Brain and C-type natriuretic peptide are expressed by the human placenta and are not altered with preeclampsia, fetal growth restriction, or placental hypoxia.

INTRODUCTION: The natriuretic peptide system is a critical regulator of hemodynamic function. Yet its expression in the human placenta has had limited investigation, despite its clear relevance to pregnancy. Preeclampsia and fetal growth restriction are serious conditions of pregnancy, whereby placental dysfunction is central to their pathophysiology. Here, we set out to characterise expression of C-type natriuretic peptide (CNP) and Brain natriuretic peptide (BNP) in human placenta. METHODS: CNP, BNP, NPR-B and NPR-C expression was assessed in placental tissue from preeclamptic (preterm n =

Fetal vascular malperfusion and other placental lesions in relation to maternal SARS-CoV-2 exposure: a case-control study.

BACKGROUND: The morphologic characteristics of placentas associated with coronavirus disease-19 (COVID-19) have been described but with varying findings. Studies conducted at the onset of the pandemic reported frequency of Fetal Vascular Malperfusion (FVM), an umbrella term that describes occlusive vasculopathies that affect fetal circulation and their associated sequelae. Morphologic manifestations include thrombosis, avascular villi and vascular stromal karyorrhexis. Associated outcomes include fetal growth restriction, thromboembolic events such as neonatal stroke and fetal demise. Placenta

Redox imbalance and mitochondrial dysfunction in gestational diabetes mellitus: Implications for maternal health, placental function, and offspring metabolic programming.

Mitochondrial dysfunction and redox imbalance are increasingly recognised as essential factors in the pathophysiology of gestational diabetes mellitus (GDM). Impaired oxidative phosphorylation in the placenta, increased production of reactive oxygen species (ROS), and insufficient antioxidant protection impair bioenergetics and affect maternal-foetal nutrient exchange. Such changes promote excessive foetal growth and metabolic programming while predisposing mothers to future type 2 diabetes. Current research indicates alterations in mitochondrial dynamics and mitophagy as additional risk facto

SLC6A8-centered placental molecular signature predicts fetal growth restriction via machine learning and single-cell analysis.

Fetal growth restriction (FGR) is a common and severe complication during pregnancy, serving as a primary cause of intrauterine fetal distress and perinatal mortality. Its pathogenesis involves placental insufficiency, impaired nutrient transport, and metabolic disturbances, yet key molecular regulators remain incompletely understood. Using machine learning, we identified a 5-placental gene signature (CLDN9, GCNT4, PIM3, SLC6A8 and TPBG) for robustly predicting FGR and validated its performances in multiple datasets. Among those, SLC6A8 expression was significantly upregulated in of FGR placen

Placental Heat Shock Protein (HSP) Expression in Fetal Growth Restriction (FGR) Pregnancies: A Case-Control Immunohistochemistry Study.

Fetal growth restriction (FGR) is frequently defined as the failure of the fetus to reach its genetically predetermined growth potential. Heat shock proteins (HSPs) are extreme-temperature-resistant molecules that help proteostasis. The aim of this prospective case-control immunohistochemistry study is to evaluate the expression of HSP90 and HSP70 in the placentas of pregnancies complicated with FGR and compare their levels with the control placentas of normal-growth pregnancies. A prospective case-control study was conducted including people undergoing singleton pregnancies who gave birth in

MyD88 signaling in trophoblasts contributes to maternal high fat diet-associated alterations in placental gene expression and lipid profiles.

INTRODUCTION: Maternal obesity contributes to programming of offspring obesity. A causal pathway for placental inflammatory changes in utero with obesity remains to be established. We examined the role of the toll-like receptor (TLR) pathway in mediating maternal obesity associated changes in placental gene expression using trophoblast-specific deletion of the TLR adapter protein, MyD88. METHODS: Female MyD88 conditional knockouts (MyD88-CKO) at 5 weeks of age were provided control (17% fat calories, CON) or high-fat diets (HFD, 45% fat calories) for 12 weeks and mated with MyD88 (flox-contro

Evaluation of a deep generative computational framework under constrained conditions for multi-regional placental single-cell transcriptomic inference.

INTRODUCTION: The placenta is a highly heterogeneous organ, and its dysfunction is central to preeclampsia (PE). Although single-cell RNA sequencing (scRNA-seq) enables high-resolution characterization, its high cost limits large-scale clinical use. scSemiProfiler integrates deep generative modeling with bulk RNA-seq to infer single-cell transcriptomes, but the original framework relies on an active learning module to select informative samples. In this study, we evaluate a simplified workflow that omits active learning, assessing its performance under these constrained conditions in complex,

Placental growth factor (PLGF)-based testing to help diagnose suspected pre-eclampsia: a systematic review and economic evaluation.

