Pain and the Brain: A Systematic Review of Methods, EEG Biomarkers, Limitations, and Future Directions.

Background: Pain is prevalent in almost all populations and may often hinder visual, auditory, tactile, olfactory, and taste perception as it alters brain neural processing. The quantitative methods emerging to define pain and assess its effects on neural functions and perception are important. Identifying pain biomarkers is one of the initial stages in developing such models and interventions. The existing literature has explored chronic and experimentally induced pain, leveraging electroencephalograms (EEGs) to identify biomarkers and employing various qualitative and quantitative approaches

The appearance of phagocytic microglia in the postnatal brain of Niemann Pick type C mice is developmentally regulated and underscores shortfalls in fine odor discrimination.

The loss of NPC1 or NPC2 function results in cholesterol and sphingolipid dyshomeostasis that impairs developmental trajectories, predisposing the postnatal brain to the appearance of pathological signs, including progressive and stereotyped Purkinje cell loss and microgliosis. Despite increasing evidence reporting the activation of pro-inflammatory microglia as a cardinal event of NPC1 disease progression at symptomatic stages both in patients and preclinical models, how microglia cells respond to altered neurodevelopmental dynamics remains not completely understood. To gain an insight on thi

Odor detection threshold, but not odor identification, is impaired in children with autism.

The aim of our study was to examine odor detection thresholds and odor identification in autistic subjects. Thirty-five patients with Asperger's syndrome and high functioning autism (mean age 10.8 ± 3.6 years; 31 boys) were compared with 35 healthy control subjects (mean age 10.4 ± 2.4 years; 28 boys). There were no significant differences between groups with regard to mean age (p = 0.598) and gender proportion (p = 0.324). Olfactory testing used the Sniffin' Sticks test (threshold and identification parts only). Participants with Asperger's syndrome and high functioning autism, in comparison

Effects of anesthetic agents on socially transmitted olfactory memories in mice.

Mice can learn a food preference from odor cues transmitted on the breath of a conspecific, even if the "demonstrator" is anesthetized. To our knowledge there are no studies examining the effect of anesthetizing the "observer" on development of memory for socially transmitted food preferences (STFP). In Experiment 1 we found that 2-4 month-old F2 C57Bl/6x129sv male and female mice demonstrated a STFP after a 5min exposure to an anesthetized demonstrator mouse when tested 24h later. In Experiment 2, observer mice anesthetized with Sagatal (60 mg/kg) prior to the "social interaction" preferentia

De Novo Assembly and Annotation from Parental and F1 Puma Genomes of the Florida Panther Genetic Restoration Program.

In the mid-1990s, the population size of Florida panthers became so small that many individuals manifested traits associated with inbreeding depression (e.g., heart defects, cryptorchidism, high pathogen-parasite load). To mitigate these effects, pumas from Texas were introduced into South Florida to augment genetic variation in Florida panthers. In this study, we report a de novo puma genome assembly and annotation after resequencing 10 individual genomes from partial Florida-Texas-F1 trios. The final genome assembly consisted of ∼2.6 Gb and 20,561 functionally annotated protein-coding genes.

Effects of olfactory enrichment on perioperative neurocognitive disorders in elderly patients undergoing orthopedic surgery: a study protocol of a multi-center randomized controlled trial.

BACKGROUND: Neurocognitive disorders are prevalent among older surgical patients, yet their underlying mechanisms and effective interventions remain largely unknown. Studies in rodents and patients suggest a connection between olfactory impairment and perioperative neurocognitive disorder (PND). Previous work in mice demonstrates that anesthesia and surgery can lead to both olfactory impairment and cognitive deficits. Olfactory enrichment, which is also referred to as olfactory training (i.e., daily exposure to multiple odorants), may prove helpful. For example, olfactory enrichment appears to

Mixture of cis-3-hexenol and trans-2-hexenal attenuates behavioral and stress responses induced by 2,5-dihydro-2,4,5-trimethylthiazoline and electric footshock stress in rats.

Green odor (GO), a mixture of cis-3-hexenol and trans-2-hexenal, attenuates stress responses and anxiety to psychological stressors in rodents; however, it remains unknown whether GO affects behavioral and stress responses to risk-related olfactory stimuli and actual noxious stimuli. The present study investigated the effects of green odor on behavioral and plasma adrenocorticotropic hormone (ACTH) responses to 2,5-dihydro-2,4,5-trimethylthiazoline (TMT), a component of fox feces, and electric footshock (FS) stress. When rats were simultaneously exposed to TMT and GO, they showed decreases in

Gut-brain axis dysfunction mediates neurotoxicity of embryonic p-phenylenediamine exposure in zebrafish.

