Two different Clostridium perfringens strains produce different levels of necrotic enteritis in broiler chickens.

Subclinical necrotic enteritis (NE) is primarily caused by the gram-positive bacterium, Clostridium perfringens (Cp). The trend towards removal of in-feed antimicrobials and subsequent increased emergence of infection in poultry has resulted in a wide interest in better understanding of the mechanism behind this disease. The virulence of NE, to a large extent, depends on the virulence of Cp strains. Thus, this study was to assess how 2 different strains of Cp affect performance and gut characteristics of broiler chickens. Ross 308 male broilers (n = 468) were assigned to a 2 × 3 factorial arra

Bacteria within the Gastrointestinal Tract Microbiota Correlated with Improved Growth and Feed Conversion: Challenges Presented for the Identification of Performance Enhancing Probiotic Bacteria.

Identification of bacteria associated with desirable productivity outcomes in animals may offer a direct approach to the identification of probiotic bacteria for use in animal production. We performed three controlled chicken trials (n = 96) to investigate caecal microbiota differences between the best and poorest performing birds using four performance measures; feed conversion ratio (FCR), utilization of energy from the feed measured as apparent metabolisable energy, gain rate (GR), and amount of feed eaten (FE). The shifts in microbiota composition associated with the performance measures w

Fibrolytic capacity of the gut microbiome in captive African elephants (Loxodonta africana): a pilot study.

African elephants (Loxodonta africana) depend on the fibrolytic capacity of their gut microbiome (GMB) to efficiently utilize fibrous plant material. However, knowledge is missing on the temporal stability and functional dynamics (especially fibrolytic capacity) of their GMB. Likewise, information on the associated volatile fatty acids (VFA) production in the elephant's hindgut is scarce. In this pilot study, we collected feces from five captive African elephants on two occasions separated by 12 months to explore the GMB composition and its changes over time. We characterized the GMB's fibroly

[Construction of a long-lived engineered yeast and its efficient catalytic synthesis of ursodeoxycholic acid].

Ursodeoxycholic acid(UDCA) is a first-line drug for the clinical treatment of hepatobiliary diseases. The bioconversion of chenodeoxycholic acid(CDCA) based on whole-cell catalysis provides an important alternative approach for its green production. However, during fermentation, cellular aging and CDCA-induced stress lead to decreased stability of the catalytic system, which severely restricts its industrial application. This study took the extension of cellular chronological lifespan as the entry point, and systematically evaluated the effects of overexpressing the chronological lifespan regu

Oral and gut microbiota dysbiosis with strengthened oral-gut connectivity in post-stroke cognitive impairment.

BACKGROUND: Microbiome studies in post-stroke cognitive impairment (PSCI) have focused on the gut, while upstream oral dysbiosis and oral-gut axis signatures remain undercharacterized. We profiled paired oral and gut microbiota in PSCI to assess coordinated alterations and cross-site associations. METHODS: This single-center cross-sectional study enrolled 133 post-stroke participants (64 PSCI). Paired tongue-coating and fecal samples underwent 16S rRNA gene sequencing. We compared α/β diversity, taxonomic composition, and predicted functional pathways at both sites, quantified within-individu

The gut microbiota metabolite isovalerate enhances the epithelial barrier function in cell monolayers derived from porcine ileum organoids.

The gut microbiota produces numerous metabolites that influence the epithelial barrier function. Bacterial catabolism of amino acids produces a wide variety of metabolites whose effects on the intestinal epithelium remain to be identified. In this study, we investigated the effects of amino acid-derived metabolites (isovalerate, isobutyrate, 2-methylbutyrate, 5-aminovalerate, cadaverine, putrescine, and tryptamine) in cell monolayers derived from porcine ileum organoids. Our results show that the branched-chain fatty acid (BCFA) isovalerate improved the epithelial barrier function, as assessed

Fecal microbiome and metabolome dynamics during immunotherapy-based total neoadjuvant therapy in rectal cancer: associations with treatment response and toxicity.

