Evidence for somatic cell expression of the c-mos protein [corrected].

The c-mos protein has been found to be enriched in germ cells of male mice, as described in a recent report from this laboratory (Herzog et al., Oncogene 3, 225, 1988). We report on further studies which indicate that the c-mos protein (a 41 to 43 kDa protein termed p43c-mos) is expressed in somatic tissues of mice and in cells grown in culture. In testes of mice, germ cell fractions have increased levels of p43c-mos relative to other cells of the testes. However, non-germ cells harbor significant levels of p43c-mos, as judged by comparison of testes from normal mice to those with mutations th

Impaired neurotransmitter uptake in PC12 cells overexpressing human Cu/Zn-superoxide dismutase--implication for gene dosage effects in Down syndrome.

Rat PC12 cells expressing elevated levels of transfected human Cu/Zn-superoxide dismutase (CuZn-SOD) gene were generated. These transformants (designated PC12-hSOD) closely resembled the parental cells in their morphology, growth rate, and response to nerve growth factor, but showed impaired neurotransmitter uptake. The lesion was localized to the chromaffin granule transport mechanism. We found that the pH gradient (delta pH) across the membrane, which is the main driving force for amine transport, was diminished in PC12-hSOD granules. These results show that elevation of CuZnSOD activity int

Product of the cellular oncogene, c-fos, observed in mouse and human tissues using an antibody to a synthetic peptide.

The transforming gene of the osteosarcoma-producing FBJ murine sarcoma virus, v-fos, is homologous to a normal cellular gene, c-fos, in vertebrate species. Transcripts from the c-fos proto-oncogene accumulate to very high levels in late gestational mouse and human extra-embryonic tissues. We now report that these RNA transcripts are translated in these tissues. Rabbits were immunized with a synthetic peptide whose sequence is common to both c-fos and v-fos. After affinity purification on an immunosorbent containing the fos peptide (a nonapeptide), the antibody reacted with a component(s) in nu

Inadequate waste management drives the spread of antimicrobial resistance across the watershed of a large African city.

Rapid urbanization in the Global South consistently outpaces sanitation infrastructure, transforming urban rivers into AMR reservoirs. Yet watershed-scale quantification of resistomes in such settings remains scarce, limiting exposure risk assessment and intervention planning. Here we present the most detailed spatial baseline for waterborne AMR in sub-Saharan Africa, using the Akaki River (Addis Ababa, Ethiopia) as a model system for rapidly urbanizing megacity watersheds with inadequate sanitation. We sampled 38 sites across an extreme pollution gradient during the dry season, when low flow

Time-resolved phenotyping of seizure evolution in developmental and epileptic encephalopathy: insights and gaps from the CYFIP2 Arg87Cys mouse model.

In developmental and epileptic encephalopathies (DEEs), the seizure phenotype is rarely static. Infantile epileptic spasms typically subside and are often replaced years later by other seizure types, and in several genetically defined DEEs seizure-free intervals precede seizure recurrence. Although these transitions are linked to long-term outcome, little is known about the molecular and cellular events that accompany them, because the interval is clinically silent and brain tissue is inaccessible. Animal studies have only partly filled this gap: most DEE models have been characterized at a si

GPCR signaling systems facilitate precision interventions for multi-oncology therapy.

Precision oncology has undergone a transformative evolution from static genomic profiling toward dynamic, multi-omics frameworks capable of capturing tumor heterogeneity and adaptive resistance in real time. Central to this paradigm shift is the emerging recognition that G protein-coupled receptors (GPCRs) constitute a vastly underexploited axis of oncogenic signaling, tumor microenvironment (TME) modulation, and non-genetic adaptive resistance. Here we delineate the effective transition to GPCR-focused precision oncology, emphasizing the integrated deployment of next-generation sequencing (NG

The inflammatory clock: how cGAS-STING ticks in the aging ovary.

Premature ovarian insufficiency (POI) is more than a fertility issue; it's a silent epidemic of accelerated systemic aging in young women, with current treatments failing to address its root cause. For too long, the relentless decline of ovarian function has been viewed as an inevitable mystery. But what if the ovary holds an internal "inflammatory clock," ticking away with each cellular insult and dictating the pace of its own decline? Here, we spotlight a surprising culprit: the cGAS-STING signaling pathway. Far beyond its day job in antiviral defense, this pathway emerges as a master integr

Versatile SMAD2 and SMAD3 epitope-tagged mouse models for genomic profiling of TGFβ signaling: Uncovering GDF9-SMAD2/3 targets.

Transforming growth factor β (TGFβ) signaling pathways are integral for a plethora of biological processes. SMAD2 and SMAD3 are the principal transcriptional effectors of TGFβ superfamily ligands, yet quantitative, genome-wide mapping of their DNA-associated complexes under physiological contexts has remained limited due to the lack of specific, robust models. Here, we generated two versatile epitope-tagged mouse models in which endogenous SMAD2 and SMAD3 proteins are globally tagged with hemagglutinin (HA) and podoplanin (PA) sequences, respectively, enabling high-fidelity profiling of SMAD2

Biobank-Scale Plasma Proteomics Identifies Novel Biomarkers in Hypertrophic Cardiomyopathy.

