Effects of Site-Specific Glycation on α-Synuclein.

Glycation of α-synuclein (αSyn) by methylglyoxal (MGO) has been implicated as a pathologic mechanism in Parkinson's disease. However, mechanistic understanding has been limited so far by the heterogeneity of chemically MGO-modified αSyn. We developed and applied two complementary semisynthetic strategies based on native chemical ligation for site-specific incorporation of Nε-carboxyethyllysine (CEL), a major advanced glycation endproduct formed by MGO. Using this approach, we generated a panel of 11 αSyn variants bearing CEL modifications at defined positions in the N-terminal region. Single C

Constitutive elevation of cAMP in Arabidopsis thaliana enhances plant growth, development, and productivity.

Cyclic adenosine 3',5'-monophosphate (cyclic AMP; cAMP) has emerged as a bona fide signalling molecule in plants. Although its regulatory roles in diverse cellular processes are increasingly recognised, the transient nature of cAMP signalling has limited our understanding of its systemic functions at the whole-plant level. In this study, we engineered Arabidopsis thaliana plants to constitutively express adenylate cyclase (AC) activity, thereby enabling sustained elevation of endogenous cAMP levels. The AC transgenic lines exhibited a 1.3- to 1.8-fold increase in endogenous cAMP content relati

C-reactive protein-related cortical thickness changes and its association with resting-state functional connectivity alterations in major depressive disorder.

BACKGROUND: C-reactive protein (CRP) has emerged as a promising peripheral biomarker of neuroinflammatory processes implicated in the pathophysiology of major depressive disorder (MDD). We combined structural magnetic resonance imaging (MRI) with resting-state functional MRI to examine plasma CRP-related cortical thickness and resting-state functional connectivity (RSFC) changes. METHODS: Seventy-six patients with MDD and 65 healthy controls (HCs) were included in this study. The association between whole-brain cortical thickness and plasma CRP levels in the total sample (MDD+ HC, n = 141) wa

Multiomic single nuclei profiling the mouse hippocampus reveals that ACSS2 confers neuronal resilience to tauopathy.

INTRODUCTION: Epigenomic dysregulation contributes to Alzheimer's disease (AD) and related tauopathies. Acetyl-CoA synthetase 2 (ACSS2), a nuclear-localized metabolic enzyme in neurons, supports histone acetylation and learning-related gene expression. We examined how ACSS2 loss affects molecular and behavioral phenotypes in a mouse model of tauopathy. METHODS: We induced tauopathy in ACSS2 knockout and control mice via injection of pathological human tau. We assessed transcriptomic, epigenomic, and behavioral changes, and tested long-term acetate supplementation as a rescue strategy. RESULT

Fine mapping and candidate gene analysis of tassel glume closure1 (tgc1) gene in maize (Zea mays L.).

The tassel glume closure1 (tgc1) mutant was identified and mapped, and ZmKAO1, which encodes an ent-kaurenoic acid oxidase, is the key candidate gene. Tassel glume timely opening is a key trait that influences cross-pollination and hybrid breeding. However, in maize (Zea mays L.), the related genes regulating this process remain largely unexplored. Here, we identified tassel glume closure1 (tgc1), a recessive mutant that exhibits tassel glume closure, anther non-extrusion, and no pollen shedding at the adult-plant stage. Anatomical and scanning electron microscopy (SEM) observations revealed t

SNP-Based KASP Markers Reveal Genetic Diversity and Population Structure Among African Sorghum Breeding Lines and Hybrids.

Sorghum is a strategic food-security and livestock-feed crop in Africa, and molecular characterization of its germplasm underpins genetic diversity assessment and hybrid parent selection. This study evaluated 106 sorghum accessions (23 A-lines, 28 B-lines, 32 R-lines, and 23 hybrids) from ICRISAT and Seed Co gene banks using 79 single nucleotide polymorphism (SNP)-based Kompetitive Allele-Specific PCR (KASP) markers. Genetic diversity, population structure, cluster analysis, and analysis of molecular variance (AMOVA) were assessed to support heterotic grouping. Gene diversity ranged from 0.23

An Epichloë fungal endophyte regulates grain size in barley.

