A Caenorhabditis elegans spatiotemporal proximity atlas reveals the MAPK p38 as a generator of phenotypic plasticity in vivo.

Phenotypic plasticity, the ability to change diverse traits without altering genomic information, is fundamental for organismal adaptations to changing environments. The mitogen-activated protein kinase (MAPK) pathway plays a key role in cellular adaptations to changing environments. To analyze the contributions of the MAPK p38 and its interaction partners to phenotypic plasticity in an animal, we established an in vivo proximity labeling proteomics method called ContinuumID. With this method, we built an atlas of PMK-1 physical interactors during development, in specific tissues, and during e

Oncogenic role of PHF23 in gastric cancer progression.

Gastric cancer (GC) poses a persistent global health burden, largely attributed to its asymptomatic early stages and limited efficacy of existing therapies. PHF23, which has been primarily studied in hematologic malignancies and degenerative joint diseases, has not been explored in gastric cancer. In this study, PHF23 expression was assessed in GC tissues through immunohistochemistry and public dataset analysis. Functional assays, including proliferation, migration, invasion, and apoptosis, were performed following PHF23 knockdown in GC cell lines. Tumorigenic effects were evaluated in nude mo

Reduced colony-stimulating factor 1 receptor expression in myeloid cells has limited impact on chronic lymphocytic leukaemia progression.

Targeting the colony-stimulating factor 1 receptor (CSF1R) to remove tumour-associated macrophages is being explored as cancer therapy. This strategy may be relevant for chronic lymphocytic leukaemia (CLL), which strongly depends on support from myeloid cells. However, it is unclear how CSF1R expression affects the CLL microenvironment and leukaemic progression. To examine this question, we created Eμ-TCL1 transgenic mice for CLL with Csf1r haploinsufficiency to investigate changes in myeloid cells, Csf1r expression and leukaemia progression. Csf1r haploinsufficiency reduced Csf1r expression o

Host-cell dependent epigenetic profiles associated with survival outcomes in T. gondii infection.

Toxoplasmosis is a widespread parasitic disease affecting roughly one-third of the global population. In immunocompromised individuals or during pregnancy, infection can result in severe complications. Following primary infection, Toxoplasma gondii forms dormant bradyzoite cysts in tissues such as the brain and eyes. These cysts can rupture, particularly in immunocompromised hosts, releasing active parasites and triggering recrudescence. Efforts to experimentally induce and study bradyzoite cyst recrudescence have been hindered by the limited capacity of cell culture adapted strains to form ti

Development and immunoprotective evaluation of a thermostable mRNA vaccine targeting Vibrio harveyi flagellin in the large yellow croaker (Larimichthys crocea).

Vibrio harveyi causes severe vibriosis in the large yellow croaker (Larimichthys crocea), resulting in high mortality rates and significant economic losses in mariculture. Currently, there is no effective commercial vaccine available, and the overuse of antibiotics raises concerns about resistance and the environment. In this study, we developed a thermostable mRNA-LNP vaccine that encodes the FlaA flagellin, which is a key virulence and immunogenic antigen of V. harveyi. Through the optimisation of UTRs, capping, lipid nanoparticle (LNP) formulation, microfluidic encapsulation and lyophilisat

Comparative Tumor Microenvironment Analysis for HCC and PDAC Using KMplotter.

Hepatocellular carcinoma (HCC) and pancreatic ductal adenocarcinoma (PDAC) are highly lethal cancers marked by profound epigenetic and metabolic reprogramming. Among the candidate biomarkers, the DNA methyltransferase DNMT3A and the guanine monophosphate synthetase (GMPS) have emerged as potential prognostic drivers, yet their roles across tumor contexts remain unclear. Here, we demonstrate the application of KMplotter to interrogated pan-cancer transcriptomic and survival datasets encompassing over 7000 patients, complemented by expression profiling of normal, tumor, and metastatic tissues, a

Analysis and identification of PTBP2 as an oncogene in hepatocellular carcinoma.

PURPOSE: Liver cancer, also known as hepatocellular carcinoma (HCC), is one of the most prevalent and lethal malignancies in the world. The management of HCC depends on the underlying hepatic function and histological stage of the neoplasm, potentially encompassing locoregional therapies, surgical resection, hepatic transplantation, targeted drug therapies, and immunotherapy. However, the outlook for advanced HCC remains poor, with global 5-year survival rates below 15%. Consequently, it is imperative to identify novel tumor biomarkers for HCC prediction. METHODS: Multiple public databases we

Frontline sigvotatug vedotin plus pembrolizumab vs pembrolizumab for non-small cell lung cancer with PD-L1 tumor proportion score ≥50%: phase III study design.

Integrin beta-6 (IB6) is a tumor-associated membrane protein involved in many cellular processes, including wound healing and tissue remodeling. While IB6 expression is constitutively low in healthy tissues, high IB6 expression in numerous cancers, including non-small cell lung cancer (NSCLC), is associated with poor outcomes. In a phase I study, the novel IB6-directed vedotin-based antibody-drug conjugate sigvotatug vedotin (SV) showed manageable safety and encouraging efficacy as a monotherapy and in combination with pembrolizumab in patients with advanced solid tumors, including NSCLC. Plasma Gelsolin Inhibits Natural Killer Cell Function and Confers Chemoresistance in Epithelial Ovarian Cancer.

