Plant sex-determining genes and the genetics of the evolution towards dioecy.

Thanks to significant advances in genomics and bioinformatics, research on plant sex-determining genes has made remarkable progress over the past decade. Since the discovery of the OGI-MeGI sex-determination system in persimmons in 2014, candidate sex-determining genes have been identified in a dozen flowering plant species. In this review, we examine these newly discovered genes and explore what they reveal about the genetic basis of the evolution of dioecy. While these genes are diverse, many belong to key developmental pathways previously described in other species. Two primary evolutionary

Temporal patterns in the mass flowering of Firmiana simplex: A new case of duodichogamy.

PREMISE: Given that insect-pollinated trees are generally characterized by mass flowering, hermaphrodites may benefit from abundant pollinators but suffer the cost of geitonogamy. To reduce such selfing caused by intraplant pollination, duodichogamy (a temporal sexual system involving three alternating sexual phases within an individual) has been suggested to be a common strategy. However, the timing of the female and male phases within individuals throughout floral phenology remains undocumented in any duodichogamous species. METHODS: Over 2 years, we quantified flower sexes at the panicle a

Down-regulation of the rice HRS1 HOMOLOG3 transcriptional repressor gene due to N deficiency directly co-activates ammonium and phosphate transporter genes.

Rice HRS1 HOMOLOG3 (OsHHO3) acts as a transcriptional repressor of AMMONIUM TRANSPORTER1 (OsAMT1) genes in rice; thus, reduced OsHHO3 expression in nitrogen (N)-deficient environments promotes ammonium uptake. In this study, we show that OsHHO3 also functions as a repressor of a specific subset of phosphate (Pi) transporter (PT) genes involved in the uptake and root-to-shoot translocation of Pi, including OsPT2, OsPT4, and OsPHO1;1. Disruption of OsHHO3 increased Pi uptake and Pi contents in shoots and roots, while overexpression of OsHHO3 caused the opposite effects. Furthermore, phosphorus (

Unraveling the Protective Role of Sulfide Signaling in Plants against Climate-Induced Abiotic Stress.

Hydrogen sulfide (H2S) is now widely recognized as a signaling molecule, playing a crucial role as a regulator of essential biological processes. This review outlines current knowledge of the endogenous H2S sources in different cellular compartments and highlights its principal mechanisms of action. In the current context of climate change, the involvement of H2S in plant stress responses has gained increasing attention. H₂S has been shown to alleviate the adverse effects of various abiotic stresses by regulating specific physiological and molecular responses, enabling stress tolerance and cro

Amyloplasts are necessary for full gravitropism in thallus of Marchantia polymorpha.

Gravitropism is a plant response to gravity that directs organ growth and development, playing a key role in the adaptation of land plants. While its molecular basis has been extensively studied in flowering plants, much less is known about this process in other plant lineages. Here, we investigated the gravitropic response of the liverwort Marchantia polymorpha, a model for early land plant evolution. In darkness, the thallus tips extended upward, forming several straight, narrow structures whose growth direction was consistently opposite to gravity and disrupted by clinostat treatment. These

The potential of silicon in crop protection against phloem feeding and chewing insect pests: a review.

As future crop production is threatened by the increasing incidence of insect pests caused by changing climate, it is crucial to focus on searching for alternative methods of insect pest management. This review summarizes the current knowledge on the effect of silicon (Si) on the incidence, growth, development, longevity, survival, feeding efficiency, and reproduction parameters of phloem sap-feeding and chewing insects of plants. While Si may negatively impact the biological parameters of both insect guilds, the effect is generally more pronounced in chewing insects and often varies under dif

Impacts of ramet density and herbivory on floral volatile emissions and seed production in Solidago altissima.

PREMISE: Plants produce an array of floral olfactory and visual cues to attract pollinators, including volatile organic compounds (VOC), which mediate plant-pollinator interactions and may be influenced by herbivory and neighboring plants. Consequently, these factors may affect plant fitness by disrupting pollination. However, most evidence comes from controlled experiments, limiting our understanding of how VOCs function in natural populations. This study investigated how herbivory and conspecific ramet density influence floral VOC profile, pollination, and seed production in a naturally occu

Role of transcription factor complex OsbHLH156-OsIRO2 in regulating manganese, copper, and zinc transporters in rice.