Grain size represents a key agronomic trait and a major determinant of barley grain yield potential. We observed that the grain size of Epichloë bromicola-inoculated barley plants were significantly larger than those of the uninoculated barley plants. However, the mechanisms and functions by which the endophyte fungus E. bromicola modulates barley grain size have not been investigated. In this study, we compared grain size between uninoculated (KE-) and E. bromicola-inoculated (KE+) barley plants to elucidate the association between E. bromicola colonization and barley grain size. Quantitative

Native Starch Granules from Unconventional Peruvian Sources: Banana, Mango and Taro.

The transition toward sustainable food systems has driven the use of agricultural materials and agroindustrial byproducts as alternative sources of functional ingredients. In this study, starches were isolated from Bellaco plantains, Criollo mango seeds, and Antiquorum taro using aqueous extraction, and their physicochemical, morphological, structural, rheological, and thermal properties were characterized. The amylose content ranged from 16.83% to 25.92%, values comparable to those of conventional starch sources. Taro starch exhibited the highest brightness (98.30) and whiteness index (96.56)

18-Year Monitoring of the Steno-Endemic Verbascum rupicola (Scrophulariaceae): Compounding Pressures and the Extinction Vortex.

The steno-endemic Verbascum rupicola faces a precarious future due to its extreme habitat specialization on tectonically active hydrothermal quartz veins. This study presents a long-term assessment based on periodic population censuses spanning 18 years (2007, 2016, and 2025) to assess the demographic and spatial trends of its global population in the Tahtalı Dam basin, Türkiye. Field surveys, GIS-based habitat mapping, and controlled pollination experiments were integrated with seed germination kinetics and ex situ cultivation trials. Results reveal a precipitous 69.12% global population decl

Elevated methionine in plants: the effect on development, stress responses, epigenetics, and nutritional biofortification.

Methionine (Met) is an essential amino acid that limits the nutritional value of many crop plants, yet its steady-state level in plant tissues is remarkably low. At the same time, Met is a central metabolic hub supporting protein synthesis and, through S-adenosylmethionine (SAM), drives the production of hormones, vitamins, polyamines, and epigenetic marks. The primary objective of this review is to understand why and how plants maintain low steady-state Met levels, and what occurs when Met content is elevated, a question of growing importance for Met biofortification. We first summarize evide

Do Quantum Kernels Improve Medical Image Classification? A Leakage-Controlled Benchmark Against Classical RBF-SVM.

Quantum kernel methods are often proposed as a route to improved medical image classification because quantum feature maps can embed data into high-dimensional Hilbert spaces. However, their practical value remains unclear when compared with strong classical kernels under leakage-controlled conditions. This study benchmarks two simulated quantum kernel support vector machines, QSVM-ZZ and QSVM-Pauli, against logistic regression, linear SVM, fixed-default RBF-SVM, and a compact multi-layer perceptron on BrainTumorMRI and BreastMNIST. All models used identical frozen ResNet18 features, train-onl

Advances In the Valorization of Avocado By-Products: Bioactive Compounds, Techno-Functional Properties, and Structure-Function Relationships.

Avocado (Persea americana Mill.) processing generates large amounts of peel and seed that remain underutilized despite their potential as functional food ingredients. While previous reviews have focused mainly on the chemical composition and antioxidant capacity of avocado by-products, less attention has given to the role of tissue-specific microstructure and ripening-related structural changes in determining their techno-functional behavior. This review critically integrates recent advances in the valorization of avocado by-products, with particular emphasis on peel and seed as sources of bio

Differential Regulation of Pre-Harvest Sprouting by OsERF1 and OsERF94 Through Hormone Signaling and Metabolic Reprogramming in Rice.