Plasma gelsolin (pGSN) overexpression in ovarian cancer (OVCA) disarms immune function, contributing to chemoresistance. The aim of this study was to investigate the immunoregulatory effects of pGSN expression on natural killer (NK) cell function in OVCA. OVCA tissues from primary surgeries underwent immunofluorescent staining of pGSN and the activated NK cell marker natural cytotoxicity triggering receptor 1 to analyze the prognostic impact of pGSN expression and activated NK cell infiltration. The immunoregulatory effects of pGSN on NK cells were assessed using apoptosis assay, cytokine secr

Identification of FOXP1 as a favorable prognostic biomarker and tumor suppressor in intrahepatic cholangiocarcinoma.

BACKGROUND: Forkhead-box protein P1 (FOXP1) has been proposed to have both oncogenic and tumor-suppressive properties, depending on tumor heterogeneity. However, the role of FOXP1 in intrahepatic cholangiocarcinoma (ICC) has not been previously reported. METHODS: Immunohistochemistry was performed to detect FOXP1 expression in ICC and normal liver tissues. The relationship between FOXP1 levels and the clinicopathological characteristics of patients with ICC was evaluated. Finally, in vitro and in vivo experiments were conducted to examine the regulatory role of FOXP1 in ICC cells. RESULTS: F

Lineage-specific requirements of Alx4 function in craniofacial and hair development.

BACKGROUND: Disruption of ALX4 causes autosomal dominant parietal foramina and autosomal recessive frontonasal dysplasia with alopecia, but the mechanisms involving ALX4 in craniofacial and other developmental processes are not well understood. Although mice carrying distinct mutations in Alx4 have been previously reported, the perinatal lethality of homozygous mutants together with dynamic patterns of Alx4 expression in multiple tissues have hindered systematic elucidation of the cellular and molecular mechanisms involving Alx4 in organogenesis and disease pathogenesis. RESULTS: We report ge

Liver-directed AAV gene therapy normalizes disease symptoms and provides cross-correction in a model of lysosomal acid lipase deficiency.

Lysosomal acid lipase deficiency (LAL-D) is caused by mutations in the LIPA gene, which encodes the lysosomal enzyme that hydrolyzes triglycerides and cholesteryl esters to free fatty acids and free cholesterol. The objective of this study was to develop a curative single-treatment therapy for LAL-D using adeno-associated virus (AAV). Treatment at both early (1-2 days) and late (8-week) timepoints with rscAAVrh74.LP1.LIPA, a liver-directed AAV gene therapy, normalized many disease measures in Lipa-/- mice when measured at 24 weeks of age, including hepatosplenomegaly, serum transaminase activi

PHF6 promotes the progression of endometrial carcinoma by increasing cancer cells growth and decreasing T-cell infiltration.

Uterine corpus endometrial carcinoma (UCEC) is the most common cancer of the female reproductive tract. The overall survival of advanced and recurrent UCEC patients is still unfavourable nowadays. It is urgent to find a predictive biomarker and block tumorgenesis at an early stage. Plant homeodomain finger protein 6 (PHF6) is a key player in epigenetic regulation, and its alterations lead to various diseases, including tumours. Here, we found that PHF6 expression was upregulated in UCEC tissues compared with normal tissues. The UCEC patients with high PHF6 expression had poor survival than UCE

Acute Myeloid Leukemia Expresses a Specific Group of Olfactory Receptors.

Acute myeloid leukemia (AML) is the most common form of acute leukemia in adults, with a 5-year overall survival rate of approximately 30%. Despite recent advances in therapeutic options, relapse remains the leading cause of death and poor survival outcomes. New drugs benefit specific small subgroups of patients with actionable therapeutic targets. Thus, finding new targets with greater applicability should be pursued. Olfactory receptors (ORs) are seven transmembrane G-protein coupled receptors preferentially expressed in sensory neurons with a critical role in recognizing odorant molecules.

RNA Sequencing Reveals Unique Transcriptomic Signatures of the Thyroid in a Murine Lung Cancer Model Treated with PD-1 and PD-L1 Antibodies.

Immune checkpoint inhibitors (ICI) are commonly associated with thyroid immune-related adverse events, yet the mechanism has not been fully elucidated. We aimed to further explore the mechanism of ICI-induced thyroid dysfunction by assessing changes induced in the thyroid transcriptome by ICI treatment (αPD-1/αPD-L1) in a lung cancer murine model. RNA-sequencing of thyroid tissues revealed 952 differentially expressed genes (DEGs) with αPD-1 treatment (|fold-change| ≥1.8, FDR < 0.05). Only 35 DEG were identified with αPD-L1, and we therefore focused on the αPD-1 group alone. Ingenuity Pathway

Necroptosis-Related miRNA Biomarkers for Predicting Overall Survival Outcomes for Endometrial Cancer.

Endometrial cancer (EC) is the gynecological tumor with the highest incidence. In recent years, it has been proved that necroptosis is a method of cell death related to EC. However, the expression of necroptosis-related miRNA in EC and its correlation with prognosis still ill-defined. Use the Cancer Genome Atlas (TCGA) cohort to obtain prognostic data and related clinical data for ECs and normal endometrium tissues. In this study, we identified three necroptotic regulatory miRNAs that are necroptosis-related and survival-related miRNAs (DENSMs) between normal endometrium tissues and EC from 13
