Effect of the oral intake of probiotic Pediococcus acidilactici BA28 on Helicobacter pylori causing peptic ulcer in C57BL/6 mice models.

Probiotic lactic acid bacteria are being proposed to cure peptic ulcers by reducing colonization of Helicobacter pylori within the stomach mucosa and by eradicating already established infection. In lieu of that, in vitro inhibitory activity of pediocin-producing probiotic Pediococcus acidilactici BA28 was evaluated against H. pylori by growth inhibition assays. Further, chronic gastritis was first induced in two groups of C57BL/6 mice by orogastric inoculation with H. pylori with polyethylene catheter, and probiotic P. acidilactici BA28 was orally administered to study the eradication and cur

Abrogation of antibody-induced arthritis in mice by a self-activating viridin prodrug and association with impaired neutrophil and endothelial cell function.

OBJECTIVE: To test a novel self-activating viridin (SAV) prodrug that slowly releases wortmannin, a potent phosphoinositide 3-kinase inhibitor, in a model of antibody-mediated inflammatory arthritis. METHODS: The SAV prodrug was administered to K/BxN mice or to C57BL/6 (B6) mice that had been injected with K/BxN serum. Ankle thickness was measured, and histologic changes were scored after a 10-day disease course (serum-transfer arthritis). Protease activity was measured by a near-infrared imaging approach using a cleavable cathepsin-selective probe. Further near-infrared imaging techniques we

Genomic and functional insights into a novel Lactococcus sp. AK05 from Cirrhinus mrigala with potent antagonism against Aeromonas hydrophila.

In aquaculture, addressing antibiotic-resistant fish pathogens underscores the immediate necessity for the identification of candidate probiotic strains with favorable safety and functional attributes as alternatives to antibiotics. We isolated and characterized a new strain of Lactococcus sp., called AK05, from the gut of Cirrhinus mrigala. The phenotypic tests showed that it can survive and grow in harsh conditions, including a pH of 4, and 0.8% bile salts. At 8% NaCl, it survived and exhibited merely a one-log rise in CFU·mL⁻¹, while NaCl levels ≤ 5% facilitated over a 4-log increase in 24

The antimicrobial peptide Microcin C7 inhibits the growth of Porphyromonas gingivalis and improves the perodontal status in a rat model.

AIMS: This study aimed to investigate the antibacterial and anti-inflammatory effects of the antimicrobial peptide Microcin C7 for Porphyromonas gingivalis-associated diseases. METHODS AND RESULTS: Reverse-phase high-performance liquid chromatography revealed that Microcin C7 could remain 25.5% at 12 h in saliva. At a concentration of <10 mg ml-1, Microcin C7 showed better cytocompatibility, as revealed by a hemolysis test and a subchronic systemic toxicity test. Moreover, the minimum inhibitory concentration and minimum bactericidal concentration of Microcin C7 were analyzed using a broth mi

Discovery, Synthesis, and Optimization of Peptide-Based Antibiotics.

The rise of multidrug resistant bacteria has significantly compromised our supply of antibiotics and poses an alarming medical and economic threat to society. To combat this problem, it is imperative that new antibiotics and treatment modalities be developed, especially those toward which bacteria are less capable of developing resistance. Peptide natural products stand as promising candidates to meet this need as bacterial resistance is typically slow in response to their unique modes of action. They also have additional benefits including favorable modulation of host immune responses and oft

Micrococcin P1, a naturally occurring macrocyclic peptide inhibiting hepatitis C virus entry in a pan-genotypic manner.

Hepatitis C virus (HCV) is considered a major public health concern worldwide. Despite recent advances in curing chronic hepatitis C, unmet medical needs still remain, especially due to the high economic burden of therapies. Accordingly, our study aimed to identify affordable novel HCV inhibitors by screening of natural product compound libraries. We identified micrococcin P1, a macrocyclic peptide antibiotic, inhibiting HCV entry in a pan-genotypic manner with an EC50 range of 0.1-0.5 μM. Micrococcin P1 interfered with HCV entry at an attachment step. Furthermore, micrococcin P1 efficiently i

Macrophage environment turns otherwise MccJ25-resistant Salmonella into sensitive.

BACKGROUND: Microcin J25 (MccJ25) is a plasmid-encoded antibiotic peptide produced by Escherichia coli (E. coli). MccJ25 enters into the sensitive E. coli strains by the outer membrane receptor FhuA and the inner membrane proteins TonB, ExbB, ExbD and SbmA. The resistance of Salmonella enterica serovar Typhimurium (S. Typhimurium) to MccJ25 is attributed to the inability of its FhuA protein to incorporate the antibiotic into the cell. RESULTS: In this work we demonstrate that S. Typhimurium becomes notably susceptible to MccJ25 when replicating within macrophages. In order to determine the po

Harnessing microvesicles from plantaricin-enhanced stem cells to trigger apoptosis in colon cancer cells.

BACKGROUND: Colorectal cancer (CRC) remains a leading cause of cancer-related mortality, necessitating the development of selective and cell-free therapeutic strategies. This study evaluated the pro-apoptotic potential of microvesicles (MVs) derived from plantaricin-preconditioned mesenchymal stem cells (MSCs) against CRC cells. METHODS: Human bone marrow-derived MSCs were preconditioned with a sub-IC50 concentration of plantaricin (10 µg/mL). MVs were isolated from conditioned media by differential ultracentrifugation and characterized using flow cytometry (CD9, CD63, CD81), dynamic light sc

An in vivo imaging approach for simultaneously assessing tumor response and cytotoxicity-induced tissue response in chemotherapy.

