Cepharanthine improves blood glucose levels and mitigates renal injury in streptozotocin-induced diabetic rats via restoring pancreatic β-cell integrity and reducing inflammation.

BACKGROUND/OBJECTIVES: We have previously demonstrated that the natural alkaloid Cepharanthine (Ceph) reduces renal injury in streptozotocin (STZ)-induced insulin-deficient diabetic rats via antioxidant and anti-inflammatory mechanisms. However, the hypoglycemic mechanism of Ceph remains unclear. We hypothesize that Ceph reduces blood glucose in STZ-induced insulin-deficient diabetic rats via restoring pancreatic β-cells function and insulin secretion. METHODS: Diabetes was induced in male Sprague-Dawley rats by a single intraperitoneal injection of STZ (50 mg/kg, i. p). Rats with blood gluco

History of pre-eclampsia negatively impacts stroke severity postpartum in rats.

BACKGROUND: Preeclampsia (PE) is a serious hypertensive disorder of pregnancy that has lifelong deleterious effects, including increased risk of stroke postpartum (PP). Here we determined if previous PE exacerbates ischemic injury in the PP period and investigated underlying mechanisms including oxidative stress and collateral perfusion. METHODS: Female Sprague-Dawley rats were studied at 4-9 months PP, after either a normal pregnancy (NormP-PP n = 7) or experimental PE (ePE) induced via high cholesterol diet during gestation (ePE-PP n = 9). Animals underwent transient middle cerebral artery

(R)-vanzacaftor potentiates BKCa channels in the absence of CFTR correction or potentiation.

We previously demonstrated the CFTR correctors VX-445 (elexacaftor) and S-VX-121 (vanzacaftor) potentiate heterologously expressed BKCa channels, as well as in primary human bronchial epithelial cells (HBEs). This potentiation of BKCa resulted in altered vasoreactivity and neuronal excitability. We postulated novel compounds could be identified that would potentiate BKCa while not affecting CFTR. Herein, we demonstrate that the enantiomer of vanzacaftor, R-VX-121, possesses these attributes. Using Fisher rat thyroid (FRT) cells expressing F508del CFTR, we demonstrate S-VX-121 corrects F508del

Myoendothelial feedback in mouse mesenteric resistance arteries is similar between the sexes, dependent on nitric oxide synthase, and independent of TPRV4.

Myoendothelial feedback (MEF), the endothelium-dependent vasodilation following sympathetic vasoconstriction (mediated by smooth muscle to endothelium gap junction communication), has been well studied in resistance arteries of males, but not females. We hypothesized that MEF responses would be similar between the sexes, but different in the relative contribution of the underlying nitric oxide and hyperpolarization mechanisms, given that these mechanisms differ between the sexes in agonist-induced endothelium-dependent dilation. We measured MEF responses (diameter changes) of male and female f

Electronic cigarette solvents, pulmonary irritation, and endothelial dysfunction: role of acetaldehyde and formaldehyde.

After more than a decade of electronic cigarette (E-cig) use in the United States, uncertainty persists regarding E-cig use and long-term cardiopulmonary disease risk. As all E-cigs use propylene glycol and vegetable glycerin (PG-VG) and generate abundant saturated aldehydes, mice were exposed by inhalation to PG-VG-derived aerosol, formaldehyde (FA), acetaldehyde (AA), or filtered air. Biomarkers of exposure and cardiopulmonary injury were monitored by mass spectrometry (urine metabolites), radiotelemetry (respiratory reflexes), isometric myography (aorta), and flow cytometry (blood markers).

Ifetroban reduces coronary artery dysfunction in a mouse model of Duchenne muscular dystrophy.

Dilated cardiomyopathy contributes to morbidity and mortality in Duchenne muscular dystrophy (DMD), an inheritable muscle-wasting disease caused by a mutation in the dystrophin gene. Preclinical studies in mouse models of muscular dystrophy have demonstrated reduced cardiomyopathy and improved cardiac function following oral treatment with the potent and selective thromboxane A2/prostanoid receptor (TPr) antagonist ifetroban. Furthermore, a phase 2 clinical trial (NCT03340675, Cumberland Pharmaceuticals) is currently recruiting subjects to determine whether ifetroban can improve cardiac functi

Inhibition of mitochondrial reactive oxygen species improves coronary endothelial function after cardioplegic hypoxia/reoxygenation.

