Safety and efficacy of the therapy with CD4 + CD25highCD127-T regulatory cells: When paediatric patient becomes adult.

AIM: CD4 + CD25highCD127-T regulatory cells (Tregs) remain a drug candidate for immunotherapy of type 1 diabetes. We completed three trials testing Tregs in recently diagnosed type 1 diabetes children. Here, we looked for long-term safety and efficacy aspects important in the authorisation of this therapy. MATERIALS AND METHODS: We re-examined 51 participants exposed to Treg therapy between 7.5 and 12 years prior to assessment. Based on previous assignments, we assessed the following subgroups treated with Tregs and anti-CD20 antibody (Tregs+antiCD20, N = 9), with 1 or 2 doses of Tregs only (

Targeting VCAM-1 with chimeric antigen receptor and regulatory T cell for abdominal aortic aneurysm treatment.

Abdominal aortic aneurysm (AAA) is a life-threatening condition characterized by the dilation of the abdominal aorta, leading to a high risk of rupture. Current treatment options are limited, particularly for patients ineligible for surgical interventions. This study explores a novel immunotherapeutic approach using chimeric antigen receptor Treg cells targeting vascular cell adhesion molecule-1 (VCAM-1) to mitigate AAA progression. By leveraging the specificity and regulatory function of CAR-Treg cells, our research aims to modulate the immune response and reduce inflammation in the aneurysma

Intravenous Immunoglobulin Elevates Regulatory T Cells in Guillain-Barré Syndrome: A Potential Biomarker of Therapeutic Response.

BACKGROUND AND AIMS: Intravenous immunoglobulin (IVIg) is the primary treatment for Guillain-Barré syndrome (GBS), yet its immunological mechanisms underlying variable clinical outcomes remain unclear. This study investigated the immunomodulatory effect of IVIg on regulatory T cells (Tregs), cytokines, and their association with treatment response and clinical outcomes. METHODS: In this prospective case-controlled study, 57 GBS patients and 57 age- and sex-matched healthy controls (HCs) were investigated. CD4+CD25+FoxP3+ Treg percentages, cytokine production (IL-10, TNF-α, IFN-γ, and IL-12),

Association of peripheral monocytic myeloid-derived suppressor cells with molecular subtypes in single-center endometrial cancer patients receiving carboplatin + paclitaxel/avelumab (MITO-END3 trial).

The MITO-END3 trial compared carboplatin and paclitaxel (CP) with avelumab plus carboplatin and paclitaxel (CPA) as first-line treatment in endometrial cancer (EC) patients and demonstrated a significant interaction between avelumab response and mismatch repair status. To investigate prognostic/predictive biomarker, 29 MITO-END3-EC patients were evaluated at pretreatment (B1) and at the end of CP/CPA treatment (B2) for peripheral myeloid-derived suppressor cells (MDSC) and Tregs. At B2, effector Tregs frequency was significantly higher in patients treated with CPA as compared to CP (p = 0.038)

Echinococcus granulosus antigen B regulates T-cell function through inhibition of signal transducer and activator of transcription 3 in experimental immune thrombocytopenia.

Dysregulated T-cell homeostasis is central to the development of immune thrombocytopenia (ITP), characterized by reduced platelet counts. Antigen B (AgB), a key protein in Echinococcus granulosus cyst fluid, modulates T-cell differentiation and reduces inflammation. Here, we explored the role of AgB in ITP and found that it enhances the generation and function of regulatory T cells (Tregs), boosting their immunosuppressive activity. In our passive ITP murine model, AgB treatment alleviated thrombocytopenia and restored the Treg-helper T-cell (Th) balance. However, the therapeutic effects of Ag

Targeting Setdb1 in T cells induces transplant tolerance without compromising antitumor immunity.

