Cardiovascular Effects of Kratom (Mitragyna speciosa).

Mitragyna speciosa (kratom) is an increasingly used product marketed for pain relief, mood enhancement, stimulation, and self-management of opioid withdrawal. Its principal alkaloids, mitragynine and 7-hydroxymitragynine, interact with μ-opioid, adrenergic, serotonergic, and dopaminergic receptors and may also influence cardiac ion channels and autonomic regulation. Experimental studies demonstrate inhibition of the human ether-à-go-go-related gene potassium channel, delayed ventricular repolarization, and potential effects on sodium and G-protein-coupled inwardly rectifying potassium channels

A novel urinary metabolic signature for the non-invasive screening of lupus nephritis in systemic lupus erythematosus.

Lupus nephritis (LN) is a severe complication of systemic lupus erythematosus (SLE) that requiring early screening. This study preliminarily explored urinary metabolomic profiles to identify potential, non-invasive candidate biomarkers for the early diagnosis of LN. Urine samples were collected from 55 participants: 20 SLE patients with LN, 20 SLE patients without LN, and 15 healthy controls. Untargeted liquid chromatography-mass spectrometry (LC-MS) was utilized to determine metabolic profiles, and support vector regression was applied for data normalization. The Least Absolute Shrinkage and

An Adaptive Multi-Scale Manifold Embedding Preprocessing Framework for High-Dimensional Data Visualization.

To improve the robustness of neighborhood construction and the separation of intra-cluster and inter-cluster structures in high-dimensional visualization, this paper proposes Adaptive Multi-Scale Manifold Embedding (AMSME), an ordinal distance based preprocessing framework for existing manifold embedding algorithms. The framework introduces ordinal distance to replace traditional Euclidean distances. Under an idealized high-dimensional Gaussian setting, our analysis shows that ordinal distance can improve the stability of neighborhood ordering between homogeneous and heterogeneous samples, the

A new system for custom helical mandibular distraction: a bench study on anatomical models.

Current mandibular distraction osteogenesis relies on stock or hybrid devices, which can only execute rectilinear or in-plane circular movements. This bench feasibility study evaluated the accuracy of a novel patient-specific helical distractor compared with commonly used rectilinear and hybrid rectilinear devices. Retrospective data from four patients with severe mandibular deformities were utilized. Computer-assisted surgical simulation software was employed to plan ideal distraction paths for each rectilinear and helical case. Three distractors-custom helical, hybrid, and stock-were fabrica

Abnormal Dynamic Functional Connectivity Within the Fronto-Limbic Network Mediates the Association Between Depressive Symptoms and Non-Suicidal Self-Injury in Adolescents.

AIM: Non-suicidal self-injury (NSSI) is a prevalent behavior among adolescents with major depressive disorder (MDD), yet the precise neural mechanisms remain unclear. This study aimed to investigate the temporal dynamics of brain connectivity associated with NSSI in adolescents with MDD using dynamic functional connectivity (dFC) analysis. METHODS: Resting-state fMRI data from 204 adolescents (154 with NSSI, 50 without) were analyzed. dFC variability within the fronto-limbic network was assessed using a seed-based dynamic conditional correlation approach. Group differences in dFC variability

[18F]-FDG-PET for the assessment of major infectious disorders of the musculoskeletal system.

Infections of the musculoskeletal system contribute substantially to global morbidity and healthcare expenditure, and conventional imaging frequently falls short of providing the sensitivity and specificity required for early diagnosis in complicated presentations. In this review, we examine the clinical role of [18F]-fluorodeoxyglucose positron emission tomography paired with computed tomography (FDG-PET/CT) or magnetic resonance imaging (FDG-PET/MRI) in the workup of these infections. Drawing on the published literature, we summarize evidence across osteomyelitis, spondylodiscitis, septic ar

Mu and Beta Oscillatory Changes During a Motor Task Following Rehabilitation in Chronic MCA Stroke: Insights from EEG.

BackgroundStroke in the middle cerebral artery (MCA) often leads to persistent upper limb disability. While clinical scales such as the Fugl-Meyer Assessment for the Upper Extremity (FM-UE) track motor recovery, high-density EEG provides insight into cortical motor-activation patterns. Event-related desynchronization/synchronization (ERD/ERS) in mu and beta bands may offer insight into recovery mechanisms.Objectivea) To characterize changes in mu and beta oscillations following upper limb rehabilitation in chronic MCA stroke patients, and b) To identify ERD features most closely associated wit

The promise of quantitative approaches to computed tomography imaging in pulmonary sarcoidosis.

PURPOSE OF REVIEW: Pulmonary sarcoidosis is characterized by marked radiologic heterogeneity and limited reproducibility of visual high-resolution computed tomography (HRCT) assessment, which together restrict standardization, phenotyping, and prognostication. This review examines the potential that quantitative and artificial intelligence-based approaches offer in solving these challenges. It then offers key insights into the solutions needed to fully realize the value of HRCT imaging in pulmonary sarcoidosis. RECENT FINDINGS: Early quantitative and artificial intelligence-driven studies dem

Protocol for a prospective single-arm feasibility study of suzetrigine for postoperative pain management in outpatient facial plastic surgery.

Despite relatively brief postoperative pain courses and well-established risks of adverse effects and dependency, opioid analgesics remain routinely prescribed following outpatient facial plastic surgery. Suzetrigine (Journavx) is a first-in-class, oral, non-opioid analgesic that selectively inhibits the voltage-gated sodium channel 1.8 (NaV1.8), a peripheral nociceptive pathway target that may avoid central nervous system adverse effects associated with opioids. While suzetrigine has demonstrated efficacy in phase III trials of acute postoperative pain, its use in facial plastic surgery has n

Applicability of the U-SENS™ assay for assessing skin sensitization potential of agrochemical active ingredients and formulations.

Skin sensitization testing is essential for the regulatory classification of agrochemicals, including active ingredients (AIs) and formulations. Although several alternative methods have been developed, their application to agrochemicals remains limited. In particular, the performance of the U-SENS™ assay, which addresses key event three (dendritic cell activation) of the adverse outcome pathway, has not yet been evaluated for these products. This study assessed the U-SENS™ applicability to predict the sensitization potential of agrochemical formulations and their AIs. Twelve formulations and

Saxitoxin-Equivalent Toxicity of Gymnodinium catenatum and Toxicity Equivalence Factors of GC Toxins Determined by the Neuro-2a Assay.

Gymnodinium catenatum is a paralytic shellfish toxin (PST)-producing dinoflagellate that produces classical PSTs and hydroxybenzoate analogues known as GC toxins. Toxicological evaluation of GC toxins remains limited because certified reference materials (CRMs) and toxicity data are unavailable. This study applied the Neuro-2a (N2a) cell-based assay to evaluate PST functional toxicity in a Korean G. catenatum strain. The algal extract was analyzed by dose-response curve fitting to estimate total saxitoxin (STX)-equivalent toxicity, while GC1 & 2, GC3, and GC4 & 5, isolated from large-scale cul
