Nanotechnology enabled radioprotectants to reduce space radiation-induced reactive oxidative species.

Interest in space exploration has seen substantial growth following recent launch and operation of modern space technologies. In particular, the possibility of travel beyond low earth orbit is seeing sustained support. However, future deep space travel requires addressing health concerns for crews under continuous, longer-term exposure to adverse environmental conditions. Among these challenges, radiation-induced health issues are a major concern. Their potential to induce chronic illness is further potentiated by the microgravity environment. While investigations into the physiological effect

Development of a graphical user interface for automatic separation of human voice from Doppler ultrasound audio in diving experiments.

Doppler ultrasound (DU) is used in decompression research to detect venous gas emboli in the precordium or subclavian vein, as a marker of decompression stress. This is of relevance to scuba divers, compressed air workers and astronauts to prevent decompression sickness (DCS) that can be caused by these bubbles upon or after a sudden reduction in ambient pressure. Doppler ultrasound data is graded by expert raters on the Kisman-Masurel or Spencer scales that are associated to DCS risk. Meta-analyses, as well as efforts to computer-automate DU grading, both necessitate access to large databases

Crash Prediction Using Deep Learning in a Disorienting Spaceflight Analog Balancing Task.

Were astronauts forced to land on the surface of Mars using manual control of their vehicle, they would not have familiar gravitational cues because Mars' gravity is only 0.38 g. They could become susceptible to spatial disorientation, potentially causing mission ending crashes. In our earlier studies, we secured blindfolded participants into a Multi-Axis Rotation System (MARS) device that was programmed to behave like an inverted pendulum. Participants used a joystick to stabilize around the balance point. We created a spaceflight analog condition by having participants dynamically balance in

Radiobiological damage by space radiation: extension of the BIANCA model to heavy ions up to iron, and pilot application to cosmic ray exposure.

Space research seems to be object of a renewed interest, also considering that human missions to the Moon, and possibly Mars, are being planned. Among the risks affecting such missions, astronauts' exposure to space radiation is a major concern. In this work, the question of the evaluation of biological damage by Galactic Cosmic Rays (GCR) was addressed by a biophysical model called BIophysical ANalysis of Cell death and chromosome Aberrations (BIANCA), which simulates the induction of cell death and chromosome aberrations by different ions. While previously BIANCA has been validated for calcu

Targeted silencing of miRNA-132-3p expression rescues disuse osteopenia by promoting mesenchymal stem cell osteogenic differentiation and osteogenesis in mice.

BACKGROUND: Skeletal unloading can induce severe disuse osteopenia that often occurs in spaceflight astronauts or in patients subjected to prolonged bed-rest or immobility. Previously, we revealed a mechano-sensitive factor, miRNA-132-3p, that is closely related to the osteoblast function. The aim of this study was to investigate whether miRNA-132-3p could be an effective target for treating disuse osteopenia. METHODS: The 2D-clinostat device and the hindlimb-unloaded (HU) model were used to copy the mechanical unloading condition at the cellular and animal levels, respectively. Mimics or inh

A framework for assessing the safety of degraded pharmaceuticals for long-duration space missions - Part 1: framework development.

The National Aeronautics and Space Administration (NASA) is planning exploration space missions to Mars, which will require plans for a wide array of medical contingencies, most of which require treatment with medications. A major challenge is that medications cannot be resupplied beyond Earth orbit, and many medications will exceed their labeled expiration date over the duration of an exploration-class mission. Furthermore, the spaceflight environment may alter the rate or pathways involved in the degradation of active pharmaceutical ingredients (APIs). However, the stability of only a handfu

Randomized, Double-Blind, Phase I Pharmacokinetic Study of Subcutaneous Recombinant Human Superoxide Dismutase (rhSOD) in Healthy Volunteers.

