Folate Deficiency Facilitates Genomic Integration of Human Papillomavirus Type 16 DNA In Vivo in a Novel Mouse Model for Rapid Oncogenic Transformation of Human Keratinocytes.

BACKGROUND: Epidemiologic and in vitro studies suggest independent linkages between poor folate and/or vitamin B-12 nutrition, genomic human papillomavirus (HPV) type 16 viral integration, and cancer. However, there is no direct evidence in vivo to support the causative role of poor folate nutrition in HPV16 integration into the cellular genome. OBJECTIVE: We tested the hypothesis that folate deficiency enables the integration of HPV16 into the genome of HPV16-harboring keratinocytes, and could thereby influence earlier transformation of these cells to cancer in an animal model. METHODS: HPV

Vimentin Modulates Infectious Internalization of Human Papillomavirus 16 Pseudovirions.

Human papillomavirus (HPV) infection is the most common viral infection of the reproductive tract, with virtually all cases of cervical cancer being attributable to infection by oncogenic HPVs. However, the exact mechanism and receptors used by HPV to infect epithelial cells are controversial. The current entry model suggests that HPV initially attaches to heparan sulfate proteoglycans (HSPGs) at the cell surface, followed by conformational changes, cleavage by furin convertase, and subsequent transfer of the virus to an as-yet-unidentified high-affinity receptor. In line with this model, we e

Potential benefits of estrogens and progestogens on breast cancer.

The mammary gland seems to be the only organ that is not fully developed at birth. Estrogens stimulate breast tissue via estrogen receptors (ERs). In the mammary gland, ER-mediated mechanisms have been shown to regulate: various growth factors, such as TGF-alpha and TGF-beta; enzymes, such as cathepsin D and plasminogen-activator; proto-oncogenes, such as c-fos, c-myc and HER-2/neu; cyclines and other regulatory substances that provide signaling systems for cell division and differentiation; other steroid receptors and epidermal growth factor receptors. Estrogen target genes contain estrogen-r

Characterization of a t(10;12)(q24;p13) in a case of CML in transformation.

We have used Southern blotting and fluorescence in situ hybridization (FISH) to define the breakpoints of a reciprocal translocation, t(10;12)(q24;p13), acquired as a secondary abnormality in a patient with Philadelphia chromosome positive chronic myeloid leukemia (CML) in transformation. A YAC clone that spanned the breakpoint at 12p13 was identified; this YAC included the CDKN1B gene but did not include ETV6. Neither ETV6 nor CDKN1B was rearranged, as determined by FISH and Southern blotting; however, a small deletion encompassing the translocated CDKN1B allele was detected. Analysis of two

Biochemical Signature in Deciphering the Metastasis-Associated Lung Adenocarcinoma Transcript 1/microRNA-145 Axis and Its Molecular Correlation with Nuclear Factor Kappa Beta/Transforming Growth Factor-Beta and Apoptotic Proteins Expression in Colorectal Cancer.

Colorectal cancer (CRC) is a multifactorial malignancy characterized by complex interactions between oncogenic signaling networks and noncoding RNA regulators. Metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) and microRNA-145 (miR-145) have been recognized as reliable diagnostic biomarkers for cancer. This study evaluates their biochemical interaction, along with their connection to the nuclear factor kappa beta (NF-κβ)/transforming growth factor-beta (TGF-β) signaling pathway and crosstalk with vascular endothelial growth factor (VEGF) and apoptotic markers in CRC. An oncogenic role for EBV-encoded BILF1 in nasopharyngeal carcinoma.

It is widely recognized that latent infection is the predominant mode of Epstein-Barr virus (EBV) infection in nasopharyngeal carcinoma (NPC) in which a specific set of latent genes is responsible for driving the development of the disease. However, the expression of these latent genes does not adequately explain the various transformed phenotypes of NPC cells. Using an established high-throughput quantitative reverse transcription PCR array, we found that several EBV lytic genes were more widely expressed in primary NPC tissues than previously recognized. We focused on a lytic gene, BILF1, wh

Ochratoxin A drives hepatocellular carcinoma progression via GRPR-mediated activation of PLCβ/IP3R/Ca²⁺ and RPS15A/p53 pathways by regulating cancer cell fate.

Ochratoxin A (OTA), a group 2B human carcinogen, has been implicated in hepatocellular carcinoma (HCC), yet its precise molecular mechanisms remain elusive. In this study, we investigated the oncogenic role of OTA using both in vitro (Huh7 and Hep-3B cell lines) and in vivo (C57BL/6 mouse models) approaches. Molecular docking, surface plasmon resonance, co-immunoprecipitation coupled with mass spectrometry (Co-IP/MS), and functional rescue experiments were employed to elucidate the underlying pathways. Our results demonstrated that OTA exposure (50 nM) significantly promoted HCC cell prolifera

A modern review of uveal melanoma: molecular insights, clinical management, and emerging therapies.

