Selective changes in the protein-turnover rates and nature of growth induced in trout liver by long-term starvation followed by re-feeding.

We report upon the effects of a cycle of long-term starvation followed by re-feeding on the liver-protein turnover rates and nature of protein growth in the rainbow trout (Oncorhynchus mykiss). We determined the protein-turnover rate and its relationship with the nucleic-acid concentrations in the livers of juvenile trout starved for 70 days and then re-fed for 9 days. During starvation the total hepatic-protein and RNA contents decreased significantly and the absolute protein-synthesis rate (A(S)) also fell, whilst the fractional protein-synthesis rate (K(S)) remained unchanged and the fracti

Intracerebral expression of gonadotropin-releasing hormone and growth hormone-releasing hormone is delayed until smoltification in the salmon.

A developmental strategy was employed to investigate the functional assembly of neuropeptidergic systems in the migratory species of chum salmon Oncorhynchus keta. Using immunocytochemistry we have demonstrated that different groups of gonadotropin-releasing hormone- (GnRH)- and growth hormone-releasing hormone- (GHRH)-synthesizing neurons emerged according to very different developmental timetables. From the eye pigmentation stage (23 +/- 2 days after fertilisation (DAF)) through to the pre-smoltification stage (136 DAF), salmon-GnRH neurons originating from the olfactory placodes remained re

Bacillus subtilis spores influence the capacity of rainbow trout leukocytes to respond to a posterior viral encounter.

Although the positive effects of probiotics on the general immune status of aquacultured fish has been widely demonstrated using different probiotic strains and fish species, whether probiotics specifically stimulate antiviral responses has only been scarcely addressed. To provide more insights into this matter, we have investigated the capacity of Bacillus subtilis to modulate the transcription of a range on genes related to antiviral responses in rainbow trout (Oncorhynchus mykiss) through two independent experiments. In the first one, fish were fed a B. subtilis-supplemented diet or a contr

Not Just a Fish Killer: Multi-Organ Toxicity and Mechanisms of 6PPD-Quinone.

6PPD-Quinone (6PPD-Q) is a tire derivative formed by the oxidation of N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine (6PPD), a commonly used antioxidant and ozone stabilizer in rubber products, and has emerged as a significant environmental concern in recent years. It is widely present in the atmosphere, surface lakes, and soil. The primary routes of exposure to 6PPD-Q are the digestive tract and respiratory tract. Studies indicate that it is a major factor causing acute mortality in coastal coho salmon (Oncorhynchus kisutch). Reports indicate that 6PPD-Q exhibits greater chemical stabilit

Stressor- and Tissue-Specific Regulation of the Corticotropin-Releasing Factor System Across Epithelial Tissues in Rainbow Trout.

The corticotropin-releasing factor (CRF) system is a major neural regulator of stress responses in vertebrates. However, stress-related roles for the CRF system in other tissues-and whether these roles vary between stressor types-remain unclear. To address this gap, we first characterized the CRF system in the gills and intestine of rainbow trout (Oncorhynchus mykiss) and then evaluated how it is transcriptionally regulated following either an immune (vaccination) or osmotic (seawater transfer) stressor. Additionally, since the CRF system is involved in food intake regulation, we also evaluate

Salmon nasal cartilage proteoglycan up-regulates Listeria monocytogenes-mediated immune response in mice.

Proteoglycan (PG) is a complex glycohydrate that is widely distributed in the extracellular matrix. We have reported that daily oral administration of PG extracted from salmon (Oncorhynchus keta) nasal cartilage modulates the severity and proinflammatory cytokine responses in Escherichia coli-stimulated macrophages, and attenuates mouse models of various inflammatory diseases. Thus, PG has shown anti-inflammatory activities. In this study, we investigated the effect of salmon nasal cartilage PG on a bacterial infection by using a mouse model of infection with Listeria monocytogenes, which is a

Ocular mucosal homeostasis of teleost fish provides insight into the coevolution between microbiome and mucosal immunity.