BACKGROUND: Excessive production of reactive oxygen species (ROS), particularly superoxide anion ( O 2 · - ), is a key mechanism in diseases such as cancer, inflammatory disorders, neurodegenerative conditions, and metabolic diseases. Evidence also suggests that microgravity-induced oxidative stress, primarily driven by elevated O 2 · - levels, may contribute to the adverse physiological effects observed in astronauts during extended space missions. Superoxide dismutase (SOD) is critical for mitigating oxidative stress, and exogenous SOD supplementation offers a potential therapeutic strat

Whole-body vibration transmission during resistance vibration exercise.

INTRODUCTION: Resistive exercise combined with whole-body vibration (WBV) and short-arm human centrifugation is being considered as a countermeasure to mitigate muscle and bone loss in astronauts during prolonged space missions. WBV may provide exercise benefits or adverse effects on organs and the lower back. These effects may result from vibration transmitted throughout the body. The objective of this study was to compare vibration transmission (VT) from the ground to the body during resistance vibration exercise (RVE) comprising squat and calf raise performed on a rotational vibration plate

Identification of gravity-responsive proteins in the femur of spaceflight mice using a quantitative proteomic approach.

Although the microgravity (μ-g) environment that astronauts encounter during spaceflight can cause severe acute bone loss, the molecular mechanism of this bone loss remains unclear. To investigate the gravity-response proteins involved in bone metabolism, it is important to comprehensively determine which proteins exhibit differential abundance associated with mechanical stimuli. However, comprehensive proteomic analysis using small bone samples is difficult because protein extraction in mineralized bone tissue is inefficient. Here, we established a high-sensitivity analysis system for mouse b

A Network approach to find poor orthostatic tolerance by simple tilt maneuvers.

The approach introduced by Network Physiology intends to find and quantify connectedness between close- and far related aspects of a person's Physiome. In this study I applied a Network-inspired analysis to a set of measurement data that had been assembled to detect prospective orthostatic intolerant subjects among people who were destined to go into Space for a two weeks mission. The advantage of this approach being that it is essentially model-free: no complex physiological model is required to interpret the data. This type of analysis is essentially applicable to many datasets where individ

Introducing the Concept of Exercise Holidays for Human Spaceflight - What Can We Learn From the Recovery of Bed Rest Passive Control Groups.

In an attempt to counteract microgravity-induced deconditioning during spaceflight, exercise has been performed in various forms on the International Space Station (ISS). Despite significant consumption of time and resources by daily exercise, including around one third of astronauts' energy expenditure, deconditioning-to variable extents-are observed. However, in future Artemis/Lunar Gateway missions, greater constraints will mean that the current high volume and diversity of ISS in-flight exercise will be impractical. Thus, investigating both more effective and efficient multi-systems counte

Ophthalmic changes in a spaceflight analog are associated with brain functional reorganization.

Following long-duration spaceflight, some astronauts exhibit ophthalmic structural changes referred to as Spaceflight Associated Neuro-ocular Syndrome (SANS). Optic disc edema is a common sign of SANS. The origin and effects of SANS are not understood as signs of SANS have not manifested in previous spaceflight analog studies. In the current spaceflight analog study, 11 subjects underwent 30 days of strict head down-tilt bed rest in elevated ambient carbon dioxide (HDBR+CO2 ). Using functional magnetic resonance imaging (fMRI), we acquired resting-state fMRI data at 6 time points: before (2),

The effects of a spaceflight analog with elevated CO2 on sensorimotor adaptation.

Aboard the International Space Station (ISS), astronauts must adapt to altered vestibular and somatosensory inputs due to microgravity. Sensorimotor adaptation on Earth is often studied with a task that introduces visuomotor conflict. Retention of the adaptation process, known as savings, can be measured when subjects are exposed to the same adaptive task multiple times. It is unclear how adaptation demands found on the ISS might interfere with the ability to adapt to other sensory conflict at the same time. In the present study, we investigated the impact of 30 days' head-down tilt bed rest c

Low-dose photon and simulated solar particle event proton effects on Foxp3+ T regulatory cells and other leukocytes.

