SHIP1 regulates TREM2 signalling and macrophage functions in a hiPSC-derived model.

Genetic and functional studies strongly implicate microglia in the pathology of Alzheimer's disease. The Triggering Receptor Expressed on Myeloid cells 2 (TREM2) pathway is an important functional regulator of microglia, promoting phagocytosis of apoptotic cells, debris and pathogenic proteins. Genome-wide association studies have identified risk-bearing polymorphisms in several members of the pathway including INPP5D, encoding the inositol phosphatase SHIP1. Recent studies utilising in vitro and in vivo disease-relevant models have identified a role for SHIP1 in modulating TREM2 signalling an

Lung microbiota dysbiosis exacerbates influenza virus infection.

BACKGROUND: Previous studies have shown that microbiota in the respiratory tract play a significant role in shaping host immunity and reducing the risk of respiratory tract infections. Influenza, an acute respiratory infectious disease caused by the influenza virus (IAV), necessitates to have an in-depth understanding of the pathogenic mechanisms of viral infection and to utilize these mechanisms to prevent and control influenza infection. RESULTS: In this study, we established models of different disease severity levels in mice infected with IAV, the relationships between changes in pulmonar

Monocytes acquire a tumor-associated IL1B program upon encountering patient-derived colon cancer organoids.

Tumor-associated macrophages (TAMs) and monocytes that accumulate in colorectal cancer (CRC) play a crucial role in shaping the tumor microenvironment (TME) and anti-tumor immune responses. Although TAMs have been linked to both pro- and anti-tumor functions, our understanding of the cues instructing their heterogeneous phenotypes and function in cancer patients remains limited. Here, we established co-cultures comprising primary human monocytes and patient-derived organoids (PDOs) from patients with microsatellite-stable CRC to emulate myeloid/tumor cell interactions in vitro. Upon encounteri

Inhibition of CD24 Expression Enhances the Anti-Melanoma Effect of Endostatin by Ameliorating Anti-Tumor Immune Responses in Melanoma-Bearing Mice.

INTRODUCTION: Melanoma is a highly aggressive skin cancer that arises from transformed epidermal melanocytes and is one of the malignancies with the fastest proliferation rates globally. Angiogenesis has been identified as a critical target for melanoma therapy, and endostatin has been verified to impede endogenous angiogenesis. However, some patients' therapeutic responses remain unsatisfactory. CD24, which functions as an anti-phagocytic signal, represents a potential target for tumor immunotherapy. Thus, in this study, we investigated the anti-melanoma effects of combined CD24 inhibition an

Fluorinated lipid nanoparticles enable in vivo CAR-macrophage therapy in solid tumor and enhance anti PD-L1 immunotherapy.

In vivo CAR-macrophage (CAR-M) therapy exploits macrophages' tumor-tropic homing, phagocytic capacity, and ability to remodel the immunosuppressive tumor microenvironment, with low risk of cytokine release syndrome and potential as an off-the-shelf therapy. However, clinical translation is currently constrained by the lack of efficient non-viral platforms for in situ macrophage engineering. To address this challenge, fluorinated ionizable lipids were rationally designed-capitalizing on fluorine's high electronegativity, low polarizability, and lipophobicity-to enhance lipid nanoparticle (LNP)

Melanosome Clusters and Apoptotic Melanocytes Internalized by Dermal Fibroblasts Impairs Cellular Function.

Skin pigmentation disorders with dermal melanin involvement are therapeutically challenging due to poor responsiveness to conventional treatments and unclear pathogenesis. While melanin in the dermis is typically cleared by macrophages, human dermal fibroblasts (HDFs) can also internalize extracellular melanosome clusters (EMC) and apoptotic melanocytes that have occasionally been observed in the dermis, which may contribute to persistent pigment-related abnormalities. However, the functional impact of phagocytosis for fibroblasts remains unclear. In this study, HDFs were incubated with EMC or

Inhibition of Pseudomonas aeruginosa Biofilm Formation by Peptidyl-Arginine Deiminases 2 and 4.

