Adaptation of the spiders to the environment: the case of some Chilean species.

Spiders are small arthropods that have colonized terrestrial environments. These impose three main problems: (i) terrestrial habitats have large fluctuations in temperature and humidity; (ii) the internal concentration of water is higher than the external environment in spiders, which exposes them continually to water loss; and (iii) their small body size determines a large surface/volume ratio, affecting energy exchange and influencing the life strategy. In this review we focus on body design, energetic, thermal selection, and water balance characteristics of some spider species present in Ch

The Host-Symbiont-Pathogen Triad in Bathymodiolus azoricus: The Multifunctional Gill at the Deep-Sea Interface.

Deep-sea hydrothermal vents and cold seeps sustain highly productive animal communities through chemosynthetic symbioses, among which bathymodioline mussels are prominent examples. Bathymodioline gill bacteriocytes accommodate intracellular chemosynthetic symbionts, including sulfur- and/or methane-oxidizing bacteria depending on the host species, while remaining sheltered in an epithelium continuously exposed to environmental microorganisms, creating a fundamental immunological problem: how can an innate defense system remain effective without eliminating the microbial partners on which host

Integrated In Silico, In Vivo, and Deep Learning Approaches in the Discovery of Novel Candidate Molecules for Aedes aegypti Control.

The mosquito Aedes aegypti is a primary vector responsible for transmitting major arboviruses, including dengue, Zika, chikungunya, and yellow fever. Increasing resistance to conventional synthetic insecticides, combined with their well-known environmental drawbacks, underscores the urgent need for more selective, sustainable, and effective strategies for vector control. Chalcones have been previously identified by our research group as a promising chemical class of larvicidal agents, with preliminary evidence for distinct mechanisms of action. More recently, an additional strategy for integra

Cellular direct conversion by cell penetrable OCT4-30Kc19 protein and BMP4 growth factor.

BACKGROUND: The number of patients suffering from osteoporosis is increasing as the elderly population increases. The demand for investigating bone regeneration strategies naturally arises. One of the approaches to induce bone regeneration is somatic cell transdifferentiation. Among the transcriptional regulators for transdifferentiation, octamer-binding transcription factor 4 (OCT4) is famous for its role in the regulation of pluripotency of stem cells. Bone morphogenetic protein 4 (BMP4) is another factor that is known to have a significant role in osteogenic differentiation. Previous studie

Leptinotoxin-h action in synaptosomes and neurosecretory cells: stimulation of neurotransmitter release.

Guinea pig brain cortex synaptosomes and neurosecretory PC12 cells were loaded with [3H]3,4-dihydroxyphenylethylamine ([3H]DA, [3H]dopamine) and then exposed to leptinotoxin-h (LPTx) (purified and partially purified preparations, obtained from the hemolymph of Leptinotarsa haldemani). In a Ca2+-containing Ringer medium the toxin induced prompt and massive release of the neurotransmitter. Half-maximal effects were obtained at concentrations estimated of approximately 3 X 10(-11) M for synaptosomes, and 1.5 X 10(-10) M for PC12 cells. Release responses in the two experimental systems investigate

DENV-2 3'UTR dumbbell structure is a critical factor for viral infection and dissemination in Aedes mosquito.

UNLABELLED: Dengue virus (DENV) is a mosquito-borne flavivirus with the capability of replicating and spreading between vertebrate and invertebrate hosts, which poses a huge threat to global public health. Understanding the molecular mechanism of its replication and transmission in mosquitoes is crucial for controlling the dengue epidemic. The 3' untranslated region (UTR) has been identified as a mediator for viral replication, pathogenicity, and transmission, but its effect on DENV in the mosquito host remains unknown. This study investigated the role of the dumbbell (DB) structure within the

Genome sequence of the Drosophila melanogaster male-killing Spiroplasma strain MSRO endosymbiont.

UNLABELLED: Spiroplasmas are helical and motile members of a cell wall-less eubacterial group called Mollicutes. Although all spiroplasmas are associated with arthropods, they exhibit great diversity with respect to both their modes of transmission and their effects on their hosts; ranging from horizontally transmitted pathogens and commensals to endosymbionts that are transmitted transovarially (i.e., from mother to offspring). Here we provide the first genome sequence, along with proteomic validation, of an endosymbiotic inherited Spiroplasma bacterium, the Spiroplasma poulsonii MSRO strain

The Anti-Biofilm Efficacy of Caffeic Acid Phenethyl Ester (CAPE) In Vitro and a Murine Model of Oral Candidiasis.

