Translatome profiling reveals opposing alterations in inhibitory and excitatory neurons of fragile X mice and identifies EPAC2 as a therapeutic target.

Symptoms of fragile X syndrome (FXS), the leading monogenic cause of intellectual disability and autism, are thought to arise from an excitation/inhibition (E/I) imbalance. Here, we leverage cell-type-specific mRNA sequencing to profile molecular alterations in cortical excitatory (Camk2) and inhibitory (Pvalb) neurons in Fmr1 knockout (KO) mice, integrating transcriptomic results with circuit and behavioral readouts to prioritize novel therapeutic targets. We uncovered significant genotype-by-cell type interactions for differential gene expression in Camk2a and Pvalb translatomes, and, striki

Regulation of PHOX2B gene expression by the long non-coding natural antisense RNA PHOX2B-AS1.

Paired mesoderm homeobox protein 2B (PHOX2B) is a transcription factor essential for autonomic nervous system development. Heterozygous mutations in the PHOX2B gene are associated with neurodevelopmental disorders, including congenital central hypoventilation syndrome and Hirschsprung's disease. Additionally, PHOX2B plays a role in the genetic landscape of neuroblastoma, with mutations detected in both familial and sporadic forms of this rare cancer. Notably, PHOX2B is highly expressed in most neuroblastoma cells. Despite its significance, little is known about the regulation of PHOX2B gene ex

Mechanical properties of the developing brain in a model of fetal alcohol spectrum disorders and relationships to perineuronal net integrity.

Neuroimaging is a useful tool for examining altered neurodevelopmental trajectories in fetal alcohol spectrum disorders (FASD). FASD affects 1 in 20 infants in the United States with higher prevalence in specific regions across the globe. Advanced neuroimaging methods, such as volumetric morphometry and diffusion-weighted imaging, are critical for determining the effectiveness of interventions that support neurodevelopment in FASD. In this study, we introduce the use of magnetic resonance elastography (MRE), a cutting-edge neuroimaging technique used to measure the mechanical properties of bra

Molecular Networks of Redox Dysregulation in Fetal Alcohol Spectrum Disorders: Mechanisms and Therapeutic Prospects.

Fetal alcohol spectrum disorders (FASD) encompass a continuum of developmental abnormalities caused by prenatal alcohol exposure, resulting in persistent neurodevelopmental and structural defects. Accumulating evidence indicates that redox dysregulation plays a central role in the pathogenesis of FASD. Ethanol disrupts cellular redox homeostasis by promoting excessive reactive oxygen species production and depleting endogenous antioxidants, thereby perturbing key redox-sensitive molecular networks. Dysregulation of these pathways leads to mitochondrial dysfunction, endoplasmic reticulum stress

Contrasting outcomes of 16p11.2 microdeletion and microduplication in prenatal diagnosis: phenotypic variability and genetic counseling strategies.

BACKGROUND: Copy number variations (CNVs) in the 16p11.2 region are associated with neurodevelopmental disorders, but they exhibit incomplete penetrance and variable expressivity. Prenatal diagnosis of these CNVs presents significant challenges because of the unpredictable phenotypic outcomes. MATERIALS AND METHODS: We retrospectively analyzed two fetal cases diagnosed prenatally via amniocentesis with CNV sequencing (CNV-seq) as having 16p11.2 CNVs, and discussed them in the context of current literature. In this research, GTG-banding karyotype analysis, CNV-seq, and whole-exome sequencing w

Activity-dependent development of vocal circuits in the neonatal rodent forebrain.

Vocal communication is fundamental for social interaction across species, yet the neural mechanisms that shape vocal circuit development remain poorly understood despite their relevance to neurodevelopmental disorders. Here, we investigate vocal circuit development in mice using isolation-induced ultrasonic vocalizations (USVs) in neonates. An activity-tagging approach identifies the ventromedial prefrontal cortex (vmPFC) as a cortical region strongly activated during USV emission. We find a predictable temporal correlation between vmPFC activity and USV emission using in vivo fiber photometry

Microglia, cerebellar inflammation, and autism spectrum disorders: Developmental mechanisms and therapeutic perspectives.

