Novel SARS-COV2 poly epitope phage-based candidate vaccine and its immunogenicity.

BACKGROUND AND PURPOSE: The global emergence of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has prompted widespread concern. Bacteriophages have recently gained attention as a cost-effective and stable alternative for vaccine development due to their adjuvant properties. This study aimed to design and validate a poly epitope composed of viral proteins. EXPERIMENTAL APPROACH: SARS-CoV-2 proteins (spike, nucleocapsid, membrane, envelope, papain-like protease, and RNA-dependent RNA polymerase) were selected for analysis. Immunoinformatic methods were employed to predict B an

Dual template (epitope) imprinted electrode for sensing bacterial protein with high selectivity.

Epitope imprinting has shown better prospects to synthesize synthetic receptors for proteins. Here, dual epitope imprinted polymer electrode (DEIP) matrix was fabricated on gold surface of electrochemical quartz crystal microbalance (EQCM) for recognition of target epitope sequence in blood samples of patients suffering from brain fever. Epitope sequences from outer membrane protein Por B of Neisseria meningitidis (MC58) bacteria predicted through immunoinformatic tools were chosen for imprinting. Self-assembled monolayers (SAM) of cysteine appended epitope sequences on gold nanoparticles were

Evaluation of a Human T Cell-Targeted Multi-Epitope Vaccine for Q Fever in Animal Models of Coxiella burnetii Immunity.

T cell-mediated immunity plays a central role in the control and clearance of intracellular Coxiella burnetii infection, which can cause Q fever. Therefore, we aimed to develop a novel T cell-targeted vaccine that induces pathogen-specific cell-mediated immunity to protect against Q fever in humans while avoiding the reactogenicity of the current inactivated whole cell vaccine. Human HLA class II T cell epitopes from C. burnetii were previously identified and selected by immunoinformatic predictions of HLA binding, conservation in multiple C. burnetii isolates, and low potential for cross-reac

Rational design of multi epitope-based subunit vaccine by exploring MERS-COV proteome: Reverse vaccinology and molecular docking approach.

Middle East respiratory syndrome (MERS-COV), first identified in Saudi Arabia, was caused by a novel strain of coronavirus. Outbreaks were recorded from different regions of the world, especially South Korea and the Middle East, and were correlated with a 35% mortality rate. MERS-COV is a single-stranded, positive RNA virus that reaches the host by binding to the receptor of dipeptidyl-peptides. Because of the unavailability of the vaccine available for the protection from MERS-COV infection, the rapid case detection, isolation, infection prevention has been recommended to combat MERS-COV infe

Identification and selection of immunodominant B and T cell epitopes for dengue multi-epitope-based vaccine.

Dengue virus (DENV) comprises four serotypes (DENV1-4) which cause 390 million global infections with 500,000 hospitalizations and 25,000 fatalities annually. Currently, the only FDA approved DENV vaccine is the chimeric live-attenuated vaccine, Dengvaxia®, which is based on the yellow fever virus (YFV) genome that carries the prM and E genes of the respective DENV 1, 2, 3, and 4 serotypes. However, it has lower efficacies against serotypes DENV1 (51%) and DENV2 (34%) when compared with DENV3 (75%) and DENV4 (77%). The absence of T cell epitopes from non-structural (NS) and capsid (C) proteins

scImmuneCo: a compendium of cell-type-specific functional modules for decoding immune responses from single-cell RNA-seq data.

Traditional, knowledge-driven pathway annotations and bulk transcriptomic analyses often fail to capture the cellular specificity and mechanistic heterogeneity of immune responses. We present scImmuneCo, a comprehensive resource of immune cell-specific co-expression modules derived from single-cell RNA sequencing across 17 immunological conditions and 1.78 million cells. Using a modified graph-based framework, we constructed 873 robust modules spanning 7 major immune cell types, providing stable, cell-type-specific interaction networks for functional inference. scImmuneCo resolves complex biol

Computational and experimental mapping of antigenic epitopes on iridovirus major capsid protein for precision vaccine design.

The major capsid protein (MCP), owing to its strong immunogenicity, has emerged as a critical target for largemouth bass ranavirus (LMBV) vaccine development. However, its key antigenic epitopes remain uncharacterized. This study integrated phage display technology with bioinformatics approaches to design a multiepitope vaccine against LMBV. By constructing a phage Ab library derived from LMBV-infected largemouth bass spleen tissues, we obtained high-affinity Abs (Ab-S1 and Ab-S2) capable of specifically binding LMBV. Through molecular docking and alanine scanning mutagenesis of MCP-Ab complex

Computational identification of conserved dengue virus envelope protein epitopes for vaccine design and immunodiagnostic platform.

Dengue virus (DENV) remains a major global health challenge, with its incidence increasing sharply over the past two decades. More than 390 million infections occur annually, affecting nearly half of the world’s population. Despite progress in vaccine research, the coexistence of four antigenically distinct dengue virus serotypes (DENV1-4) complicates vaccine and diagnostic development, emphasizing the urgent need for improved preventive and diagnostic strategies. Immunoinformatics and computational vaccinology are powerful tools to elucidate host–pathogen interactions and accelerate vaccine a

In silico Designing of a Multiepitope Vaccine using the Hallmark Proteins of Mycobacterium tuberculosis Involved in Different Aspects of Virulence.

