Mechano-bactericidal activity of cicada wing nanostructures against gram-positive bacteria.

Traditional antifouling mechanisms primarily prevent biofouling by inhibiting the initial adhesion of microorganisms to material surfaces. Conversely, the antifouling effect of cicada wings arises from their microstructured or nanostructured surface. These structures trap and rupture adherent microorganisms, which prevent biofilm formation and enable effective antifouling. This study used two typical gram-positive pathogenic strains-the rod-shaped Bacillus cereus and the coccus-shaped Staphylococcus aureus-as model microorganisms. The bactericidal efficacy of the nanopillar array on Pomponia l

Endogenous corazonin signaling modulates the post-mating switch in behavior and physiology in females of the brown planthopper and Drosophila.

Mating in insects typically triggers a post-mating response (PMR) in females, characterized by reduced receptivity to re-mating and increased oviposition, which ensures numerous and viable offspring and male paternity. This PMR is induced by male seminal factors, such as sex peptide in Drosophila melanogaster, as well as intrinsic female signaling components. The latter signaling remains poorly understood in most insects, including the devastating rice pest, the brown planthopper (BPH) Nilaparvata lugens. Here, we show that the neuropeptide corazonin (CRZ) and its receptor (CrzR) are critical

Inward Rectifier Potassium (Kir) Channel Inhibitors Protect Citrus from the Asian Citrus Psyllid by Inducing Toxicity and Inhibition of Feeding.

Huanglongbing (HLB), caused by the phloem-restricted bacterium Candidatus Liberibacter asiaticus (C. Las), is vectored by the Asian citrus psyllid (ACP) and threatens global citrus production. We demonstrate that VU041 (C19H20F3N3O), a small-molecule inhibitor of inward rectifier potassium (Kir) channels, provides a dual-mechanism approach to management of Huanglongbing (HLB) disease by combining acute toxicity with antifeedant activity. Topical and tarsal contact bioassays revealed that VU041 induced rapid mortality to ACP (24 h LD50 = 25 ng/insect) and reduced salivary gland secretion to sup

Overexpression of cytochrome P450 gene CYP402C18 contributes to the detoxification of dimpropyridaz, one novel pyridazine pyrazolecarboxamide insecticide, in the whitefly Bemisia tabaci Mediterranean cryptic species.

Bemisia tabaci is a major agricultural pest, and its widespread resistance to multiple insecticides poses a significant challenge for effective control. Dimpropyridaz, a new pyridazine-pyrazolecarboxamide insecticide, demonstrates high toxicity against sap-sucking pests, including those resistant to other class insecticides. However, the mechanisms underlying B. tabaci detoxification of dimpropyridaz remain unclear. In this study, transcriptome sequencing was performed on B. tabaci adults exposed to a lethal dose of dimpropyridaz for 48 h, identifying 973 differentially expressed genes (DEGs).

Flavonoids enhance tomato plant resistance to whitefly by interfering with the expression of a salivary effector.

Plants can deploy chemical defenses that poison herbivorous insects or deter their feeding; however, insects can counter by secreting effector proteins that modulate these defense responses. In principle, plants might therefore interfere with the expression of insect effector proteins, but knowledge about such a process remains limited. Here, we explored the mechanisms underlying how resistant tomato (Solanum lycopersicum) influences the effector proteins of whiteflies (Bemisia tabaci), thereby affecting the interaction of B. tabaci with tomato. Using two near-isogenic tomato lines that differ

Design and Synthesis of Indole-Containing Tetrahydrothiazolo[3,2-a]pyrimidine Mesoionics against Hemipteran Pests.

Mesoionic insecticides have emerged as promising pest nAChR modulators devoid of cross-resistance with neonicotinoids, demonstrating exceptional efficacy against sap-feeding insects. Nevertheless, the tetrahydrothiazolo[3,2-a]pyrimidine structure remains underexplored within this chemotype. Here, we report an indole-based scaffold hopping of tetrahydrothiazolo[3,2-a]pyrimidine, which leads to the development of a series of novel mesoionics. Their insecticidal potency is evaluated against the hemipteran pest Acyrthosiphon pisum (A. pisum) and Sogatella furcifera (S. furcifera). Derivative G1 ex

Involvement of mind the gap in the organization of the tracheal apical extracellular matrix in Drosophila and Nilaparvata lugens.

