High E2F7 Expression Indicates Pancreatic Cancer Aggressiveness and Downregulation of E2F7 Enhances Sensitivity to S-1.

BACKGROUND: Pancreatic cancer (PC) remains a highly lethal disease with few reliable biomarkers to guide chemotherapy choices. New biomarkers for selecting anticancer drugs are needed to enhance the effectiveness of current multimodal treatment approaches. This study aimed to find a new biomarker by using clinical data and specimens collected for a Japanese randomized controlled trial (RCT). METHODS: Gene expression array analysis was performed using PC tissues collected for the ancillary research of JASPAC01, a nationwide phase 3 RCT of adjuvant chemotherapy for patients with PC in Japan. [A Case Report of Luminal A Male Inflammatory Breast Cancer that Was Difficult to Treat Because of Trousseau Syndrome].

This report describes the case of a 67-year-old male with inflammatory breast cancer. He had noticed a left breast mass about seven years previously, but he had ignored it. He then visited our hospital 4 months previously when multiple small masses occurred in the left front chest wall. The tumor was diagnosed as skin metastasis of breast cancer by skin biopsy and he was referred to our department. The tumor cells were positive for estrogen receptor and progesterone receptor, and negative for HER2/neu, and the Ki67 expression was 10-15%. The subtype of his breast cancer was luminal A type. It

Oral administration of the amino acids cystine and theanine attenuates the adverse events of S-1 adjuvant chemotherapy in gastrointestinal cancer patients.

BACKGROUND: Nutritional therapy is used to reduce the adverse events (AEs) of anticancer drugs. Here, we determined whether the amino acids cystine and theanine, which provide substrates for glutathione, attenuated the AEs of S-1 adjuvant chemotherapy. METHODS: Patients scheduled to receive S-1 adjuvant chemotherapy were randomized to the C/T or the control groups. The C/T group received 700 mg cystine and 280 mg theanine orally 1 week before the administration of S-1, which then continued for 5 weeks. Each group received S-1 for 4 weeks. Blood sampling was performed and AEs were evaluated (C

High L-Type Amino Acid Transporter 1 Levels Are Associated with Chemotherapeutic Resistance in Gastric Cancer Patients.

INTRODUCTION: We investigated whether the expression of L-type amino acid transporter 1 (LAT-1) in clinical gastric cancer (GC) patients could predict patient therapeutic response to postoperative adjuvant chemotherapy. METHODS: Immunohistochemistry was used to investigate LAT-1, CD98, and phosphorylated-mammalian target of rapamycin (p-mTOR) expression in 111 GC patients. To clarify whether LAT-1 influences the therapeutic effects of chemotherapy, the correlation between disease-free survival rates and LAT-1 was determined in 2 groups: 59 patients who did not undergo postoperative adjuvant c

Salvage gastrectomy after intravenous and intraperitoneal paclitaxel (PTX) administration with oral S-1 for peritoneal dissemination of advanced gastric cancer with malignant ascites.

BACKGROUND: Peritoneal metastasis of gastric cancer has extremely poor clinical outcomes. Recently, we developed a combination chemotherapy that used intraperitoneal (IP) paclitaxel (PTX) and produced excellent antitumor effects against peritoneal lesions. However, no information is available about the benefit of gastrectomy in cases with malignant ascites. METHODS: A total of 64 patients with severe peritoneal metastasis and ascites received IP PTX at 20 mg/m(2) via implanted subcutaneous peritoneal access ports as well as intravenous (IV) PTX at 50 mg/m(2) on days 1 and 8. Safety and feasibility of S-1 after capecitabine-induced toxicity in the adjuvant treatment of colon cancer patients: A population-based real-world study.

BACKGROUND: S-1 has comparable efficacy to capecitabine/5-FU in metastatic colorectal cancer, but is associated with a lower incidence of hand-foot syndrome (HFS) and cardiovascular toxicity (CVT). The European Medicines Agency has approved its use when capecitabine or 5-FU is discontinued due to HFS or CVT. However, in the adjuvant setting, data on its safety profile are scarce. We assessed the feasibility of S-1 in patients who discontinued capecitabine due to HFS or CVT. METHODS: We included all patients with colon cancer included in the Prospective Dutch Colorectal Cancer Cohort (PLCRC),

Relative Dose Intensity and Outcomes in Elderly Patients With Resectable Pancreatic Cancer Receiving Neoadjuvant Chemotherapy.

BACKGROUND: Neoadjuvant chemotherapy (NAC) with gemcitabine and S-1 (GS) is the standard regimen for resectable pancreatic cancer (PC) in Japan. However, its efficacy in elderly patients and those with impaired renal function remains unclear. METHODS: We retrospectively analyzed patients with resectable PC who received NAC GS. Patients were classified as elderly (≥ 75 years) or non-elderly (< 75 years). The primary endpoint was relative dose intensity (RDI) of NAC. Secondary endpoints included adverse events (AEs), perioperative outcomes, RDI of adjuvant chemotherapy (AC), recurrence-free sur

The investigation of the survival time after recurrence in patients with pancreatic ductal adenocarcinoma for individualization of adjuvant chemotherapy.

PURPOSE: Pancreatic ductal adenocarcinoma (PDAC) is a lethal disease; however, the frequency of recurrence can be reduced if curative surgery following adjuvant chemotherapy is applied. At present, adjuvant chemotherapy is uniformly performed in all patients, as it is unclear which tumor types are controlled best or worst. We investigated patients with recurrence to establish the optimum treatment strategy. METHODS: Of 138 patients who underwent curative surgery for PDAC, 85 developed recurrence. Comprehensive clinicopathological factors were investigated for their association with the surviv
