Carbonic anhydrase (CA) enzyme was purified from rainbow trout brain by Sepharose-4B-L: -tyrosine-sulfanilamide affinity chromatography. The enzyme was obtained with a specific activity of 2,275 EU mg(-1) and a yield of 22.5%. The sample obtained from the affinity column was used for kinetic properties and inhibition studies. Both optimum and stable pH were found as 9.0 in 1 M Tris-SO(4) at 4 degrees C, respectively. To check the purity and subunit molecular weight of enzyme, sodium dodecyl sulfate (SDS) polyacrylamide gel electrophoresis was performed, and MW was found as approximately 29.0 k

Twenty-four hours of N(2) induced anoxia induced global perturbations on protein expression in rainbow trout hypodermal fibroblasts cell line. Anoxia was obtained by depleting the medium of O(2) by flushing with N(2), and protein changes were studied by 2-DE coupled with MS providing quantitative measurements of a large number of proteins in one single study. The anoxic insult changed the level of 33 protein spots: 22 of these were up-regulated compared to the control situation and 11 were down-regulated. Using MS/MS sequencing 19 of the 33 protein spots that changed were identified, correspon

Ammonia and urea are the primary forms of nitrogen excretion in teleost fish. There exists, however, a discrepancy between the sum of ammonia plus urea nitrogen and total nitrogen, indicating that 'Нет данных' nitrogen end products may play an important role in nitrogen metabolism. The current study analysed a wide range of nitrogen end products in both fed and fasted juvenile rainbow trout. Ammonia-N (53-68%) and urea-N (6-10%) were confirmed as the most important forms of nitrogenous waste, but an interesting finding was the considerable excretion of nitrogen as amino acids (4-10%), via the gil

Teleost fish possess discrete blocks of oxidative red muscle (RM) and glycolytic white muscle, whereas tetrapod skeletal muscles are mixed oxidative/glycolytic. It has been suggested that the anatomy of RM in teleost fish could lead to higher intramuscular O2 partial pressures (PO2) than in mammalian skeletal muscles. This study provides the first direct experimental support for this suggestion by using novel optical fibre sensors to discover a mean (+/- S.E.M., N=6) normoxic steady-state red muscle PO2 (PrmO2) of 61+/-10 mmHg (1 mmHg=133.3 Pa) in free-swimming rainbow trout Oncorhynchus mykis

In Teleost fish, development, growth, and reproduction are influenced by the daily and seasonal variations of photoperiod and temperature. Early in vivo studies indicated the pineal gland mediates the effects of these external factors, most probably through the rhythmic production of melatonin. The present investigation was aimed at determining whether melatonin acts directly on the pituitary to control GH and prolactin (PRL) secretion in rainbow trout. We show that 2-[125I]-iodomelatonin, a melatonin analog, binds selectively to membrane preparations and tissue sections from trout pituitaries

The interactive effects of ultraviolet (UV) and fire-retardant chemicals were evaluated by exposing rainbow trout (Oncorhyncus mykiss) juveniles and tadpoles of southern leopard frogs (Rana sphenocephala) to six fire-retardant formulations with and without sodium ferrocyanide (yellow prussiate of soda [YPS]) and to YPS alone under three simulated UV light treatments. Yellow prussiate of soda is used as a corrosion inhibitor in some of the fire-retardant chemical formulations. The underwater UV intensities measured were about 2 to 10% of surface irradiance measured in various aquatic habitats a

The binding of antibiotics (gentamicin, oleandomycin and chloramphenicol) to vitelline and fertilization envelopes and their extracts was investigated by immunohistochemical and immunocytochemical techniques and immunoblot analysis using mature and artificially activated eggs of the fish Oncorhynchus masou. Binding of antibiotics was detected in the vitelline and fertilization envelope outermost layers, the fertilization envelope inner surface and cortical alveolus exudates, with differences in immunoreactive intensity and deposition. The fertilization envelope outermost layer had the capacity

Pineal photoreceptor cells produce the neurohormone melatonin, a major "Zeitgeber" of the organism. This compound is involved in the control of development, growth, sexual maturation, and seasonal reproductive cycles. The present study reports that the photosensitive pineal organ and the retina of the trout express a 3.5-kb mRNA corresponding to the estradiol-17 beta receptor. The effects of estradiol-17 beta on melatonin production by cultured pineal photoreceptor cells were also studied. Under a light/dark (LD:12/12) cycle, these cells maintained a rhythmic secretion of melatonin, with highe

Studies of N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine-quinone (6PPDQ) toxicity have identified sensitive and resistant species within the subfamily Salmoninae, but thus far fewer than 10 species, all in two genera (Oncorhynchus and Salvelinus), have shown sensitivity in experiments. The Arctic grayling (Thymallus arcticus, Salmonidae, Thymallinae) was extirpated from its range in the Great Lakes region of the United States, but because of the importance of this species to the Native American tribes of Michigan, a reintroduction effort is underway. Because high 6PPDQ concentrations have

The study of flow and transformation of energy and nutrients via mathematical modelling provides an in silico tool approach for designing scientific experiments, improving precision in aquaculture production and reducing the need for experimental animals. The proposed nutritional bioenergetics model is based on the dynamic energy budget (DEB) theory, a mechanistic framework to study individual metabolism. The model is an extension of the typical DEB models in that it includes a digestion module where the protein and non-protein food components contribute to assimilation via the concept of a sy