INTRODUCTION: Pseudomonas aeruginosa is a significant pathogen associated with acute and chronic infections, particularly in immunocompromised individuals. Its capacity for biofilm formation, combined with antibiotic resistance, plays a critical role in the persistence of these infections. Peptidyl-arginine deiminases (PADs), including PAD2 and PAD4 isoforms, are involved in neutrophil phagocytic killing of P. aeruginosa. This study aimed to investigate the impact of PAD enzymes on biofilm development and virulence factor production in P. aeruginosa, with focus on the multidrug resistant strai

Impaired removal of dying brain cells by microglia in Gpr34 deficient mice.

GPR34, a G protein-coupled receptor present selectively in microglia and other myeloid cells, is highly expressed in homeostatic microglia but is downregulated in disease associated microglia (DAM) such as those found in Alzheimer’s disease brain. However, little is known about GPR34’s role in microglia function or brain development. Here, we studied Gpr34 knockout (KO) mice at postnatal 18-day (P18) and 3 months (3 mo) age. In the brains of P18 Gpr34 KO mice, there were elevated numbers of neurons, oligodendrocytes and microglia, many of which were IHC-positive for the cell death markers clea

Akt at the crossroads of microglial function: a double-edged sword in Alzheimer's disease neuroinflammation.

Alzheimer's disease (AD) pathology is increasingly understood to be driven by complex neuroinflammatory processes, with microglia-the resident immune cells of the brain playing a pivotal role. The PI3K/Akt signaling pathway serves as a critical intracellular hub, orchestrating the diverse and often opposing functions of microglia. This review synthesizes current insights into the multifaceted role of Akt signaling in modulating microglial activity in the context of AD. We explore the dualistic nature of Akt, which can promote pro-inflammatory neurotoxicity through pathways such as NF-κB while

Interweaving microglial senescence and gut microbiome dynamics in Alzheimer's disease - Mechanisms and therapeutic frontiers.

Alzheimer's disease (AD), a prevalent neurodegenerative disorder characterized by cognitive impairment and neuronal degeneration, is increasingly recognized as being driven not only by the traditional amyloid-beta and tau pathologies but also by persistent neuroinflammation and systemic immune dysregulation. Emerging evidence implicates microglia senescence and gut microbiota dysbiosis is critical contributors to the neuroinflammatory landscape. Senescent microglia marked by reduced phagocytic ability and a pro-inflammatory secretory profile, are unable to clear pathogenic stimuli, thereby int

Melanoma exosomal miR-708-5p promotes macrophage M2 polarization and cancer metastasis.

Monocyte-derived macrophages are usually recruited and play pivotal roles in establishing an immunosuppressive tumor microenvironment, and the interplay between tumor cells and tumor-associated macrophages (TAMs) is crucial for tumor development. However, the detailed mechanisms remain largely unelucidated in certain aggressive human cancers, such as melanoma. Here, through miRNA sequencing analysis, we found the microRNA miR-708-5p was highly enriched in melanoma exosomes, which was dependent on SFRS1. Treatment by melanoma exosomes facilitated M2 polarization of macrophages, while the polari

Human induced pluripotent stem cell-derived chimeric antigen receptor-macrophages eradicate IL-13Rα2-positive solid tumors.

Macrophages exhibit extensive tumor infiltration capacity across diverse solid malignancies, establishing macrophage-targeted immunotherapies as an emerging frontier in oncology. Genetic engineering of macrophages using chimeric antigen receptor (CAR) technology - enabling recognition and phagocytosis of neoplastic cells - is emerging as a potential therapeutic strategy against solid tumors. Human induced pluripotent stem cells (iPSCs) provide a renewable platform for the efficient differentiation of functionally competent macrophages. In this study, we engineered human iPSC-derived CAR macrop

Bacterial metabolites induce cell wall remodeling, antifungal resistance, and immune recognition of commensal fungi.

The fungus Candida albicans commensally colonizes mucosal surfaces in healthy individuals but can cause both superficial mucosal and life-threatening disseminated infections. The balance between commensalism and pathogenicity is complex and depends on factors including host and fungal genetic background, the host environment, and fungal interactions with local microbes. The major interaction interface of C. albicans with the host is its multilayered cell wall, which is dynamic and highly responsive to the surrounding environment. Therefore, factors that influence the fungal cell wall will dire

In Vivo Spatiotemporal Protection and Recognition of Circulating Tumor DNA for Early Cancer Diagnosis and Monitoring.

