Ethical Considerations of Whole-Eye Transplantation.

Whole eye transplantation (WET) remains experimental. Long presumed impossible, recent scientific advances regarding WET suggest that it may become a clinical reality. However, the ethical implications of WET as an experimental therapeutic strategy remain largely unexplored. This article evaluates the ethical considerations surrounding WET as an emerging experimental treatment for vision loss. A thorough review of published literature pertaining to WET was performed; ethical issues were identified during review of the articles.

Stem cell transplantation enhances endogenous brain repair after experimental stroke.

Stem cell transplantation for stroke treatment has been a promising therapy in small and large animal models, and many clinical trials are ongoing to establish this strategy in a clinical setting. However, the mechanism underlying functional recovery after stem cell transplantation has not been fully established and there is still a need to determine the ideal subset of stem cells for such therapy. We herein reviewed the recent evidences showing the underlying mechanism of functional recovery after cell transplantation, focusing on endogenous brain repair. First, angiogenesis/neovascularizatio

Enteric nervous system cell replacement therapy for Hirschsprung disease: beyond tissue-engineered intestine.

Hirschsprung disease (HD), a neurocristopathy characterized by failed migration of neural crest cells to the distal colon, requires surgical resection of the aganglionic segment. Advances in stem cell and regenerative medicine research have opened the possibility to treat HD less invasively using enteric nervous system (ENS) cell replacement therapy. This article reviews the progress to date of culturing and delivering ENS stem cells in various in vitro and in vivo models, as well as review the available evidence of functionality of the transplant-derived cells. Potential areas of future study

Waking up the sleepers: shared transcriptional pathways in axonal regeneration and neurogenesis.

In the last several years, relevant progress has been made in our understanding of the transcriptional machinery regulating CNS repair after acute injury, such as following trauma or stroke. In order to survive and functionally reconnect to the synaptic network, injured neurons activate an intrinsic rescue program aimed to increase their plasticity. Perhaps, in the attempt to switch back to a plastic and growth-competent state, post-mitotic neurons wake up and re-express a set of transcription factors that are also critical for the regulation of their younger brothers, the neural stem cells. A retrograde tracing study of compensatory corticospinal projections in rats with neonatal hemidecortication.

To examine the compensatory mechanisms in rats that underwent left decortication at postnatal day 7 (P7), we injected the retrograde tracers fluorescein isothiocyanate-cholera toxin B subunit (FITC-CTB) and Fast Blue (FB) into the right and left upper cervical spinal cord, respectively, at postoperative weeks 2, 3, 4, and 5 and counted the number of retrogradely labeled corticospinal neurons in the right cerebral cortex compared with that in normally developed rats. Significantly more ipsilaterally projecting neurons were labeled with FITC-CTB in the decorticated rats compared with normal rats

Examination of axonal injury and regeneration in micropatterned neuronal culture using pulsed laser microbeam dissection.

We describe the integrated use of pulsed laser microbeam irradiation and microfluidic cell culture methods to examine the dynamics of axonal injury and regeneration in vitro. Microfabrication methods are used to place high purity dissociated central nervous system neurons in specific regions that allow the axons to interact with permissive and inhibitory substrates. Acute injury to neuron bundles is produced via the delivery of single 180 ps duration, lambda = 532 nm laser pulses. Laser pulse energies of 400 nJ and 800 nJ produce partial and complete transection of the axons, respectively, res

Bifurcating microchannels as a scaffold to induce separation of regenerating neurites.

Many neural interfacing strategies, such as the sieve electrode and the cultured probe, rely on neurite growth to establish neural contact. But this growth is subject to natural fasciculation, compromising the effectiveness of these interfacing strategies by reducing potential selectivity. This in vitro study shows that the fasciculation mechanism can be manipulated by providing an appropriate microchannel scaffold to guide and influence growth, thereby achieving a high degree of selectivity. The microchannels employed have a bifurcation from a primary channel into two secondary channels. This

Triggered by asphyxia neurogenesis seems not to be an endogenous repair mechanism, gliogenesis more like it.

We analyzed the long-term consequences of asphyxial cardiac arrest for hippocampal cell proliferation in rats to evaluate if the ischaemia-induced degenerated CA1 region may be repopulated by endogenous (stem) cells. Studies were performed in an asphyxial cardiac arrest model with 5 minutes of asphyxiation and three different survival times: 7, 21, and 90 days. Sham-operated non-asphyxiated rats served as control. Cell proliferation was studied by labeling dividing cells with 5-bromo-2'-deoxy-uridine (BrdU). The neurodegenerative/regenerative pattern at single cell levels was monitored by immu

Therapeutic potential of olfactory ensheathing cells in neurodegenerative diseases.

