Progress in prophylaxis against nonsteroidal anti-inflammatory drug-associated ulcers and erosions. Omeprazole NSAID Steering Committee.

Four large clinical studies have shown that omeprazole, 20 mg once daily, is effective in preventing the significant gastroduodenal consequences of nonsteroidal anti-inflammatory drugs (NSAIDs). In the Scandinavian Collaborative Ulcer Recurrence (SCUR) study, patients were randomized (without initial endoscopy) to receive omeprazole or placebo for up to 3 months. Of the patients treated with omeprazole, 24.7% experienced treatment failure, compared with 50.0% of those on placebo. Fewer patients in the omeprazole group than in the placebo group developed a peptic ulcer (4.7% versus 16.7%, respe

Dynamics in the Vicinity of the Stable Halo Orbits.

BACKGROUND: Gut microbiota (GM) is closely associated with the onset of depression, in which inflammation plays an essential role. Identifying specific GM associated with depression and their mechanisms, based on the principles of predictive, preventive, and personalized medicine (PPPM), is a critical step toward achieving targeted prevention and personalized treatment for depression. WORKING HYPOTHESIS AND METHODOLOGY: We hypothesized that both gut microbiota (GM) and cytokines influence the onset of depression, with cytokines acting as mediators of GM effects on depression. To test this hyp

Microbial community characterization of multi-crop growouts in the XROOTS aeroponic-hydroponic system on the International Space Station.

Plant growth systems tested on the International Space Station (ISS) are small-area growth units that mostly use solid media. With NASA's plan to send astronauts on long-duration exploration missions, there is a need to produce larger amounts of fresh food with limited upmass and resources. The eXposed Root On-Orbit Test System (XROOTS) is an aeroponic-hydroponic nutrient delivery system designed for exploration missions and was tested on the ISS. Post-harvest samples were returned for microbiological analyses of the plant leaves, roots, and fruit from lettuce, mizuna mustard, wheat, radish, t

Gastrointestinal motility in microgravity: a critical review of multi-level mechanisms and model-dependent effects.

BACKGROUND: Gastrointestinal motility disturbances rank among the most frequently reported medical complications of spaceflight. Astronauts experience delayed gastric emptying, erratic small intestinal transit and reduced colonic propulsion. The underlying mechanisms are multifactorial. Microgravity alters intra-abdominal physical mechanics, disrupts autonomic and enteric neural circuits, shifts gastrointestinal hormone secretion profiles, inflicts oxidative stress upon effector cells, and perturbs gut microbial communities. Cross-model comparisons reveal substantial disagreement, suggesting t

Bone mineral content determined by energy X-ray absorptiometry correlates with handgrip strength in Latin American divers.

INTRODUCTION: Although a lack of gravitational stimuli decreases bone mineral content in some populations, such as astronauts' bone mineral content (BMC), little is known about the association of BMC with handgrip muscle strength (HGS) in divers of different years of diving experience. OBJECTIVES: To describe the BMC and body composition of diver workers of different years of diving experience. A second objective was to associate the level of BMC with HGS. MATERIALS AND METHODS: A descriptive and longitudinal pilot study was carried out in adult men divers of different years of diving experi

Extracellular vesicles-derived microRNAs expression as biomarkers for neurological radiation injury: Risk assessment for space exploration.

Space missions pose threats to the health of the astronauts due to long-term exposure to galactic cosmic rays and solar particle events comprised predominantly of medium to high energy protons, energetic helium ions, and energetic high atomic number particles (HZEs). While the tissue-specific effects of radiation have been studied extensively, the changes in exosomal miRNA expression levels in response to acute radiation exposure have not been assessed. Extracellular vesicles (EVs) originate from the host cells and contain nucleic acid and proteins that can modify the physiology of the receivi

Exosomes derived from cyclic mechanical stretch-exposed bone marrow mesenchymal stem cells inhibit RANKL-induced osteoclastogenesis through the NF-κB signaling pathway.

BACKGROUND: Skeletal unloading usually induces severe disuse osteoporosis (DOP), which often occurs in patients subjected to prolonged immobility or in spaceflight astronauts. Increasing evidence suggests that exosomes are important mediators in maintaining the balance between bone formation and resorption. We hypothesized that exosomes play an important role in the maintenance of bone homeostasis through intercellular communication between bone marrow mesenchymal stem cells (BMSCs) and osteoclasts under mechanical loading. METHODS: Cells were divided into cyclic mechanical stretch (CMS)-trea

Daily generation of a footward fluid shift attenuates ocular changes associated with head-down tilt bed rest.

Astronauts have presented with a constellation of visual changes referred to as spaceflight-associated neuro-ocular syndrome (SANS). However, neither have early markers of microgravity-induced optic remodeling been fully identified nor have countermeasures been developed. To identify early markers of SANS, we studied 10 subjects with optical coherence tomography and ultrasonography when upright and supine and again after 24 h of 6° head-down tilt (HDT) bed rest. Upon acute transition from the upright to the supine position, choroid area (2.24 ± 0.53 to 2.28 ± 0.52 mm2, P = 0.001) and volume (9

The psychological consequences of (perceived) ionizing radiation exposure: a review on its role in radiation-induced cognitive dysfunction.

