Alkaline phosphatase from venom of the endoparasitoid wasp, Pteromalus puparum.

Using chromogenic substrates 5-bromo-4-chloro-3'-indolyl phosphate and nitro blue tetrazolium, alkaline phosphatase (ALPase) was histochemically detected in the venom apparatus of an endoparasitoid wasp, Pteromalus puparum L. (Hymenoptera: Pteromalidae). Ultrastructural observations demonstrated its presence in the secretory vesicles and nuclei of the venom gland secretory cells. Using p-nitrophenyl phosphate as substrate to measure enzyme activity, the venom ALPase was found to be temperature dependent with bivalent cation effects. The full-length cDNA sequence of ALPase was amplified from th

Mating biology of the leaf-cutting ants Atta colombica and A. cephalotes.

Copulation behavior has often been shaped by sexually selected sperm competition or cryptic female choice. However, manipulation of previously deposited ejaculates is unknown in the social Hymenoptera and the degree to which sperm competes after insemination or is actively selected by females has remained ambiguous. We studied the mating process in the leaf-cutting ants Atta colombica and A. cephalotes, which belong to one of the few derived social insect lineages where obligate multiple mating has evolved. As copulations often occur at night and in remote places, direct observations were impo

Olfactory responses of banana weevil predators to volatiles from banana pseudostem tissue and synthetic pheromone.

As a response to attack by herbivores, plants can emit a variety of volatile substances that attract natural enemies of these insect pests. Predators of the banana weevil, Cosmopolites sordidus (Germar) (Coleoptera: Curculionidae) such as Dactylosternum abdominale (Coleoptera: Hydrophilidae) and Pheidole megacephala (Hymenoptera: Formicidae), are normally found in association with weevil-infested rotten pseudostems and harvested stumps. We investigated whether these predators are attracted to such environments in response to volatiles produced by the host plant, by the weevil, or by the weevil

Machine learning enables early risk stratification of hymenopteran stings: evidence from a tropical multicenter cohort.

BACKGROUND: Hymenopteran stings (from bees, wasps, and hornets) can trigger severe systemic reactions, especially in tropical regions, risking patient safety and emergency care efficiency. Accurate early risk stratification is essential to guide timely intervention. OBJECTIVE: To develop and validate an interpretable machine learning model for early prediction of severe outcomes following hymenopteran stings. METHODS: We retrospectively analyzed 942 cases from a multicenter cohort in Hainan Province, China. Questionnaires with >20% missing data were excluded. Mean substitution was applied fo

Rapid Change in FcεRI Occupancy on Basophils After Venom Immunotherapy Induction.

Specific venom immunotherapy (VIT) in patients with hymenoptera venom allergy (HVA) represents a well-studied approach to reduce the severity of a possible anaphylactic reaction. Currently, data on mechanisms of tolerance induction at the cellular level within the first hours of therapy are lacking. To address this, total and unoccupied high-affinity IgE receptor (FcεRI) numbers per basophil, soluble FcεRI (sFcεRI) and serum tryptase levels were measured before and after the first day of VIT induction in HVA patients. Additionally, basophil activation tests (BATs) were performed at those time

Epidemiology of insect venom sensitivity.

The prevalence of insect sting reaction and of venom sensitization in adults is unknown. We report the results of intake evaluation of a stratified random sample of a large adult population previously studied for the determinants of atopic disease. In 269 subjects, the prevalence of systemic allergic sting reactions was 3.3% and 26.5% had IgE antibodies to venom demonstrated by skin test or radioallergosorbent test. Asymptomatic sensitization (positive venom skin test) was observed in 15% of subjects with no history of an allergic sting reaction. Positive venom skin tests were more frequent in

Phenology of Asian Citrus Psyllid (Hemiptera: Liviidae), With Special Reference to Biological Control by Tamarixia radiata , in the Residential Landscape of Southern California.

Since its discovery in 2008, the pestiferous Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Liviidae), has become widely established in residential citrus trees throughout southern California. In 2011, Tamarixia radiata (Waterston) (Hymenoptera: Eulophidae), a host-specific parasitoid of D. citri , sourced from Punjab Pakistan, was introduced into California as part of a classical biological program aimed at suppressing D. citri populations in urban areas. Despite these release efforts, little is known about the population dynamics of D. citri in urban citrus or the efficacy of T.

Effect of Leaf Maturity on Host Habitat Location by the Egg-Larval Parasitoid Ascogaster reticulata.

Adoxophyes honmai, a serious pest of tea plants, prefers to lay eggs on mature tea leaves rather than young leaves. Here, we examined a hypothesis that Ascogaster reticulata, an egg-larval parasitoid of A. honmai, increases the likelihood of encountering host egg masses by searching mature tea leaves when host-derived cues are not available. In a dual-choice bioassay using a four-arm olfactometer, A. reticulata preferred odor from intact, mature leaves versus young leaves. Based on volatile analysis with gas chromatography-mass spectrometry (GC-MS), we identified 5 and 10 compounds from mature

Gene expression analysis predicts insect venom anaphylaxis in indolent systemic mastocytosis.

