Photobiomodulation with near-infrared laser for tinnitus management: preliminary animal experiments and randomized clinical trials.

This study investigates the effectiveness of photobiomodulation therapy (PBMT) in treating chronic high-frequency tinnitus with the TINI device, a near-infrared (830 nm) laser. The study includes preliminary animal experiments with 28 mice and a randomized controlled trial with 56 participants to examine the functional and molecular changes in the auditory system that PBMT may cause. The animal model used sodium salicylate to induce tinnitus, followed by PBMT, which showed promising reductions in the behavioral evidence of tinnitus and a reversal of tinnitus-associated upregulation of vesicula

A Case of Vestibular Schwannoma with Deafness Showing Remarkable Hearing Recovery Following Hearing-Preserving Surgery.

Surgery for vestibular schwannoma can be divided into hearing-preserving and nonhearing-preserving surgeries. Hearing-preserving surgery is usually not considered in patients with deafness due to vestibular schwannoma, because hearing is unlikely to improve, and surgery aims to maximize the tumor resection at the expense of hearing. We report an extremely rare case of a 46-year-old man with unilateral profound hearing loss due to a vestibular schwannoma with marked cystic degeneration in the left cistern, which significantly recovered to near-normal hearing levels after hearing-preserving surg

Susceptibility weighted imaging can be a sensitive sequence to detect brain damage in neonates with kernicterus: a case report.

BACKGROUND: Kernicterus in the acute phase is difficult to diagnose. It depends on a high signal on T1 at the globus pallidum and subthalamic nucleus level. Unfortunately, these areas also show a relatively high signal on T1 in neonates as an expression of early myelination. Therefore, a less myelin-dependent sequence, like SWI, may be more sensitive to detecting damage in the globus pallidum area. CASE PRESENTATION: A term baby developed jaundice on day three following an uncomplicated pregnancy and delivery. Total bilirubin peaked at 542 μmol/L on day four. Phototherapy was started, and an

In utero exposure to valproic acid disrupts ascending projections to the central nucleus of the inferior colliculus from the auditory brainstem.

Prenatal exposure to the antiepileptic valproic acid (VPA) is associated with an increased risk of autism spectrum disorder (ASD) in humans. Accordingly, in utero exposure to VPA is a validated and biologically relevant animal model of ASD. The majority of individuals with ASD exhibit some degree of auditory dysfunction, ranging from deafness to hypersensitivity. Animals exposed to VPA in utero have abnormal tonotopic maps and responses in the cerebral cortex and hyperactivation, hypoplasia, abnormal neuronal morphology and reduced calcium binding protein expression throughout the auditory bra

Tinnitus and temporary hearing loss result in differential noise-induced spatial reorganization of brain activity.

Loud noise frequently results in hyperacusis or hearing loss (i.e., increased or decreased sensitivity to sound). These conditions are often accompanied by tinnitus (ringing in the ears) and changes in spontaneous neuronal activity (SNA). The ability to differentiate the contributions of hyperacusis and hearing loss to neural correlates of tinnitus has yet to be achieved. Towards this purpose, we used a combination of behavior, electrophysiology, and imaging tools to investigate two models of noise-induced tinnitus (either with temporary hearing loss or with permanent hearing loss). Docosahexaenoic acid (cervonic acid) incorporation into different brain regions in the awake rat.

A quantitative method is presented to examine the localization, in individual brain regions of awake rats, of docosahexaenoic acid (22:6 n-3 or cervonic acid), the main polyunsaturated fatty acid of the nervous system together with arachidonic acid. Following the intravenous injection of 10 microCi [14C]22:6 n-3 (around 0.2 mumol/rat). 0.11-0.28% of the initial radioactivity was located in specific brain areas after detection from 10 to 240 min. Brain regional radioactivity determined by quantitative autoradiography indicated that 60 min after injection, [14C]22:6 n-3 concentrations ranged fro

Effect of Donepezil on Speech Recognition in Adult Cochlear Implant Users: A Pilot Randomized Clinical Trial.

OBJECTIVES: Speech recognition outcomes following cochlear implantation vary widely among users, potentially due to differences in neuroplasticity. The cholinergic system plays a critical role in cortical reorganization and sensory learning; yet, its potential to enhance auditory plasticity in cochlear implant (CI) users has not been explored. The aim of this study is to investigate the effects of donepezil, a cholinergic agonist, on speech perception and cortical plasticity in adults with CIs. DESIGN: This pilot randomized clinical trial was conducted at a single tertiary academic center fro

Management of vestibular schwannomas (acoustic neuromas): the value of neurophysiology for intraoperative monitoring of auditory function in 200 cases.

OBJECTIVE: The present study investigated the significance of the presence or absence of auditory brain stem response (ABR) Waves I, III, and V as functional representatives of the cochlea, the nucleus cochlearis, and the colliculus inferior, respectively, and attempted to identify the microsurgical maneuvers that were especially likely to cause isolated or combined component losses and subsequent hearing losses. METHODS: Based on the previously described ABR classification system, 201 patients with preserved Waves I, III, and V or Waves I and V were investigated for the peak latencies and am

A three-dimensional digital neurological atlas of the mustached bat (Pteronotus parnellii).

