Inducible, split base editors for in vivo cancer functional genomics.

Cancer functional genomics using CRISPR base editors (BEs) holds great promise for molecular characterization and new target discovery. However, traditional BEs, using intact DNA deaminases as mutators, are often constrained by limited control and nonspecific toxicities. Here we developed a small-molecule-controllable system using split-engineered BEs (seBEs). By placing deaminase activity under small-molecule control, seBEs significantly reduced cellular toxicity and enabled robust and inducible in vivo functional genomics screens. High-density seBE genetic screens using ~11,000 single guide

Targeting Mitochondria for Postoperative Cognitive Dysfunction: From Mechanisms to Therapeutics.

Postoperative cognitive dysfunction (POCD) is a frequent neurological complication in older surgical patients, leading to substantial declines in quality of life and placing a considerable burden on society. This review provides a comprehensive overview of how mitochondrial dysfunction contributes to POCD, including disrupted energy metabolism, excessive reactive oxygen species production, calcium imbalance, and abnormalities in mitochondrial dynamics and quality control processes. These mitochondrial impairments further trigger neuroinflammation and activate multiple cell death pathways. In a

The estrogen-ferroptosis axis in postmenopausal osteoporosis, osteoarthritis, and intervertebral disc degeneration: shared mechanisms and emerging evidence.

Estrogen deficiency is a major risk factor for degenerative disorders of the musculoskeletal system in postmenopausal women, while ferroptosis, an iron-dependent form of programmed cell death, has been implicated in the pathogenesis of various osteoarticular diseases. Recent studies indicate an interplay between estrogen signaling and ferroptosis, with evidence relatively well supported in postmenopausal osteoporosis (PMOP), emerging in osteoarthritis (OA), and still limited in intervertebral disc degeneration (IVDD). Estrogen deficiency may disrupt iron homeostasis, lipid metabolism, and anti

Ewing sarcoma - current diagnostic and therapeutic approaches.

BACKGROUND: Ewing sarcoma belongs to the family of undifferentiated small round cell sarcomas of bone and soft tissue. It is characterized by a gene fusion involving EWSR1 and an ETS-family transcription factor gene. In 85-96% of cases, a specific chromosomal translocation results in the EWSR1-FLI1 fusion gene, whose product functions as an oncogene essential for tumorigenesis. Ewing sarcoma is most common in adolescents and young adults. It primarily affects the diaphyses of long bones, the pelvis, and the axial skeleton, although extraosseous involvement is not uncommon. This is a highly mal

Nanomedicine targeting vascular endothelial dysfunction in metabolic diseases: Mechanisms and therapeutic opportunities.

Vascular endothelial dysfunction is a central pathophysiological mechanism underlying a broad spectrum of metabolic diseases, including type 2 diabetes mellitus, atherosclerosis, obesity, and non-alcoholic fatty liver disease. As a metabolically active interface, the endothelium regulates vascular tone, inflammation, oxidative stress, and nutrient exchange; its impairment contributes to the initiation and progression of these disorders. This review synthesizes current understanding of the molecular mechanisms linking endothelial dysfunction to metabolic disease progression. Particular focus is

Standardized Isolation, Culture, and Imaging of Contracting Cardiomyocytes from Neonatal Mouse Hearts.

Cardiomyocyte maturation represents the final stage of functional development in the heart. During this critical process, fetal cardiomyocytes gradually acquire adult phenotypes and become capable of sustaining billions of contraction-relaxation cycles throughout an organism's lifespan. This transition is driven by gene expression changes that remodel cardiomyocyte structure, metabolism, and functions. In mice, cardiomyocyte maturation takes place during embryonic day (E)18.5 to postnatal two weeks, during which the heart loses its regenerative capacity in response to injury. Hence, characteri

Transdermal Drug Delivery Systems: Methods for Enhancing Skin Permeability and Their Evaluation.

Transdermal drug delivery (TDD) is an increasingly important non-invasive method for administering active pharmaceutical ingredients (APIs) through the skin barrier, offering advantages such as improved therapeutic efficacy and reduced systemic side effects. As demand increases for patient-friendly and minimally invasive treatment options, TDD has attracted substantial attention in research and clinical practice. This review summarizes recent advances enhancing skin permeability through chemical enhancers (e.g., ethanol, fatty acids, terpenes), physical (e.g., iontophoresis, microneedles, sono

Implantable Cardioverter-Defibrillators for Secondary Prevention in Giant Cell Myocarditis.

Giant cell myocarditis (GCM) presents significant challenges in clinical management, particularly regarding the role of implantable cardioverter-defibrillators (ICDs) for secondary prevention of ventricular tachycardia (VT). We present the case of a 42-year-old female patient with histologically confirmed GCM who underwent orthotopic heart transplantation and subsequently developed VT due to biopsy-proven recurrent GCM within one year after transplant. An ICD was placed for secondary prevention following multiple episodes of monomorphic nonsustained VT. The patient would then present to the ho

Chromoplexy: A Pathway to Genomic Complexity and Cancer Development.

