Orienting-like reaction after ibotenic acid injections into the thalamic centre median nucleus in the cat.

The excitotoxin ibotenic acid was injected bilaterally into the intralaminar centre median nucleus of chronically implanted cats in order to study the effects of early excitation of centre median population on electrographic correlates of behavioral states and to compare them to those induced by injection into intralaminar centralis lateralis nucleus, previously shown. Immediately and during the first 24h after injection, highly aroused behavior with electrocortical activation, myoclonic jerks, enhancement of ocular movements and ponto-geniculo-occipital waves were observed. Surprisingly, in c

Selective vulnerability of dorsal raphe-medial prefrontal cortex projection neurons to corticosterone-induced hypofunction.

Glucocorticoid hormones and serotonin (5-HT) are strongly associated with the development and treatment of depression, respectively. Glucocorticoids regulate the function of serotonergic neurons in the dorsal raphe nucleus (DR), which are the major source of 5-HT to the forebrain. DR 5-HT neurons are electrophysiologically heterogeneous, though whether this phenotypic variation aligns with specific brain functions or neuropsychiatric disease states is largely unknown. The goal of this work was to determine if chronic exogenous glucocorticoid administration differentially affects the electrophy

Auditory processing and morphological anomalies in medial geniculate nucleus of Cntnap2 mutant mice.

Genetic epidemiological studies support a role for CNTNAP2 in developmental language disorders such as autism spectrum disorder, specific language impairment, and dyslexia. Atypical language development and function represent a core symptom of autism spectrum disorder (ASD), with evidence suggesting that aberrant auditory processing-including impaired spectrotemporal processing and enhanced pitch perception-may both contribute to an anomalous language phenotype. Investigation of gene-brain-behavior relationships in social and repetitive ASD symptomatology have benefited from experimentation on

Visual FMRI responses in human superior colliculus show a temporal-nasal asymmetry that is absent in lateral geniculate and visual cortex.

Eye patching has revealed enhanced saccadic latencies or attention effects when orienting toward visual stimuli presented in the temporal versus nasal hemifields of humans. Such behavioral advantages have been tentatively proposed to reflect possible temporal-nasal differences in the retinotectal pathway to the superior colliculus, rather than in the retinogeniculate pathway or visual cortex. However, this has not been directly tested with physiological measures in humans. Here, we examined responses of the human superior colliculus (SC) to contralateral visual field stimulation, using high sp

Evaluation of a surgical method for immobilizing the eye of an alert monkey.

One eye of a macaque monkey was immobilized by severing the third, fourth, and sixth cranial nerves. The residual movements of the operated eye were measured by repeatedly mapping the position of a visual receptive field. Movements of several degrees were caused by contraction of the orbital musculature during the closure blink. The amount of movement was dramatically reduced by paralysis of the superficial orbital muscles with a local anesthetic. Side effects of surgical immobilization include increased risk of corneal clouding and eye infection. In one monkey intraocular pressure was lowered

Distribution of bismuth in the brain after intraperitoneal dosing of bismuth subnitrate in mice: implications for routes of entry of xenobiotic metals into the brain.

Bismuth (Bi) can produce neurotoxic effects in both humans and animals under certain dosing conditions, but little else is known about the effects of Bi in the brain. In the present study we determined the distribution of Bi in the brains of adult female Swiss-Webster mice 4, 7, 14, 21 and 28 days after a single 2500 mg/kg i.p. injection of Bi subnitrate (BSN), which establishes a depot of absorbable Bi and produces morphological signs of neurotoxicity. Sections of brains were processed by autometallographic (AMG) procedures that produced silver grains at the site of Bi localization (AMGBi). V

Assessment of auditory pathway microstructure in vestibular schwannoma patients: a quantitative diffusion tensor tractography study.

OBJECTIVE: Vestibular schwannomas (VSs) are cerebellopontine angle tumors that can result in cranial nerve dysfunction, most commonly sensorineural hearing loss. Conventional structural MRI is unable to provide correlative information on cranial nerve function. In this study, the authors used multitensor tractography to study the white matter microstructural properties of the auditory neural pathway as a correlate of cranial nerve function in a cohort of VS patients. They evaluated the relationship between the auditory neural pathway microstructural properties using pure-tone audiometry (PTA)

Abnormal lateral geniculate nucleus and optic chiasm in human albinism.

Our objective was to measure how the misrouting of retinal ganglion cell (RGC) fibers affects the organization of the optic chiasm and lateral geniculate nuclei (LGN) in human albinism. We compared the chiasmal structures and the LGN in both pigmented controls and patients with albinism by using high-resolution structural magnetic resonance imaging (MRI). We studied 12 patients with oculocutaneous albinism and 12 age-matched pigmented controls. Using a 3T MRI scanner, we acquired a T1 -weighted three-dimensional magnetization-prepared rapid gradient-echo (MPRAGE) image of the whole brain, orie

Automated morphometry of the visual pathway in primary open-angle glaucoma.

PURPOSE: To establish whether primary open-angle glaucoma (POAG) is associated with a change in volume of the visual pathway structures between the eyes and the visual cortex. METHODS: To answer this question, magnetic resonance imaging (MRI) was used in combination with automated segmentation and voxel-based morphometry (VBM). Eight patients with POAG and 12 age-matched control subjects participated in the study. Only POAG patients with bilateral glaucomatous visual field loss were admitted to the study. The scotoma in both eyes had to include the paracentral region and had to, at least part

Sensory contributions to impaired emotion processing in schizophrenia.

