The lesions of Alzheimer disease include accumulation of proteins, losses of neurons and synapses, and alterations related to reactive processes. Extracellular Abeta accumulation occurs in the parenchyma as diffuse, focal or stellate deposits. It may involve the vessel walls of arteries, veins and capillaries. The cases in which the capillary vessel walls are affected have a higher probability of having one or two apoepsilon 4 alleles. Parenchymal as well as vascular Abeta deposition follows a stepwise progression. Tau accumulation, probably the best histopathological correlate of the clinical

Distributions of arg3.1 and c-fos immunoreactive neurons (IRN) in gerbil auditory cortex (AC) and amygdala showed characteristic differences when comparing systemic application of the tinnitus-eliciting drug salicylate with acoustic stimulation or saline injections. In AC, arg3.1 IRN induced by stimulation focused in regions corresponding to the frequency content of the stimulus. Injections of salicylate (350 mg/kg body weight) led to accumulation of arg3.1 IRN in the high frequency domain, while saline injections produced a diffuse distribution. After all treatments, c-fos IRN outnumbered arg

Treadmill training and clonidine, an alpha-2 noradrenergic agonist, have been shown to improve locomotion after spinal cord injury. We speculate that transmission in load pathways, which are involved in body support during stance, is specifically modified by training. This was evaluated by comparing two groups of spinal cats; one group (n = 11) was trained to walk until full-weight-bearing (3-4 weeks), and the other (shams; n = 7) was not. During an acute experiment, changes in group I pathways, monosynaptic excitation, disynaptic inhibition, and polysynaptic excitation were investigated by me

Tyrosine kinase B (TrkB) levels in the adult guinea pig cochlear nucleus (CN) were determined from Western blots for up to 60 days after unilateral cochlear ablation (UCA). Compared to TrkB levels on the intact side, those on the lesioned side were elevated in the anteroventral CN (AVCN) at 7 and 60 days and in the posteroventral CN (PVCN) at 30 days. TrkB levels were depressed in the AVCN and the dorsal CN (DCN) at 30 days. Elevations in the AVCN on the lesioned side at 7 days coincided with a period of synaptogenesis. Other changes were temporally related to up- or downregulations of transmi

BACKGROUND: Healthy aging alone can lead to cognitive decline, decreased brain size, protein aggregation, accumulation of senescent cells and neuroinflammation. Furthermore, age is the primary risk factor for several neurodegenerative disorders such as Parkinson's and Alzheimer's disease. Age-related neuroinflammation, as known as inflammaging, is thought to restrict brain plasticity. Perineuronal nets (PNNs), specialized extracellular matrix structures surrounding fast-spiking parvalbumin (PV) interneurons, regulate plasticity and protect neurons from oxidative stress. Given the known impact

Autism spectrum disorder (ASD) is a severe life-long neuropsychiatric disorder. Alterations and imbalance of several neurochemical systems may be involved in ASD pathophysiology, of them, serotonergic neurotransmission dysfunction and deficiency may underlie behavioral abnormalities associated with ASD. However, the functional importance of serotonergic receptors, particularly 5HT7 receptors in ASD pathology remains poorly defined. Serotonin receptor subtype 7 (5-HT7R) plays a direct regulatory role in the development and also for the mature function of the brain, therefore, further studies ar

Humans have a distinguishing ability for fine motor control that is subserved by a highly evolved cortico-motor neuronal network. The acquisition of a particular motor skill involves a long series of practice movements, trial and error, adjustment and refinement. At the cortical level, this acquisition begins in the parieto-temporal sensory regions and is subsequently consolidated and stratified in the premotor-motor cortex. Task-specific dystonia can be viewed as a corruption or loss of motor control confined to a single motor skill. Using a multimodal experimental approach combining neuroima

Functional neurological recovery remains the primary objective when treating ischemic stroke. However, current therapeutic approaches often fall short of achieving optimal outcomes. One of the most significant challenges in stroke treatment is the effective delivery of neuroprotective agents across the blood-brain barrier to ischemic regions within the brain. The blood-brain barrier, while essential for protecting the brain from harmful substances, also restricts the passage of many therapeutic compounds, thus limiting their efficacy. In this review, we summarizes the emerging role of nanopart

Immediate-early gene (IEG) induction after administration of amphetamine or cocaine has been used to trace the signaling pathways that mediate neuronal plasticity required for the short- and long-term behavioral effects of these psychostimulants. We recently reported that a novel cyclic AMP (cAMP)-dependent Rap guanine nucleotide exchange factor-2 (RapGEF2)-ERK signaling pathway is required for Egr-1 induction in D1 medium-spiny neurons (MSNs) of the nucleus accumbens (NAc) after cocaine treatment, and that its deletion from the NAc neurons attenuates cocaine-induced locomotor sensitization an

Hyperactivity of the hypothalamic-pituitary-adrenal (HPA) axis is accepted as one of the fundamental biological mechanisms that underlie major depression. This hyperactivity is caused by diminished feedback inhibition of glucocorticoid (GC)-induced reduction of HPA axis signaling and increased corticotrophin-releasing hormone (CRH) secretion from the hypothalamic paraventricular nucleus (PVN) and extra-hypothalamic neurons. During chronic stress-induced inhibition of systemic feedback, cytosolic glucocorticoid receptor (GR) levels were significantly changed in the prefrontal cortex (PFC) and h

