Noradrenergic modulation of brainstem nuclei alters cochlear neural output.

The peripheral auditory sense organ, the cochlea, receives innervation from lateral and medial olivocochlear neurons in the brainstem. These neurons are able to modulate cochlear neural output. Anatomical studies have shown that one of the neurotransmitters which is present in varicosities surrounding the olivocochlear neurons in the brainstem is noradrenaline and previous work on brainstem slices has demonstrated a generally excitatory effect of noradrenaline on medial olivocochlear neurons. In order to assess in vivo the function of the noradrenergic inputs to olivocochlear neurons, we injec

Functional maturation of the medial efferent olivocochlear system in human neonates.

The effect of contralateral acoustic stimulation on transient evoked otoacoustic emissions (TEOAE) was used to investigate the functional maturity of the medial efferent olivocochlear system in full term neonates and a 6-week-old baby. The technique used was to record the TEOAE alternately with and without contralateral noise. The result was a reduction in TEOAE amplitude and latency. This was best illustrated with a difference waveform obtained from the subtraction of the TEOAE waveform recorded with contralateral noise, from the waveform recorded without contralateral noise. All the full ter

Observation in inner ear of tree shrew using scanning electron microscope and the Atoh1 distribution in cochlea.

This study aimed to observe the ultrastructure on the surface of the inner ear of a normal tree shrew using scanning electron microscope (SEM). The specimens of cochlea, macula utriculi, macula sacculi, and crista ampullaris of the normal adult tree shrew were collected and observed by SEM. We used immunofluorescence for cochlear protein Atoh1 staining. We observed that cochlea of the tree shrew is centered on the cochlear axis, circling about 3.5 times from bottom to top of the cochlea. The organ of Corti is located between medial and lateral grooves, including inner and outer hair cells as w

Improved electrically evoked auditory steady-state response thresholds in humans.

Electrically evoked auditory steady-state responses (EASSRs) are EEG potentials in response to periodic electrical stimuli presented through a cochlear implant. For low-rate pulse trains in the 40-Hz range, electrophysiological thresholds derived from response amplitude growth functions correlate well with behavioral T levels at these rates. The aims of this study were: (1) to improve the correlation between electrophysiological thresholds and behavioral T levels at 900 pps by using amplitude-modulated (AM) and pulse-width-modulated (PWM) high-rate pulse trains, (2) to develop and evaluate the

Investigating the mechanisms of ageing and neuroinflammation in age-related hearing loss: A proteomic analysis.

BACKGROUND: Age-related hearing loss (ARHL) is a prevalent neurodegenerative condition commonly linked to aging and chronic inflammation. However, there is currently a lack of substantial proteomic evidence elucidating the underlying mechanisms within the brain. METHODS: The study employed proteomic techniques to identify proteins as biomarkers for ARHL and to investigate and predict their underlying pathogenic mechanisms and potential intervention targets. RESULTS: In studying the impact of aging on ARHL, we found significant expression of 7 hearing-related proteins. including Mbp, Mag, Plp

Objective Detection of Tinnitus Based on Electrophysiology.

Tinnitus, a common disease in the clinic, is associated with persistent pain and high costs to society. Several aspects of tinnitus, such as the pathophysiology mechanism, effective treatment, objective detection, etc., have not been elucidated. Any change in the auditory pathway can lead to tinnitus. At present, there is no clear and unified mechanism to explain tinnitus, and the hypotheses regarding its mechanism include auditory plasticity theory, cortical reorganization theory, dorsal cochlear nucleus hypothesis, etc. Current theories on the mechanism of tinnitus mainly focus on the abnorm

Neuronal differentiation of dental pulp stem cells from human permanent and deciduous teeth following coculture with rat auditory brainstem slices.

Sensorineural hearing loss is a common disability found worldwide which is associated with a degeneration of spiral ganglion neurons (SGN). It is a challenge to restore SGN due to the permanent degeneration and viability of SGN is requisite for patients to receive an advantage from hearing aid devices. Human dental pulp stem cells (DPSC) and stem cells from human exfoliated deciduous teeth (SHED) are self-renewing stem cells that originate from the neural crest during development. These stem cells have a high potential for neuronal differentiation. This is primarily due to their multilineage d

Changes in auditory thalamus neural firing patterns after acoustic trauma in rats.

Tinnitus is an abnormal phantom perception associated with cochlear trauma, and is thought to cause changes in the rates and patterns of firing neurons in the central auditory pathway. Recent studies have suggested a key role for the auditory thalamus, the medial geniculate nucleus (MGN), in the generation of tinnitus as it may serve a gating function for information en route to cortex. Dysfunctional gating would lead to abnormal activity reaching cortex and hence inappropriate perception, tinnitus, would occur. In this study we compared spontaneous MGN firing rates and burst firing parameters

The anti-inflammatory selective melanocortin receptor subtype 4 agonist, RO27-3225, fails to prevent acoustic trauma-induced tinnitus in rats.

In preliminary studies we have observed a massive microglial activation in the cochlear nucleus following acoustic trauma-induced tinnitus in rats, which suggests that inflammatory responses within the central auditory system may be involved in the development and maintenance of tinnitus. Recently, the anti-inflammatory properties of melanocortins (MCs), have gained increasing interest in pharmacology due to their promising therapeutic potential in the treatment of inflammatory-mediated diseases. Among the five subtypes of the MC receptor, MC3 and MC4 receptors are the predominant brain recept

The relation between perception and brain activity in gaze-evoked tinnitus.

