Novel transcriptional regulator OxtR1 regulates potential ferrodoxin in response to oxygen stress in Treponema denticola.

OBJECTIVE: Treponema denticola has been strongly implicated in the pathogenesis of chronic periodontitis. Previously, we reported that the potential transcriptional regulator TDE_0259 (oxtR1) is upregulated in the bacteriocin ABC transporter gene-deficient mutant. OxtR1 may regulate genes to adapt to environmental conditions during colonization; however, the exact role of the gene in T. denticola has not been reported. Therefore, we investigated its function using an oxtR1-deficient mutant. METHODS: The growth rates of the wild-type and oxtR1 mutant were monitored under anaerobic conditions;

Janus PEGylated CuS-engineered Lactobacillus casei combats biofilm infections via metabolic interference and innate immunomodulation.

Bacterial implant-associated infections predominantly contribute to the failure of prosthesis implantation. The local biofilm microenvironment (BME), characterized by its hyperacidic condition and high hydrogen peroxide (H2O2) level, inhibits the host's immune response, thereby facilitating recurrent infections. Here, a Janus PEGylated CuS nanoparticle (CuPen) armed engineered Lactobacillus casei (L. casei) denoted as LC@CuPen, is proposed to interfere with bacterial metabolism and arouse macrophage antibiofilm function. Once LC@CuPen reached the BME, NIR irradiation-activated mild heat damage

Characterisation of the bacterial microbiota of the vagina of dairy cows and isolation of pediocin-producing Pediococcus acidilactici.

BACKGROUND: Uterine infections in dairy cows lower profitability of dairy operations. Infections of the reproductive tract are related to the overgrowth of pathogenic bacteria during the first three weeks after parturition. However, alterations in the vaginal microbiota composition in the first weeks after parturition remain poorly documented. RESULTS: In this study, bacteria isolated from the vagina of healthy pregnant, and infected postpartum cows were characterised by random amplification of polymorphic DNA (RAPD) analysis and partial 16S ribosomal RNA (rDNA) gene sequencing. Populations o

Fluoresceination of FepA during colicin B killing: effects of temperature, toxin and TonB.

We studied the reactivity of 35 genetically engineered Cys sulphydryl groups at different locations in Escherichia coli FepA. Modification of surface loop residues by fluorescein maleimide (FM) was strongly temperature-dependent in vivo, whereas reactivity at other sites was much less affected. Control reactions with bovine serum albumin showed that the temperature dependence of loop residue reactivity was unusually high, indicating that conformational changes in multiple loops (L2, L3, L4, L5, L7, L8, L10) transform the receptor to a more accessible form at 37 degrees C. At 0 degrees C colici

Distribution and persistence of probiotic Streptococcus salivarius K12 in the human oral cavity as determined by real-time quantitative polymerase chain reaction.

The bacteriocin producer Streptococcus salivarius K12 is used as a probiotic targeting the oral cavity, so our study aimed to assess whether its dispersal and persistence could be monitored using real-time quantitative polymerase chain reaction. To this end, we designed polymerase chain reaction primers and a hybridization probe specifically targeting salA, which encodes for the prepropeptide of salivaricin A. Using a single individual as our subject, we administered four lozenges of K12 Throat Guard per day over 3 days, then measured salA gene levels for 16 different oral sites at six differe

Genetic and phenotypic differences among Enterococcus faecalis clones from intestinal colonisation and invasive disease.

This study investigated the differences among Enterococcus faecalis isolates from the intestinal compartment of healthy volunteers (n = 36), intensive care unit (ICU) patients (n = 29) and blood isolates (n = 31) from the same institution, in comparison with seven epidemic clones from other institutions. In general, isolates from colonised ICU patients and from bacteraemic patients showed higher rates of antimicrobial resistance than isolates from colonised healthy volunteers, particularly for erythromycin and aminoglycosides. The proportion of isolates/clone was 1.05 in the community, 2.63 in

Probiotic and safety characterization of Lactiplantibacillus plantarum ZZM01 isolated from mink.