OBJECTIVE: Cardioplegic ischemia-reperfusion and diabetes mellitus are correlated with coronary endothelial dysfunction and inactivation of small conductance calcium-activated potassium channels. Increased reactive oxidative species, such as mitochondrial reactive oxidative species, may contribute to oxidative injury. Thus, we hypothesized that inhibition of mitochondrial reactive oxidative species may protect coronary small conductance calcium-activated potassium channels and endothelial function against cardioplegic ischemia-reperfusion-induced injury. METHODS: Small coronary arteries and e

Clinical trial readiness to solve barriers to drug development in FSHD (ReSolve): protocol of a large, international, multi-center prospective study.

BACKGROUND: Facioscapulohumeral muscular dystrophy (FSHD) is a dominantly-inherited progressive muscular dystrophy caused by de-repression of the DUX4 gene, which causes disease by a toxic-gain-of-function. As molecularly targeted drugs move from preclinical testing into human trials, it is essential that we validate clinical trial tools and methodology to facilitate the drug development process. METHODS/DESIGN: The primary goal of this study is to hasten drug development for FSHD by validating two novel clinical outcome assessments (COAs) and refining clinical trial strategies. We will perfo

Protein kinase Cδ and pharmacomechanical coupling: Re-envisioning cerebral vascular control.

Constrictor stimuli set arterial tone through coupling processes dependent (electromechanical) and independent (pharmacomechanical) of VM. As the latter receives limited attention, we conducted a focused examination of the cerebral circulation to identify key signalling kinases involved in tone development and their role in regulating blood flow. A multiscale approach was implemented extending from cells to live brain and including myography, western blotting, immunolabelling, two-photon microscopy and modelling. We began by superfusing a G protein-coupled receptor agonist (U46619) onto isolat

The Beneficial Effects of Berberine on Vascular Dysfunction in Type 2 Diabetes Are Enhanced by HSP70 Inhibition.

Type 2 diabetes (T2D) is a chronic metabolic disorder leading to increased cardiovascular risk and vascular dysfunction. Hyperglycemia, a hallmark of T2D, drives hypercontractility, thereby compromising vascular function. Heat shock protein 70 (HSP70) has emerged as an important player in vascular reactivity under physiological conditions via its interaction with calcium mobilization, and in T2D, blocking this protein prevents hypercontractility. Circulating extracellular HSP70 (eHSP70) has also been proposed as a biomarker in chronic diseases, as it can function as a damage-associated molecul

Galectin-3 Regulates Smooth Muscle Contraction and Blood Pressure by Modulating CaV1.2 Channel Function.

BACKGROUND: Fine-tuning of CaV1.2 calcium channel activity by binding proteins represents a novel mechanism for regulating smooth muscle contraction and blood pressure (BP). This study aimed to elucidate the role of Gal-3 (galectin-3), a newly identified CaV1.2-binding protein, in the pathogenesis of hypertension. METHODS: In vitro, ex vivo, and in vivo experiments involving molecular and biochemical assays, in silico prediction, patch-clamp electrophysiologic recordings, immunohistochemistry, pressure myography, and tail-cuff BP measurements were used to evaluate the molecular mechanisms by

CGRP-targeted migraine treatment and early pathophysiology in experimental subarachnoid hemorrhage.

BACKGROUND: Calcitonin gene-related peptide (CGRP) is a central mediator in migraine and an endogenous vasodilator in the trigeminovascular system. Its role in subarachnoid hemorrhage (SAH), remains uncertain, particularly in patients receiving CGRP-targeted migraine therapy at the time of hemorrhage. This study examined whether CGRP blockade administered shortly before experimental SAH affects outcome and whether early SAH is associated with altered trigeminovascular CGRP signaling. METHODS: CGRP-mediated vasodilation was first assessed ex vivo in rat and mouse cerebral arteries using wire m

Predominant α2-adrenoceptor control of chelonian systemic resistance arteries provides a mechanistic basis for the cardiovascular effects of α2-agonist sedation.