Spinocerebellar ataxia type 1 (SCA1) is a dominantly inherited disorder characterized by progressive loss of coordination, motor impairment and the degeneration of cerebellar Purkinje cells, spinocerebellar tracts and brainstem nuclei. Many dominantly inherited neurodegenerative diseases share the mutational basis of SCA1: the expansion of a translated CAG repeat coding for glutamine. Mice lacking ataxin-1 display learning deficits and altered hippocampal synaptic plasticity but none of the abnormalities seen in human SCA1; mice expressing ataxin-1 with an expanded CAG tract (82 glutamine resi

This study investigated whether there are changes in the excitability of motor axons in peripheral nerves of patients with corticospinal lesions, reflecting plasticity of the motoneuron due to altered descending drives and/or changes in afferent feedback. The excitability of motor and sensory axons in peripheral nerves of the affected limb of 11 patients with unilateral hemiparesis due to stroke was compared with that for the unaffected limbs and with data for 12 age-matched controls. There was significantly less accommodation to hyperpolarizing currents in motor axons on the affected side. Th

BACKGROUND: Stroke disrupts cortical excitability, inhibition, and network connectivity, contributing to persistent motor deficits. OBJECTIVE: To determine whether acupuncture modulates cortical reactivity, functional connectivity, and corticospinal output after stroke using an integrated transcranial magnetic stimulation–electroencephalography–motor evoked potential (TMS–EEG–MEP) framework in a retrospective cohort. METHODS: Clinical and neurophysiological data from 200 patients with first-ever ischemic stroke (100 acupuncture; 100 control) were analyzed at baseline, 4 weeks, and 8-week follo

This study investigated the effects of moderate chronic exercise on hippocampal integrity, motor learning, and neuroplasticity markers in diabetic aged (16-month-old) female Wistar rats. Forty Wistar rats were divided into four groups: Control (n = 10), Exercise Control (n = 10), Diabetes (n = 10), and Diabetes + Exercise (n = 10). Diabetes was induced via streptozotocin (40 mg/kg, i.p.), while exercise groups performed 45 min daily treadmill sessions for four weeks. Motor coordination was assessed using the rotarod test. Hippocampal Nogo-A, KLOTHO, and NZF-2b gene expressions were analyzed by

Spinal cord injury (SCI) remains a severe condition that leads to permanent motor and sensory impairments, significantly affecting patients' quality of life. In recent years, neuromodulation techniques such as spinal cord stimulation (SCS), transcranial magnetic stimulation (TMS), and transcranial direct current stimulation (tDCS) have shown promising results in promoting neural plasticity and functional recovery. However, the limitations of single-modality approaches have spurred the development of multimodal neuromodulation strategies. This review systematically analyzes the integrated appli

Ageing is associated with neural alterations that impair cognitive abilities, particularly working memory (WM), which is the capacity to temporarily store and manipulate information essential for daily functioning. Transcranial direct current stimulation (tDCS) has shown promise in enhancing WM in older adults by modulating cortical excitability and promoting neuroplasticity. However, findings on the combined effects of tDCS and WM training remain inconsistent, especially regarding transfer of training gains to nontrained WM tasks (near-transfer effects). This study investigated the behavioura

INTRODUCTION: Stroke stands as one of the worldwide leading causes of disability, highlighting the need to develop new therapies. Although placebo effects in motor recovery have been reported to produce clinically meaningful improvements, the neural substrates underlying these effects, particularly across different modalities such as open-label and hidden placebo, remain unclear. This study aims to characterise the neurophysiological signatures of hidden and open placebo during motor-cortex stimulation in chronic stroke survivors and its correlation to motor recovery.

Recent studies in human motor learning have documented the involvement of higher-order somatosensory regions, specifically the rostral parietal cortex (rPC), in learning and retention, with limited participation of primary motor cortex (M1). The absence of M1 involvement suggests the recruitment of alternative frontal regions to support learning. The dorsal premotor cortex (PMd) is a primary candidate given its role in movement planning and its anatomical connectivity with rPC. However, the functional contribution of PMd to retention and the nature of its interaction with the parietal cortex-w

Ischaemic stroke is one of the leading causes of disability and death worldwide, yet effective treatment options remain limited. Bilateral hemisphere theta burst stimulation (TBS), a non-invasive brain stimulation technique, has shown considerable therapeutic potential in stroke; however, relevant research is still limited, and the specific mechanisms remain unclear. This study used a middle cerebral artery occlusion (MCAO) model in non-human primates to explore the application of bilateral hemispheric TBS in stroke rehabilitation. By integrating behavioural assessments, electrophysiology, fun

Insomnia disorder (ID) is often accompanied by cognitive deficits and disturbances in brain networks and metabolism. As a potential marker of central metabolism and oxidative stress, the role of 27-hydroxycholesterol (27-OHC) in ID pathophysiology and dCBT-I-induced neuroplasticity remains unclear. This study investigated the relationships among alterations in brain function, 27-OHC levels, and the therapeutic effects of dCBT-I. We collected resting-state fMRI data and serum samples from 32 ID patients (at baseline and after six weeks of dCBT-I) and 32 healthy controls. We compared group diffe
