[Tinidazole: a classical anaerobical drug with multiple potential uses nowadays].

Tinidazole is a 5-nitroimidazole active in vitro against a wide variety of anaerobic bacteria and protozoa. Tinidazole is an effective treatment against anaerobic microorganisms based on its pharmacokinetic characteristics (C(max) 51 microg/ml, t(1/2) 12.5 h) and its excellent in vitro activity. Its long half-life allows once a day regimens. Tinidazole is as effective as metronidazole in the treatment of infections caused by T. vaginalis, giardiasis and amebiasis and bacterial vaginosis, malaria, odontogenic infections, anaerobic bacterial infections (pelvic inflammatory disease, diabetic foot

A possible cluster of sexually transmitted Entamoeba histolytica: genetic analysis of a highly virulent strain.

BACKGROUND: Transmission of Entamoeba histolytica generally occurs by fecal excretion of cysts followed by oral ingestion of contaminated food or water. However, fecal-oral transmission may occur within households and long-term care institutions, and sexual transmission occurs among men who have sex with men. Epidemiologically linked clusters of E. histolytica infection are rare in industrialized countries. We report such a sexually linked cluster in Canada. METHODS: An index case involving a young female with an amebic liver abscess led to an epidemiological investigation of sexual contacts.

Persistence of Entamoeba histolytica infection in CBA mice owes to intestinal IL-4 production and inhibition of protective IFN-gamma.

The mechanisms whereby certain mouse strains develop persistent intestinal infection with Entamoeba histolytica remain unclear. In this work, we characterized the kinetic pattern of cytokine responses during the course of natural infection in CBA mice and showed that intracecal amebic infection led to a rapid and sustained upregulation of Th2 (IL-4, IL-5, IL-13) and Th17 cytokine responses while Th1 cytokines, IL-12p35 and interferon (IFN)-gamma, were suppressed. Depletion of IL-4 cleared infection by 14 days post-challenge, and this clearance correlated with and was mediated by IFN-gamma. The

The use of an ELISPOT assay to evaluate intestinal and systemic antibody responses to locally administered Entamoeba histolytica antigen in mice.

The local induction of humoral immune response to Entamoeba histolytica trophozoites in the gut has not been studied. This work reports some of our recent studies aimed to induce optimal immune responses against E. histolytica in mice and to describe a novel approach for monitoring mucosal immune responses induced in the gastrointestinal tract and expressed locally and systemically. We have compared the kinetics of both mucosal and systemic primary antibody responses to E. histolytica in the Peyer's patches (PP) and the spleen in Balb/c mice after a single dose of glutaraldehyde fixed amebas (

A survey of Entamoeba histolytica and Entamoeba dispar (Brumpt) infections on Mahé, the Seychelles.

E. histolytica, pathogenic, and E. dispar, non-pathogenic, being morphologically identical are, when recovered as cysts in feces, diagnosed as E. histolytica. A survey on The Seychelles demonstrated that when culture and zymodeme characterization was used to compare against microscopy alone, the risk of overdiagnosis of E. histolytica infections was drastically reduced. From a total of 313 subjects tested 21 cultures grew amebas. By zymodeme analysis eight were E. histolytica, 40 were E. dispar and the remainder were various species of non-pathogenic intestinal amebas. Two further small compar

Dot-enzyme-linked immunosorbent assay (Dot-ELISA) of anti-Entamoeba histolytica antibodies in human serum and colostrum.

Seroepidemiologic and clinical studies of amoebiasis require improved methods for the detection and titration of anti-Entamoeba histolytica (Eh) trophozoite antibodies. We have developed a Dot-enzyme-linked immunosorbent assay (Dot-ELISA) to titrate anti-Eh antibodies in human serum and colostrum. The anti-Eh serum IgG antibody titers from healthy individuals and from patients with amoebic liver abscess obtained, with our Dot-ELISA were similar to those obtained with a standard ELISA, but the background values were lower. Significant anti-Eh sIgA antibody titers were found in seven out of 42 c

Patients treated for amebic liver abscess develop cell-mediated immune responses effective in vitro against Entamoeba histolytica.

We studied the afferent and efferent cell-mediated immune response in 15 patients treated for amebic liver abscess. Patients had a lower T4 to T8 ratio (1.25 +/- 0.65) compared with age- and sex-matched controls (1.89 +/- 0.44, p less than 0.01) due to a decrease in T4-"helper" cells and an increase in T8-"suppressor" cells (p less than 0.01). The in vitro proliferative response of patient T lymphocytes to the plant mitogen concanavalin A (Con A) was depressed; responses to phytohemagglutinin were not. The proliferative response of patient lymphocytes to an amebic soluble protein preparation (

Case report: multiple and atypical amoebic cerebral abscesses resistant to treatment.

BACKGROUND: The parasite Entamoeba histolytica is the causal agent of amoebiasis, a worldwide emerging disease. Amebic brain abscess is a form of invasive amebiasis that is both rare and frequently lethal. This condition always begins with the infection of the colon by E. histolytica trophozoites, which subsequently travel through the bloodstream to extraintestinal tissues. CASE PRESENTATION: We report a case of a 71-year-old female who reported an altered state of consciousness, disorientation, sleepiness and memory loss. She had no history of hepatic or intestinal amoebiasis. A preliminary

Longitudinal evaluation of enteric protozoa in Haitian children by stool exam and multiplex serologic assay.

