Unravelling SLC7 family members in thyroid cancer and translational barriers.

SLC7 family transporters are an important group of membrane proteins that facilitate the transport of different amino acids across the cell membrane. They have a crucial role in oncogenic signalling pathways, cellular metabolism, and redox balance. Previous literature had revealed the functions of SLC7 in breast, liver, prostate, and lung cancers, but the role of SLC7 family members in thyroid cancer remains unclear. Therefore, this review investigated the emerging roles of SLC7, the molecular mechanisms underlying thyroid cancer progression, and strategies to improve its therapeutic delivery.

Multiplexed Profiling of Individual Extracellular Vesicles Reveals Lung Cancer-Associated Protein Signatures.

Lung cancer is frequently diagnosed after curative treatment windows have narrowed, creating a need for minimally invasive biomarkers that report tumor development at earlier stages. Extracellular vesicles (EVs) are promising analytical targets because they carry molecular cargo from their originating cells, but tumor-associated EV signals can be rare in blood and obscured in bulk measurements. Here, we present Cygnus (Cyclic-imaging gateway to nanovesicles underlying signature), an end-to-end workflow that integrates cyclic immunofluorescence imaging with multiscale analysis of individual EVs

Armored Chimeric Antigen Receptor T-cell Therapy Targets Antigen-Heterogeneous Glioma.

UNLABELLED: Chimeric antigen receptor (CAR) T-cell therapy has shown early promise against glioblastoma, which lacks effective treatment options. However, two key challenges curtail efficacy: tumor-antigen heterogeneity and an immunosuppressive tumor microenvironment. CAR T cells engineered to secrete combinations of immunomodulatory proteins can reverse immune suppression and engage endogenous immunity. Through head-to-head in vivo comparisons of potentially synergistic armor combinations, we demonstrated that T cells expressing a CAR plus IL12 and the decoy-resistant form of IL18 (CAR-12.DR1

Guardians of T-Cell Ca2+ Stores: SERCA Pumps Integrated Within Complex Functional and Disease-State Signaling Dynamics.

T cells are the central regulators of the adaptive immune system, guiding both the cell-mediated and antibody-based elements of the immune response. Crucial to T-cell activation and differentiation, the T-cell receptor must transduce antigen exposure using a sustained elevated Ca2+ signal. A substantial body of research has identified and characterized multiple players in the Ca2+ signaling pathway, yet the sarcoplasmic/endoplasmic reticulum Ca2+-ATPase (SERCA) transporters, which intervene actively to regulate Ca2+ signal patterning and duration, remain relatively poorly characterized in the

Multiparatopic antibodies overcome tyrosine kinase inhibitor resistance by inducing lysosomal degradation of EGFR mutants.

Acquired resistance to epidermal growth factor receptor (EGFR) tyrosine kinase inhibitors (TKIs) remains a major challenge in the treatment of non-small cell lung cancer (NSCLC). To address this issue, we developed a novel therapeutic strategy based on multiparatopic antibodies that induce targeted degradation of EGFR, independent of driver mutations typically residing in the cytosolic domain. We engineered nanobodies (Nbs) recognizing four distinct epitopes within the EGFR extracellular domain into biparatopic and triparatopic antibody formats. These antibodies effectively promoted EGFR clust

Glucose Transporter 1 in Health and Disease.

Glucose Transporter 1 (GLUT1) is the quintessential facilitator of basal glucose uptake, indispensable for maintaining cellular energy homeostasis, particularly across the blood-brain barrier. Beyond physiological necessity, GLUT1 dysregulation drives a broad pathological spectrum. While genetic haploinsufficiency precipitates severe neurological energy crises like Glut1 deficiency syndrome (Glut1DS), oncogenic networks hyperactivate GLUT1 as the central executor of the Warburg effect to fuel malignant proliferation. Despite its immense therapeutic potential, severe on-target toxicity in norma

CD40 Agonist Therapy in Melanoma: Translating Preclinical Promise Into Clinical Practice.