Iron (Fe), manganese (Mn), copper (Cu), and zinc (Zn) are essential micronutrients that are necessary for plant growth and development, but can be toxic at supra-optimal levels. Plants have evolved a complex homeostasis network that includes uptake, transport, and storage of these metals. It was shown that the transcription factor (TF) complex OsbHLH156-OsIRO2 is activated under Fe deficient conditions and acts as a central regulator on Strategy II Fe acquisition. In this study, the role of the TF complex on Mn, Cu, and Zn uptake was evaluated. While Fe deficiency led to significant increases

The effect of local habitat and spatial connectivity on urban seed predation.

PREMISE: During the last centuries, the area covered by urban landscapes is increasing all over the world. Urbanization can change local habitats and decrease connectivity among these habitats, with important consequences for species interactions. While several studies have found a major imprint of urbanization on plant-insect interactions, the effects of urbanization on seed predation remain largely unexplored. METHODS: We investigated the relative impact of sunlight exposure, leaf litter, and spatial connectivity on predation by moth and weevil larvae on acorns of the pedunculate oak across

Low sucrose availability reduces basal spikelet fertility by inducing abscisic acid and jasmonic acid synthesis in wheat.

Within a spike of wheat, the central spikelets usually generate three to four fertile florets, while the basal spikelets generate zero to one fertile floret. The physiological and transcriptional mechanism behind the difference in fertility between the basal and central spikelets is unclear. This study reports a high temporal resolution investigation of transcriptomes, number and morphology of floret primordia, and physiological traits. The W6.5-W7.5 stage was regarded as the boundary to distinguish between fertile and abortive floret primordia; those floret primordia reaching the W6.5-W7.5 st

Effects of anther-stigma position on cross-pollination efficiency in a hermaphroditic plant.

PREMISE: Evolution of cross-pollination efficiency depends on the genetic variation of flower traits, the pollen vector, and flower trait matching between pollen donors and recipients. Trait matching has been almost unexplored among nonheterostylous species, and we examined whether the match of anther length in pollen donors and stigma length in pollen recipients influences the efficiency of cross-pollination. To explore potential constraints for evolutionary response, we also quantified genetic variation and covariation among sepal length, petal length and width, stamen length, style length,

Widespread male sterility and trioecy in androdioecious Mercurialis annua: Its distribution, genetic basis, and estimates of morph-specific fitness components.

PREMISE: Angiosperms range in sexual system from hermaphroditism through gynodioecy and androdioecy to dioecy. Trioecy, where females and males coexist with hermaphrodites, is rare. Recently, trioecy was documented in hexaploid populations of the wind-pollinated herb Mercurialis annua in Spain. METHODS: We surveyed the frequency of males, hermaphrodites, and females in M. annua across its distribution in the Iberian Peninsula, tracked sex-ratio variation in several populations over consecutive generations, and assessed evidence for pollen limitation. In a common garden, we estimated male, fem

Why are triploid quaking aspen (Populus tremuloides) common?

PREMISE: Quaking aspen is a clonal tree species that has mixed ploidy, often with high relative abundance of both diploids and triploids but no haploids or tetraploids. Triploids typically have low fertility, leaving their occurrence apparently unlikely from an evolutionary perspective, unless they provide a "triploid bridge" to generating higher-fitness tetraploids-which are not observed in this species. This study focused on how triploidy can be maintained in quaking aspen. METHODS: A computational model was used to simulate gamete production, sexual reproduction, asexual reproduction, pare

CRISPR/Cas9-mediated multiplex gene editing of gamma and omega gliadins: paving the way for gliadin-free wheat.

