Automated Segmentation and Morphometric Analysis of Thioflavin-S-Stained Amyloid Deposits in Alzheimer's Disease Brains and Age-Matched Controls Using Weakly Supervised Deep Learning.

Alzheimer's disease (AD) involves the accumulation of amyloid-β (Aβ) plaques, whose quantification plays a central role in understanding disease progression. Automated segmentation of Aβ deposits in histopathological micrographs enables large-scale analyses but is hindered by the high cost of detailed pixel-level annotations. Weakly supervised learning offers a promising alternative by leveraging coarse or indirect labels to reduce the annotation burden. We evaluated a weakly supervised approach to segment and analyze thioflavin-S-positive parenchymal amyloid pathology in AD and age-matched br

Puerariae Lobatae Radix-Gastrodiae Rhizoma ameliorates nitroglycerin-induced chronic migraine by modulating p38 MAPK/CREB signaling pathway in the prefrontal cortex-thalamus neural circuit.

ETHNOPHARMACOLOGICAL RELEVANCE: Puerariae Lobatae Radix (PLR) and Gastrodiae Rhizoma (GR) are classic "medicine-food homology" pairs in Traditional Chinese Medicine (TCM). For millennia, they have been used in formulas to treat headache and vertigo, as recorded in Compendium of Materia Medica and Shennong's Classic of Materia Medica. Clinical practices confirm their efficacy in alleviating migraine, but the underlying mechanisms remain unclear. AIM OF THE STUDY: This study aimed to evaluate the therapeutic effect of PLR-GR on chronic migraine and elucidate its potential mechanism of improving

Survey of hard ticks in high-incidence area of Severe Fever with Thrombocytopenia Syndrome in China.

Anhui Province in China bears the highest burden of Severe Fever with Thrombocytopenia Syndrome (SFTS) nationwide; however, comprehensive surveys of tick vectors remain limited. We aimed to characterize tick species composition and to evaluate spatial patterns of vector distribution across SFTS-endemic areas in the province. In this study, a combined morphological and molecular approach was utilized for tick identification. Ticks were initially identified based on their morphological characteristics. Subsequently, gene fragments of the large subunit ribosomal RNA (16 S rRNA) and the cytochrome

Paired single-cell and spatial transcriptional profiling reveals a central osteopontin macrophage response mediating tuberculous granuloma formation.

Granulomas are classic manifestations of tuberculosis pathogenesis. They result from an ensemble of immune responses to Mycobacterium tuberculosis infection, but the identities, arrangement, cellular interactions, and regulation of the cells that comprise them have thus far been incompletely understood. To better understand the composition of granulomas, we conducted spatial and single-cell RNA sequencing of granulomas in biopsy specimens from patients with tuberculosis. We found that granulomas consist of concentric transcriptional laminae surrounding areas of central necrosis. We identified

The Positional Relationship between Lacunae and White Matter Hyperintensity in Patients with Cerebral Small Vessel Disease.

BACKGROUND: Lacunae and white matter hyperintensity (WMH) are two crucial imaging biomarkers of cerebral small vessel disease (CSVD). Multiple studies have revealed a close relationship between WMH and lacunae and found that a double penumbra existed at the edge of WMH that affects lacunae formation. The study aimed to explore the spatial distribution characteristics and possible influencing factors of lacuna in relation to white matter hyperintensity in patients with CSVD. METHODS: A total of 480 CSVD patients with WMH and with or without lacunae were included. Data about blood biochemical i

OBJECTIVE: Electroconvulsive therapy (ECT) is endorsed as a principal treatment approach for major depressive disorder (MDD) worldwide. Despite prior studies highlighting potential short-term cognitive deficits post-ECT, the debate regarding its long-term implications persists. This study endeavors to elucidate the reasons for this contention using an evidence-based approach. METHODS: This investigation, meticulously aligned with PRISMA guidelines, was prospectively enlisted on PROSPERO (CRD42023439259). A comprehensive search was performed across various databases, including PubMed, Cochrane

Characterization of Forearm Muscle Activation in Duchenne Muscular Dystrophy via High-Density Electromyography: A Case Study on the Implications for Myoelectric Control.

Duchenne muscular dystrophy (DMD) is a genetic disorder that results in progressive muscular degeneration. Although medical advances increased their life expectancy, DMD individuals are still highly dependent on caregivers. Hand/wrist function is central for providing independence, and robotic exoskeletons are good candidates for effectively compensating for deteriorating functionality. Robotic hand exoskeletons require the accurate decoding of motor intention typically via surface electromyography (sEMG). Traditional low-density sEMG was used in the past to explore the muscular activations of

Chronic Undernutrition Differentially Changes Muscle Fiber Types Organization and Distribution in the EDL Muscle Fascicles.

