DDX3X/DED1 cause defects in the translational response to TORC1 inhibition.

Medulloblastoma is the most common pediatric brain cancer, but current treatments are largely non-specific, often causing developmental side effects. Genomic sequencing identified the RNA helicase DDX3X as one of the most frequently mutated genes in this cancer and a potential treatment target, yet its role in tumor progression remains elusive. Prior studies have indicated that the mutations cause specific defects in translation; however, both DDX3X and its yeast ortholog Ded1 have also been associated with cellular stress responses, suggesting that the contribution of the DDX3X mutations to m

The PP2A phosphatase regulatory subunit SsRts1 regulates morphogenesis, mitosis, sexual mating and virulence in Sporisorium scitamineum.

Sugarcane smut, caused by S. scitamineum, results in substantial economic losses across global sugarcane growing areas. Elucidating the pathogenic mechanisms of S. scitamineum is therefore crucial for developing effective disease control strategies. Our previous work has shown that the protein phosphatase SsPpe1 regulates mating and pathogenicity in S. scitamineum. In this study, we identified SsRts1 as a novel interacting protein of SsPpe1, which contains a PP2A regulator subunit B56 domain. Deletion of Ssrts1 gene impaired sporidia growth, resulting in pseudohyphal sporidia with multiple nuc

[Cholesterol in the ciliary membrane as a therapeutic target of polycystic kidney].

Primary cilia are non-motile, microtubule-based cellular protrusions that sense extracellular chemical and mechanical cues, converting them into intracellular signals. Ciliary dysfunction causes ciliopathies including polycystic kidney disease and retinal degeneration. The ciliary membrane is highly enriched in cholesterol, which is essential for the physiological function of ciliary-localized receptors, including G-protein-coupled receptors (GPCRs) and ion channels. Defective de novo cholesterol synthesis, as seen in Smith-Lemli-Opitz syndrome (SLOS), causes typical ciliopathy-like symptoms,

Elaboration of Cortical Pyramidal Dendrites Requires Dephosphorylation of Signal Regulatory Protein α by Protein Tyrosine Phosphatase δ.

Protein tyrosine phosphatase δ (PTPδ) is involved in Sema3A-induced dendritic elaboration of cortical pyramidal neurons through the activation of Fyn tyrosine kinase. However, the in vivo substrates of PTPδ remain largely unknown. Phosphotyrosine proteome analysis of Ptpδ -/- mice brains from both male and female revealed that signal regulatory protein α (SIRPα) was hyperphosphorylated at the carboxy-terminal Tyr501 residue in the knock-outs. Immunohistochemistry with anti-phospho-Tyr501 SIRPα antibody showed the hyperphosphorylation of SIRPα in various regions including the olfactory nerve la

Propagation of gut-injected AAV2/1-Htt171-82Q to the brain induces Huntington's disease-like pathology.

Huntington's disease (HD) is a progressive neurodegenerative disorder that leads to thbe production of pathogenic mutant huntingtin (mHtt) protein and is characterized by motor and psychiatric symptoms. While HD has classically been studied as a brain-centered condition, emerging research indicates that the gut-brain axis may play a role in disease initiation and progression. However, direct experimental evidence demonstrating a peripheral-to-central route for mHtt propagation remains limited and unclear. Here, we tested whether mHtt expressed in the gastrointestinal tract could migrate to the

Computational analysis of missense mutations in squalene epoxidase associated with terbinafine resistance in clinically reported dermatophytes.

Dermatophyte infections, as a significant public health threats, are increasingly associated with antifungal drug resistance, particularly to terbinafine. Mutations in the squalene epoxidase (SQLE) gene have been linked to resistance by altering amino acid residues and interfering with drug-protein interactions. This study applied computational tools including I-Mutant, ConSurf, HOPE, DynaMut2, STRING, and molecular docking to assess the structural and functional impact of clinically reported SQLE missense mutations in terbinafine-resistant dermatophyte isolates. Twelve out of fourteen mutatio

Overexpression of Wild-Type TMEM43 Improves Cardiac Function in Arrhythmogenic Right Ventricular Cardiomyopathy Type 5.

BACKGROUND: Arrhythmogenic right ventricular cardiomyopathy type 5 (ARVC5) is the most aggressive type of ARVC, caused by a fully penetrant missense mutation (p.S358L) in TMEM43 (transmembrane protein 43). Pathologically, the disease is characterized by dilation of the cardiac chambers and fibrofatty replacement of the myocardium, which results in heart failure and sudden cardiac death. Current therapeutic options are limited, and no specific therapies targeting the primary cause of the disease have been proposed. METHODS: We investigated whether overexpression of wild-type (WT) TMEM43 could

A systems genetics approach identifies roles for proteasome factors in heart development and congenital heart defects.

Congenital heart defects (CHDs) occur in about 1% of live births and are the leading cause of infant death due to birth defects. While there have been remarkable efforts to pursue large-scale whole-exome and genome sequencing studies on CHD patient cohorts, it is estimated that these approaches have thus far accounted for only about 50% of the genetic contribution to CHDs. We sought to take a new approach to identify genetic causes of CHDs. By combining analyses of genes that are under strong selective constraint along with published embryonic heart transcriptomes, we identified over 200 new c

Novel HSPB8 mutations in severe early-onset myopathy with involvement of respiratory and cardiac muscles cause proteostasis defects in cell models.

Heat shock protein family B (small) member 8 (HSPB8) promotes chaperone-assisted selective autophagy (CASA), which assures proteostasis in muscles and neurons. HSPB8 frameshift mutations found in neuromyopathies are translated on the same frame, generating the same C-terminal extension, which causes HSPB8 aggregation and proteostasis defects. Here, we describe three novel HSPB8 frameshift variants, translated to protein using the third alternative frame to stop codons downstream to the canonical one and to the one used by other known HSPB8 frameshift mutants. Therefore, these variants are pred

Role of biotin carboxyl carrier protein subunit 2 (BCCP2) in resistance to multiple stresses in Arabidopsis thaliana.

Abiotic stresses, including drought, salinity, and temperature extremes, are serious constraints to plant growth and agricultural development. These stresses that plants face in nature are often multiple and complex. Biotin carboxyl carrier protein subunit 2 (BCCP2) is one of the two subunits of biotin carboxyl carrier protein, which is a functional subunit of acetyl coenzyme A carboxylase, primarily studied for its role in fatty acid synthesis. In this study, we identified the expression pattern of AtBCCP2 under various stress conditions, including 4 mM NaHCO₃, 2 mM Na₂CO₃, 150 mM NaCl, 300 m

Kif1a and intact microtubules maintain synaptic-vesicle populations at ribbon synapses in zebrafish hair cells.

Sensory hair cells of the inner ear utilize specialized ribbon synapses to transmit sensory stimuli to the central nervous system. This transmission necessitates rapid and sustained neurotransmitter release, which depends on a large pool of synaptic vesicles at the hair-cell presynapse. While previous work in neurons has shown that kinesin motor proteins traffic synaptic material along microtubules to the presynapse, the mechanisms of this process in hair cells remain unclear. Our study demonstrates that the kinesin motor protein Kif1a, along with an intact microtubule network, is essential fo