Wheat is a staple cereal in the human diet. Despite its significance, an increasing percentage of the population suffers adverse reactions to wheat, which are triggered by wheat gluten, particularly the gliadin fractions. In this study, we employed CRISPR/Cas [clustered regularly interspaced palindromic repeats (CRISPR)/CRISPR-associated protein] multiplexing to introduce targeted mutations into γ- and ω-gliadin genes of wheat, to produce lines deficient in one or both immunogenic gliadin fractions simultaneously. For this study, eight single guide RNAs (sgRNAs) were designed and combined into

A cornucopia of diversity-Ranunculales as a model lineage.

The Ranunculales are a hyperdiverse lineage in many aspects of their phenotype, including growth habit, floral and leaf morphology, reproductive mode, and specialized metabolism. Many Ranunculales species, such as opium poppy and goldenseal, have a high medicinal value. In addition, the order includes a large number of commercially important ornamental plants, such as columbines and larkspurs. The phylogenetic position of the order with respect to monocots and core eudicots and the diversity within this lineage make the Ranunculales an excellent group for studying evolutionary processes by com

Down-regulation of tomato STEROL GLYCOSYLTRANSFERASE 1 perturbs plant development and facilitates viroid infection.

Potato spindle tuber viroid (PSTVd) is a plant pathogen naturally infecting economically important crops such as tomato (Solanum lycopersicum). Here, we aimed to engineer tomato plants highly resistant to PSTVd and developed several S. lycopersicum lines expressing an artificial microRNA (amiRNA) against PSTVd (amiR-PSTVd). Infectivity assays revealed that amiR-PSTVd-expressing lines were not resistant but instead hypersusceptible to the viroid. A combination of phenotypic, molecular, and metabolic analyses of amiRNA-expressing lines non-inoculated with the viroid revealed that amiR-PSTVd was

Homeostasis of Arabidopsis R protein RPS2 is negatively regulated by the RING-type E3 ligase MUSE16.

The homeostasis of resistance (R) proteins in plants must be tightly regulated to ensure precise activation of plant immune responses upon pathogen infection, while avoiding autoimmunity and growth defects when plants are uninfected. It is known that CPR1, an F-box protein in the SCF E3 complex, functions as a negative regulator of plant immunity through targeting the resistance (R) proteins SNC1 and RPS2 for degradation. However, whether these R proteins are also targeted by other E3 ligases is unclear. Here, we isolated Arabidopsis MUSE16, which encodes a RING-type E3 ligase, from a forward

Fifty years of sperm cell isolations: from structural to omic studies.

The fusion of male and female gametes is a fundamental process in the perpetuation and diversification of species. During the last 50 years, significant efforts have been made to isolate and characterize sperm cells from flowering plants, and to identify how these cells interact with female gametes to achieve double fertilization. The first techniques and analytical approaches not only provided structural and biochemical characterizations of plant sperm cells but also paved the way for in vitro fertilization studies. Further technological advances then led to unique insights into sperm biology

Hierarchical contribution of Argonaute proteins to antiviral protection.

Antiviral RNAi is the main protective measure employed by plants in the fight against viruses. The main steps of this process have been clarified in recent years, primarily relying on the extensive genetic resources of Arabidopsis thaliana. Our knowledge of viral diseases of crops, however, is still limited, mainly due to the fact that A. thaliana is a non-host for many agriculturally important viruses. In contrast, Nicotiana benthamiana has an unparalleled susceptibility to viruses and, since it belongs to the Solanaceae family, it is considered an adequate system for modeling infectious dise

Overexpression of maize ZmLOX6 in Arabidopsis thaliana enhances damage-induced pentyl leaf volatile emissions that affect plant growth and interaction with aphids.

Pentyl leafy volatiles (PLV) are C5 volatiles produced from polyunsaturated fatty acids by plant 13-lipoxygenases (13-LOX) in concert with other lipid metabolizing enzymes. Unlike related C6 volatiles (GLV, green leafy volatiles), little is known about the biosynthesis and physiological function of PLV in plants. Zea mays LOX6 (ZmLOX6) is an unusual plant LOX that lacks lipid oxygenation activity but acts as a hydroperoxide lyase hypothesized to be specifically involved in PLV synthesis. We overexpressed ZmLOX6 in Arabidopsis thaliana and established that it indeed produces PLVs. Overexpressio