Fiber type composition, organization, and distribution are key elements in muscle functioning. These properties can be modified by intrinsic and/or extrinsic factors, such as undernutrition and injuries. Currently, there is no methodology to quantitatively analyze such modifications. On one hand, we propose a fractal approach to determine fiber type organization, using the fractal correlation method in software Fractalyse. On the other hand, we applied the kernel methodology from machine learning to build radial-basis functions for the spatial distribution of fibers (distribution functions), b

El Niño Southern Oscillation as an early warning tool for dengue outbreak in India.

BACKGROUND: Dengue is rapidly expanding climate-sensitive mosquito-borne disease worldwide. Outbreaks of dengue occur in various parts of India as well but there is no tool to provide early warning. The current study was, therefore, undertaken to find out the link between El Niño, precipitation, and dengue cases, which could help in early preparedness for control of dengue. METHODS: Data on Oceanic Niño Index (ONI) was extracted from CPC-IRI (USA) while the data on monthly rainfall was procured from India Meteorological Department. Data on annual dengue cases was taken from the website of Nat

The Metabolic Pattern of Idiopathic REM Sleep Behavior Disorder Reflects Early-Stage Parkinson Disease.

Idiopathic REM sleep behavior disorder (iRBD) is considered a prodromal stage of Parkinson disease (PD) and other Lewy body disorders. Spatial covariance analysis of 18F-FDG PET data has disclosed a specific brain pattern of altered glucose metabolism in PD. In this study, we identify the metabolic pattern underlying iRBD and compare it with the known PD pattern. To understand the relevance of the iRBD pattern to disease progression, we studied the expression of the iRBD pattern in de novo PD patients. Methods: The iRBD-related pattern was identified in 18F-FDG PET scans of 21 patients with po

Neural bases of personal and extrapersonal neglect in humans.

Human awareness of left space may be disrupted by cerebral lesions to the right hemisphere (hemispatial neglect). Current knowledge on the anatomical bases of this complex syndrome is based on the results of group studies that investigated primarily the best known aspect of the syndrome, which is visual neglect for near extrapersonal (or peripersonal) space. However, another component-neglect for personal space-is more often associated with, than double-dissociated from, extrapersonal neglect, especially, in chronic patients. The present investigation aimed at exploring the anatomical substrat

Movement Anywhere: An Open-Source Distributed 2D Video-Based Movement Analysis Platform Empowered by Active Learning.

Movement analysis plays a pivotal role in diagnosing and monitoring neurodegenerative and musculoskeletal diseases. Traditional tools, such as 3D motion capture systems and electronic walkways, though effective, are costly and spatially demanding, limiting their accessibility. To address these challenges, we introduce Movement Anywhere, an open-source, distributed plat form for 2D video-based movement analysis. This plat form features advanced tracking algorithms that robustly handle scenarios where patients require assistance, and it rigorously establishes the conditions necessary for precise

RXR agonist increases newly characterized GM-CSF-producing B220+CD8αα intraepithelial T cells.

Context-dependent cytokine granulocyte-macrophage colony-stimulating factor (GM-CSF) plays a crucial role in maintaining intestinal immune homeostasis and exacerbating inflammation. Although multiple cell types produce GM-CSF in the gut, whether intraepithelial lymphocytes (IELs) contribute to GM-CSF production during the steady state remains unclear. In the small intestinal epithelial layer, we identified B220-expressing CD8αα+ T cells that selectively produced GM-CSF but not interleukin-17A or interferon-γ. Transcriptional analysis combined with weighted parametric gene set analysis revealed

Genome-wide analysis in over 1.6 million participants uncovers 147 loci associated with obstructive sleep apnoea.

We conducted the largest GWAS meta-analysis for obstructive sleep apnoea (OSA; Ncases= 230,657; Ncontrols= 1,377,442) using European ancestry genetic data from five countries. We identified 147 independent loci associated with OSA, and estimated SNP-based heritability at 16%. We report six independent loci in a separate African population meta-analysis (Ncases= 46,834; Ncontrols= 149,192). We observed spatially resolved gene enrichment involving GABAergic and glutamate pathways, synaptic transmission, and cytoskeletal remodelling. Automated brain atrophy quantification and evaluation using spatial resolution enhancement.

Brain atrophy is one of the most common features of neurodegenerative diseases and is particularly critical in the early diagnosis of conditions like Alzheimer's and multiple sclerosis. Automated segmentation and quantification are highly desirable in brain atrophy evaluation but existing methods require high-quality MRI scans with isotropic resolution. However in practice, clinicians usually choose to reduce the number of slices to save time, and because of their anisotropic resolution, the morphometric analysis cannot be implemented. Here we propose the use of an inter-slice interpolation ne