In recent years, probiotics have attracted increasing attention due to their roles in gut health regulation, antimicrobial activity, and metabolic benefits. Given the high-fat dietary characteristics of mink, their intestinal microbiota represents a potential source of functionally valuable lactic acid bacteria. In this study, we evaluated the probiotic properties, safety profile, and functional potential of Lactiplantibacillus plantarum ZZM01 isolated from mink intestine. ZZM01 exhibited good tolerance to simulated gastrointestinal conditions, with survival rates of 71.92% at pH 2.0 and 84.71

The MarR family regulator RmaH underlies the trade-off between acid sensitivity and antibiotic tolerance in Lactococcus lactis.

UNLABELLED: Microorganisms exhibit remarkable adaptability to diverse environmental stresses through transcriptional reprogramming. Here, we focused on the MarR family regulator RmaH in Lactococcus lactis, which orchestrated a pivotal trade-off: enhancing tolerance to the bacteriocin nisin and the antibiotic vancomycin at the expense of acid resistance. RmaH directly activated genes for cell wall and membrane lipid biosynthesis, leading to a thicker cell wall, and repressed the arginine deiminase pathway, attenuating acid tolerance. Integrated transcriptomic, chromatin immunoprecipitation-sequ

Nisin improves the storage quality of refrigerated pre-packaged fermented soybean whey-tofu-food safety, texture, and flavor.

INTRODUCTION: Fermented soybean whey-based tofu, a traditional and popular food product, faces significant spoilage issues during storage, including texture softening, elevated volatile basic nitrogen (TVB-N) levels, and accumulation of biogenic amines, which can compromise sensory attributes and pose health risks. Current preservation strategies, such as physical sterilization and chemical preservatives, have limitations due to potential nutritional loss and safety concerns. Therefore, there is an urgent need for safe, efficient, and eco-friendly preservation technologies. This study evaluate

Yersinia pestis BipA is a novel regulator of pesticin and a type 6 secretion system.

Yersinia pestis is a gram-negative bacterium and the causative agent of bubonic, septicemic, and pneumonic plague. Y. pestis is most commonly transmitted to humans by infected fleas that deposit the bacteria into the dermis at the bite site, leading to bubonic plague. The bacteria ultimately access the bloodstream, and after deposition in the lung, can be transmitted person-to-person through infectious respiratory droplets, resulting in primary pneumonic plague, a highly lethal and rapidly progressing pneumonia. Pathogenesis is mediated by a suite of chromosomally encoded and plasmid-borne vir

Impact of the calcium concentration on the efficacy of phage phiIPLA-RODI, LysRODIΔAmi and nisin for the elimination of Staphylococcus aureus during lab-scale cheese production.

Staphylococcus aureus is a Gram-positive human opportunistic pathogen that may also cause food poisoning because of the ability of some strains to produce heat stable enterotoxins that can persist in food even after the pathogen is successfully eliminated. In this context, biopreservation may be a forward-looking strategy to help eliminate staphylococcal contamination in dairy products by using natural compounds. However, these antimicrobials exhibit individual limitations that may be overcome by combining them. This work investigates the combination of a virulent bacteriophage, phiIPLA-RODI,

Double-Edged Nanobiotic Platform with Protean Functionality: Leveraging the Synergistic Antibacterial Activity of a Food-Grade Peptide to Mitigate Multidrug-Resistant Bacterial Pathogens.

While persistent efforts are being made to develop a novel arsenal against bacterial pathogens, the development of such materials remains a formidable challenge. One such strategy is to develop a multimodel antibacterial agent which will synergistically combat bacterial pathogens, including multidrug-resistant bacteria. Herein, we used pediocin, a class IIa bacteriocin, to decorate Ag° and developed a double-edged nanoplatform (Pd-SNPs) that inherits intrinsic properties of both antibacterial moieties, which engenders strikingly high antibacterial potency against a broad spectrum of bacterial

Genomic analysis of Indian strains of Salmonella enterica subsp. enterica serovar Typhi indicates novel genetic repertoire for pathogenicity and adaptations.