Tinnitus is a phantom sound percept that can be severely disabling. Its pathophysiology is poorly understood, partly due to the inability to objectively measure neural correlates of tinnitus. Gaze-evoked tinnitus (GET) is a rare form of tinnitus that may arise after vestibular schwannoma removal. Subjects typically describe tinnitus in the deaf ear on the side of the surgery that can be modulated by peripheral eye gaze. This phenomenon offers a unique opportunity to study the relation between tinnitus and brain activity. We used functional magnetic resonance imaging in humans to show that in n

[Neurofibromatosis type 2 and auditory brainstem implantation].

BACKGROUND: Neurofibromatosis type 2 (NF2) is a rare and severe autosomal dominant disorder caused by mutations in a tumour suppressor gene. This article reviews NF2 and its treatment with auditory brainstem implantation. MATERIAL AND METHODS: The review is based on the authors' experience with the disease and literature identified through a non-systematic search of PubMed. RESULTS: NF2 is caused by loss-of-normal function of the tumour suppressor protein merlin. Merlin normally suppresses cell growth and proliferation. The clinical picture is dominated by neurological symptoms, caused by mu

Effects of sex, gonadal hormones, and augmented acoustic environments on sensorineural hearing loss and the central auditory system: insights from research on C57BL/6J mice.

Mice of the C57BL/6J (B6) inbred strain exhibit genetic progressive sensorineural hearing loss and have been widely used as a model of adult-onset hearing loss and presbycusis. Males and females exhibit similar degrees of hearing loss until about 3 months of age, after which, the loss accelerates in females. This paper reviews research on how the B6 auditory system is affected by sex, gonadectomy (i.e., a reduction of gonadal hormone levels), and nightly exposure to moderately intense augmented acoustic environments (AAEs) - a low-frequency noise band (LAAE) or high-frequency band (HAAE). Expression of c-fos in auditory and non-auditory brain regions of the gerbil after manipulations that induce tinnitus.

Subjective tinnitus is a phantom sound sensation that does not result from acoustic stimulation and is audible to the affected subject only. Tinnitus-like sensations in animals can be evoked by procedures that also cause tinnitus in humans. In gerbils, we investigated brain activation after systemic application of sodium salicylate or exposure to loud noise, both known to be reliable tinnitus-inductors. Brains were screened for neurons containing the c-fos protein. After salicylate injections, auditory cortex was the only auditory area with consistently increased numbers of immunoreactive neur

The effect of acoustic overexposure on the tonotopic organization of the nucleus magnocellularis.

We assessed the effect a sound-induced cochlear lesion had on the tonotopic organization of the nucleus magnocellularis (NM) immediately after acoustic overexposure and following a twelve day recovery period. The acoustic overexposure was a 0.9 kHz tone at 120 dB sound pressure level (SPL) for 48 h. Initially after the acoustic overexposure, the tonotopic organization of the NM was statistically different from that of age-matched controls. Specifically, it appeared that the center frequencies of units in the frequency region of the NM associated with the acoustic overexposure had higher center

[Correlation between electrically-induced stapedius reflex and discomfort threshold in cochlear implant patients].

Electrically elicited stapedius reflex thresholds are an objective criteria for the fitting of speech processors in very young children. Reflex thresholds generally fit well within the upper third of a subject's individual dynamic range, the difference between electrical threshold of hearing, (T) and maximum comfort (C) levels and can be used to predict ultimate behavioral maximum comfort levels. In acoustically elicited stapedius reflexes a saturation of impedance amplitude can be registered at approximately 110 dB, which is 90% of the dynamic range. Assuming a similar relationship for the el

Enhancement of Endocannabinoid-dependent Depolarization-induced Suppression of Excitation in Glycinergic Neurons by Prolonged Exposure to High Doses of Salicylate.

The Dorsal Cochlear Nucleus (DCN) is a region which has been traditionally linked to the genesis of tinnitus, the constant perception of a phantom sound. Sodium salicylate, a COX-2 inhibitor, can induce tinnitus in high doses. Hyperactivity of DCN neurons is observed in several animal models of tinnitus, including salicylate-induced tinnitus. The DCN presents several forms of endocannabinoid (EC)-dependent synaptic plasticity and COX-2 can also participate in the oxidative degradation of ECs. We recently demonstrated that short-term perfusion of sodium salicylate and other inhibitors of both o

Cochlear Implant Rate Pitch and Melody Perception as a Function of Place and Number of Electrodes.

Six Nucleus cochlear implant recipients participated in a study investigating the effect of place of stimulation on melody perception using rate-pitch cues. Each stimulus was a pulse train delivered on either a single electrode or multiple electrodes sequentially. Four spatial stimulation patterns were used: a single apical electrode, a single mid electrode, a pair of electrodes (apical and mid), and 11 electrodes (from apical to mid). Within one block of trials, all stimuli had the same spatial stimulation pattern, with pulse rate varying from 131 to 262 pps. An additional pulse rate range of

Effects of aromatic solvents on acoustic reflexes mediated by central auditory pathways.

From previous in vivo investigations, it has been shown that toluene can mimic the effects of cholinergic receptor antagonists and may thereby modify the response of protective acoustic reflexes. The current study aimed to define the relative effects of aromatic solvents on the middle ear and inner ear acoustic reflexes. Toward this end, the cochlear microphonic (CMP) elicited with a band noise centered at 4 kHz, and the compound action potential (CAP) elicited with 4-kHz tone pips was measured in rats. Both potentials were recorded before, during, and after triggering the protective reflexes
