Comparative genome analysis of Pseudomonas chlororaphis strains reveals biosynthetic diversity underlying plant growth promotion and soil health improvement.

BACKGROUND: The growing need for sustainable farming methods has led to increased efforts to understand plant-beneficial microbes that can serve as alternatives to chemical fertilizers and pesticides. Among these, bacteria from the Pseudomonas chlororaphis group have emerged as particularly promising candidates due to their metabolic versatility and ability to produce a diverse arsenal of bioactive compounds. RESULTS: In this study, we conducted an extensive comparative genomic analysis of two Pseudomonas chlororaphis strains, FS-2 (endophytic) and RP-4 (rhizoplane-associated), to explore thei

Minimal domain peptides derived from enterocins exhibit potent antifungal activity.

The antimicrobial peptide (AMP) circularized bacteriocin enterocin AS-48 produced by Enterococcus sp. exhibits broad-spectrum antibacterial activity via dimer insertion into the plasma membrane to form membrane pore structures, compromising membrane integrity and leading to bactericidal activity. A specific alpha-helical region of enterocin AS-48 has been shown to be responsible for the membrane-penetrating activity of the peptide. The canon syn-enterocin peptide library, generated using rational design techniques to have ninety-five synthetic peptide variants from the truncated, linearized, m

Overcoming Microbial Inhibition of S. Sonnei Through the Exploitation of Genomically Predicted Antibiotic Resistance Profiles for the Development of Food Enrichment Media.

Linking outbreaks of Shigella spp. to specific foods is challenging due to poor selectivity of current enrichment media. We have previously shown that enrichment media, tailored to the genomically-predicted antimicrobial resistance (AMR) of Shiga toxigenic E. coli strains, enhances their isolation from foods. This study investigates the application of this approach for Shigella isolation. The AMR gene profiles of 21,908 published S. sonnei genomes indicated a high prevalence of genes conferring resistance to streptomycin (aadA, aph(3″)-Ib, aph(6)-Id, 92.8%), sulfonamides (sul1, sul2, 74.8%), a

Docking on Lipid II-A Widespread Mechanism for Potent Bactericidal Activities of Antibiotic Peptides.

Natural product antibiotics usually target the major biosynthetic pathways of bacterial cells and the search for new targets outside these pathways has proven very difficult. Cell wall biosynthesis maybe the most prominent antibiotic target, and ß-lactams are among the clinically most relevant antibiotics. Among cell wall biosynthesis inhibitors, glycopeptide antibiotics are a second group of important drugs, which bind to the peptidoglycan building block lipid II and prevent the incorporation of the monomeric unit into polymeric cell wall. However, lipid II acts as a docking molecule for many

Characterization of functional, safety, and gut survival related characteristics of Lactobacillus strains isolated from farmhouse goat's milk cheeses.

A set of 80 Lactobacillus strains (36 Lactobacillus plantarum and 44 Lactobacillus paracasei) isolated from Spanish farmhouse cheeses have been studied as to their functional and safety properties and their survival under gut-related conditions. None of these 80 Lactobacillus strains were able to hydrolyse starch. A high percentage of L. plantarum and L. paracasei strains were, however, capable of hydrolysing casein (86.1% and 68.2% respectively). For the other characteristics investigated, L. plantarum strains generally had more positive responses than L. paracasei. The latter strains tested

Culture-free supernatant of potential probiotic Bacillus albus attenuates the expression of biofilm-associated virulence determinants in Burkholderia thailandensis and Staphylococcus aureus.

The role of native bacterial probiotics in the Chinese traditional fermentation starter, Qubing, has not been previously studied. In this investigation, 22 bacterial strains were isolated from Qubing, and potential probiotic candidates were selected for their antibiofilm activity against Burkholderia thailandensis and Staphylococcus aureus. The most resistant bacterial isolates were highly tolerant of simulated harsh conditions, including acid, bile salts, and gastric juice. Many isolates exhibited bile-salt hydrolase activity, suggesting that they reduce cholesterol levels by deconjugating bi

Orally ingested Lactobacillus crispatus VPC177 enhances total vaginal Lactobacillus counts-a cross-over exploratory study.

INTRODUCTION: The vaginal microbiome plays a crucial role in vaginal health, and disturbances in the microbial balance can negatively affect female wellbeing. In recent years, there has been growing interest in the use of probiotics and prebiotics to support a healthy vaginal microbiota. However, effects are often strain-specific and should be clinically demonstrated to influence the vaginal microbiota. This study aimed at evaluating the ability of four different orally administered probiotic strains to affect the fecal, and more specifically, the vaginal microbiota in healthy women. METHODS:

Integrated Phenotypic and Whole-Genome Analysis of Enterococcus hirae HI3 Isolated from Hu Sheep Jejunum Supports Its Potential as a Ruminant Probiotic Candidate.

The ban on antibiotic growth promoters in livestock production has created an urgent demand for safe and effective probiotic alternatives derived from indigenous animal microbiota. Here, we isolated Enterococcus hirae HI3 from Hu sheep jejunum and confirmed its species assignment via whole-genome sequencing, with ANI analysis showing 98.92% identity to the E. hirae reference genome. HI3 showed moderate acid tolerance, with survival rates of 19.31% (95% CI: 15.83-22.78%) at pH 2 and 18.40% (95% CI: 17.87-18.94%) at pH 3 after 4 h, and maintained > 50% survival in 2% bile salts. Intra-individual genetic diversity of vaginal Lactobacillus crispatus revealed through citizen science-driven isolation and pangenome analysis.

AIMS: A vaginal microbiome dominated by Lactobacillus crispatus is associated with positive reproductive and sexual health outcomes, yet intra-individual genetic diversity within this species remains largely unexplored. This study characterized inter- and intra-individual genomic variation in L. crispatus strains isolated through a citizen science initiative and assessed implications for multi-strain probiotic development. METHODS AND RESULTS: Fifty-three women participated in this citizen science project. Self-sampling resulted in 48 shotgun metagenomes. Twenty-two participants isolated thei

Influenza A Virus Coinfection Alters Streptococcus pneumoniae Gene Expression during Upper Respiratory Tract Colonization.

Streptococcus pneumoniae (Spn) asymptomatically colonizes the upper respiratory tract (URT), a niche from which it can transmit to another host or cause invasive disease in the same host. The in vivo transcriptional adaptations that Spn undergoes during nasopharyngeal colonization, particularly during influenza A virus (IAV) coinfection, are poorly understood. Here, we leveraged an established infant mouse model of colonization, shedding, and transmission to perform genome-wide transcriptomic profiling of Spn during mono- and during IAV co-infection. A Critical Review of Risk Assessment Models for Listeria monocytogenes in Seafood.

Invasive listeriosis, due to its severe nature in susceptible populations, has been the focus of many quantitative risk assessment (QRA) models aiming to provide a valuable guide in future risk management efforts. A review of the published QRA models of Listeria monocytogenes in seafood was performed, with the objective of appraising the effectiveness of the control strategies at different points along the food chain. It is worth noting, however, that the outcomes of a QRA model are context-specific, and influenced by the country and target population, the assumptions that are employed, and th
