Tick-borne bacterial and protozoal pathogens in horses in Austria - A cross-sectional study on current and past infections and potential risk factors.

Tick-borne bacterial and protozoal pathogens are a major threat to equine health worldwide. In Austria, studies on pathogens such as piroplasms, Anaplasma phagocytophilum and Borrelia spp. in horses are scarce and do not cover the countrywide horse population. To address this, blood samples from horses without prior record of infection with tick-borne pathogens (n = 588) were tested for the presence of Theileria equi, Babesia caballi or A. phagocytophilum DNA by PCR and for specific antibodies against these pathogens and Borrelia burgdorferi with commercially available assays. While no DNA of

Molecular detection of Rickettsia helvetica in lesion and blood samples after tick bites in Serbia: two cases with One Health implications.

Rickettsia helvetica is a spotted fever group rickettsia associated mainly with Ixodes ricinus and increasingly detected in European ticks. We describe two tick-bite-associated human cases in Serbia in May 2025. A 65-year-old woman (Case 1) developed pruritic erythematous lesions after two adult female I. ricinus attachments acquired in the Valjevo area. Rickettsia spp. DNA was detected in capillary blood from both lesions, and sequencing of the PCR amplicons identified R. helvetica, while whole blood was PCR-negative and serology did not show reproducible seroconversion. An 8-year-old girl (C

Striking tick-borne virus diversity and potential reservoirs documented during One-Health-based cross-sectional screening in Anatolia.

BACKGROUND: An expansion of recently described human pathogenic tick-borne viruses from Central Asia toward Europe has been documented. Located on important bird migration routes, Anatolia is an intercontinental crossing hub with various climactic zones and with an abundance of endemic tick species. We sought to investigate tick-borne viruses utilizing a One Health approach encompassing host-removed ticks and host samples. METHODS: We collected host-attached ticks and accompanying plasma in 2023-2024 at locations in 20 provinces representing the 7 distinct geographical regions in Anatolia. Th

Patient-based evidence for amyotrophic lateral sclerosis prognostic health communication: "the clock is ticking…how long do I have?".

Objectives: Prognostic health communication is a critical challenge for amyotrophic lateral sclerosis (ALS) health-care professions, however patient-based evidence for best practice remains limited. We investigated how the experiences of ALS patients and caregivers can inform prognostic communication and whether patient-based evidence supports clinical use of predictive tools. Methods: Data were drawn from ALS Talk, an asynchronous, online focus group study. Patients and family caregivers were recruited from across Canada. Seven groups interacted in a threaded web-forum structure. Sixty-four p

A qualitative approach to life with neurofibromatosis type 1 based on Mishel's Uncertainty in Illness Theory: "My body is a ticking time bomb".

Neurofibromatosis type 1 (NF1) is a genetic disease that follows an autosomal dominant inheritance pattern, characterized by significant phenotypic variability and unpredictable clinical progression. Affected individuals can present from mild cosmetic involvement with café-au-lait macules to benign and malignant neoplasms, among other comorbidities that can diminish well-being or pose a threat to it. The transmission risk is 50%, and it is impossible to predict the extent of manifestations in the offspring. This qualitative study explored emotional and psychological meanings attributed by indi

Remdesivir is active in vitro against tick-borne encephalitis virus and selects for resistance mutations in the viral RNA-dependent RNA polymerase.

BACKGROUND: Tick-borne encephalitis (TBE) is a neurological disease caused by the tick-borne encephalitis virus (TBEV). Despite available vaccines, breakthrough infections occur, some fatal. OBJECTIVES: As no antiviral therapy for TBE is currently approved, this study evaluated the in vitro activity of already licenced remdesivir (RDV) and sofosbuvir (SOF) for possible drug repurposing against TBEV. METHODS: TBEV was cultured in A549 cells, and the inhibitory effects of RDV (GS-5734), its parent nucleotide GS-441524, and SOF (GS-7977) were assessed. RESULTS: After 78 h, RDV demonstrated sig

Identifying hotspots and risk factors for tick-borne encephalitis virus emergence at its range margins to guide interventions, Great Britain.

BackgroundTick-borne encephalitis virus (TBEV) is expanding its range in Europe, with increasing human cases reported. Since the first detection of TBEV in ticks in the United Kingdom in 2019, one possible, two probable and two confirmed autochthonous cases in humans have been reported.AimWe aimed to understand the environmental and ecological factors limiting TBEV foci at their range edge and predict suitable areas for TBEV establishment across Great Britain (GB) by modelling patterns of exposure to TBEV in deer.MethodsWe developed spatial risk models for TBEV by integrating data between 2018

Tick-Borne Encephalitis and West Nile Virus Antibody Prevalence in the City of Hamburg, a Flavivirus Non-Risk/Low-Risk Area.