PURPOSE: Exposure to ionizing radiation following environmental contamination (e.g., the Chernobyl and Fukushima nuclear accidents), radiotherapy and diagnostics, occupational roles and space travel has been identified as a possible risk-factor for cognitive dysfunction. The deleterious effects of high doses (≥1.0 Gy) on cognitive functioning are fairly well-understood, while the consequences of low (≤0.1 Gy) and moderate doses (0.1-1.0 Gy) have been receiving more research interest over the past decade. In addition to any impact of actual exposure on cognitive functioning, the persistent psyc

Comparative RNA-Seq transcriptome analyses reveal dynamic time-dependent effects of 56Fe, 16O, and 28Si irradiation on the induction of murine hepatocellular carcinoma.

BACKGROUND: One of the health risks posed to astronauts during deep space flights is exposure to high charge, high-energy (HZE) ions (Z > 13), which can lead to the induction of hepatocellular carcinoma (HCC). However, little is known on the molecular mechanisms of HZE irradiation-induced HCC. RESULTS: We performed comparative RNA-Seq transcriptomic analyses to assess the carcinogenic effects of 600 MeV/n 56Fe (0.2 Gy), 1 GeV/n 16O (0.2 Gy), and 350 MeV/n 28Si (0.2 Gy) ions in a mouse model for irradiation-induced HCC. Simulated Microgravity Accelerates Alloy Corrosion by Aspergillus sp. via the Enhanced Production of Organic Acids.

Metal corrosion caused by Aspergillus sp. was shown to be significantly enhanced on a space station, but its mechanism is still unknown. To simulate this on earth, the corrosion capability of A. carbonarius on five metal sheets was investigated under simulated microgravity. Also, the effect of metal ions on growth and organic acid production was determined. Results showed that A. carbonarius could corrode all five types of metal, including Ti alloy, aluminum alloy, iron, and aluminum and copper sheet, and the corrosion was intensified under simulated microgravity. Energy dispersive X-ray spect

Multiple simulated spaceflight stressors impact cardiac fibrosis, calcium dynamics, immune function, cytokines and gene variants in rat, Rattus norvegicus.

Space radiation and microgravity pose significant health risks for astronauts conducting deep space exploration. The impact of galactic cosmic radiation (GCR) and weightlessness alone, and their combined effects on cardiac disease, the immune system, cytokines and calcium dynamics should be understood before undertaking such missions. Male WAG/RijCmcr rats, Rattus norvegicus, 8-9 months old were hind limb unloaded (HLU) to simulate exposure to microgravity for 5 days before, during, and 25 days after whole-body exposure to a simplified GCR simulation (simGCRsim) dose of 1.5 Gy that was adminis

Ankle Somatosensation and Lower-Limb Neuromuscular Function on a Lunar Gravity Analogue.

Background/Objectives: The adverse effects of low gravity on human physiology are well documented; however, much of the literature is directed at changes which occur in microgravity (µg: weightlessness) with relatively less documented on changes in hypogravity (<1 g; >µg: gravity less than Earth's but more than microgravity). Somatosensation and neuromuscular control may be of particular importance for astronauts as they prepare for future missions to walk on the Moon. This study aimed to explore the effect of reduced weight bearing (to simulate conditions of hypogravity) on ankle somatosensat

Inflammasome-Inhibiting Nanoligomers Are Neuroprotective against Space-Induced Pathology in Healthy and Diseased Three-Dimensional Human Motor and Prefrontal Cortex Brain Organoids.

The microgravity and space environment has been linked to deficits in neuromuscular and cognitive capabilities, hypothesized to occur due to accelerated aging and neurodegeneration in space. While the specific mechanisms are still being investigated, spaceflight-associated neuropathology is an important health risk to astronauts and space tourists and is being actively investigated for the development of appropriate countermeasures. However, such space-induced neuropathology offers an opportunity for accelerated screening of therapeutic targets and lead molecules for treating neurodegenerative

Biological sex divergence in transcriptomic profiles during the onset of hindlimb unloading-induced atrophy.

Disuse-induced muscle atrophy is a common clinical problem observed mainly in older adults, intensive care units patients, or astronauts. Previous studies presented biological sex divergence in progression of disuse-induced atrophy along with differential changes in molecular mechanisms possibly underlying muscle atrophy. The aim of this study was to perform transcriptomic profiling of male and female mice during the onset and progression of unloading disuse-induced atrophy. Male and female mice underwent hindlimb unloading (HU) for 24, 48, 72, and 168 h (n = 8/group). Muscles were weighed for

Visuomotor Adaptation Brain Changes During a Spaceflight Analog With Elevated Carbon Dioxide (CO2): A Pilot Study.

Astronauts on board the International Space Station (ISS) must adapt to several environmental challenges including microgravity, elevated carbon dioxide (CO2), and isolation while performing highly controlled movements with complex equipment. Head down tilt bed rest (HDBR) is an analog used to study spaceflight factors including body unloading and headward fluid shifts. We recently reported how HDBR with elevated CO2 (HDBR+CO2) affects visuomotor adaptation. Here we expand upon this work and examine the effects of HDBR+CO2 on brain activity during visuomotor adaptation. Eleven participants (34