BACKGROUND: Anaphylaxis to insect venom (Hymenoptera) is most severe in patients with mastocytosis and may even lead to death. However, not all patients with mastocytosis suffer from anaphylaxis. The aim of the study was to analyze differences in gene expression between patients with indolent systemic mastocytosis (ISM) and a history of insect venom anaphylaxis (IVA) compared to those patients without a history of anaphylaxis, and to determine the predictive use of gene expression profiling. METHODS: Whole-genome gene expression analysis was performed in peripheral blood cells. RESULTS: Twen

Venom of Euplectrus separatae causes hyperlipidemia by lysis of host fat body cells.

Although the lepidopteran larva Pseudaletia separata is attacked by the gregarious ectoparasitoid Euplectrus separatae, it continues to feed and grow. Lipid concentration in the hemolymph of the parasitized host was higher than that of the nonparasitized host from 3 to 8 days after parasitization. Artificial injection of parasitoid venom also elevated lipid concentration in the host hemolymph. One day after venom injection the host's fat body contained many lipid particles, but most of the lipid particles disappeared 7 days later. Light microscopy and transmission electron microscopy showed th

Evaluating the effectiveness of caffeine-based baits against two pest ant species.

The black cocoa ant, Dolichoderus thoracicus (Smith, 1860) (Hymenoptera: Formicidae) is a pest species capable of reaching high population densities in diverse habitats. In Taiwan, infestations are associated with intrusions into residential homes, damage to electrical equipment, and decreased agricultural productivity. Although baiting programs using borax-based or boric acid-based ant baits have been implemented, these efforts often yield unsatisfactory results. This study evaluated the potential of caffeine, a naturally occurring bioactive compound, as an active ingredient in ant baits for

The angiotensinogen AGT p.M235T gene polymorphism may be responsible for the development of severe anaphylactic reactions to insect venom allergens.

BACKGROUND: Insect venom allergy (IVA) is present in 1-3% of the population. A group of patients with high specific IgE do not react to stings. In contrast, a proportion of patients with IVA have low specific IgE levels. These findings indicate that factors other than specific IgE may also be involved in IVA. Dysfunction of the renin-angiotensin system (RAS) has been described as a potential factor in IVA. The objective of this study was to determine the prevalence of angiotensin AGT p.M235T and angiotensin-converting enzyme ACE I/D, I/I, D/D gene polymorphisms in patients with IVA and to rela

Epidemiology of anaphylaxis.

PURPOSE OF REVIEW: The prevalence of allergic disorders has more than doubled in the last two decades leading to increased community concern and anxiety, and unprecedented demand for allergy-specialist services. However, although allergic reactions are common, anaphylaxis is uncommon and fatal anaphylaxis is rare. This review examines recent developments in the epidemiology of anaphylaxis, focusing on new information that may assist in identifying those at increased risk of severe reactions and adverse outcomes. RECENT FINDINGS: Recent studies suggest an increase in prevalence of anaphylaxis

Allergic reactions to vespids: comparison of sensitivities to two species in a Mediterranean area.

We have studied a group of twenty-seven patients who suffer allergic reactions to vespids stings. Specific IgE antibodies to venom extracts from Polistes gallicus and Vespula germanica were measured by RAST, and the crossreactivity between the two venoms was compared using the RAST inhibition technique. We concluded that, in southern Spain, sensitization to P. gallicus was more prevalent than that to V. germanica, with 44% of the subjects in this study reacting to P. gallicus compared with 33% to V. germanica. However, there was a considerable degree of crossreactivity between the two species.

A sterility-associated long noncoding RNA involved in honey bee caste determination and adult queen and worker fertility.

Insect sociality is a major evolutionary transition based on the suppression of worker reproduction in favor of a queen's reproductive monopoly. Although sophisticated insect societies have arisen independently on multiple occasions in the Hymenoptera (bees, ants, and wasps), how worker sterility evolved through natural selection is not fully understood. In honey bees (Apis mellifera), dietary cues (royal or worker jelly) and social chemical signals (e.g., queen pheromones) promote caste differentiation by modulating the levels of juvenile hormone (JH), the key physiological orchestrator of ca

The Platelet Activating Factor-Platelet Activating Factor Acetylhydrolase Enzyme Axis in Anaphylaxis: Current Evidence and Future Perspectives.

Anaphylaxis is a severe systemic hypersensitivity reaction characterized by rapid onset, unpredictable clinical course, and potentially fatal outcomes. Although its diagnosis remains primarily clinical, the identification of biomarkers capable of improving risk stratification and understanding disease mechanisms remains an important unmet need. Among the mediators implicated in anaphylaxis, platelet-activating factor (PAF) has emerged as a key effector molecule involved in vascular permeability, bronchoconstriction, platelet activation, and cardiovascular dysfunction. The biological activity o

Mastoparan as a Potential Cause of Hemolytic Anemia Due to a Suspected Single Wasp Sting in a Dog.

OBJECTIVE: In people, severe Hymenoptera stings can lead to systemic allergic reactions, acute kidney injury, rhabdomyolysis, hemolysis, shock, and even death. Hemolytic anemia as a result of multiple honeybee stings has been reported in dogs; however, there are no prior reports of hemolytic anemia due to wasp stings in the dog. In this report, we present a case of a dog that developed hemolytic anemia after a single wasp sting. We also report the vulnerability of RBCs to the wasp venom using an in vitro hemolysis assay.
