Application of representational difference analysis to identify sequence tags expressed by Metarhizium anisopliae during the infection process of the tick Boophilus microplus cuticle.

Metarhizium anisopliae is a well-characterized biocontrol agent of a wide range of plagues, including insects and acari. To identify genes involved in the infection process, representational difference analysis was performed using cDNA generated from germinated conidia of M. anisopliae in the tick Boophilus microplus cuticle, and cDNA generated during fungal growth in glucose-rich medium. Sequence determination of approximately 135 clones and comparison analysis using public databases led to the identification of 34 sequences and 14 expressed sequence tags with known orthologs. As expected, al

Identification of Trombiculid Chigger Mites Collected on Rodents from Southern Vietnam and Molecular Detection of Rickettsiaceae Pathogen.

Trombiculid "chigger" mites (Acari) are ectoparasites that feed blood on rodents and another animals. A crosssectional survey was conducted in 7 ecosystems of southern Vietnam from 2015 to 2016. Chigger mites were identified with morphological characteristics and assayed by polymerase chain reaction for detection of rickettsiaceae. Overall chigger infestation among rodents was 23.38%. The chigger index among infested rodents was 19.37 and a mean abundance of 4.61. A total of 2,770 chigger mites were identified belonging to 6 species, 3 genera, and 1 family, and pooled into 141 pools (10-20 chi

Borreliae in Ixodes ricinus ticks feeding on humans.

A total of 298 Ixodes ricinus (L.) ticks (Acari: Ixodidae) feeding on humans in the Czech Republic were tested for borreliae (Borrelia burgdorferi sensu lato) by darkfield microscopy between 1997 and 2003. A majority (68%) of the supplied I. ricinus ticks were nymphs, 25% were females and 7% were larvae. Overall, 20% of 74 examined females and 9% of 203 examined nymphs (but none of 21 examined larvae) were infected with borreliae. The proportion of ticks with a high infection load (>100 spirochetes) was 4% in females and 2% in nymphal I. ricinus. During the year, the highest numbers and propor

Chemical and Plant-Based Insect Repellents: Efficacy, Safety, and Toxicity.

Most emerging infectious diseases today are arthropod-borne and cannot be prevented by vaccinations. Because insect repellents offer important topical barriers of personal protection from arthropod-borne infectious diseases, the main objectives of this article were to describe the growing threats to public health from emerging arthropod-borne infectious diseases, to define the differences between insect repellents and insecticides, and to compare the efficacies and toxicities of chemical and plant-derived insect repellents. Internet search engines were queried with key words to identify scient

The occurrence of Borrelia burgdorferi sensu lato in certain ectoparasites (Mesostigmata, Siphonaptera) of Apodemus flavicollis and Myodes glareolus in chosen localities in the Czech Republic.

Ticks represent the primary vectors of several serious diseases, including the Lyme disease caused by Borrelia burgdorferi sensu lato (Bbsl). In this study two dominant ectoparasitic groups of arthropods (Mesostigmata, Siphonaptera) were investigated for the presence of borrelian DNA in order to determine their potential role of vectors (or carriers) of this bacterium. All individuals (235) were collected from wild-living rodents obtained in three localities in the Czech Republic (Bažantula, Baba and Křižovice). The majority of parasites were members of the families Parasitidae and Dermanyssid

[Sensitization to the acarus Blomia tropicalis in patients with respiratory allergy].

House dust mites are important environmental allergens in Southern Brazil. Some allergic patients do not respond to specific immunotherapy for Dermatophagoides ssp. For that reason we decided to evaluate the skin test sensitivity to tropical mites, Blomia tropicalis and Blomia kulagini. Patients with perennial allergic rhinitis and/or asthma were selected for tests. (N = 135). The wheals cross diameter were recorded. The frequency of strong reactions were 82% for B. tropicalis, 69% for D. pteronyssinus, 56% for D. farinae and 46% for B. kulagini. Wheals were larger with Dermatophagoides sp in

Peromyscus maniculatus, a possible reservoir host of Borrelia garinii from the Gannet Islands off Newfoundland and Labrador.

Thirty-five deer mice, Peromyscus maniculatus, were trapped on Gannet Cluster 2 (GC-2), one of a group of islands numbered by convention in the Gannet Island Archipelago, and examined for ectoparasites. One species each of Acari (Ixodes uriae) and Siphonaptera (Orchopeas leucopus) were recovered. Samples of mice favored males to females (3.4∶1). Twenty-nine percent (10) of the mice were free of ectoparasites. Males were more heavily parasitized than females when both parasites were considered. No ticks were recovered from the female mice, while the males that were parasitized carried adult Ixo

Geographical distribution of Demodex ursi in black bears from Florida.

From May 1990 to February 1996, 55 black bears (Ursus americanus floridanus) (38 males, 17 females) from 21 counties in Florida (USA), were examined for Demodex ursi (Acari: Demodecidae). The sample included cubs < 1-yr-old (n = 12), juveniles (1 to < 3 yr, n = 20), and adults (3 to 12 yr, n = 23). Demodex ursi was collected from four bears by deep scrapings of 15 sites on the head and forelimb of each bear. All four bears with demodicosis had crusty, scaly skin lesions, and moderate alopecia. No D. ursi were seen in any of the 750 scrapings from the other 51 bears, nor did any of those bears

Spider genomes provide insight into composition and evolution of venom and silk.

Spiders are ecologically important predators with complex venom and extraordinarily tough silk that enables capture of large prey. Here we present the assembled genome of the social velvet spider and a draft assembly of the tarantula genome that represent two major taxonomic groups of spiders. The spider genomes are large with short exons and long introns, reminiscent of mammalian genomes. Phylogenetic analyses place spiders and ticks as sister groups supporting polyphyly of the Acari. Complex sets of venom and silk genes/proteins are identified. We find that venom genes evolved by sequential

[Biological pollution and allergic diseases].

House dust mites, pets, microorganisms such as fungi and bacteria are the main causes of indoor allergens. The diseases correlated to the presence of these allergens are of increasing importance in public health as well as in occupational medicine. Indoor allergens are widespread in residential buildings as well as in public and in office buildings. Surveys conducted in Italian office buildings demonstrated detectable allergen concentrations in most of these buildings. In some cases, the concentrations were higher than the proposed risk threshold for allergenic sensitisation or for the elicita

Psoroptes ovis mange in a dog in Brazil.

Psoroptes ovis (Acari: Psoroptidae, Hering, 1838) is an ectoparasite that poses a high risk to herd animals and causes a severe dermal disease called psoroptic mange. This work aimed to report P. ovis parasitism in a dog. A male Pinscher that lives in rural areas and had free access to sheep farming showed hair loss in the dorsal region and mild itching. Dermatological examination demonstrated irregular alopecia, scabs, dry desquamation and erythema around the dorsal region of the thorax and pelvis. Hemogram has shown mild eosinophilia and the fungal culture was negative. In skin scrapings, P.

Integrated metatranscriptomics and metaproteomics for the characterization of bacterial microbiota in unfed Ixodes ricinus.

An innovative metaomics approach integrating metatranscriptomics and metaproteomics was used to characterize bacterial communities in the microbiota of the Lyme borreliosis spirochete vector, Ixodes ricinus (Acari: Ixodidae). Whole internal tissues and salivary glands from unfed larvae and female ticks, respectively were used. Reused I. ricinus RNA-sequencing data for metranscriptomics analysis together with metaproteomics provided a better characterization of tick bacterial microbiota by increasing bacteria identification and support for identified bacteria with putative functional implicatio