Autism spectrum disorders (ASD) are neurodevelopmental conditions characterized by deficits in social interaction and communication, as well as restricted and repetitive behaviors. These conditions often co-occur with medical issues linked to synaptic alterations, which compromise synaptic integrity and are associated with brain circuit dysfunction. Both human and animal studies have identified cerebellar structural and functional alterations in subsets of ASD cases, with evidence of abnormal morphology and disrupted connectivity correlating with symptom severity. Numerous studies further sugg

Diagnosis and Management of Loeys-Dietz Syndrome: Evidence Gaps and Future Directions.

PURPOSE OF REVIEW: This review summarizes current knowledge on Loeys-Dietz syndrome (LDS), including its genetic basis, multisystem manifestations, diagnosis, surveillance, and management. It also highlights key evidence gaps in risk stratification, medical therapy, imaging surveillance, surgical thresholds, and care of pregnant and pediatric patients. RECENT FINDINGS: Although understanding of the genetic basis and clinical spectrum of LDS has improved, LDS-specific evidence remains limited. LDS is increasingly recognized as a multisystem disorder with aggressive aortic disease, extra-aortic

Biallelic VPS41 Variants in Autosomal Recessive Spinocerebellar Ataxia 29 Resolved by Long-Read Sequencing and RNA Analysis.

BACKGROUND: Biallelic variants in VPS41, encoding a subunit of the HOPS complex, cause autosomal recessive spinocerebellar ataxia 29 (SCAR29), a rare neurodevelopmental disorder with an incompletely defined phenotypic and molecular spectrum. METHODS: We investigated a 24-year-old man with cerebellar ataxia, hypotonia, and intellectual disability. Exome sequencing identified four candidate VPS41 variants. Because maternal DNA was unavailable, long-read genome sequencing was performed to determine allelic configuration, followed by RNA and protein analyses. RESULTS: In addition to typical SCAR

Distinct developmental trajectories shape human sensitivity to rhythms in the environment.

Rhythm is an omnipresent feature of our environment. Repetitive temporal patterns in sound and vision influence how we pay attention to the world, move, and speak. Grasping these regularities is critical for development. Humans can track surrounding rhythms explicitly-like dancing to the beat of music-or implicitly, when rhythms guide perception and behavior without deliberate attention. Whether these abilities follow different developmental trajectories remains unknown. Here, we tested 98 children aged 7-13 using a novel gamified task measuring implicit rhythm processing, alongside assessment

PTEN regulates starburst amacrine cell dendrite morphology during development.

Neurons are subject to extensive developmental regulation to ensure precise subtype-specific morphologies that are intimately tied to their function. Starburst amacrine cells (SACs) in the mammalian retina have a highly stereotyped, radially symmetric dendritic arbor that is essential for their role in direction-selective circuits in the retina. We show that PTEN, the primary negative regulator of the PI3K-AKT-mTOR pathway that is highly implicated in neurodevelopmental disorders, regulates SAC morphology in a cell-autonomous manner. Pten-deficient SACs show a nearly twofold increase in the nu

Rare Case with Pathogenic Variant in DHX16 Gene Causing Neuromuscular Disease and Oculomotor Anomalies.

The DEAD/DExD/H-box RNA helicases are a group of RNA-binding proteins involved in the metabolism of mRNAs. They coordinate gene expression programs and play a role in cellular signaling, fate, and survival. We describe a case of a 36-year-old female with neuromuscular disease, sensorineural hearing loss, retinitis pigmentosa, and primary ovarian insufficiency harboring a heterozygous de novo missense pathogenic variant in the DEAH-box helicase 16 (DHX16) gene. This is the first case exhibiting a high intellectual level and the highest survival outcome so far. Eight previous cases of DHX16 dise

Reframing Dental Anxiety: Cognitive Behavioral Therapy and Its Role in Phobia Treatment-A Narrative Review.