BACKGROUND: The visual organ plays a crucial role in sensing environmental information. However, its mucosal surfaces are constantly exposed to selective pressures from aquatic or airborne pathogens and microbial communities. Although few studies have characterized the conjunctival-associated lymphoid tissue (CALT) in the ocular mucosa (OM) of birds and mammals, little is known regarding the evolutionary origins and functions of immune defense and microbiota homeostasis of the OM in the early vertebrates. RESULTS: Our study characterized the structure of the OM microbial ecosystem in rainbow

Farmed fish welfare during slaughter in Italy: survey on stunning and killing methods and indicators of unconsciousness.

Information on slaughter procedures for farmed fish in aquaculture is limited, both in Europe and in Italy, due to a general lack of field data. The aim of this study was to gather information on the procedures used to slaughter fish in Italy and to discuss them considering the WOAH and EFSA recommendations on fish welfare. Using a questionnaire survey, data were collected by official veterinarians in 64 slaughtering facilities where 20 different species of fish were slaughtered. The main species slaughtered were rainbow trout (Oncorhynchus mykiss; 29/64), followed by European sea bass (Dicent

Bacillus subtilis Expressing the Infectious Pancreatic Necrosis Virus VP2 Protein Retains Its Immunostimulatory Properties and Induces a Specific Antibody Response.

Bacillus subtilis has been documented in the past years as an effective probiotic for different aquacultured species, with recognized beneficial effects on water quality, fish growth and immune status. Furthermore, its potential as a vaccine adjuvant has also been explored in different species. In the current work, we have used B. subtilis spores as delivery vehicles for the presentation of the VP2 protein from infectious pancreatic necrosis virus (IPNV). For this, the VP2 gene was amplified and translationally fused to the crust protein CotY. The successful expression of VP2 on the spores was

Acute physiological stress down-regulates mRNA expressions of growth-related genes in coho salmon.

Growth and development in fish are regulated to a major extent by growth-related factors, such as liver-derived insulin-like growth factor (IGF) -1 in response to pituitary-secreted growth hormone (GH) binding to the GH receptor (GHR). Here, we report on the changes in the expressions of gh, ghr, and igf1 genes and the circulating levels of GH and IGF-1 proteins in juvenile coho salmon (Oncorhynchus kisutch) in response to handling as an acute physiological stressor. Plasma GH levels were not significantly different between stressed fish and prestressed control. Plasma IGF-1 concentrations in

Computational estimation of rainbow trout estrogen receptor binding affinities for environmental estrogens.

Environmental estrogens have been the subject of intense research due to their documented detrimental effects on the health of fish and wildlife and their potential to negatively impact humans. A complete understanding of how these compounds affect health is complicated because environmental estrogens are a structurally heterogeneous group of compounds. In this work, computational molecular dynamics simulations were utilized to predict the binding affinity of different compounds using rainbow trout (Oncorhynchus mykiss) estrogen receptors (ERs) as a model. Specifically, this study presents a c

Extracellular carbonic anhydrase in the dogfish, Squalus acanthias: a role in CO2 excretion.

In Pacific spiny dogfish (Squalus acanthias), plasma CO(2) reactions have access to plasma carbonic anhydrase (CA) and gill membrane-associated CA. The objectives of this study were to characterise the gill membrane-bound CA and investigate whether extracellular CA contributes significantly to CO(2) excretion in dogfish. A subcellular fraction containing membrane-associated CA activity was isolated from dogfish gills and incubated with phosphatidylinositol-specific phospholipase C. This treatment caused significant release of CA activity from its membrane association, a result consistent with

Evaluation of projection patterns in the primary olfactory system of rainbow trout.

Topographic projections are important for coding sensory information in the visual, auditory, and somatosensory systems but are of uncertain importance in the coding of olfactory information. We searched for topographic projections between olfactory receptor cells and the olfactory bulb of the rainbow trout Oncorhynchus mykiss. Anterograde axonal tracing with HRP revealed that the olfactory axons arising from discrete regions of the olfactory epithelium travel together within the olfactory nerve. The abrupt resorting and redistribution of these axons at the interface between the olfactory nerv

Comparative evaluation of three protein nanoscaffolds for IHNV G ectodomain display in rainbow trout (Oncorhynchusmykiss).