Suppressing immune responses promotes allograft survival but also favours tumour progression and recurrence. Selectively suppressing allograft rejection while maintaining or even enhancing antitumor immunity is challenging. Here, we show loss of allograft-related rejection in mice deficient in Setdb1, an H3K9 methyltransferase, while antitumor immunity remains intact. RNA sequencing shows that Setdb1-deficiency does not affect T-cell activation or cytokine production but induces an increase in Treg-cell-associated gene expression. Depletion of Treg cells impairs graft acceptance in Setdb1-defi

GPR15 differentially regulates the effects of cigarette smoke exposure on Crohn's disease and ulcerative colitis.

Inflammatory bowel disease (IBD), which includes Crohn's disease (CD) and ulcerative colitis (UC), is a chronic disorder characterized by gastrointestinal inflammation. Cigarette smoke is a well-established risk factor for the development and exacerbation of CD while exerting a paradoxical protective effect against the onset of UC. The exact mechanisms by which cigarette smoke influences IBD, as well as the opposite effects in UC and CD, have long remained unexplained. Here, we demonstrated the detrimental impact of cigarette smoke on CD progression while highlighting its beneficial effects on

Cancer Stem Cells Shift Metabolite Acetyl-Coenzyme A to Abrogate the Differentiation of CD103+ T Cells.

CD103+ T cells mediate potent anti-tumor immune responses and correlate with favorable clinical outcomes in cancer patients. However, the mechanisms by which cancer cells influence the differentiation of these cells remain elusive. Herein, we demonstrate that cancer stem cells (CSCs) play a pivotal role in suppressing CD103+ T cell differentiation in patients with non-small cell lung cancer (NSCLC). Specifically, CSCs facilitate the transfer of the metabolite acetyl-coenzyme A (acetyl-CoA) into interacting T cells via an exosome-dependent pathway. This process enhances the acetylation of B lym

H3K18 lactylation in cancer-associated fibroblasts drives malignant pleural effusion progression via TNFR2+ Treg recruitment.

Tumor necrosis factor receptor 2-positive regulatory T (TNFR2+ Treg) cells, the most suppressive subset of Treg cells, are enriched in malignant pleural effusion (MPE), contributing to disease progression. However, the underlying mechanisms responsible for their accumulation remain unclear. Here we demonstrate that the C-X-C motif chemokine ligand 16 (CXCL16)/C-X-C chemokine receptor type 6 (CXCR6) axis plays a critical role in recruiting TNFR2+ Treg cells to MPE, with cancer-associated fibroblasts serving as the primary source of CXCL16. Mechanistically, under the hypoxic conditions prevailin

Evaluating Activated Regulatory T Cells as a Biomarker of Chronic Allograft Inflammation in Pediatric Kidney Transplant Recipients.

BACKGROUND: There is a need for noninvasive immunological biomarkers that can identify stable kidney allograft immune quiescence to inform individualized immunosuppression. METHODS: We conducted a cross-sectional, pilot cohort study evaluating the relative abundance of regulatory T cells (Tregs) to effector T-cell (Teff) populations as a surrogate marker of long-term graft tolerance. We obtained fresh peripheral blood mononuclear cell samples from stable pediatric kidney transplant recipients, most with recent surveillance biopsies to identify the presence or absence of chronic inflammation.

SREBF1 mediates immunoparalysis of dendritic cells in sepsis by regulating lipid metabolism and endoplasmic reticulum stress.

BACKGROUND: Lipid metabolic reprogramming is a key feature of sepsis, with increased lipid storage contributing to disease progression. Although lipid metabolism dysregulation has been implicated in sepsis pathogenesis, how lipid biosynthesis, particularly mediated by sterol regulatory element-binding transcription factor 1 (SREBF1), leads to dendritic cell (DC) immunoparalysis remains unclear. METHODS: Intracellular lipid accumulation was assessed by Oil Red O and BODIPY staining. Gene and protein expression levels were analyzed via qPCR, Western blot, and immunofluorescence. SREBF1 activity

Coupling IL-2 with IL-10 to mitigate toxicity and enhance antitumor immunity.