BACKGROUND: Predicting a diagnosis of pre-eclampsia is based on a combination of clinical assessment of blood pressure, presence of protein in the urine, symptoms and laboratory test abnormalities. Accurately detecting pre-eclampsia is important to avoid false-positive diagnoses which could lead to unnecessary antenatal admissions and/or preterm delivery. Four blood tests that measure the biomarkers of placental growth factor or the ratio of soluble fms-like tyrosine kinase-1 to placental growth factor are available (known as Triage, Elecsys, DELFIA Xpress, and BRAHMS Kryptor tests). Abnormal

Mercury chloride exposure disrupts placental function and embryonic methionine metabolism leading to neural tube defects in ICR mice.

The aim of the study was to explore the precise mechanisms involved in maternal mercury exposure induced neural tube defects (NTDs). We established a mouse model of mercury chloride (HgCl2)-induced embryo NTDs using ICR mice, and investigated the roles of amino acid transport and associated methionine metabolic disorders in the placenta and fetus. Oral administration of 0, 1.5, 3, and 4.5 mg/kg/d HgCl2 to pregnant ICR mice was performed between gestation days 5.5 and 9.5, with assessments conducted on gestation day 10.5. The incidence of NTDs was significantly elevated in the 3 mg/kg/d group w

Low-molecular-weight components of placental extract inhibit degranulation by blocking Ca2+ influx in rat basophilic leukemia cells.

BACKGROUND: Placental extract (PE) is rich in peptides, amino acids, nucleic acids, vitamins, and minerals. Although PE is used for skin care and as a health food, it is also considered effective in the treatment of various diseases. Mast cells play an essential role in allergic diseases by secreting various inflammatory mediators, such as histamine, leukotrienes, and prostaglandins. PE has inhibitory effects on mast cell activation; however, the molecular mechanisms underlying these inhibitory effects remain unclear. The aim of this study was to investigate the mechanisms underlying the inhib

Stepwise breach of maternal-fetal fortress: an adhesion-degradation-immune evasion cascade underpinning Treponema pallidum placental crossing.

Congenital syphilis, a severe maternal-fetal disease caused by vertical transmission of Treponema pallidum (T. pallidum) via the placenta, affects over one million pregnancies annually and remains a leading cause of preventable adverse fetal outcomes. While the clinical consequences are well recognized, the molecular mechanisms underlying placental penetration are incompletely understood. This review synthesizes current evidence into a unified conceptual framework known as the adhesion-degradation-immune evasion cascade. Established findings, supported by direct placental studies, indicate tha

Analysis of drug transporter expression in syncytiotrophoblast derived from human placental stem cells: Expression and function of efflux transporters.

OBJECTIVE: The placenta is a vital organ for exchanging nutrients, endogenous substances, and xenobiotics between mother and fetus. The syncytiotrophoblast (ST) is crucial in maintaining the placental barrier. Human trophoblast stem cells (hTSCs) have been recently established; however, their utility in studying placental transport functions has not been fully elucidated. This study investigated the expression and function of transporters in hTSC-derived ST cells. METHODS: TSCT cells, as hTSCs, were differentiated into ST-like cells (ST-TSCT), and the gene expression of 84 transporters in ST-

27-hydroxycholesterol impairs placental development via p53/p21/Cdk6 pathway: Implications for nutrient transport and cellular senescence.

Aberrant placental development and function contribute to various pregnancy complications. 27-hydroxycholesterol (27-OHC), a recognized mediator linking hypercholesterolemia and metabolic diseases, has an undefined role in placental development. This study investigates the impact of 27-OHC on placental development and its underlying mechanisms, particularly in relation to cellular senescence. Pregnant mice were subcutaneously administered either 27-OHC (27-OHC group) or normal saline (control group) during gestation. Subsequently, placentas underwent spatial transcriptome (ST) sequencing. Vertical transfer of bacterial symbionts via a placental analogue in the cyclostome bryozoan Patinella verrucaria (Stenolaemata): Ultrastructural and molecular evidence.

Symbiotic associations with prokaryotes are common among marine filter-feeding invertebrates. In the almost exclusively colonial phylum Bryozoa, however, such associations have only been recorded in some species of the order Cheilostomata (class Gymnolaemata). Here we describe for the first time symbiotic bacteria in the colonies, larvae and developing ancestrulae of the bryozoan Patinella verrucaria from the order Cyclostomata (class Stenolaemata) using transmission electron and fluorescent microscopy. Ultrastructural and molecular data suggest the existence of two distinct bacterial species,

Aberrant placental structure is corrected with repeated nanoparticle-mediated IGF1 treatments in a Guinea pig model of fetal growth restriction.

BACKGROUND: Fetal growth restriction (FGR) is most commonly due to placental insufficiency. There are currently no treatments for placental insufficiency or FGR, and the only intervention is iatrogenic pre-term delivery. We have previously shown efficacy of repeated placental nanoparticle-mediated insulin-like 1 growth factor (IGF1) treatment in improving placental efficiency (increased fetal-placental weight ratio) and correcting fetal growth in a maternal nutrient restriction (MNR) guinea pig model of FGR. We hypothesize placenta structural changes (reduced exchange area, altered vascular st