Circulating tumor DNA (ctDNA) offers a promising avenue for noninvasive cancer diagnosis and prognosis, yet its effective utilization is fundamentally limited by rapid in vivo clearance and nuclease-mediated degradation, resulting in extremely low bioavailability. In addition, conventional in vitro recognition-based ctDNA detection means may potentially compromise the diagnostic accuracy. Here, we report an in vivo spatiotemporal protection and recognition strategy that actively enhances ctDNA bioavailability, enabling accurate early cancer diagnosis and quantitative monitoring of disease prog

Microglial handling of myelin debris and remyelination after ischaemic stroke: recognition, intracellular processing, lipid fate, and oligodendroglial repair.

Post-ischaemic white-matter injury involves axonal dysfunction, myelin disorganization, delayed accumulation of myelin-derived debris, and an often incomplete regenerative response. Microglia provide a cellular link between debris clearance and white-matter repair through a sequence of functionally distinct processes encompassing selective debris recognition, internalization, intracellular degradation, lipid handling, inflammatory regulation, and oligodendroglial support. Importantly, increased myelin uptake should not be equated with successful clearance, and clearance itself does not establi

Convergent evolution of immune evasion in ESKAPE pathogens: A cross-pathogen architecture of conserved host-defense checkpoints.

Antimicrobial resistance in ESKAPE pathogens is primarily attributed to resistance genes, yet persistent infections despite appropriate therapy implicate immune evasion as an independent driver of treatment failure. Although immune-evasion mechanisms have been extensively characterized in individual pathogens, their shared architecture across the ESKAPE group remains insufficiently integrated. This review synthesizes current evidence to show that phylogenetically diverse ESKAPE pathogens have convergently evolved conserved strategies to evade host immunity under comparable selective pressures.

Co-activating the intrinsic FcRγ/TLR4 signaling axis enhances the antitumor activity of NKG2D CAR-macrophages against prostate cancer.

Prostate cancer presents a major therapeutic challenge due to its immunosuppressive "cold" tumor microenvironment. To address this, we focused on harnessing the inherent phagocytic killing activity and immunomodulatory capacities of macrophages. Here, we explore an innovative strategy to enhance the therapeutic function of chimeric antigen receptor macrophages (CAR-Ms) through targeted engineering. We designed and constructed three distinct CAR-M variants targeting NKG2D ligands: a truncated control (Truncated-CAR-M), a construct containing the Fc receptor gamma chain (FcRγ) signaling domain (

Lacticaseibacillus paracasei LPB27: Effectively Enhances the Immune System by Improving the Intestinal Environment.

Lacticaseibacillus paracasei LPB27 is a novel probiotic isolated from the intestines of healthy infants and young children. In this study, healthy mice were used as a model to systematically investigate the immune regulatory effects of LPB27 mediated by the microbiota-gut-immune axis, with a focus on evaluating its immune-enhancing potential. The safety evaluation showed that, at a dose of 0.50 g/kg/day, LPB27 caused no observable toxicity or adverse reactions. The evaluation of immune function indicated that in terms of cellular immunity, high-dose LPB27 increased the delayed-type hypersensit

Extraction, structural characterization, and immunomodulatory activity of a high-molecular-weight polysaccharide from cultured ophiocordyceps.

Ophiocordyceps (Ophiocordyceps sinensis) is a precious medicinal and edible fungus widely utilized in nutraceuticals and functional foods. However, the structural intricacies and biological potential of its high-molecular-weight polysaccharides remain largely unknown. In the present study, a novel high-molecular-weight neutral polysaccharide, designated as CSP1b, was isolated and purified from cultured ophiocordyceps via water extraction, ethanol precipitation, and sequential column chromatography. Characterization revealed that CSP1b possesses an extraordinary molecular weight of 2.65 × 107 D

TSC-associated microglial hyperactivity: enhanced calcium signaling, metabolism, and phagocytosis.

Tuberous sclerosis complex (TSC) is a multisystem genetic disorder with prominent neurological manifestations, most notably epilepsy, and is frequently accompanied by a wide range of neuropsychiatric comorbidities. Hyperactivation of the mechanistic target of rapamycin (mTOR) pathway plays a central role in TSC pathology, disrupting both general brain development and specific molecular processes such as metabolism. While much attention has focused on neurons and astrocytes in these TSC-related alterations, the contribution of microglia remains relatively underexplored. In this study, we first
