The molecular mechanisms of muscle weakness and sarcopenia in postmenopausal women are largely unknown. To determine the effect of a new estrogen receptor, GPR30, in the maintenance of exercise capacity and skeletal muscle function in females, the selective GPR30 agonist, G1 (100 μg/kg/day), or vehicle (V, soybean oil) was administered subcutaneously daily (n = 7 per group) to ovariectomized (OVX) 27-month-old Fischer 344 × Brown Norway (F344BN) female rats. Following 8 weeks of treatment, the exercise capacity (treadmill walk time to exhaustion) was reduced in OVX vs. sham rats (5.1 ± 1.4 vs.

Alterations within the RAS (renin-angiotensin system) are pivotal for the development of renal disease. ACE2 (angiotensin-converting enzyme 2) is expressed in the kidney and converts the vasoconstrictor AngII (angiotensin II) into Ang-(1-7), a peptide with vasodilatory and anti-fibrotic actions. Although the expression of ACE2 in the diabetic kidney has been well studied, little is known about its expression in non-diabetic renal disease. In the present study, we assessed ACE2 in rats with acute kidney injury induced by STNx (subtotal nephrectomy). STNx and Control rats received vehicle or ram

Bradykinin is associated with infections and inflammation, which given the strong correlation between uterine infection and preterm labour may imply that it could play a role in this process. Therefore, we investigated bradykinin signalling, and the roles that arrestin proteins play in their regulation in human myometrial cells. Bradykinin induced rapid, transient intracellular Ca(2+) increases that were inhibited following B2 receptor (B2R) antagonism. Arrestin2 or arrestin3 depletion enhanced and prolonged bradykinin-stimulated Ca(2+) responses, and attenuated B2R desensitisation. Knockdown

BACKGROUND: Gintonin is a novel non-saponin glycolipoprotein complex that is derived from ginseng. Its functional components are lysophosphatidic acids (LPAs), which act as ligands for six G protein-coupled LPA receptor subtypes. Thus, gintonin exerts it activity through the LPA receptor signaling pathway. Previous studies have shown that crude gintonin derived from Korean Red ginseng marc (KRGM) contains approximately 0.2-0.3% LPAs. The aim of this study was to develop a method for concentrating the LPA content in KRGM-derived gintonin.
METHODS: First, we prepared a gintonin-enriched fractio

Chronic hepatitis B (CHB) remains a major global public health challenge. Current therapies based on nucleos(t)ide analogues and interferon mainly achieve long-term viral suppression, whereas only a small proportion of patients attain a functional cure, defined as sustained hepatitis B surface antigen loss with hepatitis B virus (HBV) DNA below the limit of quantification for at least 24 weeks after treatment discontinuation, with or without anti-HBs seroconversion. Emerging evidence from the gut-liver axis indicates that gut microbiota-derived metabolites, particularly short-chain fatty acids

Neurons typically release both a neurotransmitter and one or more neuropeptides, but how these signals are integrated within neural circuits to generate and tune behaviors remains poorly understood. We studied how the two hermaphrodite-specific neurons (HSNs) activate the egg-laying circuit of Caenorhabditis elegans by releasing both the neurotransmitter serotonin and NLP-3 neuropeptides. Egg laying occurs in a temporal pattern with approximately 2-min active phases, during which eggs are laid, separated by approximately 20-min inactive phases, during which no eggs are laid. To understand how

BACKGROUND & AIMS: Bile diversion to the ileum (GB-IL) has strikingly similar metabolic and satiating effects to Roux-en-Y gastric bypass (RYGB) in rodent obesity models. The metabolic benefits of these procedures are thought to be mediated by increased bile acids, although parallel changes in body weight and other confounding variables limit this interpretation.
METHODS: Global G protein-coupled bile acid receptor-1 null (Tgr5-/-) and intestinal-specific farnesoid X receptor null (FxrΔ/E) mice on high-fat diet as well as wild-type C57BL/6 and glucagon-like polypeptide 1 receptor deficient (G

BACKGROUND: To understand the effect of the long intracellular loop 3 (ICL3) on the intrinsic dynamics of human β2-adrenergic receptor, molecular dynamics (MD) simulations were performed on two different models, both of which were based on the inactive crystal structure in complex with carazolol (after removal of carazolol and T4-lysozyme). In the so-called loop model, the ICL3 region that is missing in available crystal structures was modeled as an unstructured loop of 32-residues length, whereas in the clipped model, the two open ends were covalently bonded to each other. The latter model wi

Calcitonin inhibits bone resorption by acting on osteoclasts via a specific receptor. The calcitonin receptor (CTR) is also found in many other normal and malignant tissues and cell lines. It has been cloned and sequenced in several species including humans. It belongs to a subclass of seven-transmembrane G protein-coupled receptors. Four human CTR (H-CTR) isoforms generated by alternatively spliced mRNA have previously been described. Two H-CTR encoding DNAs containing an unidentified 50-bp insert are now reported from T47D cells. The 50-bp insert corresponds to a DNA region located between e

The mu-opioid receptor (MOR), a G-protein-coupled receptor, is internalized after endogenous agonist binding. Although receptor activation and internalization are separate events, internalization is a good assay for activation because endogenous opioid peptides all induce internalization. Estrogen treatment of ovariectomized rats induces MOR internalization, providing a neurochemical signature of estrogen activation of the medial preoptic nucleus. MOR activation appears to be the mechanism via which estrogen acts in the medial preoptic area to prevent the display of female reproductive behavio