The tracheal apical extracellular matrix (aECM) is vital for expansion of the tracheal lumen and supports the normal structure of the lumen to guarantee air entry and circulation in insects. Although it has been found that some cuticular proteins are involved in the organization of the aECM, unidentified factors still exist. Here, we found that mind the gap (Mtg), a predicted chitin-binding protein, is required for the normal formation of the apical chitin matrix of airway tubes in the model holometabolous insect Drosophila melanogaster. Similar to chitin, the Mtg protein was linearly arranged

Field evaluation of Cucurbit yellow stunting disorder virus transmission by Bemisia tabaci.

Cucurbit yellow stunting disorder virus (CYSDV) is a whitefly-transmitted Crinivirus (Closteroviridae) that impacts melon production in many parts of the world including the USA. It has been responsible for melon crop loss in the southwestern U.S. since 2006 when it was first identified. Control strategies have revolved mainly around chemical control, but research to identify suitable products and approaches to implementing them have lagged. The current study investigated the performance of four systemic insecticides in the field while concurrently tracking CYSDV disease progression after cont

Genomic divergence and lack of introgressive hybridization between two 13-year periodical cicadas support life cycle switching in the face of climate change.

Life history evolution spurred by post-Pleistocene climatic change is hypothesized to be responsible for the present diversity in periodical cicadas (Magicicada), but the mechanism of life cycle change has been controversial. To understand the divergence process of 13-year and 17-year cicada life cycles, we studied genetic relationships between two synchronously emerging, parapatric 13-year periodical cicada species in the Decim group, Magicicada tredecim and M. neotredecim. The latter was hypothesized to be of hybrid origin or to have switched from a 17-year cycle via developmental plasticity

Whiteflies glycosylate salicylic acid and secrete the conjugate via their honeydew.

During insect feeding, a complex interaction takes place at the feeding site, with plants deciphering molecular information associated with the feeding herbivore, resulting in the upregulation of the appropriate defenses, and the herbivore avoiding or preventing these defenses from taking effect. Whiteflies can feed on plants without causing significant damage to mesophyll cells, making their detection extra challenging for the plant. However, whiteflies secrete honeydew that ends up on the plant surface at the feeding site and on distal plant parts below the feeding site. We reasoned that thi

Host manipulation by the orange leafhopper Cicadulina bipunctata: gall induction on distant leaves by dose-dependent stimulation.

The evolution of the gall-inducing ability in insects and the adaptive significance of the galling habit have been addressed by many studies. Cicadulina bipunctata, the maize orange leafhopper, is an ideal study organism for evaluating these topics because it can be mass-reared and it feeds on model plants such as rice (Oryza sativa) and maize (Zea mays). To reveal differences between gall inductions by C. bipunctata and other gall inducers, we conducted four experiments concerning (a) the relationship between the feeding site and gall-induction sites of C. bipunctata on maize, (b) the effects

Morphological and molecular characterization of Cordyceps javanica (Frieder. & Bally) Kepler and its in-vitro and in-vivo efficacy against tea mosquito bug, Helopeltis theivora waterhouse.

Tea mosquito bug, Helopeltis theivora Waterhouse (Hemiptera: Miridae), is a major pest of tea, Camellia sinensis (L.) O. Kuntze, causing up to 50% yield losses in the North Eastern region of India. The management of this pest is challenging owing to its rapid multiplication and development of resistance to conventional insecticides. Therefore, the present study was made to assess the efficacy of naturally occurring native entomopathogenic fungus (EPF), which was isolated from Archips, a leaf feeder pest of Lac host plant, Flemingia semialata. In vitro efficacy of this EPF against H. theivora c

Population Genomics Approaches Identify a Cryptic, Emerging Generalist Pest Complex.