Wild-type interleukin (IL)-2 induces anti-tumor immunity and toxicity, predominated by vascular leak syndrome (VLS) leading to edema, hypotension, organ toxicity, and regulatory T cell (Treg) expansion. Efforts to uncouple IL-2 toxicity from its potency have failed in the clinic. We hypothesize that IL-2 toxicity is driven by cytokine release syndrome (CRS) followed by VLS and that coupling IL-2 with IL-10 will ameliorate toxicity. Our data, generated using human primary cells, mouse models, and non-human primates, suggest that coupling of these cytokines prevents toxicity while retaining cyto

Acsbg1 maintains intestinal immune homeostasis and controls inflammation by regulating ST2+ Tregs.

The immune balance in mucosal tissues depends on a delicate interplay between inflammatory T helper 17 (Th17) cells and immunosuppressive regulatory T cells (Tregs). But what happens when this balance is disturbed? In this study, we uncovered a critical role for Acyl-CoA synthetase bubblegum family member 1 (Acsbg1) in shaping Th17 and Treg dynamics. Using Acsbg1-deficient mice, we show that while its absence does not disrupt homeostasis under steady-state conditions, it significantly alters Treg populations, particularly in gut-associated tissues. Under high-fat diet-induced metabolic stress,

SpaCBA-OVA Liposome Reprograms Immune Tolerance to Ameliorate Allergic Rhinitis via DC-T Cell Crosstalk.

Allergic rhinitis (AR) is a prevalent immune disorder driven by Th2-polarised inflammation, with current therapies primarily providing symptomatic relief rather than addressing the underlying immune dysfunction. Allergen-specific immunotherapy (AIT) holds promise for inducing long-term tolerance but is limited by poor mucosal delivery, enzymatic degradation of antigens, and inconsistent efficacy. A liposomal formulation co-encapsulating SpaCBA (probiotic protein) and ovalbumin (OVA) was engineered. Characterisation included in vitro/in vivo evaluations of immune modulation and therapeutic effi

Clostridium butyricum attenuates radiation-induced bone loss through gut microbiota and immune regulation in mice.

Radiation-induced bone loss remains clinically challenging due to limitations of existing treatments. Using a mouse model of fractionated total abdominal irradiation (TAI), we demonstrate that TAI triggers severe osteoporosis characterized by trabecular bone loss, suppressed osteogenesis, and elevated osteoclast activity. Through 16S rRNA sequencing and immune profiling, we found TAI induced gut dysbiosis, immune dysregulation such as Th17/Treg imbalance, and systemic inflammation. Oral supplementation with C. butyricum reversed bone loss, activated osteogenic signaling, suppressed osteoclasto

iTr35 cells: a novel immunoregulatory subset involved in bone loss in rheumatoid arthritis and clinical implications.

BACKGROUND: Rheumatoid arthritis (RA) is an autoimmune disorder characterized by chronic inflammation and progressive bone destruction. While classical regulatory T (Treg) cells are known to inhibit osteoclastogenesis, the role of non-classical subsets such as inducible IL-35-producing regulatory T cells (iTr35) in RA-related bone loss remains unexplored. OBJECTIVE: This prospective case-control study aimed to evaluate the expression of iTr35 cells in RA patients and assess their association with bone mineral density (BMD) and serum bone turnover markers. METHODS: Peripheral blood mononuclea

Immunopathological and microbial signatures of inflammatory bowel disease in partial RAG deficiency.

Partial RAG deficiency (pRD) can manifest with systemic and tissue-specific immune dysregulation, with inflammatory bowel disease (IBD) in 15% of the patients. We aimed at identifying the immunopathological and microbial signatures associated with IBD in patients with pRD and in a mouse model of pRD (Rag1w/w) with spontaneous development of colitis. pRD patients with IBD and Rag1w/w mice showed a systemic and colonic Th1/Th17 inflammatory signature. Restriction of fecal microbial diversity, abundance of pathogenic bacteria, and depletion of microbial species producing short-chain fatty acid we
