Population-Specific Probabilistic Risk Assessment of PFOA for Ovarian Function Using PBTK Translation of Data from Subacute Exposure of Mice.

We assessed the risk of perfluorooctanoic acid (PFOA) to ovarian function by integrating in vivo mouse data with physiologically based toxicokinetic (PBTK) modeling and population-level analysis. Adult mice received PFOA (0.06, 1.15, 22 mg/kg bw/day; 14 days), followed by analysis of ovarian histology and whole-ovary transcriptomics. Effect doses were converted to human equivalent doses (HEDs) and propagated through a PBTK model to estimate ovarian bioactive concentrations (OBCs) in human ovaries. Median histology HEDs were 41 and 501 mg/week, whereas transcriptomic HED was 322 mg/week values

Chamuangone purified from Garcinia cowa inhibits inflammation, angiogenesis, epithelial-mesenchymal transition, and metastasis in breast cancer.

BACKGROUND: Breast cancer (BC) is a life-threatening disease, with approximately 60% of cases progressing to breast cancer lung metastasis (BCLM). Chamuangone, a key bioactive constituent derived from the leaves of the Thai plant Garcinia cowa Roxb., has demonstrated significant potential as an anticancer agent. To evaluate its anticancer effects, chamuangone was purified from the leaves and assessed through in silico, in vitro, and in vivo studies using a mouse model of BC. RESULTS: The results revealed that chamuangone (10 mg L-1) inhibited cancer cell proliferation by inducing G0/G1 phase

Deriving a Health-Based Guidance Value for 9,10-Anthraquinone via integrating PBTK modeling-based reverse dosimetry and In Vitro bioassays.

Anthraquinones, both naturally occurring and synthetic, are widely distributed in the environment. Recent years, human exposure to 9,10-anthraquinone (9,10-AQ) through contaminated food has been raising significant health concerns due to its potential toxicity upon chronic exposure. Among these, 9,10-AQ has been studied in traditional toxicology, with few of established Points of Departure (PoDs) and Health-Based Guidance Values (HBGV). However, toxicological data for other anthraquinones remain severely limited. Traditional animal experiments are resource-intensive and time-consuming, restric

Autopsy, thanatopraxy, cemeteries and crematoria as hotspots of toxic organic contaminants in the funeral industry continuum.

The occurrence and health risks of toxic organic contaminants (TOCs) in the funeral industry are relatively under-studied compared to other industries. An increasing body of literature reports TOCs including emerging contaminants in the funeral industry, but comprehensive reviews of the evidence are still lacking. Hence, evidence was analysed to address the proposition that, the funeral industry constitutes several hotspot reservoirs of a wide spectrum of TOCs posing ecological and human health risks. TOCs detected include embalming products, persistent organic pollutants, synthetic pesticides

Cohort study of workers at a New Zealand agrochemical plant to assess the effect of dioxin exposure on mortality.

OBJECTIVES: To describe how the exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) influenced mortality in a cohort of workers who were exposed more recently, and at lower levels, than other cohorts of trichlorophenol process workers. DESIGN: A cohort study. SETTING: An agrochemical plant in New Zealand PARTICIPANTS: 1,599 men and women working between 1 January 1969 and 1 November 1988 at a plant producing the herbicide 2,4,5-trichlorophenoxyacetic acid (2,4,5-T) with TCDD as a contaminant. Cumulative TCDD exposure was estimated for each individual in the study by a toxicokinetic model.

Could the environmental toxicity of diclofenac in vultures been predictable if preclinical testing methodology were applied?

Diclofenac, a non-steroidal anti-inflammatory pharmaceutical agent was responsible for the death of millions of Gyps vulture's in the Indian sub-region with the safety of the other non-steroidal anti-inflammatory drugs (NSAIDs) being questionable. With preclinical safety testing not well established for avian species unlike for mammalian and environmental toxicity, we ask the question if a preclinical model could have predicted the toxic effect of the drug. For this study, we test an Organisation for Economic Co-operation and Development (OECD) guideline 223 for assessing the acute toxic poten

Trichloroethylene risk assessment: a review and commentary.

Trichloroethylene (TCE) is a widespread environmental contaminant that is carcinogenic when given in high, chronic doses to certain strains of mice and rats. The capacity of TCE to cause cancer in humans is less clear. The current maximum contaminant level (MCL) of 5 ppb (microg/L) is based on an US Environment Protection Agency (USEPA) policy decision rather than the underlying science. In view of major advances in understanding the etiology and mechanisms of chemically induced cancer, USEPA began in the late 1990s to revise its guidelines for cancer risk assessment. TCE was chosen as the pil

Assessment of developmental toxicity of vorinostat, a histone deacetylase inhibitor, in Sprague-Dawley rats and Dutch Belted rabbits.