BACKGROUND: Hypertrophic cardiomyopathy (HCM) is characterized by substantial heterogeneity in both clinical phenotype and risk of adverse outcomes, including heart failure and sudden cardiac death. This highlights the need for robust biomarkers for risk stratification, and while previous studies have identified the role of select plasma proteins, comprehensive large-scale proteomic analyses have been limited in HCM. METHODS: We performed a case-control analysis of 2922 plasma proteins in 49 588 UK Biobank participants (100 HCM cases) to identify proteins associated with HCM. External replica

Integrated FTIR and Whole-Genome Sequencing Reveal Scale-Dependent Genotype-Phenotype Relationships in Multidrug-Resistant Pseudomonas aeruginosa.

Multidrug-resistant Pseudomonas aeruginosa is a major cause of healthcare-associated infections, particularly in high-burden clinical settings where rapid tools to capture clinically relevant resistance and virulence phenotypes are needed. In this study, we applied an integrated whole-genome sequencing (WGS) and Fourier-transform infrared (FTIR) spectroscopy approach to evaluate genotype-phenotype relationships in multidrug-resistant P. aeruginosa isolates collected during the COVID-19 pandemic. High-quality WGS data were used to characterize antimicrobial resistance determinants, mobile genet

Total Thrombus-Formation Analysis System (T-TAS) in Aortopathies: A Conceptual and Potential Framework to Spatial Heterogeneity and Regional Context.

Thoracic aortopathies, including aneurysm and dissection, are complex vascular disorders characterized by structural alterations of the aortic wall that disrupt normal haemodynamics. Altered shear stress, turbulent flow, and endothelial dysfunction promote thrombus formation and modulate systemic hemostasis via platelet activation and the von Willebrand factor-ADAMTS13 axis. The Total Thrombus-Formation Analysis System (T-TAS) is a microfluidic, flow-dependent assay that quantitatively evaluates thrombus formation under physiological shear conditions. Although studied in various cardiovascular

Harm reduction in paediatric off-label therapy: ethical analysis of early CFTR modulator use in an infant with cystic fibrosis.

Cystic fibrosis (CF) is a life-limiting genetic disease frequently associated with pancreatic insufficiency (PI), traditionally considered irreversible. CF transmembrane conductance regulator modulators such as elexacaftor/tezacaftor/ivacaftor (ETI; Trikafta) have transformed CF care yet remain FDA-approved only for children aged 2 years and older. We present the case of a 9-month-old female with CF and PI whose mother, highly health-literate and resourceful, pursued early ETI exposure through breastmilk transfer and later direct off-label dosing, despite medical guidance and regulatory restri

Understanding the Cellular Spatiotemporal Dogma by Spatial and Single-cell Omics.

The biological complexity of plants arises from highly coordinated cellular activities. We propose that a "cellular spatiotemporal dogma" governs the zygote's programmed development into a complete plant and its adaptation to various environmental stresses, representing the set of principles describing how gene expression, cell identity, and function are coordinated across physical spatial contexts and temporal developmental progressions. Historically, our understanding of this dogma remains limited because traditional bulk tissue sequencing provides only a homogenized average of gene expressi

Pollution duration drives divergent pathways of antibiotic and metal resistance gene dissemination in coastal mangrove sediments.

Coastal mangrove sediments are critical reservoirs of antibiotic and heavy metal resistance genes (ARGs/HMRGs). Yet, how pollution history shapes their dissemination remains unclear. We elucidated this by comparing sedimentary resistomes across three distinct ecosystems: a pristine site (BH), a site under mid-term copper stress (BHCu), and a chronically multi-polluted site (FT). Chronic multi-pollution generated a persistent co-resistance reservoir, significantly enriching ARGs, HMRGs, mobile genetic elements (MGEs), and transferable ARG-MGE clusters. Bioavailable metal fractions cooperated wi

Plant immune co-receptors: bridging gaps toward next-generation crop defense.

Plant innate immunity is a crucial, multi-layered defense system that protects crops from a wide range of bacterial, fungal, and viral pathogens, all of which pose a significant threat to global food security. Co-receptors function as central molecular modulators that amplify and fine-tune immune signaling. This signaling is initiated by cell-surface pattern-recognition receptors (PRRs) and intracellular nucleotide-binding leucine-rich repeat (NLR) proteins. Critical co-receptors for strong immune signal propagation include the SOMATIC EMBRYOGENESIS RECEPTOR KINASE (SERK) protein family, repre

Therapeutic antibody delivery: vector tools to boost efficacy and affordability.

Antibody (Ab)-based therapeutics have become powerful tools across diverse disease areas, with advances in bioengineering giving rise to next-generation molecules designed to outperform conventional Abs. Yet, large-scale production and purification of such complex proteins remain costly and can restrict patient access. A promising alternative is to improve in vivo expression capabilities, which will reduce manufacturing burdens and improve safety and tolerability. Multiple gene delivery platforms - ranging from mRNA and viral vectors to engineered cell therapies - have matured considerably, as