Candida albicans is the main fungal species associated with the development of oral candidiasis. Currently, therapeutic options for these infections are limited by the adverse effects of antifungal drugs and by the emergence of drug resistant strains. Thus, the development of new antifungal agents is needed for the prevention and treatment of oral Candida infections. Caffeic acid phenethyl ester (CAPE) is a natural compound from propolis polyphenolic groups that exhibits many pharmacological properties. In this study, we investigated whether CAPE can have antifungal and immunomodulatory effect

The phenoloxidase activity and antibacterial function of a tyrosinase from scallop Chlamys farreri.

Tyrosinase (TYR), also known as monophenol monooxygenase, is a ubiquitous binuclear copper-containing enzyme which catalyzes the hydroxylation of phenols to catechols and the oxidation of catechols to quinones. In the present study, the cDNA of a tyrosinase (CfTYR) was identified from scallop Chlamys farreri, which encoded a polypeptide of 486 amino acids. The CfTYR mRNA transcripts were expressed in all the tested tissues, including haemocytes, adductor muscle, kidney, hepatopancreas, gill, gonad and mantle, with the highest level in mantle. The expression level of CfTYR mRNA in haemocytes de

Oxidative stress and genotoxic effect of deltamethrin exposure on the Chinese mitten crab, Eriocheir sinensis.

The oxidative stress and genotoxic effect of deltamethrin on the Chinese mitten crab Eriocheir sinensis were assessed using several commonly used biomarkers in this study. The results showed that the 48 h and 96 h LC50 values of deltamethrin to E. sinensis were 2.319 and 1.164 μg/L, respectively, and the safe concentration was 0.293 μg/L. According to these results, deltamethrin was applied at concentrations of 1/16, 1/8, 1/4, 1/2 and 1/1 96 h LC50 for 8 d in an exposure experiment. The activity of superoxide dismutase (SOD) increased remarkably at 1 d, but decreased at 4 d in concentration gr

Sequence-structure-function relations of the mosquito leucine-rich repeat immune proteins.

BACKGROUND: The discovery and characterisation of factors governing innate immune responses in insects has driven the elucidation of many immune system components in mammals and other organisms. Focusing on the immune system responses of the malaria mosquito, Anopheles gambiae, has uncovered an array of components and mechanisms involved in defence against pathogen infections. Two of these immune factors are LRIM1 and APL1C, which are leucine-rich repeat (LRR) containing proteins that activate complement-like defence responses against malaria parasites. In addition to their LRR domains, these

Bioaccumulative potential and toxicity of endosulfan insecticide to non-target animals.

1. Endosulfan insecticide is a polychlorinated compound used for controlling a variety of insects; it is practically water-insoluble, but readily adheres to clay particles and persists in soil and water for several years. Its mode of action involves repetitive nerve-discharges positively correlated to increase in temperature. This compound is extremely toxic to most fish and can cause massive mortalities. In fish, it causes marked changes in Na and K concentrations, decrease in blood Ca(2+) and Mg levels and inhibits Na, K and Mg-dependent ATPase (in brain). 2. Bioaccumulation of endosulfan is

Monitoring fatty acid trafficking during Drosophila melanogaster oogenesis reveals a role for the triglyceride synthase DGAT1 in protecting mitochondrial integrity.

UNLABELLED: Successful oogenesis requires the precise coordination of nutrient uptake, storage, and utilization to meet the high metabolic demands of egg production. In mammals, fatty acid (FA) metabolism has emerged as a key driver of oocyte maturation; however, the mechanisms by which follicles regulate FA trafficking and utilization remain poorly understood across all systems. To address these issues, we leverage the genetic tractability of Drosophila melanogaster oogenesis. We found that nurse cell mitochondria are metabolically active and catabolize FA in a stage-dependent manner, with fa

A tailless Dscam from Eriocheir sinensis diversified by alternative splicing.

Dscam (Down syndrome cell adhesion molecule), a member of the immunoglobulin superfamily (IgSF), plays an essential role in pathogen recognition and further involves in the innate defense of invertebrates. In the present study, the cDNA of a Dscam from Chinese mitten crab Eriocheir sinensis (designated EsDscam) was cloned and characterized. It contained a 5-terminal untranslated region (UTR) of 60 bp, a 3-UTR of 216 bp with a poly (A) tail, and an open reading frame (ORF) of 4848 bp encoding a polypeptide of 1615 amino acids with the putative molecular mass of 178.4 kDa and theoretical isoelec

Rhipicephalus sanguineus: vector of a new spotted fever group rickettsia in the United States.

A rickettsia related to but distinct from the spotted fever agent, Rickettsia rickettsii, has been detected in 167 (18.9%) of 884 Rhipicephalus sanguineus taken off dogs in central and northern Mississippi. The organisms could readily be isolated in male meadow voles (Microtus pennsylvanicus), where it produced massive infections in the tissues of tunica vaginalis. It was practically nonpathogenic for male guinea pigs, although inoculation of these animals with infected tunica vaginalis of voles afforded in 30 of 38 instances solid immunity to challenge with virulent R. rickettsii.