Pre-harvest sprouting (PHS), the premature germination of grains on the mother plant, causes substantial yield loss and grain-quality deterioration in rice under humid conditions. Although seed dormancy and germination are largely controlled by hormonal balance, the transcriptional mechanisms linking hormone signaling with metabolic adaptation during PHS remain unclear. In this study, we investigated the roles of two ethylene-responsive factor transcription factors, OsERF1 and OsERF94, in rice PHS regulation using CRISPR/Cas9-mediated knockout lines, together with physiological, gene-expressio

Hermaphroditism in Cannabis sativa L.: Impacts, Inducers, and Industry Implications.

Cannabis sativa L. is a predominantly dioecious species, but sex expression is highly plastic and can be modified by genetic, hormonal, developmental, and environmental factors. This plasticity has major implications for commercial production because hermaphroditic expression in female plants can cause unintended pollination, seed formation, reduced floral quality, and losses in cannabinoid yield. This review summarizes current understanding of sex determination and sex-expression instability in C. sativa, with emphasis on hermaphroditism and its agronomic significance. We examine the genomic

Hybrid CNN-embedding fusion with MFCC-SVM for speech emotion recognition: Random vs actor-wise evaluation on CREMA-D.

Speech Emotion Recognition (SER) is an important component of human-centered intelligent systems, yet robust performance remains challenging when speaker identities differ between training and testing. This study presents a protocol-aware and reproducible comparison on the CREMA-D corpus using three pipelines: (i) a classical MFCC-based Support Vector Machine (SVM), (ii) a log-mel Convolutional Neural Network (CNN), and (iii) a lightweight hybrid model that concatenates handcrafted acoustic descriptors with CNN-derived embeddings and uses an SVM classifier. The methodological contribution is n

Aerosol and bioaerosol generation from toilet flushing: A systematic review and risk factor analysis.

Toilet flushing is a daily activity that can generate bioaerosols in indoor spaces. This exposure pathway is increasingly important in the context of emerging pathogens, changing water systems, and high indoor occupancy. Stronger empirical evidence is needed to support risk-based decision-making to protect human health. This systematic literature review examined factors influencing aerosol and bioaerosol concentrations generated through toilet flushing, with a focus on toilet design, target microorganisms, and air sampling methods relevant to exposure and risk assessment. Twenty-two studies me

Generation of a Mouse Prostate Organoid-Based Model for Studying Host-Pathogen Interactions.

Aging men are highly affected by prostate diseases, such as benign prostatic hyperplasia, prostatitis, and prostate cancer. Together, these conditions contribute significantly to the global health burden and are a leading cause of illness in men. Organoid models have transformed disease modeling by allowing the replication of key features of epithelial structure and function in vitro. However, the closed lumen structure of organoids limits experimental access to the apical surface, hindering studies of host-pathogen interactions. To overcome these challenges, we generated a physiologically rel

Conditional Diffusion-Augmented Cascaded Multi-View Attention BiLSTM for Non-Invasive Blood Glucose Estimation from Photoplethysmography.

Photoplethysmography (PPG)-based blood glucose estimation is attractive for non-invasive monitoring, but its development is constrained by limited paired PPG-glucose data and the weak representation of glucose-related waveform variations. This study proposes a framework combining conditional diffusion-based data augmentation with a cascaded multi-view attention bidirectional long short-term memory (BiLSTM) network. Blood glucose level, heart rate, and body mass index were used as physiological conditions to generate synthetic PPG segments, while channel and temporal attention together with cas

Patient-Derived 3D Heart-On-a-Chip Model of Dilated Cardiomyopathy With Embedded Bead-Based Mapping of Tissue Contractility.

Dilated cardiomyopathy (DCM) is the leading cause of heart transplantation, with a 50% risk of progression to heart failure within 5 years. Conventional disease modeling approaches fail to recapitulate the sophisticated function of the human heart. Alternatively, heart-on-a-chip (HOC) platforms enable real-time monitoring of disease progression and drug responses using miniaturized engineered heart tissues. Here, we developed a functional HOC model using patient-specific human induced pluripotent stem cells (hiPSCs), reprogrammed from the patients' blood samples. The chip contains two cell-see