Melanoma is an aggressive skin cancer that, once metastatic, accounts for a disproportionate share of skin-cancer mortality. Contemporary management includes surgical excision with sentinel-node assessment, adjuvant or neoadjuvant systemic therapy, radiotherapy in selected settings, targeted inhibition for BRAF/MEK-mutant disease, and intralesional modalities; nevertheless, many patients require effective systemic options. Immunotherapy has transformed outcomes by restoring antitumor T-cell activity. Within this paradigm, activation of CD40, a pivotal costimulatory receptor on antigen-presenti

Fabrication of oxidized starch/whey protein isolate complexes for fluidized powder coating of starch: A novel preparation strategy of resistant starch granules.

Exploring methods to retard starch digestion is essential for achieving related health benefits, including metabolic regulation. In this study, we proposed a novel strategy to reduce starch digestibility via fluidized bed coating. Initially, coating materials were developed based on the electrostatic complexation between oxidized starch (OS) and whey protein isolate (WPI), with epigallocatechin gallate (EGCG) employed to strengthen intermolecular interactions. This combination synergistically decreased apparent viscosity and improved film-forming properties. Furthermore, it influenced the mult

Unraveling the Mechanistic Spectrum of Myhre Syndrome: SMAD4 Signaling Disruption, Skeletal Phenotypes, and Translational Innovation.

Myhre syndrome (MYHRS) is an ultra-rare, progressive multisystem disorder caused by recurrent heterozygous missense variants in the SMAD4 gene, a central mediator of TGF-β and BMP signaling. Skeletal abnormalities-including postnatal short stature, brachydactyly, thickened calvarium, and craniofacial dysmorphism-are cardinal features, often accompanied by joint contractures and progressive soft tissue fibrosis. Extensive cellular and genetic evidence supports a gain-of-function (GoF) mechanism wherein mutant SMAD4 displays increased protein stability and prolonged nuclear localization, enhanci

Immune checkpoints in a genetically engineered mouse model of spontaneous autoimmune uveitis.

Engineered mutant AireGW/+Lyn-/- C57BL/6 mice are a model of spontaneous autoimmunity in which 50% of mice develop highly destructive uveitis due to compromised central and peripheral T cell tolerance. The key retinal autoantigen in these mice is interphotoreceptor retinoid-binding protein (IRBP). The CD4+ T cells recognizing the dominant epitope of IRBP, called P2, from eye-draining lymph nodes of mice with or without uveitis and from the retinas of mice with uveitis, were characterized by single-cell RNA sequencing (scRNA-seq) and flow cytometry. Mice with uveitis had autoantigen-specific T

Immune Pressure From Plasma From Immunocompromised and Healthy Individuals Drives Differential SARS-CoV-2 Evolution and Escape Mutation Emergence.

Immunocompromised individuals with prolonged SARS-CoV-2 infections are recognized reservoirs for viral evolution and the emergence of immune escape variants. We investigated intra-host evolutionary dynamics of SARS-CoV-2 under distinct immune pressures exerted by plasma from kidney transplant recipients (KTRs). Wild-type (WT) SARS-CoV-2 (B.6.6) and Omicron subvariants (BA.5.2 and XBB.1.5.57) were serially passaged in the presence of convalescent plasma. KTR plasma exerted selective pressure on the spike protein without compromising replicative capacity, thereby facilitating viral diversificati

Parallelised Cloning, Mammalian Cell Expression, and Purification of Nanobodies Identified by Phage Display.

Nanobodies are recombinant single-domain antibodies (VHHs) derived from the heavy chain-only subset of camelid immunoglobulins that can be reverse-engineered into bivalent antibodies by fusion to immunoglobulin Fc constant regions. Mammalian cells are the system of choice to produce VHH-Fcs to ensure authentic folding and post-translation glycosylation of the expressed VHH-Fcs. In a recent project to find neutralising VHH-Fc binders to the spike proteins of SARS-CoV-2 viruses, we identified a need for rapid expression and purification of multiple VHH-Fc fusions from nanobodies selected by phag

Pollen-inspired multifunctional nanocarrier for porcine circovirus 2 vaccine delivery.

Traditional vaccines targeting porcine circovirus type 2 (PCV2) have several limitations, such as low antigen delivery efficiency and inadequate immunogenicity. To address these challenges, this study developed a bionic nanocarrier (Fe₃O₄@MnSiO₃) that synergistically integrates antigen enrichment and immune stimulation functions. Inspired by the hierarchical structure of sunflower pollen, the design constructs composite particles featuring a superparamagnetic Fe₃O₄ core and a mesoporous MnSiO₃ shell, allowing efficient loading and delivery of the PCV2 Cap protein. Owing to its sophisticated bi

Engineered Peptide Self-Assembled Nanofibers for Enhanced STING Activation: A Promising Nanodelivery Platform for Immunotherapy of Pediatric Solid Tumors.