BACKGROUND: The developmental toxicity potential of vorinostat (suberoylanilide hydroxamic acid [SAHA], ZOLINZA), a potent inhibitor of histone deacetylase (HDAC), was assessed in Sprague-Dawley rats and Dutch Belted rabbits. HDAC inhibitors have been shown to mediate the regulation of gene expression, induce cell growth, cell differentiation, and apoptosis of tumor cells. Range-finding studies established oral dose levels of 5, 15, or 50 mg/kg/day and 20, 50, or 150 mg/kg/day in rats and rabbits, respectively. METHODS: Animals were dosed on Gestation Days 6-20 or 7-20, respectively, with lit

An intervention analysis for the reduction of exposure to methylmercury from the consumption of seafood by women of child-bearing age.

A previously developed exposure model was used [Risk Anal. 22 (2002) 689] to assess the effectiveness of various advisory scenarios on minimizing mercury (Hg) blood levels via the consumption of commercial seafood, both finfish and shellfish. This exposure model was developed to predict levels of Hg in blood in women of child-bearing age in the US based on the frequency of seafood consumption, the amount of seafood consumed per serving, and the types of seafood consumed. Steady-state relationships that employed descriptive statistics to account for toxicokinetic variation were used to predict

The effects of diet, ad libitum overfeeding, and moderate dietary restriction on the rodent bioassay: the uncontrolled variable in safety assessment.

Ad libitum (AL) overfeeding is the most significant, uncontrolled variable affecting the outcome of the current rodent bioassay. There is a highly significant correlation between AL food consumption, the resultant obesity and body weight, and low 2-yr survival in rodents. AL feeding of diets with lowered protein, metabolizable energy (ME), and increased fiber does not improve survival. Only dietary restriction (DR) of all diets tested significantly improves survival and delays the onset of spontaneous degenerative disease (i.e., nephropathy and cardiomyopathy) and diet-related tumors. Moderate

From Stable to Table: Tracing the "Forever Chemical" Vector Through the Food Animal Supply Chain.

Per- and polyfluoroalkyl substances (PFAS) represent a formidable class of persistent synthetic chemicals whose contamination of the global food supply, particularly through animal products, constitutes a major and direct route of human exposure. This review synthesizes the current scientific understanding of PFAS entry into food animal production systems including livestock, poultry, and aquaculture via contaminated feed, water, and soil. We detail the subsequent toxicokinetics in animals, which lead to pronounced tissue-specific bioaccumulation. Two distinct and non-interchangeable rankings

Human Mercury Biomonitoring Under the Minamata Convention: Blood, Urine, Hair, and Tissue Biomarkers (2000-2026).

Mercury (Hg) is a globally ubiquitous environmental pollutant classified by the World Health Organization among the chemicals of major public health concern. It occurs as elemental mercury, inorganic mercury, and organic mercury, each with distinct toxicokinetics and target organs. Human exposure arises mainly from fish and seafood consumption. Additional important sources include dental amalgam restorations, occupational settings, mercury-containing skin-lightening cosmetics, artisanal and small-scale gold mining (ASGM), and environmental remobilization of Hg naturally stored in soils and sed

Pharmacokinetics and Enterohepatic Circulation of 2-(Quinoline-8-carboxamido)benzoic Acid (2-QBA) in Mice.

The quinoline alkaloid 2-(quinoline-8-carboxamido)benzoic acid (2-QBA), which is isolated from Aspergillus sp. SCSIO06786, a deep sea-derived fungus, has been suggested as a therapeutic candidate for the treatment of Parkinson's disease. We developed an analytical method for 2-QBA using a liquid chromatography-tandem mass spectrometry (LC-MS/MS) in mouse plasma, in which a protein precipitation method for the sample preparation of 2-QBA in mouse plasma was used due to its simplicity and good extraction recovery rates (80.49-97.56%). The linearity of the calibration standard sample, inter- and

Novel Bacterial Topoisomerase Inhibitor Gepotidacin Demonstrates Absence of Fluoroquinolone-Like Arthropathy in Juvenile Rats.

Fluoroquinolone use in children is limited due to its potential toxicity and negative effects on skeletal development, but the actual effects/risks of fluoroquinolones on bone growth and the mechanisms behind fluoroquinolone-driven arthropathy remain unknown. Gepotidacin is a novel, bactericidal, first-in-class triazaacenaphthylene antibiotic with a unique mechanism of action that is not anticipated to have the same risks to bone growth as those of fluoroquinolones. Gepotidacin is in phase III clinical development for uncomplicated urinary tract infections (ClinicalTrials.gov identifiers NCT04

Pharmacokinetic considerations in clinical toxicology: clinical applications.

Pharmacokinetic and pharmacodynamic principles should be regarded in the assessment and proper management of patients exposed to a poison. Clinicians must apply these principles to make rational clinical decisions regarding the significance of the poisoning (risk assessment) and to formulate an appropriate management plan. However, pharmacokinetic processes and parameters may be changed in the patient with acute poisoning. This may result from saturation of the capacity of a number of physiological processes due to the high dose, or the toxic effects of the poison may change these processes di