Infectious hematopoietic necrosis virus (IHNV) causes severe losses in rainbow trout (Oncorhynchus mykiss) aquaculture. Vaccination is a critical control strategy, and the surface glycoprotein (G) is the primary target for protective immunity. However, recombinant G protein alone is poorly immunogenic, prompting the use of self-assembling nanoparticle scaffolds to display its ectodomain and enhance immunogenicity. The optimal scaffold for anti-IHNV vaccination remains unclear. We constructed three nanovaccines by fusing the IHNV G ectodomain to ferritin (Fe), I3-01, and E2p scaffolds, and comp

Subyearling Chinook salmon diets in Lower Columbia River estuarine habitats.

The Lower Columbia River and Estuary is critical rearing habitat for juvenile Pacific salmon. Extending from the river mouth to Bonneville Dam 235 river kilometers upstream, the Estuary has been altered by dams, dikes, and habitat loss due to deforestation and wetland removal. Since 2008, five sites have been monitored to identify the long-term status and trends in Lower Columbia River and Estuary juvenile salmon rearing habitat. Here, we address predominantly natural origin juvenile Chinook Oncorhynchus tshawytscha diets and identify spatial and temporal trends in prey consumption, stomach fu

Mycobacterium salmoniphilum and M. chelonae in Captive Populations of Chinook Salmon.

Chinook Salmon Oncorhynchus tshawytscha is a keystone fish species in the Pacific Northwest. In 2019, unusual mortalities occurred in two different populations of cultured fingerlings from the same facility in California, USA. The systems consist of outdoor, enclosed, flow-through freshwater tanks that are maintained at 18 ± 1°C. Clinical signs and gross findings were only observed in one population and included abnormal swimming, inappetence, lethargy, skin discoloration, and the presence of multifocal nodular and ulcerative skin lesions. Microscopic lesions were infrequent and consisted of s

Declining muscle hyperplasia in juvenile trout is associated with a significant impairment of the supportive function of the myogenic progenitor niche.

BACKGROUND: Unlike mammals and birds, where new muscle fiber formation (hyperplasia) ceases around birth, large and fast-growing fish such as trout undergo a spectacular post-hatching surge of hyperplasia, followed by a considerably delayed hyperplasia decline. This study investigated the role of muscle stem cells (MuSCs) and their niche in this process by assessing changes in their abundance, myogenic potential and niche functionality. METHODS: Hyperplasia kinetics were investigated by measuring the total number of fibers and their cross-sectional area (CSA) in white muscle across juvenile s

Temporal dynamics of hepatic mRNA levels of carbohydrate and lipid metabolism-related genes in rainbow trout: Impact of a plant-based diet from first feeding.

Early-life nutrition plays a critical role in shaping metabolic trajectories in fish, particularly in carnivorous species like rainbow trout (Oncorhynchus mykiss), which displays limited capacity to utilize plant-based diets. This study investigated the temporal changes in mRNA abundance of key hepatic genes involved in carbohydrate and lipid metabolism from first feeding to 100 days post-first meal in rainbow trout fry fed either a commercial-like or fully plant-based diet. mRNA abundance was quantified by RT-qPCR at six time points, revealing marked ontogenetic shifts in metabolic gene profi

Characterization of volatile compounds in the water samples from rainbow trout aquaculture ponds eliciting off-odors: understanding locational and seasonal effects.

The quality of water used in aquaculture ponds is one of the crucial factors influencing the smell and sensory properties of fish. The water samples were taken from the rainbow trout fish ponds from three different fish farms in three provinces in Türkiye in four different seasons. The samples were analyzed for the volatile components by employing HS-SPME/GC-MS. Seven different volatile groups including aldehydes, ketones, esters, alcohols, volatile phenols, terpenes and other aromatic substances were identified in the samples. Among these, aldehydes were found to be the most dominant. (E)-2-H
