Marine Guanidine Alkaloids Inhibit Malaria Parasites Development in In Vitro, In Vivo and Ex Vivo Assays.

Malaria is a disease caused by pathogenic protozoa Plasmodium spp., with a significant global impact on human health. Increasing resistance of P. falciparum strains to drugs treating malaria highlights the urgent need for the discovery of new antimalarial candidates. Batzelladines are marine guanidine alkaloids that exhibit potent antiparasitic activity. Herein, results of the parasitological profiling assessment of batzelladines F and L are reported. Both compounds exhibited potent antiplasmodial activity, moderate cytotoxicity, and suitable selectivity indexes. Batzelladines F and L are fast

Alternative transmission routes in the malaria elimination era: an overview of transfusion-transmitted malaria in the Americas.

BACKGROUND: Transfusion-transmitted (TT) malaria is an alternative infection route that has gained little attention from authorities, despite representing a life-threatening condition. There has been no systematic review of this health problem in American countries. The aim of this study was to describe the clinical and epidemiological characteristics of TT malaria in the Americas and identify factors associated with lethality based on the studies published in the literature. METHODS: Potentially relevant papers in all languages were retrieved from MEDLINE and LILACS. Additional articles were

Hematozoa in two populations of the threatened red-billed chough in Spain.

The prevalence of hematozoa in two populations of red-billed choughs (Pyrrhocorax pyrrhocorax) was sampled in 1992 and 1994 in Spain. Two blood parasites infected red-billed choughs. A species of Plasmodium, possibly Plasmodium relictum, and the piroplasm Babesia frugilegica, are described for the first time from this host. Low prevalence (1/178, < 1%) of hematozoa in these populations, was evidence for a lack of effects of blood parasites on the life history and conservation of this threatened species in at least the two populations studied.

Studies with induced malarias in Aotus monkeys.

Aotus monkeys have been shown to be susceptible to the three most important species of human malaria as well as to simian, ape, and rodent malaria. The parasites natural to man were maintained by serial passages using trophozoite infected blood. It was then possible to transfer the infections to, and among, other New World monkeys which previously were refractory. The parasites retained their viability in the monkey, were infective to mosquitos, and were transmitted between monkeys and back to man. Exoerythrocytic stages, similar to those demonstrated in man, occurred in the Aotus liver. Drugs

Malaria-Associated Acute Kidney Injury: A Key Driver of Mortality in Endemic Regions.

BACKGROUND: Malaria remains a major global health challenge, particularly in endemic regions where severe disease contributes substantially to morbidity and mortality. Among its systemic complications, malaria-associated acute kidney injury (MAKI) is increasingly recognized as a significant yet underappreciated contributor to poor clinical outcomes. Despite advances in antimalarial therapy, MAKI continues to be associated with high mortality and growing evidence suggests a link to long-term renal sequelae, including chronic kidney disease (CKD). This review synthesizes current evidence on the

Validation of a Semiquantitative Real-Time PCR Assay for Malaria Diagnosis and Treatment Monitoring.

Microscopic examination of blood smears is the current gold standard for laboratory confirmation of malaria. However, it is time-consuming and lacks sensitivity while requiring highly skilled laboratory professionals. This study validated a multiplex real-time PCR test for the detection, speciation, and semiquantification of Plasmodium species in whole blood. Analytical results showed the assays were highly specific and sensitive, with a PCR efficiency of 100.21% and 101.99% for Plasmodium falciparum and Plasmodium vivax, respectively. The limit of detection was 0.0001% parasitemia for both P.

Synthesis of β-fluoropyrrolidines designed to reduce hERG affinity in a class of antiparasitic compounds.

Malaria is a parasitic disease caused by the parasite Plasmodium which results in hundreds of thousands of deaths per year. The need for new drugs for the treatment of this disease is imperative as new resistant strains of Plasmodium emerge. Previously, our group reported two related series of pyrrolidine-based antimalarial compounds. While both series were found to be orally efficacious in a mouse infection model, some analogs also showed modest affinities for the hERG ion channel. Herein, we describe the attempted synthesis of β-fluoro substituted pyrrolidine analogs of both chemotypes. In t

Random Forest Model Predictions Afford Dual-Stage Antimalarial Agents.

The need for novel antimalarials is apparent given the continuing disease burden worldwide, despite significant drug discovery advances from the bench to the bedside. In particular, small-molecule agents with potent efficacy against both the liver and blood stages of Plasmodium parasite infection are critical for clinical settings as they would simultaneously prevent and treat malaria with a reduced selection pressure for resistance. While experimental screens for such dual-stage inhibitors have been conducted, the time and cost of these efforts limit their scope. Here, we have focused on leve

Reinventing malaria control: hierarchical intervention strategies for African settings based on the microbiota-barrier-innate immunity axis.

Malaria control is challenged by parasite drug resistance and inadequate vaccine protection. Although mosquito gut microbes regulate Plasmodium development, integrated analyses of microbiota-Plasmodium-host interactions are lacking. Innate immunity imbalance is the primary driver of disease pathogenesis. This review proposes a unified "microbiota-barrier-innate immunity" axis hypothesis. Under physiological conditions, gut microbiota modulates host innate immunity via metabolites and maintains barrier integrity. However, Plasmodium infection disrupts this axis, causing microbiota imbalance, me

Atovaquone, chloroquine, primaquine, quinine and tetracycline: antiproliferative effects of relevant antimalarials on Neospora caninum.

Neospora caninum is an apicomplexan parasite that causes abortion in cattle, resulting in significant economic losses. There is no commercial treatment for neosporosis, and drug repositioning is a fast strategy to test possible candidates against N. caninum. In this article, we describe the effects of atovaquone, chloroquine, quinine, primaquine and tetracycline on N. caninum proliferation. The IC50 concentrations in N. caninum were compared to the current information based on previous studies for Plasmodium and Toxoplasma gondii, correlating to the described mechanisms of action of each teste

Discovery and Structural Optimization of Acridones as Broad-Spectrum Antimalarials.

Malaria remains one of the deadliest diseases in the world today. Novel chemoprophylactic and chemotherapeutic antimalarials are needed to support the renewed eradication agenda. We have discovered a novel antimalarial acridone chemotype with dual-stage activity against both liver-stage and blood-stage malaria. Several lead compounds generated from structural optimization of a large library of novel acridones exhibit efficacy in the following systems: (1) picomolar inhibition of in vitro Plasmodium falciparum blood-stage growth against multidrug-resistant parasites; (2) curative efficacy after