In the era of emerging antibiotic resistance, Salmonella enterica subsp. enterica serovar Typhi the causative agent of typhoid, is a threat for healthcare systems in developing countries especially India, where the disease is highly endemic. Genetic diversity among different strains may be the cause of variable severity of disease in different regions of the world. To explore this genetic diversity, genome annotation by rapid annotation using subsystem technology (RAST) was carried out for genomes of four Salmonella Typhi strains from two distinct areas available in the public domain. Two clin

Beneficial effect of lantibiotic nisin in rabbit husbandry.

Nisin is a bacteriocin marketed as Nisaplin. The aim of our work was to test its in vivo effect in a rabbit model; its effect on phagocytic activity (PA) and morphometry has not so far been studied. Post-weaning rabbits (48), 5 weeks old (both sexes, Hycole breed), were divided into the experimental (E) and the control groups (C), 24 animals in each. They were fed a commercial diet with access to water ad libitum. Rabbits in E had nisin additionally administered to their drinking water (500 IU-20 μg per animal/day) for 28 days. The experiment lasted 42 days. On day 28, significant decrease in

Effect of new alleles of the histidine kinase gene ciaH on the activity of the response regulator CiaR in Streptococcus pneumoniae R6.

The two-component regulatory system CiaRH of Streptococcus pneumoniae affects β-lactam susceptibility, autolysis, bacteriocin production, competence development, host colonization and virulence. The system was discovered in a screen for S. pneumoniae R6 mutants resistant to the β-lactam antibiotic cefotaxime. A mutation in the histidine kinase gene ciaH led to this phenotype by enhancing CiaR-mediated gene expression. Additional mutations in ciaH have been described in other spontaneous β-lactam-resistant mutants of S. pneumoniae R6, but their influence on CiaR-mediated gene regulation has not

Transcriptional regulation of the sil locus by the SilCR signalling peptide and its implications on group A streptococcus virulence.

In the last two decades an increasing number of local outbreaks of invasive group A streptococcus (GAS) infections including necrotizing fasciitis (NF) have been reported. We identified the streptococcal invasion locus (sil) which is essential for virulence of the M14 strain JS95 isolated from an NF patient. This locus contains six genes: silA/B and silD/E encoding two-component system (TCS) and ABC transporter, respectively, homologous to the corresponding entities in the regulon of Streptococcus pneumoniae involved in genetic competence. Situated between these two units are silC and silCR, w

Functional and Genomic insights into probiotic Lactic Acid Bacteria isolated from the Gastrointestinal microbiota of domestic rabbits (Oryctolagus cuniculus).

Probiotics play vital roles in maintaining gut health and overall well-being of animals, and have been isolated from the guts of several animals. The caecotrophic and dynamic nature of rabbit gut make them a promising site for probiotic prospecting. This study therefore, investigated the probiotic properties and whole genome sequence profile of Lactic Acid Bacteria (LAB) isolated from the gut microflora of domestic rabbit. Gastrointestinal contents of rabbits were cultured anaerobically on de Mann Rogosa Sharpe (MRS) agar using the pour-plate method. Isolates obtained were initially identified

Beyond commensalism: genomic insights into micrococcin P1-producing Staphylococcus chromogenes.

UNLABELLED: Staphylococcus chromogenes (S. chromogenes) is a predominant non-aureus staphylococcal species colonizing the teat skin and mammary gland of dairy ruminants. Although often linked to mild or subclinical mastitis, specific strains may also play protective roles against major udder pathogens. In this study, we characterized two S. chromogenes isolates (4S77 and 4S90) that displayed antimicrobial activity against Gram-positive bacteria. Complete genome sequencing revealed a conserved, plasmid-encoded biosynthetic gene cluster for the thiopeptide bacteriocin micrococcin P1 (MP1). All g
