Diselenide-bridged dual-action nanoscavengers drive fasting-boosted BBB penetration and ROS-responsive resveratrol delivery for synergistic ischemic stroke therapy.

The pathogenesis of ischemic stroke (IS) is critically driven by oxidative stress and the ensuing cytokine storm. While the potent reactive oxygen species (ROS) scavenger resveratrol offers neuroprotection, its targeted delivery across the blood-brain barrier (BBB) remains a substantial challenge. To address this, we developed a BBB-targeting, resveratrol-loaded nanoscavenger (Man@Res@Se-NCP; referred to as M@R@NCP) based on diselenide-bridged nanoscale coordination polymers (NCPs). This system leverages the fasting-induced membrane translocation of GLUT1 to enhance brain accumulation and sele

Cannabidiol Protects Against 1-Methyl-4-Phenylpyridinium and Manganese-Induced Neurotoxicity via Nod-Like Receptor Protein 3 Inflammasome Suppression.

Parkinson's disease (PD) is a neurodegenerative disorder characterized by dopaminergic neurodegeneration, alpha-synuclein (α-Syn) accumulation, and neuroinflammation. The NOD-Like Receptor (NLR) family pyrin domain containing 3 NLRP3 inflammasome has recently been identified as a central mediator of PD-associated inflammatory responses. Cannabidiol (CBD), a non-psychoactive phytocannabinoid, exhibits anti-inflammatory and neuroprotective properties; however, its effects on NLRP3 inflammasome in PD remain insufficiently understood. This study investigated the neuroprotective effects of CBD-rich

The neuroprotective role of coenzyme Q10 against endoplasmic reticulum stress in the olfactory bulb caused by 5 G electromagnetic field exposure.

The widespread use of 5 G mobile communication technology has raised the possibility of neurosensory effects arising from exposure to electromagnetic fields (EMF). This study investigated the effects of EMF exposure in the 5 G FR1 band on the olfactory bulb and the potential protective role of coenzyme Q10 (CoQ10). Male Sprague-Dawley rats were randomly divided into three groups. Group 1 (Control) received only saline via oral gavage. Group 2 (EMF) was exposed to EMF in the 5 G FR1 band for 2 h per day (09:00-11:00) and received oral saline. Group 3 (EMF + CoQ10) underwent the same EMF exposur

A nature-inspired peptide from the Boana cordobae frog as a potent and reversible AChE inhibitor with anti-amyloid and neuroprotective activities.

Alzheimer's disease (AD) is a multifactorial and progressive neurodegenerative disorder for which no effective treatment currently exists. The development of multitarget-directed ligands (MTDLs) capable of simultaneously modulating several pathological pathways represents a rational strategy to address its complex etiology. In this study, we report the isolation, chemical synthesis, and functional characterization of BcI-4, a short cationic peptide identified from the skin secretion of the Argentinean frog Boana cordobae. The peptide exhibited potent and reversible inhibitory activity against

Discovery of a novel Nrf2 activator that modulates mitochondrial function in neurons by regulating DHRS3-Nrf2 interaction after ischemic stroke.

Rationale: Given the crucial role of the Nrf2 pathway in cellular adaptability to stress, targeting small-molecule activation of Nrf2 represents a promising therapeutic strategy for acute ischemic stroke (AIS). However, the clinical translation of existing Nrf2 activators is hindered by adverse effects, such as liver damage, and none are currently approved for AIS. Therefore, we aimed to develop a novel Nrf2 activator that specifically activates neuronal Nrf2 while mitigating adverse effects, with the goal of providing a lead compound for AIS. Methods: We validated the anti-AIS effects and mit

Novel 2-indolinone benzylpiperidine- thiosemicarbazone hybrids as cholinesterase inhibitors: Design, synthesis and biological evaluation.

The design of this study was based on donepezil, a selective acetylcholinesterase (AChE) inhibitor used in the treatment of AD, consisting of an 1-indanone ring and a benzylpiperidine moiety. For this purpose, the 1-indanone ring was replaced with a 2-indolinone ring and new hybrid compounds were designed by connecting the benzylpiperidine residue to the 2-indolinone ring via the thiosemicarbazone bridge. Based on the results of in silico studies, 29 new 2-indolinone benzylpiperidine-thiosemicarbazone hybrid compounds (10a-n and 11a-o) were selected and synthesized. The in vitro anti-cholinest

N-Acetylcysteine protects the developing brain in neonatal sepsis-like inflammation via a redox-neurovascular pathway.

