Immunohistochemical demonstration of enamel proteins in odontogenic tumors.

Immunohistochemical localization of two enamel proteins, amelogenin and enamelin, in comparison with that of keratin, was determined in odontogenic tumors and the allied lesions in order to verify functional differentiation of the tumor cells as ameloblasts. Amelogenin and enamelin were demonstrated in small mineralized foci and in the tumor cells surrounding them in adenomatoid odontogenic tumor (AOT), calcifying epithelial odontogenic tumor (CEOT), and calcifying odontogenic cyst (COC). Hyaline droplets in AOT showed positive staining for both enamel proteins. These mineralized and hyaline m

Tissue engineering of complex tooth structures on biodegradable polymer scaffolds.

Tooth loss due to periodontal disease, dental caries, trauma, or a variety of genetic disorders continues to affect most adults adversely at some time in their lives. A biological tooth substitute that could replace lost teeth would provide a vital alternative to currently available clinical treatments. To pursue this goal, we dissociated porcine third molar tooth buds into single-cell suspensions and seeded them onto biodegradable polymers. After growing in rat hosts for 20 to 30 weeks, recognizable tooth structures formed that contained dentin, odontoblasts, a well-defined pulp chamber, puta

Craniofacial tissues including tooth buds in fetal hypohidrotic ectodermal dysplasia.

OBJECTIVE: Hypohidrotic ectodermal dysplasia (HED) comprises defects in hair, teeth, and sweat glands. Disturbances in other ectodermal tissues have been associated with the condition. Our objective was to examine ectodermal craniofacial structures histologically in a fetus with HED and to compare the findings to similar structures in normal control fetuses. MATERIALS AND METHODS: A male fetus diagnosed with HED was therapeutically aborted in the 15th week of gestation. One male and two female healthy fetuses were used as normal controls. All fetuses were examined with parental consent, and h

Comparing the Biofilm Removal Capacity of NaOCl, Povidone-Iodine, Chlorhexidine, Curcumin, and Triphala as Endodontic Irrigants.

BACKGROUND AND AIM: A sessile multicellular organism that is immersed in a self-produced matrix of extracellular polymeric substances and has its cells firmly attached to a surface is referred to as a microbial biofilm. When it comes to pulp and periradicular pathosis, biofilms are crucial. To reduce the number of microorganisms in the root canal and assist in treating periapical pathosis, endodontic therapy must include decontamination of the system of tooth root canals through biomechanical preparation and irrigation of the root canal. This study compares sodium hypochlorite (NaOCl), povidon

Regeneration of Bone, Cartilage, and Tooth Following Lower Jaw Amputation in Newts.

Background/Objectives: In humans, diseases such as oral cancer may require surgical amputation of the jaw. This severe disruption causes impairments in eating, swallowing, and speech, leading to a significant decline in quality of life. In contrast, newts, a group of urodele amphibians, can regenerate their jaws even in adulthood. This study explored how adult newts reconstruct lower jaws after substantial loss and clarified how this process contributes to rapid functional recovery when feeding becomes impossible. Methods: Adult Japanese fire-bellied newts (Cynops pyrrhogaster) underwent surgi

Adverse effects of splinting of luxated primary teeth: A systematic review.

Traumatic dental injuries of primary teeth are considered as dental emergencies and public health problems because of the higher global prevalence. Dental literature in regard to optimal treatment options for periodontal injuries of primary teeth (luxation injuries) is quite limited and perplexing in terms of treatment outcomes/prognosis due to several relevant factors. To date, there has been no systematic review published in context of exploring and analyzing the unfavorable outcomes/adverse effects of splinting the luxated primary teeth and its effect on permanent successors teeth. Hence, t

Defining the Stem Cell Niche in Developing Pericoronal Dental Follicles.

OBJECTIVE: The pericoronal dental follicle is a connective tissue structure encased in epithelium present around the developing tooth germ before erupting and emerging in the oral cavity. Stem cells from pericoronal follicles can be obtained and cultured under suitable conditions in vitro. This study builds upon our previous research, with the primary objective of identifying stem cell subpopulations in the oral cavity based on marker expression and differentiation potential. MATERIALS AND METHODS: Dental follicle stem cells (DFSCs) were obtained and cultured under standard cell culturing con

Mosaicism for GJB1 mutation causes milder Charcot-Marie-Tooth X1 phenotype in a heterozygous man than in a manifesting heterozygous woman.

Charcot-Marie-Tooth (CMT) disease is a heterogeneous disorder of the peripheral nervous system that collectively affects approximately 1 in 2,500 individuals, thus making it the most common inherited neurologic disorder. X-linked inheritance may account for 10-20 % of CMT neuropathy. We report a Czech family with a 30-year-old woman affected by CMT since the age of 10 years, originally as an isolated case. Nerve conduction study (NCS) showed demyelinating neuropathy, and DNA testing revealed a novel heterozygous gap junction beta-1 protein (GJB1) mutation c.784_786delTA. The same mutation, but

Dental abnormalities in Schimke immuno-osseous dysplasia.

Described for the first time in 1971, Schimke immuno-osseous dysplasia (SIOD) is an autosomal-recessive multisystem disorder that is caused by bi-allelic mutations of SMARCAL1, which encodes a DNA annealing helicase. To define better the dental anomalies of SIOD, we reviewed the records from SIOD patients with identified bi-allelic SMARCAL1 mutations, and we found that 66.0% had microdontia, hypodontia, or malformed deciduous and permanent molars. Immunohistochemical analyses showed expression of SMARCAL1 in all developing teeth, raising the possibility that the malformations are cell-autonomo

Comparative Evaluation of Rotary and Hand File Systems for Optimal Obturation in Primary Molar Root Canal Treatment: A Pediatric Perspective.

Background/objectives Rotary instrumentation has gained significant importance in the endodontic preparation of primary teeth. Shorter appointment durations improve pediatric patient compliance while maintaining the original root canal anatomy is essential to preserve the integrity of the developing permanent tooth germ. Rotary files facilitate this process by reducing treatment time. This study aimed to compare the obturation quality achieved using hand files versus a rotary file system in the root canal treatment of primary molars. Materials and methods A total of 316 patients aged 4-10 year

Inherited neuropathies.

Charcot-Marie-Tooth neuropathy (CMT) type 1 is a genetically heterogeneous group of chronic demyelinating polyneuropathies with loci mapping to chromosome 17 (CMT1A), chromosome 1 (CMT1B), the X chromosome (CMTX), and to another unknown autosome (CMT1C). CMT1A is most often associated with a tandem 1.5-Mb duplication in chromosome 17p11.2-12, or in rare patients may result from a point mutation in the peripheral myelin protein-22 (PMP22) gene. CMT1B is associated with point mutations in the myelin protein zero (P0) gene. The molecular defect in CMT1C is unknown. CMTX is associated with mutatio

Hereditary spastic paraplegia is a novel phenotype for germline de novo ATP1A1 mutation.

Dominant mutations in ATP1A1, encoding the alpha-1 isoform of the Na+ /K+ -ATPase, have been recently reported to cause an axonal to intermediate type of Charcot-Marie-Tooth disease (ie, CMT2DD) and a syndrome with hypomagnesemia, intractable seizures and severe intellectual disability. Here, we describe the first case of hereditary spastic paraplegia (HSP) caused by a novel de novo (p.L337P) variant in ATP1A1. We provide evidence for the causative role of this variant with functional and homology modeling studies. This finding expands the phenotypic spectrum of the ATP1A1-related disorders, a