BACKGROUND: Mycobacterium tuberculosis ( M.tb ) remains a leading global cause of mortality. The current Bacillus Calmette-Guérin vaccine lacks efficacy in adults and fails to generate a long-term memory response. With the rise of multidrug-resistant strains, there is an urgent need for novel vaccines that can provide broader protection. This study aimed to design a multiepitope vaccine (MEV) targeting hallmark proteins involved in different aspects of M.tb virulence. METHODS: Four unique M.tb proteins, Rv1507A (role in memory response), Rv1509 (role in phagolysosomal escape), Rv1954A (role i

Systems Immunology Approaches to Understanding Immune Responses in Acute Infection of Yellow Fever Patients.

In the 2018 yellow fever (YF) outbreak in Brazil, we generated new transcriptomic data and combined it with clinical and immunological data to decode the pathogenesis of YF. Analyzing 79 patients, we found distinct gene expression patterns between acute YF, other viral infections, and the milder YF-17D vaccine infection. We identified a critical role for low-density, immature neutrophils in severe outcomes, marked by the downregulation of genes essential for neutrophil migration and maturation, such as PADI4, CSF3R, and ICAM1, in deceased patients. Our study also revealed complex interactions

A novel multiepitope peptide vaccine with intrinsic adjuvant confers protective immunity in mice against tuberculosis.

BACKGROUND: Tuberculosis remains a global health challenge with the only licensed vaccine, Bacillus Calmette-Guérin (BCG). Latent tuberculosis serves as a reservoir for TB infection. Over the past few years, protein subunit vaccines have gained research attention due to their safety and versatility. This study aimed to assess the protective efficacy of an immunoinformatically designed, expressed, and purified synthetic peptide vaccine candidate, VC28, incorporating epitopes from late-stage DosR proteins. The immunogenicity and protective efficacy of the VC28 vaccine were assessed in BALB/c mic

Design of a Multi-Epitope Vaccine Based on Fasciola gigantica Cathepsin B and Evaluation of Immunological Responses in Mice.

Fasciola gigantica (F. gigantica) is a vital parasite that causes fasciolosis. Liver fluke infections affect livestock animals, and the Fasciola species (Fasciola spp.) vaccine has been tested for many types of these diseases. Currently, computer-based vaccine design represents an attractive alternative for constructing vaccines. Thus, this study aimed to design the epitopes of linear B-cells (BCL) and helper T lymphocytes (HTL) using an immunoinformatic approach and to investigate in silico and the mice's immune response. A non-conserved host region, overlapping F. gigantica cathepsin B prote

In silico analysis for the development of multi-epitope vaccines against Mycobacterium tuberculosis.

As Bacille Calmette-Guérin (BCG) vaccine's effectiveness is limited to only children, the development of new tuberculosis (TB) vaccines is being studied using several platforms, and a novel TB vaccine that overcomes this limitation is required. In this study, we designed an effective multi-epitope vaccine against Mycobacterium tuberculosis using immunoinformatic analysis. First, we selected 11 highly antigenic proteins based on previous research: Ag85A, Ag85B, Ag85C, ESAT-6, MPT64, Rv2660c, TB10.4, HspX, GlfT2, Fas, and IniB. Among these antigens, 10 linear B-cell epitopes, 9 helper T-cell epi

Identification of a Potential Vaccine against Treponema pallidum Using Subtractive Proteomics and Reverse-Vaccinology Approaches.

Syphilis, a sexually transmitted infection, is a deadly disease caused by Treponema pallidum. It is a Gram-negative spirochete that can infect nearly every organ of the human body. It can be transmitted both sexually and perinatally. Since syphilis is the second most fatal sexually transmitted disease after AIDS, an efficient vaccine candidate is needed to establish long-term protection against infections by T. pallidum. This study used reverse-vaccinology-based immunoinformatic pathway subtractive proteomics to find the best antigenic proteins for multi-epitope vaccine production. Six essenti

Bioinformatics-Driven Design and Evaluation of Recombinant Multi-Epitope Immunogens Derived From Snake Venom Toxins as Potential Antivenom Candidates.

Snakebite envenomation is a major public health concern, particularly in low- and middle-income regions where access to safe and effective antivenoms is limited. Traditional antivenoms, derived from immunization with crude venom, often trigger adverse reactions and lack specificity against key venom components. This study presents a bioinformatics-driven approach to design, construct, and evaluate a novel panel of recombinant multi-epitope toxin-derived immunogens targeting the most clinically significant snake venom toxin families. Five principal toxin families-three-finger toxins (3FT), phos

Germline based SARS-CoV-2 specific B cell repertoire motif identified with novel sequence based bioinformatic pipeline.

Human antibody diversification, achieved through gene selection and somatic hypermutation (SHM), is critical for protecting against diverse pathogens. This study investigates whether specific immune responses possess distinct receptor sequence patterns that differentiate them from the general immune repertoire. Utilizing data from an anti-SARS-CoV-2 vaccination study, we analyzed two properties of SARS-CoV-2 specific memory B-cells and compared them to the background immune repertoire. Driven by somatic hypermutation (SHM), B cells exhibit a highly dynamic nature. Consequently, groups sharing