Chelonians are commonly sedated with α2-adrenoceptor agonists, such as medetomidine, and bradycardia is consistently reported following administration. This may reflect a baroreflex response to α2-mediated peripheral vasoconstriction and increased systemic vascular resistance, but the vascular contribution to these cardiovascular effects has not been directly investigated. Here, we tested the hypothesis that α2-adrenoceptors contribute predominantly to vascular contraction in chelonians, contrasting with the α1-adrenoceptor-dominant regulation described in mammals. Mesenteric resistance arteri

Piezo1 in PASMCs: Critical for Hypoxia-Induced Pulmonary Hypertension Development.

BACKGROUND: Pulmonary hypertension (PH) is a life-threatening and progressive yet incurable disease. The hallmarks of PH comprise (1) sustained contraction and (2) excessive proliferation of pulmonary arterial smooth muscle cells (PASMCs). A major stimulus to which PASMCs are exposed during PH development is altered mechanical stress, originating from increased blood pressure, changes in blood flow velocity, and a progressive stiffening of pulmonary arteries. Mechanosensitive ion channels, including Piezo1 (Piezo-type mechanosensitive ion channel component-1), perceive such mechanical stimuli

Cystathionine γ Lyase Deletion Enhances Corpus Cavernosum Contraction via Thromboxane A 2 and Neurogenic Pathways Without Affecting Endothelial Function.

UNLABELLED: Cystathionine γ-lyase (CSE) produces hydrogen sulfide (H₂S), a vasodilator critical for vascular function. While its systemic effects are well-documented, its role in erectile physiology remains unclear. This study investigated the impact of CSE deletion on vascular and erectile tissue reactivity. We hypothesized that CSE knockout (CSE-KO) mice would exhibit endothelial dysfunction. A total of 22 CSE-KO and 22 age-matched wild-type (WT) controls were studied at one year of age. Cardiovascular outcome in 12-month-old male and female offspring of metformin-treated obese mice.

Metformin is increasingly used to treat diabetes in pregnancy, but the effects on adult offspring health remain under-explored. The present study investigated the long-term cardiovascular effects in male and female offspring of maternal metformin treatment using a well-established mouse model of obese glucose intolerant pregnancy. Female mice were given chow, or an obesogenic diet with/without 300 mg kg-1 day-1 oral metformin during gestation. At 3, 6 and 12 months of age, male and female offspring were studied longitudinally with tail-cuff plethysmography and echocardiography. At 12 months, t

Functional outcome of the anterior vaginal wall in a pelvic surgery injury rat model after treatment with stem cell-derived progenitors of smooth muscle cells.

BACKGROUND: Stem-cell-derived therapy is a promising option for tissue regeneration. Human iPSC-derived progenitors of smooth muscle cells (pSMCs) have limited proliferation and differentiation, which may minimize the risk of in vivo tumor formation while restoring smooth muscle cell deficiencies. Up to 30 % of women who suffer from recurrence of vaginal prolapse after prolapse surgery are faced with reoperation. Therefore, there is an unmet need for therapies that can restore vaginal tissue function. We hypothesize that human pSMCs can restore vaginal function in a vaginal-injury rat model.

Arrest of mouse preterm labor until term delivery by combination therapy with atosiban and mundulone, a natural product with tocolytic efficacy.

Currently, there is a lack of FDA-approved tocolytics for the management of preterm labor (PL). In prior drug discovery efforts, we identified mundulone and its analog mundulone acetate (MA) as inhibitors of in vitro intracellular Ca 2+ -regulated myometrial contractility. In this study, we probed the tocolytic and therapeutic potential of these small molecules using myometrial cells and tissues obtained from patients receiving cesarean deliveries, as well as a mouse model of PL resulting in preterm birth. In a phenotypic assay, mundulone displayed greater efficacy in the inhibition of intrace