P-phenylenediamine antioxidants (PPDs) are widely used in the rubber industry and are increasingly recognized as environmental contaminants, raising concerns about their potential risks to wildlife and human health. Our present study investigated the neurotoxicity of embryonic exposure to a sublethal concentration (0.05 mg/L) of three PPDs (CPPD, IPPD and 77PD) in zebrafish. Our results showed that PPDs impaired the fish growth and induced distinct behavioral alterations. Specifically, CPPD exposure induced hyperactivity, aggression and social deficit. IPPD caused hyperactivity and loose shoal

Caenorhabditis elegans as a Diagnostic Aid for Pancreatic Cancer.

OBJECTIVES: Early detection of pancreatic cancer is notoriously difficult. A novel cancer diagnostic method using the ability of nematodes to detect odor of urine samples has been developed (N-NOSE). This method has a high sensitivity and specificity for various cancers; however, it has not yet been verified in pancreatic cancer. We examined the usefulness of this method to aid early diagnosis of pancreatic cancer in a cancer center. METHODS: We collected urine samples and clinical data from patients hospitalized in our division, between July 2017 and February 2019. We excluded patients with

Alzheimer's disease and cerebrovascular biomarkers in relation to odor identification in a naturalistic clinical cohort.

INTRODUCTION: Olfactory deficits, especially in odor identification (OID), have been linked to Alzheimer's disease (AD), likely due to regional proteinopathy and atrophy in the olfactory brain circuit. Their cognitive and biological correlates across the clinical spectrum, particularly in individuals with no evident cognitive impairment, have been underexplored. This examination is relevant because many individuals of this group will not progress to dementia, and olfactory deficits may reflect ongoing pathological processes and could enrich risk stratification. METHODS: We analyzed data from

Brain activation in older adults during odor identification is related to ApoE, t-tau/Aβ1-42, and hippocampal volume.

Despite altered odor identification preceding and predicting Alzheimer's disease (AD) cognitive decline, an inadequate understanding of how AD pathology affects odor memory functions limits its use as a preclinical biomarker. Multivariate linear regression was applied to whole-brain blood-oxygen-level-dependent (BOLD) activations during odor identification task (OID) responses in older adults without dementia (N = 36, 44.4 % ε4 carriers, MAge= 76.61). Apolipoprotein-E ε4 allele status, cerebrospinal fluid levels of total-tau to Amyloid-β1-42, and MRI-derived hippocampal volume measures were us

Neuroimaging biomarkers and impaired olfaction in cognitively normal individuals.

OBJECTIVE: There is a need for inexpensive noninvasive tests to identify older healthy persons at risk for Alzheimer disease (AD) for enrollment in AD prevention trials. Our objective was to examine whether abnormalities in neuroimaging measures of amyloid and neurodegeneration are correlated with odor identification (OI) in the population-based Mayo Clinic Study of Aging. METHODS: Cognitively normal (CN) participants had olfactory function assessed using the Brief Smell Identification Test (B-SIT), underwent magnetic resonance imaging (n = 829) to assess a composite AD signature cortical thi

Olfactory Function is Affected in Patients with Cirrhosis Depending on the Severity of Hepatic Encephalopathy.

INTRODUCTION AND AIM: Olfactory functions are altered to a variable degree by chronic liver disease. Few studies including only small populations of patients emphasized the possibility of hepatic encephalopathy (HE) influencing olfactory nervous tasks. So far, no study has explicitly focused on olfactory function depending on the severity of HE as assessed by objective diagnostic procedures. Thus we performed a study using the "Sniffin' Sticks" test system, critical flicker-fusion frequency (CFF) and clinical West Haven criteria. MATERIAL AND METHODS: 54 cirrhotic patients with liver cirrhosi

Human Leukocyte Antigen similarity decreases partners' and strangers' body odor attractiveness for women not using hormonal contraception.

The Human Leukocyte Antigen (HLA) is a gene complex that encodes important elements of the human immune system. HLA profile is communicated via olfaction and interindividual diversity is assumed to be advantageous for mate choice. Additionally, HLA diversity appears to enhance satisfaction and sexual attraction in existing romantic partnerships. However, whether this effect is transmitted via body odors and whether it results in an attraction towards HLA-dissimilar individuals and/or an avoidance of HLA-similar ones remains unclear. In the present study, we genotyped couples and asked each par

Metabotropic Glutamate Receptor Subtype 7 in the Bed Nucleus of the Stria Terminalis is Essential for Intermale Aggression.

Metabotropic glutamate receptor subtype 7 (mGluR7) is a member of group III mGluRs, which localize to the presynaptic active zones of the mammalian central nervous system. Although histological, genetic, and electrophysiological studies ensure the importance of mGluR7, its roles in behavior and physiology remain largely unknown. Using a resident-intruder paradigm, we found a severe reduction in intermale aggressive behavior in mGluR7 knockout (KO) mice. We also found alterations in other social behaviors in male mGluR7 KO mice, including sexual behavior toward male intruders. Because olfaction