The liver plays a significant role in regulating a wide range of metabolic, homeostatic, and host-defense functions. However, the impact of liver injury on the host's ability to control bacteremia and morbidity in sepsis is not well understood. Leukocyte recruitment and activation lead to cytokine and chemokine release, which, in turn, trigger hepatocellular injury and elevate nucleotide levels in the extracellular milieu. P2Y2 purinergic receptors, G protein-coupled and activated by extracellular ATP/UTP, are expressed at the cell surface of hepatocytes and nonparenchymal cells. We sought to

The neurotransmitters adrenaline and noradrenaline are non-selective adrenergic agonists which interact with both subtypes of alpha- and beta-receptors. Clonidine, an alpha 2-adrenergic drug with a selectivity ratio of 200/1 for alpha 2/alpha 1 has been used in clinical practice for more than 20 years. Although alpha 2-agonists have vasoconstrictor properties, sympatholytic effects on the central nervous system predominate. As a result, the sympathetic outflow from the medullary pressor centres is decreased mediating the hypotensive effects of the alpha 2-agonists. These compounds also exhibit

This study investigates the poor reversibility of salmon calcitonin (sCT) binding to rat and human calcitonin receptors. Efficacy of CT and analogue peptides in (125)I-sCT binding competition and cAMP assays was compared with the dissociation kinetics of (125)I-labelled peptides. Assessment was performed on cells stably expressing either rat or human calcitonin receptors. Dissociation kinetics of the antagonists, sCT(8-32) and AC512, revealed that binding was rapidly and completely reversible at the receptors, despite high affinity binding, suggesting that poor reversibility required the activ

Allergic rhinitis (AR) is a chronic inflammatory disease of the upper respiratory tract mediated by immunoglobulin E (IgE). Alpha-linolenic acid (ALA), an essential dietary omega-3 polyunsaturated fatty acid, has been proposed to possess anti-inflammatory and anti-allergic properties. However, research on the effects of ALA on AR and the underlying mechanisms remains limited. Therefore, we evaluated oral ALA intervention in an ovalbumin (OVA)-sensitized AR mouse model. Nasal symptoms were recorded, and nasal mucosal pathology was evaluated by hematoxylin and eosin staining. Goblet cell hyperpl

This study investigated the therapeutic effects of PEP-2, a 348.45 kDa pectin isolated from Phyllanthus emblica, against metabolic-associated fatty liver disease (MAFLD). Characterized as a homogalacturonan with minor rhamnogalacturonan I, PEP-2 consists of galacturonic acid, galactose, rhamnose, and arabinose. In high-fat diet-induced mice, PEP-2 significantly improved metabolic phenotypes, reducing obesity, insulin resistance, and hepatic steatosis. Mechanistically, PEP-2 modulated the gut-liver axis by enriching Akkermansia and enhancing short-chain fatty acid (SCFA) production. These chang

BACKGROUND: Chronic atrophic gastritis (CAG) severity risk assessment remains challenging due to limitations of current static histologic/endoscopic tools. We evaluated the leucine-rich repeat-containing G-protein coupled receptor 5 (LGR5)/trefoil factor 2 (TFF2) ratio - quantifying gastric stem cell niche imbalance- as a dynamic biomarker for high-risk CAG (OLGA/OLGIM III-IV).
METHODS: In a retrospective cohort (n = 60 CAG patients), we constructed four prediction models (Baseline: demographics; Clinical: +H. pylori/family history; Endoscopic; Full: +LGR5/TFF2 ratio) using Firth penalized re

Information on the maintenance of tissue homeostasis is important for developing effective therapeutic methods. However, reports on the cellular composition and tissue stem cells of the larynx are scarce. Therefore, we analyzed mouse laryngeal mucosa using single-cell RNA- sequencing and spatial transcriptomics by photo-isolation chemistry, and we also generated laryngeal organoids as an in vitro model. Consequently, we found a SOX9-positive basal cell subpopulation and a Lgr5-positive cell subpopulation in the mouse vocal fold, and obtained three types of epithelial organoids from laryngeal e

BACKGROUND: Gastric cancer (GC) is a leading cause of cancer-related deaths due to late diagnosis. Altered glycolytic metabolism, notably the Warburg effect, plays a critical role in tumorigenesis, offering potential for early detection and targeted therapy.
METHODS: We combined single-cell omics and computational modeling to map and validate glycolytic biomarkers in GC. A novel interquartile range (IQR)-based approach was developed to assess glycolytic activity at the single-cell level, focusing on glycolysis-related genes (GRGs). The results were verified by in vitro and in vivo experiments

BACKGROUND AND AIMS: The succinate receptor GPR91 is highly expressed in HSCs, with its expression further elevated during metabolic dysfunction-associated steatohepatitis (MASH)-induced fibrotic progression. However, convincing in vivo data on whether blocking GPR91 signaling leads to fibrotic regression in MASH are lacking.
APPROACH AND RESULTS: MASH models were induced by choline-deficient amino acid-defined diet feeding along with lipopolysaccharide injection (CDAA-LPS) plus i.p. injection of succinate or by high-fat and high-calorie diet plus high fructose and glucose in drinking water (

BACKGROUND: Succinate, a metabolite produced by both the gut microbiota and the host, has emerged as a key player in chronic inflammation. In patients with Crohn's disease (CD), increased succinate in the intestinal lumen correlates with both dysbiosis and greater disease activity.
AIM: To investigate circulating succinate as a biomarker of CD activity and its associations with gut microbiota, immune, and clinical features.
METHODS: This study with the prospective inclusion of patients with CD in remission, active CD, and non-inflammatory bowel disease controls matched by age, sex, and body