Information about biological traits essential for pest management, such as species identity, diet and movement often require laborious and time-intensive studies on pest natural history, in both laboratory and field settings. However, new agricultural pest threats are continually emerging, often requiring prompt responses with limited information. Using a combination of molecular gut content analysis and RAD-seq, we examined the species identities, plant diet composition, and population genetic structure of an emerging and important agricultural pest in the US, the peanut burrower bug, Pangaeu

Developmental and Reproductive Impacts of Arsenophonus Symbiont on the Population of Nilaparvata lugens.

The Brown Planthopper, Nilaparvata lugens (Stål.) (Hemiptera: Delphinidae), is one of the most destructive pests of rice. Its reproductive and developmental traits are influenced by various environmental and biological factors including endosymbiotic microorganisms. Arsenophonus, a widespread endosymbiotic bacterium of insects, can affect host fitness and metabolic processes. This study investigates the role of Arsenophonus in modulating the developmental and reproductive traits of N. lugens fed on transgenic cry30Fa1 rice (KF30-14) and its parent variety Minghui 86 (MH86). Unravelling genetic diversity, population connectivity, and evolutionary relationships of Indian mealybugs through mtCO1-based phylogenetic and haplotype analyses.

BACKGROUND: Mealybugs (Hemiptera: Pseudococcidae) are economically important invasive pests that cause substantial losses in agricultural and horticultural crops worldwide. However, information on their diversity, genetic variation, and population structure in India remains limited. This study investigated the diversity, phylogenetic relationships, and population genetic structure of economically important mealybugs across different agroclimatic regions of India using mitochondrial cytochrome c oxidase subunit I (mtCOI) sequences. METHODS: A total of 134 mealybug specimens were analysed using

Effects of exogenous methyl jasmonate and salicylic acid on rice resistance to Oebalus pugnax.

BACKGROUND: After herbivore attack, plants express inducible resistance-related traits activated by hormones, mainly jasmonic acid (JA) and salicylic acid (SA). Methyl jasmonate (MeJa) is a biologically active methyl ester of JA. Exogenous applications of JA, SA, and MeJa induce responses similar to herbivory by insects. In this study, rice, Oryza sativa L. (Poaceae), plants were treated with two concentrations of MeJa (2 and 5 mmol L-1 ), two concentrations of SA (8 and 16 mmol L-1 ) and herbivory to evaluate effects of elicitation and herbivory on resistance to the rice stink bug (RSB) Oebal

Eutrophication and predator presence overrule the effects of temperature on mosquito survival and development.

Adequate predictions of mosquito-borne disease risk require an understanding of the relevant drivers governing mosquito populations. Since previous studies have focused mainly on the role of temperature, here we assessed the effects of other important ecological variables (predation, nutrient availability, presence of conspecifics) in conjunction with the role of temperature on mosquito life history parameters. We carried out two mesocosm experiments with the common brown house mosquito, Culex pipiens, a confirmed vector for West Nile Virus, Usutu and Sindbis, and a controphic species; the har

A novel needleless delivery of Cas9 ribonucleoprotein complex in Bemisia tabaci embryos for gene editing.

The CRISPR/Cas9 system, particularly the Cas9-sgRNA ribonucleoprotein (RNP) complex, offers a highly efficient platform for gene editing. However, traditional microinjection methods for RNP delivery in insect embryos are labor-intensive, technically demanding, and often reduce embryo viability, especially in species with fragile, microscopic embryos. In this study, a novel, non-invasive delivery method for the RNP complex in tiny insect embryos has been optimized. Whitefly, Bemisia tabaci, an invasive insect pest of agricultural importance and vector of plant diseases, was considered as a mode

Gene expression and plant hormone levels in two contrasting rice genotypes responding to brown planthopper infestation.

BACKGROUND: The brown planthopper (BPH; Nilaparvata lugens Stål) is a destructive piercing-sucking insect pest of rice. The plant hormones salicylic acid (SA) and jasmonic acid (JA) play important roles in plant-pest interactions. Many isolated rice genes that modulate BPH resistance are involved in the metabolism or signaling pathways of SA, JA and ethylene. 'Rathu Heenati' (RH) is a rice cultivar with a high-level, broad-spectrum resistance to all BPH biotypes. Here, RH was used as the research material, while a BPH-susceptible rice cultivar 'Taichung Native 1' (TN1) was the control. A cDNA