Chromoplexy is a phenomenon of complex genome rearrangement, occurring during a single cell event and characterized by the formation of chain rearrangements affecting multiple chromosomes. Unlike other genomic rearrangements such as chromothripsis, which involves a single chromosome, chromoplexy affects several chromosomes at once, creating patterns of complex, balanced translocations, and leading to the formation of fusion genes and the simultaneous disruption of several genes. Chromoplexy was first identified in prostate cancers, but it is now observed in various cancers where gene fusions t

Levodopa and the dopamine receptor D1-Hippo/yes-associated protein axis: A novel therapeutic avenue for liver fibrosis.

Liver fibrosis remains a major global health challenge with limited therapeutic options. In their recent study, Wang et al report that levodopa, a dopamine precursor widely used in Parkinson's disease, significantly attenuates carbon tetrachloride-induced liver fibrosis in rats by enhancing dopamine receptor D1 expression and activating the Hippo signaling pathway, leading to phosphorylation and inactivation of yes-associated protein 1. This discovery links G-protein-coupled receptor signaling to Hippo pathway regulation in hepatic fibrosis. The work highlights the dopamine receptor D1-Hippo/y

[Risks of acute kidney injury with intravascular administration of an iodine-containing contrast agent in patients with acute pyelonephritis].

The occurrence of acute kidney damage associated with the introduction of an iodine-containing contrast agent into the vascular bed, which leads to transient or persistent structural and functional changes in nephrons, endothelial cells, and renal tubule epithelium, is an urgent problem in modern instrumental diagnostics of various pathological processes, since iodine-containing preparations have a strong advantage over "native" scanning. A researcher who uses different techniques depending on the tasks set opens up great prospects in studying many aspects of the structure and function of tiss

Regenerative Cartilage Treatment for Focal Chondral Defects in the Knee: Focus on Marrow-Stimulating and Cell-Based Scaffold Approaches.

Focal chondral defects of the knee are a common cause of pain and functional limitation in active individuals and may predispose to early degenerative joint changes. Given the limited regenerative capacity of hyaline cartilage, biologically based surgical strategies have emerged to promote tissue repair and restore joint function. This narrative review critically examines current treatment approaches that rely on autologous cell sources and scaffold-supported regeneration. Particular emphasis is placed on techniques that stimulate endogenous repair or support chondrocyte-based tissue restorati

RNA methylation: A new perspective in osteoarthritis research.

Osteoarthritis (OA) is a prevalent degenerative joint disease characterized by cartilage degradation, osteophyte formation, and joint dysfunction, significantly impairing the quality of life in the elderly population. Recently, RNA modifications, as a dynamic and reversible epigenetic modification, have emerged as critical players in the onset and progression of OA. This review systematically summarizes the major types of RNA modifications involved in OA, including N6-methyladenosine (m6A), 5-methylcytosine (m5C), and 7-methylguanosine (m7G), and explores their roles in regulating chondrocyte

A Mouse Model of Sporadic Alzheimer's Disease with Elements of Major Depression.

After olfactory bulbectomy, animals are often used as a model of major depression or sporadic Alzheimer's disease and, hence, the status of this model is still disputable. To elucidate the nature of alterations in the expression of the genome after the operation, we analyzed transcriptomes of the cortex, hippocampus, and cerebellum of the olfactory bulbectomized (OBX) mice. Analysis of the functional significance of genes in the brain of OBX mice indicates that the balance of the GABA/glutamatergic systems is disturbed with hyperactivation of the latter in the hippocampus, leading to the devel

Multiple measures for self-identification improve matching donors with patients in unrelated hematopoietic stem cell transplant.

BACKGROUND: Questions persist around whether and how to use race or geographic ancestry in biomedical research and medicine, but these forms of self-identification serve as a critical tool to inform matching algorithms for human leukocyte antigen (HLA) of varying levels of resolution for unrelated hematopoietic stem cell transplant in large donor registries. METHODS: Here, we examined multiple self-reported measures of race and ancestry from a survey of a cohort of over 100,000 U.S. volunteer bone marrow donors alongside their high-resolution HLA genotype data. APP dyshomeostasis in the pathogenesis of Alzheimer's disease: implications for current drug targets.

The Amyloid precursor protein (APP) is a transmembrane glycoprotein from which amyloid-β (Aβ) peptides are generated after proteolytic cleavage. Aβ peptides are the main constituent of amyloid plaques in Alzheimer's Disease (AD). The physiological functions of APP in the human adult brain are very diverse including intracellular signaling, synaptic and neuronal plasticity, and cell adhesion, among others. There is growing evidence that APP becomes dysfunctional in AD and that this dyshomeostasis may impact several APP functions beyond Aβ generation. The vast majority of current anti-amyloid ap