Haitian children were monitored longitudinally in a filariasis study. Included were stool samples examined for Giardia intestinalis and Entamoeba histolytica cysts, and serum specimens analyzed for immunoglobulin G (IgG) responses to eight recombinant antigens from G. intestinalis (variant-specific surface protein [VSP1-VSP5]), E. histolytica (lectin adhesion molecule [LecA]), and Cryptosporidium parvum (17- and 27-kDa) using a multiplex bead assay. The IgG responses to VSP antigens peaked at 2 years of age and then diminished and were significantly lower (P < 0.002) in children > 4.5 years th

A Multilocus Sequence Typing System (MLST) reveals a high level of diversity and a genetic component to Entamoeba histolytica virulence.

BACKGROUND: The outcome of an Entamoeba histolytica infection is variable and can result in either asymptomatic carriage, immediate or latent disease (diarrhea/dysentery/amebic liver abscess). An E. histolytica multilocus genotyping system based on tRNA gene-linked arrays has shown that genetic differences exist among parasites isolated from patients with different symptoms however, the tRNA gene-linked arrays cannot be located in the current assembly of the E. histolytica Reference genome (strain HM-1:IMSS) and are highly variable. RESULTS: To probe the population structure of E. histolytica

Efficacy of a Gal-lectin subunit vaccine against experimental Entamoeba histolytica infection and colitis in baboons (Papio sp.).

To determine the efficacy of a Gal-lectin based intranasal synthetic peptide vaccine, we developed a new experimental primate model of Entamoeba histolytica intestinal infection. Release of xenic E. histolytica trophozoites (5×10(6)) into the small bowel of baboons (Papio sp.) resulted in a rapid intestinal anti-amebic antibody response and a brief infection; however, release of trophozoites directly into the cecum (5 baboons) elicited a sustained E. histolytica infection, as determined by quantitative fecal PCR, and an ulcerative, inflammatory colitis observed on colonoscopy and histopatholog

Immunology of Entamoeba histolytica in human and animal hosts.

Although Entamoeba histolytica induces humoral and cellular immune responses in both human and animal hosts, there is no indication of postinfection immunity in humans; in contrast, several other mammals are protected by prior infection or immunization. The exacerbation of the disease by immunosuppression suggests a protective function of still-unknown defense mechanisms. Specific local and circulating antibodies are produced regularly during invasive amebiasis. Although serum antibodies, together with complement, are lytic to the trophozoites in vitro, the poor correlation of these antibodies

Amebiasis: host-pathogen biology.

Invasive amebiasis caused by Entamoeba histolytica, and particularly amebic liver abscess, is a major public health problem in Mexico and some other countries because of the high incidence and mortality of this disease. This paper first discusses the pathogenic effect of E. histolytica and the defensive response of the host and then reports studies concerning the experimental induction of protective immunity to amebic infection. The pathogenic effect of E. histolytica is probably initiated by a lectin-mediated adhesion of trophozoites to target cells; the adhesion is followed by cytopathic act

Assessment of three major intestinal protozoan infections in Kermanshah, Iran: a pre- and post-COVID-19 study.

Intestinal parasitic infections continue to be a significant public health concern worldwide. This study examines the impact of the COVID-19 pandemic on the prevalence of three intestinal protozoa—Blastocystis, Entamoeba coli, and Giardia lamblia—among patients at the Imam Reza Health Complex in Kermanshah, Iran, from March 2017 to March 2024. Patient data were obtained from the Health Information System and analyzed across seven age groups using descriptive and inferential statistics. A total of 15,018 individuals participated in the survey, consisting of 7169 males and 7849 females. The over

Application of real-time polymerase chain reaction in detection of Entamoeba histolytica in pus aspirates of liver abscess patients.

Entamoeba histolytica is the second major cause of liver abscess disease in humans, particularly in developing countries. Recently, DNA molecular-based methods have been employed to enhance the detection of E. histolytica in either pus or stool specimens. In this study, the results of real-time polymerase chain reaction (PCR) to detect E. histolytica DNA in pus from liver abscess cases were compared with those of indirect hemagglutination assay on the corresponding serum samples. Bacterial cultures were also performed on the pus samples for the diagnosis of pyogenic liver abscess. The real-tim

Comparison of the proteome profiles of Entamoeba histolytica and its close but non-pathogenic relative Entamoeba dispar.

The microaerophilic protist Entamoeba histolytica is a source of considerable morbidity and mortality in many countries of the world by acting as a causative agent of amoebic dysentery and liver abscess. Its close relative, Entamoeba dispar, also colonises the human colon but is non-pathogenic. In the present study, we wanted to assess if the close relatedness of these two organisms could be used to identify virulence factors in E. histolytica through a comparison of the protein expression profiles of the pathogenic and the non-pathogenic species. We applied two-dimensional polyacrylamide gel