Metabolic scope represents the aerobic energy budget available to an organism to perform non-maintenance activities (e.g., escape a predator, recover from a fisheries interaction, compete for a mate). Conflicting energetic requirements can give rise to ecologically relevant metabolic trade-offs when energy budgeting is constrained. The objective of this study was to investigate how aerobic energy is utilized when individual sockeye salmon (Oncorhynchus nerka) are exposed to multiple acute stressors. To indirectly assess metabolic changes in free-swimming individuals, salmon were implanted with

The aim of this study was to determine the kynurenine (KYN) to tryptophan (TRP) ratio (KTR) in fish tissue to assess its usefulness as a biomarker of acute stress. Laboratory held rainbow trout (Oncorhynchus mykiss) were subjected to an acute stressor and KYN, TRP and cortisol were measured in liver and brain tissues at 4- and 48-h post-stress. The analytical method used to determine our analytes was based on lyophilization, and liquid-solid extraction followed by isotope dilution high-performance liquid chromatography positive ion electrospray tandem mass spectrometry. The [KYN]/[TRP] ratio (

Despite the strong demand for orally-delivered fish vaccines and the deficient response of those currently available in the market, little is known about how teleost B cells differentiate to antibody secreting cells (ASCs) in response to antigens delivered to the intestinal mucosa. To fill this gap, in the current study, we have studied the dynamics of B cell differentiation in spleen and kidney of rainbow trout (Oncorhynchus mykiss) anally immunized with antigens catalogued in mammals as thymus dependent (TD) or thymus-independent (TI). Our results show that, in the absence of additional adju

The nematodes of the genus Anisakis are among the most relevant parasitic hazards in fishery products since they are responsible for human infection and allergy cases. In a food safety and epidemiological perspective, several marine hosts from different locations around Japan were examined to characterize the parasitism of Anisakis larvae. Chum salmon (Oncorhynchus keta) and Alaska pollock (Gadus chalcogrammus) showed the highest overall prevalence (100%), followed by blue mackerel (Scomber australasicus) (97.5%), Pacific cod (Gadus macrocephalus) (80%), chub mackerel (Scomber japonicus) (60.1

Triclosan (TRC), chloroxylenol (PCMX) and methylisothiazolinone (MIT) have been commonly used as an antimicrobial in soaps while borax (BRX) is used in household cleaning. After using these chemicals, they are washed down drains and getting into the aquatic ecosystem in which they may affect aquatic living organisms. In the present study, the chronic effects of TRC, PCMX, MIT and BRX on genotoxicity, gene expression and histopathology of rainbow trout (Oncorhynchus mykiss) were evaluated for 40 days under semi static condition. The comet assay results indicated that MIT, TRC and PCMX caused si

The globiferous pedicellariae of the venomous sea urchin Toxopneustes pileolus contains several biologically active proteins. We have cloned the cDNA of one of the toxin components, SUL-I, which is a rhamnose-binding lectin (RBL) that acts as a mitogen through binding to carbohydrate chains on target cells. Recombinant SUL-I (rSUL-I) was produced in Escherichia coli cells, and its carbohydrate-binding specificity was examined with the glycoconjugate microarray analysis, which suggested that potential target carbohydrate structures are galactose-terminated N-glycans. rSUL-I exhibited mitogenic

Melatonin contributes to synchronizing major biological and behavioral functions with cyclic changes in the environment. Arylalkylamine N-acetyltransferase (AANAT) is responsible for a daily rhythm in melatonin secretion. Teleost possess two enzyme forms, AANAT1 and AANAT2, preferentially expressed in the retina and the pineal gland, respectively. The concomitant action of light and temperature shapes the daily and seasonal changes in melatonin secretion: the former controls duration while the latter modulates amplitude. Investigating the respective roles of light and temperature is particular

Three salmonid fish species, Oncorhynchus mykiss, Salmo salar and Salmo trutta, were infected experimentally with the parasitic nematode Anisakis simplex (A. simplex) and the difference between in vivo behaviour of the nematode in the three fish species was investigated. Infection success rate differed between species. S. salar (Baltic salmon) showed the highest number of successfully established nematodes, whereas S. trutta (brown trout) and O. mykiss (rainbow trout) had a higher natural resistance. Microhabitat selection of nematodes differed according to fish species. In brown trout, A. sim

The purpose of this study was to determine adequate dose ranges and to test for side effects associated with chronic treatment of fish with indomethacin. Rainbow trout Oncorhynchus mykiss were orally treated with indomethacin at various nominal concentrations: a negative control (0 mg/kg) and low (5 mg/kg), medium (10 mg/kg), and high doses (15 mg/kg) daily for 30 d. A dose-associated mortality was observed, as cumulative mortality was 3, 13, and 33%, respectively, in the three indomethacin dose groups. No lesions were observed grossly or with histopathology in the control and low-dose treatme

The Pacific bonito, Sarda chiliensis, is anatomically intermediate between mackerel and tuna. The specialisations exhibited by tuna are present in the bonito, but to a lesser degree. Slow-twitch muscle strain and activity patterns were determined during steady swimming (tailbeat frequency 1.2-3.2 Hz) at four locations on the body of Sarda chiliensis using sonomicrometry and electromyography. Both strain and the phase of electromygraphic activity were independent of tailbeat frequency. The strain of superficial slow-twitch muscle increased from +/-3.1 % l(0) at 0.35FL to +/-5.8 % l(0) at 0.65FL