Substantial knowledge of auditory processing within mammalian nervous systems emerged from neurophysiological studies of the mustached bat (Pteronotus parnellii). This highly social and vocal species retrieves precise information about the velocity and range of its targets through echolocation. Such high acoustic processing demands were likely the evolutionary pressures driving the over-development at peripheral (cochlea), metencephalic (cochlear nucleus), mesencephalic (inferior colliculus), diencephalic (medial geniculate body of the thalamus), and telencephalic (auditory cortex) auditory pr

A glucocorticoid-regulating molecule, Fkbp5, may interact with mitogen-activated protein kinase signaling in the organ of Corti of mice cochleae.

FKBP5 is a 51-Da FK506-binding protein and member of the immunophilin family involved in controlling the signaling of glucocorticoid receptor from the cytosol to nucleus. Fkbp5 has previously been shown to be expressed in murine cochlear tissue, including the organ of Corti (i.e., the sensory epithelium of the cochlea). Fkbp5-/- mice as used in this study show hearing loss in the low-frequency (8-kHz) range and click-evoked auditory brainstem response (ABR) threshold compared to wild-type mice. Both Fkbp5-/- and wild-type mice showed hearing loss at all frequencies and click-ABR thresholds at

Do tDCS and TMS influence tinnitus transiently via a direct cortical and indirect somatosensory modulating effect? A combined TMS-tDCS and TENS study.

Tinnitus is usually defined as an intrinsic sound percept that cannot be attributed to an external sound source that tinnitus can be suppressed by neuromodulation techniques such as transcranial direct current stimulation (tDCS), transcranial magnetic stimulation (TMS), and transcranial electrical nerve stimulation (TENS). It is thought that TMS and tDCS modulate tinnitus directly by targeting the frontal and/or auditory cortex of the brain, whereas TENS most likely influences tinnitus indirectly via cervical nerve-cochlear nucleus interactions. It is unknown whether part of the tinnitus modul

Conditional Deletion of CB1 Receptor in Parvalbumin-Expressing GABAergic Neurons Results in Hearing Loss and Abnormal Auditory Brainstem Response in Mice.

Cannabinoid receptors 1 (CB1Rs) are widely expressed throughout the central auditory system and play important roles in synaptic plasticity, auditory function, and development. However, the specific functions of CB1Rs in distinct neuronal subtypes in the central auditory system remain largely unclear. In the current study, we investigated whether CB1R deficiency in the parvalbumin (PV)-expressing interneurons, a major class of GABAergic interneurons, affects hearing function. We first systematically examined the neuronal localization and distribution of CB1Rs in the mouse central auditory syst

Factors affecting hearing in mice, rats, and other laboratory animals.

The auditory system of rodents and other animals is affected by numerous genetic and environmental variables. These include genes that cause hearing loss, exposure to noise that induces hearing loss, ameliorative effects of an augmented acoustic environment on hearing loss, and effects of background noise on arousal. An understanding of genetic and environmental influences on hearing and auditory behavior is important for those who provide, use, and care for laboratory animals.

Maturation of the auditory system: 2. Transient otoacoustic emission suppression as an index of the medial olivocochlear bundle maturation.

Contralateral suppression of transient otoacoustic emissions in 42 premature babies (84 ears; post-conceptional age [PCA] 30-36 weeks) was compared to that of 39 full-term babies (78 ears; PCA: 37-45 weeks). Eighteen healthy adults and ten young children (5-14 years old) were studied as controls. Risk factors for hearing loss were registered in both preterm and full-term groups. An ILO-92 otoacoustic emission recording system was used to deliver linear clicks to the ear examined and broadband noise to the contralateral ear in an alternating on and off mode. Suppression in full-term babies was

Noradrenergic modulation of brainstem nuclei alters cochlear neural output.

The peripheral auditory sense organ, the cochlea, receives innervation from lateral and medial olivocochlear neurons in the brainstem. These neurons are able to modulate cochlear neural output. Anatomical studies have shown that one of the neurotransmitters which is present in varicosities surrounding the olivocochlear neurons in the brainstem is noradrenaline and previous work on brainstem slices has demonstrated a generally excitatory effect of noradrenaline on medial olivocochlear neurons. In order to assess in vivo the function of the noradrenergic inputs to olivocochlear neurons, we injec

Functional maturation of the medial efferent olivocochlear system in human neonates.

The effect of contralateral acoustic stimulation on transient evoked otoacoustic emissions (TEOAE) was used to investigate the functional maturity of the medial efferent olivocochlear system in full term neonates and a 6-week-old baby. The technique used was to record the TEOAE alternately with and without contralateral noise. The result was a reduction in TEOAE amplitude and latency. This was best illustrated with a difference waveform obtained from the subtraction of the TEOAE waveform recorded with contralateral noise, from the waveform recorded without contralateral noise. Comparative Analysis of Gene Regulatory Network Components in the Auditory Hindbrain of Mice and Chicken.

The neurons in the mammalian and avian auditory hindbrain nuclei share a number of significant morphological and physiological properties for fast, secure and precise neurotransmission, such as giant synapses, voltage-gated K+ channels and fast AMPA receptors. Based on the independent evolution of the middle ear in these two vertebrate lineages, on different embryonic origins of the nuclei and on marked differences on the circuit level, these similarities are assumed to reflect convergent evolution. Independent acquisition of similar phenotypes can be produced by divergent evolution of genetic