STING agonists (e.g., cGAMP), which function as immunotherapeutic agents activating the stimulator of interferon genes (STING) pathway, hold promise for treating pediatric solid tumors including neuroblastoma. However, their clinical translation is hindered by limitations including burst release, short half-life, poor targeting, and low intracellular delivery efficiency. In this study, we engineered a peptide self-assembled nanofiber (NFSA) based on the specific interaction mechanism between STING protein and cGAMP. By hydrophobically modifying the cGAMP-binding amino acid sequences on STING,

Mitigation of pre-existing immunogenicity in ADA method development.

BACKGROUND: Immunogenicity assessments are critical for evaluating biological therapeutics. Preexisting immune reactivity can affect clinical safety and efficacy and confound anti-drug antibody (ADA) assay cut point determination, complicating identification of treatment-emergent ADA. In the ALXN-X program, approximately 50% of naïve human serum samples showed preexisting reactivity, with some signals exceeding background by more than 100-fold, challenging ADA method development (cut points, sensitivity, drug tolerance). METHODS: Competitive inhibition experiments were conducted to map the bi

Natural killer cell encapsulation in core-shell proteinic adhesive microcapsules for persistent and localized release of cytotoxic factors.

Natural killer (NK) cells have attracted considerable interest as a readily available product for cancer immunotherapy, particularly because of their low incidence of graft-versus-host disease in allogeneic settings. Nevertheless, the effectiveness of this strategy is often compromised by the limited in vivo persistence of NK cells and suboptimal targeting efficiency toward malignant tissues. In this study, we engineered adhesive microcapsules laden with NK cells, comprising an aqueous core that houses NK cells and an outer shell fabricated from bioengineered mussel adhesive protein (MAP) for

Immunogenicity of induced pluripotent stem cell-derived smooth muscle cells results from a reduction in the expression of indoleamine 2,3 dioxygenase (IDO-1).

BACKGROUND: Smooth muscle cells (SMCs) derived from allogeneic induced pluripotent stem cells (iPSC) hold significant potential in cellular therapy for many cardiovascular diseases. However, their immunological profile is poorly characterized, limiting their clinical progression. AIMS: This study aimed to investigate the immunogenicity of iPSC-SMC in comparison with naturally derived vascular SMCs (v-SMCs). RESULT: We found iPSC-SMCs, in contrast to naturally derived vascular SMCs (v-SMC), triggered T effector memory (TEM) cell proliferation. However, expression of TEM activation-related pro

Intrinsically Mitochondria-Targeting Nanozyme via Coordination-Assembly of Natural Quercetin for Cascade Antioxidant Therapy of Cerebral Ischemia-Reperfusion Injury.

Mitochondrial dysfunction, culminating in oxidative stress-driven release of mitochondrial DNA (mtDNA) and subsequent inflammatory activation, constitutes a central pathogenic axis in cerebral ischemia-reperfusion injury. Disrupting this axis requires precise antioxidant delivery to neuronal mitochondria, a major therapeutic hurdle. Here, we uncover that the natural flavonoid quercetin (Quer) possesses an intrinsic ability to bind mitochondrial outer membrane proteins, revealing its unexploited potential as a natural mitochondrial-targeting ligand. Leveraging this discovery, we engineered an u

Low-temperature-tolerant Cas12a enables instrument-free detection of fungal pathogens at mushroom cultivation temperatures.

Reliable molecular diagnosis at the point of care remains challenging owning to its dependence on specialized instrumentation and temperature control. However, clustered regularly interspaced short palindromic repeats (CRISPR) and the associated Cas12a protein have shown promise for point-of-care detection. Therefore, an engineered Cas12a variant from Lachnospiraceae bacterium, namely L.b. Cas12a Ultra, was developed. It exhibited markedly enhanced functional stability and low-temperature tolerance, maintaining robust activity across typical mushroom cultivation temperatures (21 °C-29 °C), com