BACKGROUND: Neonatal sepsis can disrupt brain development through oxidative stress, blood-brain barrier (BBB) dysfunction, peripheral leukocyte infiltration, and white matter injury. N-acetylcysteine (NAC), a glutathione precursor with antioxidant and immunomodulatory properties, is a promising neuroprotective candidate, but its effects in neonatal sepsis-like brain injury remain incompletely defined. The purpose of this study was to investigate whether early NAC pretreatment, followed by continued treatment, was associated with protection in a neonatal LPS model and examine the principal mech

Structure characteristics of a polysaccharide isolated from safflower and its neuroprotective effects against cerebral ischemia/reperfusion injury by LASSO regression analysis of transcriptomic and metabolomic data.

Multi-omics allows for the systematic analysis of molecular information for each biological layer, while also posing the challenge of extracting valuable insights from exponentially growing multi-omics data. Here, we present a roadmap for multi-omics integrated analysis using a least absolute shrinkage and selection operator (LASSO) strategy. In this study, one novel purified safflower polysaccharide (SP1) was successfully obtained. The main chain of the polysaccharide was →4)-α-D-GalpA. Infarct size measurements and histopathology analysis results revealed the significant neuroprotective effe

Dual-acting molecular hybrid strategy: A dopamine D2 receptor agonist with synergistic anti-ferroptosis activity for the treatment of Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder. Accumulating evidence suggests iron homeostasis in the substantia nigra pars compacta and the subsequent induction of ferroptosis play pivotal roles in PD pathogenesis. In this study, we proposed a dual-acting molecular hybrid strategy and identified a promising lead compound 27e. 27e exhibited potent agonistic activity on dopamine D2 receptor (EC50 = 0.0325 nM), demonstrating 10-fold superior to clinical dopamine D2 agonist rotigotine (EC50 = 0.301 nM). Moreover, 27e showed selective iron-chelating ability (Kb = 3.3 × 1022

Lactobacillus acidophilus exerts neuroprotective effects in MPTP mice: potential links to the gut microbiota and GLP-1.

Previous studies have shown that Lactobacillus acidophilus (LA) improves gut microbiota, which is believed to play a role in Parkinson's disease pathogenesis by modulating neuroinflammation. Therefore, we investigated the role of LA in mice with PD induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP). To explore microbial-based interventions for Parkinson's disease (PD), we examined whether Lactobacillus acidophilus (LA) protects against neurodegeneration via the gut-brain axis. In MPTP-induced PD mice, LA treatment improved motor function, increased TH+ neurons, and suppressed neuro

Thiorphan improves motor function after spinal cord injury by inhibiting neuronal ferroptosis in female mice.

Spinal cord injury (SCI) causes severe motor dysfunction, yet effective pharmacological treatments remain scarce, underscoring the urgent need for novel therapeutic agents. Thiorphan has shown therapeutic potential in central nervous system disorders due to its ability to modulate oxidative stress and neuroinflammation; however, its role in SCI remains largely unknown. Therefore, we selected Thiorphan and investigated whether it improves motor recovery after SCI. Our study found Thiorphan treatment significantly enhanced motor function, as evidenced by increased BMS scores (increased from 2.2

Vitamin D and hypoxia-inducible factors signaling interplay: A hypothesis-driven review of therapeutic strategies for acute cerebral infarction.

Acute cerebral infarction (ACI) is a severe cerebrovascular disease characterized by a steadily rising global incidence rate, emerging as a leading cause of disability and mortality worldwide. Hypoxia-inducible factors (HIFs) play a pivotal role in the pathophysiological process of ACI. In recent years, mounting evidence has suggested a potential interplay between vitamin D (Vit D) and HIFs. Preclinical studies (e.g., rat cerebral ischemia models) have demonstrated that Vit D can stabilize HIF-1α, a key member of the HIFs family, which in turn reduces cerebral infarct volume by up to 50%.