Uveal melanoma is the most frequent primary intraocular malignancy in adults and is characterized by an aggressive clinical course, high propensity for hepatic metastasis, and poor survival after metastatic progression. Improvements in ocular imaging and local therapies have improved primary tumor control, but approximately half of patients will ultimately develop metastatic disease with poor treatment options. Recent advances in genomics and molecular biology have dramatically improved our understanding of uveal melanoma pathogenesis, with the discovery of activating mutations in GNAQ , GNA11

A novel multiplex approach for the comprehensive analysis of the Epstein-Barr virus-specific humoral immune response.

BACKGROUND: The Epstein-Barr virus (EBV) is an oncogenic herpesvirus that establishes lifelong infections in more than 90% of the human population. EBV infection is typically diagnosed using serological assays, which also distinguish between acute and persistent infection. However, current diagnostic approaches rely on a limited set of viral proteins and may therefore fail to detect disease-specific antibody signatures associated with EBV-related cancers or autoimmune diseases. Comprehensive profiling of humoral responses against the complete EBV proteome could improve the sensitivity and spec

Analysis of the coding transcriptome in NSCLC highlights variant-specific gene expression and signaling in CD74-ROS1 fusions.

The CD74-ROS1 fusion protein is an aggressive oncogenic driver detected in non-small cell lung cancer (NSCLC) patients from stages I-IV. Despite its rare occurrence, CD74-ROS1 has a notable clinical value, with ROS1 inhibitors used as first-line therapeutics in precision medicine. Upon targeting the cancer with ROS1 tyrosine kinase inhibitors (TKIs), resistance occasionally emerges for reasons that are not well-understood. Therefore, analyzing the individual roles of CD74 and ROS1 in the context of the CD74-ROS1 fusion may reveal mechanistic insights that would guide the scientific community t

A novel aurora kinase molecular glue-based degrader sp-2-067 inhibits prostate cancer cell growth and alters immune profile.

Aurora Kinase A (AKA) is overexpressed in prostate cancer and promotes disease progression through oncogenic pathways and immune modulation. AKA inhibition faces challenges in clinical trials, and alternate methods, such as degradation, may overcome some of these barriers. We evaluated SP-2-067, a novel selective AKA degrader, in C4-2 prostate cancer cells using viability assays, Western blot analysis, and siRNA knockdown. SP-2-067's dose- and time-dependent effects on AKA, related proteins (AUNIP and NINEIN) and immune checkpoint (IC) proteins (PD-L1 and LAG3) were quantified. In silico analy

Promotion of hepatocellular carcinoma by small nuclear ribonucleoprotein polypeptide F through upregulation of transmembrane P24 trafficking protein 2 and triggering of the cGAS-STING pathway.

Spliceosome abnormalities are implicated in hepatocellular carcinoma (HCC) pathogenesis, but the role of the core component small nuclear ribonucleoprotein polypeptide F (SNRPF) remains unclear. This study aims to elucidate the role and mechanism of SNRPF in HCC progression. SNRPF expression and clinical significance were analyzed using the DepMap and TCGA databases, alongside clinical HCC specimens. The results revealed that SNRPF was essential for HCC cell proliferation (CERES score = -3.02) and aberrantly overexpressed in HCC, correlating with poor prognosis. Functional validation via overe

H3K4me3 and H3K27ac Promote ccRCC Proliferation Through the CDC6-EXOSC5 Axis.

Renal cell carcinoma (RCC) is one of the most common malignant tumors of the urinary system, with clear cell renal cell carcinoma (ccRCC) accounting for more than 75% of RCC cases and representing the primary cause of mortality in renal cancer patients. CDC6 exhibits oncogenic characteristics and plays a significant role in tumor grading and prognosis prediction. Analysis of The Cancer Genome Atlas (TCGA) data shows that the CDC6 gene is significantly overexpressed in 97.22% (70/72) of paired clinical samples in ccRCC tissues compared to adjacent normal tissues. Consistent with this, elevated

NF1 and SPRED1/2 cooperate through RAS-MAPK-independent functions.

Neurofibromin, the protein product of the neurofibromatosis type 1 (NF1) gene, requires the direct binding interaction with SPRED to negatively regulate the RAS-MAPK pathway. Although the region of neurofibromin that stimulates the intrinsic GTPase activity of RAS represents only a small percentage of the entire protein, a large degree of the NF1 structural domains and their correlating mechanistic functions remain elusive. Here, we demonstrate RAS-independent biochemical and signaling functions regulated by the coordinate control of NF1 and SPRED1/2. Utilizing CRISPR-Cas9 methods to ablate NF