Dental phobia is a disabling yet underdiagnosed condition that prevents many patients from seeking essential oral healthcare, leading to avoidable pain, disease progression, and reduced quality of life. Cognitive Behavioral Therapy (CBT) is the most widely supported psychological intervention for specific phobias and has demonstrated significant efficacy in reducing dental anxiety and avoidance. This narrative review synthesizes recent evidence on CBT applications for phobia management, with particular emphasis on dental settings. In addition to reviewing established mechanisms of CBT, this pa

Neural Orchestration of Mandibular Development.

The mandible, exclusively innervated by the trigeminal nerve and influenced by intricate biomechanical factors, serves as a vital model for studying neuro-osseous interactions in craniofacial development. However, research on its neural regulation is comparatively weak relative to the appendicular skeleton. This review aims to consolidate emerging evidence regarding the neural regulation of mandibular morphogenesis, emphasizing mechanisms that differ from those of long bones, and to highlight its clinical significance. A comprehensive review was conducted across a broad array of databases, inc

The meningeal-cerebellar axis: a new perspective on cerebellar development.

The cerebellum is a highly organized brain structure best known for its roles in motor control and sensorimotor integration. While cerebellar development has traditionally been attributed to intrinsic genetic programs and local cell-cell interactions, emerging evidence indicates that extrinsic cues particularly signals from the meninges also play a critical role in shaping its maturation. Studies indicate that the meninges release cytokines, chemokines, and growth factors including CXCL12, IGF-1, IL-33, FGF2, TGF-β, and retinoic acid that influence granule cell precursor (GCPs) proliferation,

Methods to Assess Neuronal Primary Cilia Electrochemical Signaling.

Primary cilia are polymodal sensory organelles which project from the apical side of polarized cells. They are found in all brain hemispheres but are most pronounced in neurons, which comprise the granular layers of the hippocampus and cerebellum. Pathogenic variants in genes which encode primary cilia components are responsible for neuronal ciliopathies-a group of central nervous system disorders characterized by neurodevelopmental conditions such as intellectual disability, seizure, ataxia, and sensory deficits. In the hippocampus, neuronal primary cilia form chemical synapses with axons and

Postnatal downregulation of Fmr1 in microglia promotes microglial reactivity and causes behavioural alterations in female mice.

BACKGROUND: Fragile X syndrome is caused by the loss of the Fmr1 gene expression. Deletion of Fmr1 in various neuronal and non-neuronal subpopulations in the brain of mice leads to cell-type-specific effects. Microglia, immune cells critical for the refinement of neuronal circuits during brain development, have been implicated in various neurodevelopmental disorders, including fragile X syndrome. However, it is unknown whether reduced Fmr1 expression in microglia leads to molecular and behavioral phenotypes. METHODS: We downregulated Fmr1 in microglia during early and late postnatal developme

Altered social proximity in adult mice following prenatal stress Exposure: An exploratory link to cortical neurogenesis.

Prenatal stress has been implicated in the etiology of neurodevelopmental disorders, yet its impact on cortical morphogenesis and long-term social behavior remains insufficiently characterized. This study aimed to evaluate the effects of prenatal restraint stress on prenatal neurogenesis and social behavior in adult offspring using behavioral and histological approaches. Pregnant mice were subjected to restraint stress (0.5 or 2 h daily from embryonic day (E) 10.5 to 14.5). Histological analysis at E15.5 revealed reduced neurogenesis and intermediate zone formation. Behavioral assessments incl

MODY5 and 17q12 Microdeletion Syndrome: Phenotype Variability, Prenatal and Postnatal Counseling.

Maturity-Onset Diabetes of the Young Type 5 (MODY5) is caused by heterozygous pathogenic variants in the HNF1B gene, encoding the transcription factor hepatocyte nuclear factor-1β. HNF1B haploinsufficiency typically leads to young-onset non-immune diabetes and highly variable renal involvement, whose more frequent features are bilateral kidney cysts and renal hypodysplasia. Kidney cysts or echogenic kidneys can be identified by ultrasonography in the prenatal period, but the renal involvement can also start in childhood or later. Notably, a recurrent microdeletion syndrome at 17q12 (deleting H