In Hamburg, Northern Germany, no locally acquired human Tick-borne encephalitis virus (TBEV) or West Nile Virus (WNV) infections have been recorded as of to date. Since 2018, several WNV infections have been diagnosed in birds/horses. As a majority of TBEV and WNV infections are asymptomatic, diagnoses may be missed. Our objectives were to investigate TBEV- and WNV-antibody prevalence and TBE-protection rates through vaccination in Hamburg. In May 2023, we conducted a TBEV- and WNV-seroprevalence study in 1,321 blood donors at a large donation service; additionally, the percentage of TBE vacci

Assessing tick attachments to humans with citizen science data: spatio-temporal mapping in Switzerland from 2015 to 2021 using spatialMaxent.

BACKGROUND: Ticks are the primary vectors of numerous zoonotic pathogens, transmitting more pathogens than any other blood-feeding arthropod. In the northern hemisphere, tick-borne disease cases in humans, such as Lyme borreliosis and tick-borne encephalitis, have risen in recent years, and are a significant burden on public healthcare systems. The spread of these diseases is further reinforced by climate change, which leads to expanding tick habitats. Switzerland is among the countries in which tick-borne diseases are a major public health concern, with increasing incidence rates reported in

Applying MALDI-TOF MS to resolve morphologic and genetic similarities between two Dermacentor tick species of public health importance.

Hard ticks (Acari: Ixodidae) have been historically identified by morphological methods which require highly specialized expertise and more recently by DNA-based molecular assays that involve high costs. Although both approaches provide complementary data for tick identification, each method has limitations which restrict their use on large-scale settings such as regional or national tick surveillance programs. To overcome those obstacles, the matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) has been introduced as a cost-efficient method for the ident

Involving lived experience in regional efforts to address gambling-related harms: going beyond 'window dressing' and 'tick box exercises'.

BACKGROUND: Lived Experience (LE) involvement has been shown to improve interventions across diverse sectors. Yet LE contributions to public health approaches to address gambling-related harms remain underexplored, despite notable detrimental health and social outcomes linked to gambling. This paper analyses the potential of LE involvement in public health strategy to address gambling-related harms. It focuses on the example of a UK city-region gambling harms reduction intervention that presented multiple opportunities for LE input. METHODS: Three focus groups and 33 semi-structured interview

Molecular analysis of human tick-bitten skin yields signatures associated with distinct spatial and temporal trajectories - A proof-of-concept study.

Tick-associated diseases present challenges due to tridirectional interactions among host-specific responses, tick toxins and salivary proteins as well as microbes. We aimed to uncover molecular mechanisms in tick-bitten skin samples (cases) and contralateral skin samples (controls) collected simultaneously from the same participants, using spatial transcriptomics. Cases and controls analysed using NanoString GeoMx Digital Spatial Profiler identified 274 upregulated and 840 downregulated differentially expressed genes (DEGs), revealing perturbations in keratinization and immune system regulati

Qinghaienin, a novel anticoagulation protein from the hard tick Haemaphysalis qinghaiensis, inhibits the activation of factor XII by competing for anionic surfaces.

Salivary proteins of ticks can inhibit host hemostatic and inflammatory responses during the blood-sucking process of the parasites. A cDNA sequence, Hq021, was identified from a cDNA library of Haemaphysalis qinghaiensis. Hq021 encodes a mature protein containing 182 amino acids with a molecular mass of 20.5 kDa. The protein is rich in basic amino acids (lysine and arginine) at its C-terminus. Identification of no homologous proteins in databases suggested its novelty. A recombinant protein expressed with the cDNA in Escherichia coli could prolong the activated partial thromboplastin time (aP

Recurrent tick bites induce high IgG1 antibody responses to α-Gal in sensitized and non-sensitized forestry employees in Luxembourg.

BACKGROUND: The α-Gal syndrome (AGS) is characterized by the presence of specific IgE-antibodies to the carbohydrate galactose-α-1,3-galactose (α-Gal). Sensitization to α-Gal has been associated with tick bites and individuals exposed to ticks have an elevated risk of sensitization. The aim of this study was to analyze IgG and IgE antibody responses to α-Gal in a high-risk cohort of forestry employees (FE) in Luxembourg. METHODS: Questionnaires and serum samples of FE from Luxembourg (n = 219) were retrospectively analyzed. α-Gal specific IgE was quantified by ImmunoCAP, α-Gal specific IgG an

Different receptor models show differences in ligand binding strength and location: a computational drug screening for the tick-borne encephalitis virus.

The tick-borne encephalitis virus (TBE) is a neurotrophic disease that has spread more rapidly throughout Europe and Asia in the past few years. At the same time, no cure or specific therapy is known to battle the illness apart from vaccination. To find a pharmacologically relevant drug, a computer-aided drug screening was initiated. Such a procedure probes a possible binding of a drug to the RNA Polymerase of TBE. The crystal structure of the receptor, however, includes missing and partially modeled regions, which rendered the structure incomplete and of questionable use for a thorough drug s