Both emotion and visual processing deficits are documented in schizophrenia, and preferential magnocellular visual pathway dysfunction has been reported in several studies. This study examined the contribution to emotion-processing deficits of magnocellular and parvocellular visual pathway function, based on stimulus properties and shape of contrast response functions. Experiment 1 examined the relationship between contrast sensitivity to magnocellular- and parvocellular-biased stimuli and emotion recognition using the Penn Emotion Recognition (ER-40) and Emotion Differentiation (EMODIFF) test

Muscarinic regulation of dendritic and axonal outputs of rat thalamic interneurons: a new cellular mechanism for uncoupling distal dendrites.

Inhibition is crucial for sharpening the sensory information relayed through the thalamus. To understand how the interneuron-mediated inhibition in the thalamus is regulated, we studied the muscarinic effects on interneurons in the lateral posterior nucleus and lateral geniculate nucleus of the thalamus. Here, we report that activation of muscarinic receptors switched the firing pattern in thalamic interneurons from bursting to tonic. Although neuromodulators switch the firing mode in several other types of neurons by altering their membrane potential, we found that activation of muscarinic su

Electroacupuncture activation of the CaMKII/CREB pathway improves dLGN synaptic plasticity in monocular deprivation amblyopic rats.

OBJECTIVE: This study aimed to investigate the therapeutic potential of electroacupuncture(EA) for amblyopia, a common childhood visual disorder caused by early abnormalities such as monocular deprivation(MD). We examined the role of the calcium/calmodulin-dependent protein kinase II/cAMP response element-binding protein(CaMKII/CREB) pathway and enhanced synaptic plasticity in visual function improvement following EA. METHODS: An amblyopia model was established using 14-day-old rats by inducing MD during a critical developmental period. MD rats received EA stimulation at the Cuanzhu(BL2) and

Structural brain alterations in primary open angle glaucoma: a 3T MRI study.

Glaucoma is not only an eye disease but is also associated with degeneration of brain structures. We now investigated the pattern of visual and non-visual brain structural changes in 25 primary open angle glaucoma (POAG) patients and 25 age-gender-matched normal controls using T1-weighted imaging. MRI images were subjected to volume-based analysis (VBA) and surface-based analysis (SBA) in the whole brain as well as ROI-based analysis of the lateral geniculate nucleus (LGN), visual cortex (V1/2), amygdala and hippocampus. While VBA showed no significant differences in the gray matter volumes of

Axonal Terminals Exposed to Amyloid-β May Not Lead to Pre-Synaptic Axonal Damage.

BACKGROUND: Synaptic deficits and neuronal loss are the major pathological manifestations of Alzheimer's disease. However, the link between the early synaptic loss and subsequent neurodegeneration is not entirely clear. Cell culture studies have shown that amyloid-β (Aβ) applied to axonal terminals can cause retrograde degeneration leading to the neuronal loss, but this process has not been demonstrated in live animals. OBJECTIVE: To test if Aβ applied to retinal ganglion cell axonal terminals can induce axonal damage in the optic nerve and optic tract in mice. METHODS: Aβ was injected into

Early (E12) cortical progenitors can change their fate upon heterotopic transplantation.

BACKGROUND: The trend in breast cancer surgery is toward more conservative operative procedures. The new staging technique of sentinel node biopsy facilitates the identification of pathological node-negative patients in whom axillary dissection may be avoided. However, patients with a positive sentinel node biopsy would require a thorough examination of their nodal status. An axillary -dissection provides good local control, and accurate staging and prognostic information to inform decisions about adjuvant therapy. In addition, the survival benefit of axillary treatment is still debated. The o

Diffusion tensor fiber tracking of human brain connectivity: aquisition methods, reliability analysis and biological results.

We present a description, biological results and a reliability analysis for the method of diffusion tensor tracking (DTT) of white matter fiber pathways. In DTT, diffusion-tensor MRI (DT-MRI) data are collected and processed to visualize the line trajectories of fiber bundles within white matter (WM) pathways of living humans. A detailed description of the data acquisition is given. Technical aspects and experimental results are illustrated for the geniculo-calcarine tract with broad projections to visual cortex, occipital and parietal U-fibers, and the temporo-calcarine ventral pathway. NGF and BDNF are differentially modulated by visual experience in the developing geniculocortical pathway.

Neuronal activity and trophic factors have been implicated in shaping the connectivity of functional synaptic circuits. We studied the development and regulation by sensory input of the neurotrophins NGF, BDNF and NT-3 in the developing rat visual system. In the occipital cortex, NT-3 mRNA was transiently expressed in the neonate. In contrast, BDNF and NGF mRNA's increased during postnatal development, and reached mature levels around 3 weeks of age. BDNF mRNA was ten times more abundant than NGF mRNA. In the lateral geniculate nucleus (LGN), NT-3 mRNA was also transiently expressed, whereas N

Changes in both top-down and bottom-up effective connectivity drive visual hallucinations in Parkinson's disease.

Visual hallucinations are common in Parkinson's disease and are associated with a poorer quality of life and a higher risk of dementia. An important and influential model that is widely accepted as an explanation for the mechanism of visual hallucinations in Parkinson's disease and other Lewy body diseases is that these arise due to aberrant hierarchical processing, with impaired bottom-up integration of sensory information and overweighting of top-down perceptual priors within the visual system. This hypothesis has been driven by behavioural data and supported indirectly by observations deriv