In chemotherapeutic treatments, while cancer cells are the primary target, cytotoxic side effects are an important consideration. In the current study, we applied an in vivo imaging tool for characterizing chemotherapeutic response in a preclinical setting. The study focused on simultaneously examining the tumor and tissue response as a result of treatment with bortezomib, a mainstay proteasome inhibitor for treating multiple myeloma, in a preclinical model. OPM-2 tumor-bearing SCID-beige mice were designated as control or treated with bortezomib (1 mg/kg, i.v., every 4 days) (n = 8 per group)

[Streptococcal respiratory infection in large organized children's collectives and experience in optimizing its prevention].

Epidemiological analysis of the manifestations of respiratory streptococcal infection in the all-year-round pioneer camp "Ocean" over 1984-1990 and in similar large organized groups of children revealed some common regularities manifested as consecutively repeating morbidity rises in acute respiratory diseases, tonsillitis, scarlet fever over a year (rotations 1-3 and 9). Time course of the disease incidence over each rotation (month) had a specific pattern as well. Trials of our newly developed system of prevention and treatment with tomicid showed its high effectiveness, which manifested by

Identification, phenotypic characterization, and assessment of potential probiotic properties of lactic acid bacteria isolated from the Algerian traditional cheese "El-Klila".

El-Klila is a popular, traditional, spontaneously fermented Algerian cheese. Here, we report the identification and characterization of lactic acid bacteria isolated from El-Klila. A total of 26 isolates were identified, typed, and several phenotypic traits were determined to assess their potential as putative probiotics using a combination of pheno- and genotypic methods. Using initial phenotypic characterization followed by rep-PCR-fingerprinting, as well as 16S rRNA and recA gene sequencing, the isolates were identified as Lactiplantibacillus plantarum (73% of isolates), Lacticaseibacillus

Quorum sensing in oral multispecies biofilms: from interkingdom communication to therapeutic targeting.

Oral biofilms are polymicrobial ecosystems where quorum sensing (QS) governs community assembly, virulence and homeostasis. This narrative review synthesizes evidence on QS pathways in key periodontal and cariogenic pathogens, emphasizing interspecies synergy, cross-kingdom interactions, and disease progression. We surveyed English-language publications up to 2026 across PubMed, Web of Science, and Scopus. Our analysis reveals three major QS circuits: the widely conserved LuxS/AI-2 system facilitating cooperative and competitive behaviors, such as Fusobacterium nucleatum-red complex coaggregat

Identification of poultry commensal bacteria with the ability to inhibit the growth of Salmonella enterica serovar Typhimurium in co-culture.

BACKGROUND: The widespread increase of antibiotic resistant bacteria (ARB) in agriculture has led to an interest in improving the competitive exclusion ability of the microbiota in livestock through the application of probiotics. In this study, we examined a collection of gram-negative bacteria isolated from healthy chickens for their ability to inhibit the growth of Salmonella enterica serovar Typhimurium. METHODS: Twenty-five bacteria isolated from healthy chickens were mixed at a 1:1 or a 10:1 ratio with S. enterica ser. Typhimurium and grown overnight in co-culture on a solid media that c

Characterization of the probiotic properties of Lacticaseibacillus rhamnosus LR6 isolated from the vaginas of healthy Korean women against vaginal pathogens.

The human microbiome exhibits intricate populations across the body, with the vaginal tract serving as an ecosystem characterized by the prevalence of the genus Lactobacillus. Disruptions in the vaginal microbiota, which are frequently linked to variables such as sexual activity, hormonal fluctuations, and excessive use of antibiotics, can result in vaginal dysbiosis and the development of diseases such as bacterial vaginosis (BV) and candidiasis. Lactobacillus species, owing to their capacity to create an acidic environment through the production of lactic acid, have a key function within thi

Serratia marcescens.

Over the last 30 years, Serratia marcescens has become an important cause of nosocomial infection. There have been many reports concerning the identification, antibiotic susceptibility, pathogenicity, epidemiological investigations and typing of this organism. Accurate identification is important in defining outbreaks. The API 20E system has been used widely, but is not individually satisfactory. The growth of S. marcescens in the environment has been investigated in relation to water, disinfectants and plastics such as blood bags. Certain extracellular products are unique to S. marcescens. Assessment of gut microbiota dynamics and probiotic impact on HPV-associated head and neck cancer using 16S rRNA sequencing and cytokine profiling.

INTRODUCTION: Human papillomavirus (HPV) is a major etiological factor in head and neck cancers, primarily through the oncogenic activities of viral proteins E6 and E7, which disrupt the p53 and retinoblastoma (Rb) tumor suppressor pathways. This disruption promotes uncontrolled cell proliferation, immune dysregulation, chronic inflammation, and malignant transformation. Although probiotics have emerged as promising modulators of the microbiome and host immune responses, their mechanistic role in HPV-associated head and neck cancer remains insufficiently understood. Therefore, this study inves

Lactobacilli Isolated From Wild Boar (Sus scrofa) Antagonize Mycobacterium bovis Bacille Calmette-Guerin (BCG) in a Species-Dependent Manner.

Background: Wildlife poses a significant burden for the complete eradication of bovine tuberculosis (bTB). In particular, wild boar (Sus scrofa) is one of the most important reservoirs of Mycobacterium bovis, the causal agent of bTB. Wild boar can display from mild TB lesions, usually found in head lymph nodes, to generalized TB lesions distributed in different anatomical regions; but rarely clinical signs, which complicates the diagnosis of Mycobacterium bovis infection and bTB control. Among the possibilities for this variability in lesion distribution is the influence of the host-beneficial