The regenerative capacity of the olfactory system has generated interest in potential clinical application of cells from the olfactory epithelium in the treatment of neurodegenerative diseases. Experimental evidence from animal models and clinical studies suggest that transplantation of olfactory ensheathing cells (OEC), specialized glia in the olfactory system, may be therapeutically useful in neurodegenerative diseases such as spinal cord injury and stroke. This review article describes the different experimental approaches in OEC transplantation. We also discuss the possible effects of OEC

[Adult neurogenesis in physiological and pathological conditions].

Generation of new neurons persists in the two restricted regions of adult brain, the dentate gyrus (DG) of the hippocampus and the subventricular zone (SVZ). Newly generated neurons are functionally integrated into the neuronal circuits, which is involved in regulation of brain plasticity. Endogenous neuronal production in the DG and SVZ is expected to provide a continuous source of new neurons that replace degenerated neurons in the injured brain. Recent studies indicate that adult neurogenesis is modified by various brain insults including stroke, epilepsy and neurodegenerative disorders. Wh

A comparative framework for understanding the biological principles of adult neurogenesis.

Adult neurogenesis has been identified in all vertebrate species examined thus far. However, an evolutionary trend towards a reduction in both the number of proliferation zones and the overall number of newborn cells has been revealed in more recent lineages of vertebrates, such as mammals. Adult neurogenesis, and in particular the characterization of adult neural stem cells in mammals has been the focus of intense research with the goal of developing new cell-based regenerative treatments for neurodegenerative diseases, spinal cord injury, and acute damage due to stroke. Conversely, most othe

Homotransplant of pelvic ganglion into bladder wall in adult rats.

In these experiments a large portion of the pelvic ganglion of adult female rats was transplanted into the wall of the urinary bladder of the same animals. The morphology and fine structure of the transplants were studied in whole-mounts and in sections for light and electron microscopy, from two days up to four months after operation. The general architecture of the ganglion was preserved in all the transplants. The vascularization was re-established. Nerves grew out of the transplant and connections with the original intramural nerves of the bladder wall were established. All the synapses de

[Plastic remodeling of the synaptic apparatus of skeletal muscles under physiological and pathological conditions].

The synaptic apparatus of skeletal muscles undergoes continuous remodelling, by adapting to the functioning level of the peripheral neuromuscular apparatus under normal conditions, compensating and restoring its impaired function in pathology. This reorganization has specific features depending on the factors that influence it, such as increased function, decreased or no function, neuronal and nerve damage, synaptic toxin intoxication, autoantibody action, disturbances of trophic influences, etc. Damage to any link of the neuromuscular apparatus due to the above factors is likely to be an esse

Changes in the morphology of sympathetic preganglionic neurons parallel the development of autonomic dysreflexia after spinal cord injury in rats.

Following spinal cord transection (SCT), sensory input to the spinal cord causes increases in arterial pressure that are small in rats 1 week after SCT, but become large and well established by 2 weeks. Moreover, sympathetic preganglionic neurons (SPNs) undergo atrophy by 1 week after SCT, and regeneration of these neurons may be an important factor in the etiology of this autonomic dysreflexia. Therefore, we examined the morphology of SPNs 2 weeks after SCT using retrograde transport of the cholera toxin subunit B. The dendritic arbors of SPNs were re-established by 2 weeks after SCT. This re

Enhancement of central nervous system remyelination in immune and non-immune experimental models of demyelination.

Studies in both humans and experimental animals indicate that there is potential for full remyelination following CNS demyelination, but the factors that control the degree of myelin repair are unknown. In the Theiler's virus model of demyelination CNS remyelination can be promoted either by global immunosuppression or by selective immunoglobulin therapy. In this paper we discuss whether immunoglobulin-mediated remyelination is a characteristic of immune-mediated demyelination, or whether immunoglobulin-mediated remyelination also occurs in the toxic-traumatic model of lysolecithin-induced dem

Increased expression of nitric oxide synthase in hypothalamic neuronal regeneration.

This investigation deals with the histochemical and scanning electron microscopic (SEM) correlates that depict regeneration of the neurohypophyseal system that may be nitric oxide dependent following hypophysectomy in the rodent hypothalamus. NOS histochemistry and correlative SEM were employed to establish the rates of regrowth and appearance of NOS-positive supraoptic (SON) and paraventricular (PVN) neurites and their cell bodies following hypophysectomy. NOS activity increased significantly in SON and PVN neuronal perikarya and regenerating axons by 2 weeks. NOS-positive neurites were obser

[Corneal sensibility following epikeratophakia].

The postoperative rate of reinnervation following corneal surgery is widely considered to be a useful indicator of the healing process. This study reports the corneal sensitivity of 14 patients following epikeratophakia for different indications (myopia, aphakia and keratoconus) after time periods ranging from 7 to 104 weeks. All transplants were clear at the time of measurement. A newly developed aesthesiometer (using a low electric current for stimulation) was employed. Nine positions on the operated eye were chosen for the measurements, five locations of the non-operated fellow eye served a