Radiation is a major factor in the spaceflight environment that has carcinogenic potential. Astronauts on missions are continuously exposed to low-dose/low-dose-rate (LDR) radiation and may receive relatively high doses during a solar particle event (SPE) that consists primarily of protons. However, there are very few reports in which LDR photons were combined with protons. In this study, C57BL/6 mice were exposed to 1.7 Gy simulated SPE (sSPE) protons over 36 h, both with and without pre-exposure to 0.01 Gray (Gy) LDR g-rays at 0.018 cGy/h. Apoptosis in skin samples was determined by immunohi

Artificial gravity during spaceflight prevents gait and performance deficits in a gravity-dose dependent manner.

Long-term adaptations to spaceflight outside of low earth orbit (LEO), during deep space transit, and readaptations to partial gravity environments upon reaching a destination, are unclear. The combined effect of these adaptations to the LEO environment can result in declines in mobility and balance in astronauts during and after spaceflight. This study aimed to determine if there is a gravity threshold that protects from deficits in gait and performance. To do so, we exposed mice to four gravitational loading conditions (µg; 0.33 g; 0.67 g; and 1 g), induced by centrifugation, relative to pre

Microgravity as a translational model for metabolic dysfunction: implications for obesity, insulin resistance, steatotic liver and pancreatic diseases.

PURPOSE: The long-term effects of physical inactivity, whether due to microgravity in spaceflight or an inactive lifestyle on Earth, can lead to a range of metabolic problems. This review examines the physiological response to reduced gravity and contemporary inactivity, with a focus on obesity, insulin resistance, type 2 diabetes, and hepatic and pancreatic steatosis, all of which are associated with metabolic dysfunction (MASLD, MASPD). METHODS: A narrative review approach was used, integrating evidence from spaceflight research, bed rest studies, and clinical trial data to examine the meta

From Collapse to Active Self-Repair: Integrative Multi-Omics and Machine Learning Analysis Map the Hepatic Metabolic Adaption in Response to Simulated Spaceflight Stress.

Long-term spaceflight poses substantial challenges to human physiology, with the liver being highly susceptible due to its central metabolic role. To determine whether hepatic alterations represent transient stress or sustained remodeling, we performed an integrated multi-omics analysis in a rat model simulating chronic space radiation and microgravity. Herein, we applied an integrated multi-omics and AI-driven analytical framework combining histopathology, cytokine and miRNA profiling, proteomics, metabolomics and Western blot validation. After 21 days of simulated space conditions, rats exhi

Telomere Length and Telomerase Activity as Biomarkers in the Diagnostics and Prognostics of Pathological Conditions.

Telomere biology still remains a topic of interest in life sciences. Analysis of several thousand clinical samples from healthy individuals performed in recent years has shown that the telomere length (TL) in peripheral blood leukocytes correlates with the TL in cells of internal organ and reflects their condition. TL decreases under the influence of damaging factors and can serve as an indicator of health status. The telomere shortening leads to the cell proliferation arrest and is considered as a marker of replicative aging of proliferating cells. A decrease in the TL in peripheral blood leu

Gravity's effect on biology.

Gravity is a fundamental interaction that permeates throughout our Universe. On Earth, gravity gives weight to physical objects, and has been a constant presence throughout terrestrial biological evolution. Thus, gravity has shaped all biological functions, some examples include the growth of plants (e.g., gravitropism), the structure and morphology of biological parts in multicellular organisms, to its effects on our physiological function when humans travel into space. Moreover, from an evolutionary perspective, gravity has been a constant force on biology, and life, to our understanding, sh

Prevention of the gastrointestinal adverse effects of nonsteroidal anti-inflammatory drugs: the role of proton pump inhibitors.

The associations between nonsteroidal anti-inflammatory drugs (NSAIDs) and the presence and complications of gastroduodenal erosions and ulcers are well established. Evidence that acid aggravates NSAID-induced injury provides a rationale for minimising such damage by acid suppression. Other strategies discussed include avoidance of NSAIDs or minimising their dosage, selecting NSAIDs known to cause less damage, and co-prescription of various agents. Cytoprotection with misoprostol, a prostaglandin analogue, has been shown to be effective in reducing NSAID-related peptic ulcers and their complic