BACKGROUND: Immunotherapy-based total neoadjuvant therapy (iTNT) is a promising strategy for microsatellite-stable locally advanced rectal cancer (LARC), yet therapeutic response and treatment-related toxicity remain heterogeneous. Integrated fecal microbiome and metabolome profiling may provide non-invasive biomarkers and functional clues for optimizing iTNT. METHODS: We conducted a longitudinal fecal multi-omics study using samples from patients with microsatellite-stable LARC enrolled in the TORCH trial (NCT04518280). A total of 102 fecal samples were collected before treatment, during tre

The Effect of Lentinus edodes Stem Powder on the Growth Performance Immune Responses and Gut Microbiota of Sika Deer Fawns.

As the main byproduct of edible mushroom processing, mushroom stems are often discarded due to their rough texture and poor palatability, resulting in resource waste. However, as a ruminant animal, sika deer has good fiber digestion ability, providing the possibility for the resource utilization of mushroom stems. This study evaluated the effects of dietary supplementation with Lentinus edodes stem powder (LESP) on growth performance, nutrient digestibility, serum biochemical parameters, immune responses, and gut microbiota composition in sika deer fawns. A total of 120 weaned fawns (120 ± 10

Multi-omics analyses unveil gut microbiota and metabolites signatures in deoxycholic acid-associated intestinal inflammation.

OBJECTIVE: High-fat diet (HFD) is closely related to the increased incidence of inflammatory bowel disease (IBD), and excessive fecal deoxycholic acid (DCA) induced by HFD makes significant contribution to the colonic inflammation. However, the precise mechanisms remain unclear. This study aims to explore the association between DCA-induced alteration of gut microbiota as well as related metabolites and intestinal inflammation. METHODS: Wild-type C57BL/6 J mice were orally administrated with or without 0.2% DCA for 12 weeks, then the alteration of gut microbiota signature and fecal metabolite

Admission Gut and Plasma-Derived Signatures Associated With Severity and 90-day Outcome in Hepatitis A-Related and Drug-Induced Acute Liver Failure.

Acute liver failure (ALF) due to hepatitis A (ALF-A) has high mortality, but admission-day markers associated with disease severity and outcome are unknown. We aimed to define gut microbiome and plasma multi-omics signatures associated with severity and 90-day outcomes in ALF-A, compared to drug-induced ALF (ALF-D) as non-viral ALF group. ALF patients (aged 26 ± 9 years; 50.7% male, 49.3% female) with IgM HAV positive (ALF-A, n = 33), ALF-D (n = 38) were recruited along with acute viral hepatitis (AVH-A, n = 27) and healthy subjects (HC, n = 20). Stool bacteria profiling was done at D0; plasma

The Hidden Link Between Intestinal Helminthiasis, Gut Microbiome Alterations, and Colorectal Cancer Risk: A Systematic Review.

Colorectal cancer (CRC) is an increasing health concern in low- and middle-income countries (LMICs), especially in Africa, driven by dietary shifts, urbanisation, infections, and limited treatment access. The gut microbiome plays a central role in CRC, while soil-transmitted helminths (STHs) exert complex effects that can promote or mitigate risk depending on species, infection intensity, and host context. This systematic review synthesised 17 human studies (2000-2026) examining helminth impacts on gut microbial diversity, revealing a dualistic pattern. Several studies reported that chronic or

Gut microbiota causally affects ulcerative colitis by potential mediation of plasma metabolites: A Mendelian randomization study.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease with a multifactorial etiology, including genetic, immunological, and environmental factors, as well as alterations in the gut microbiome and plasma metabolites. The interplay between these factors is complex and not fully elucidated, particularly regarding the potential mediation of metabolites in the relationship between gut microbiota and UC. We performed a Mendelian randomization (MR) study to investigate the causal associations between gut microbiota, plasma metabolites, and UC. The study utilized a two-sample MR approach to

Integrative analysis of pancreatic microbiota and metabolome in patients with pancreatic ductal adenocarcinoma.

The importance of microbiota-associated metabolites in pancreatic ductal adenocarcinoma (PDAC) tumorigenesis highlights the need to investigate the microbiota and metabolome along the normal-adjacent-carcinoma sequence to explore their mechanistic implications in PDAC tumorigenesis. We characterized the pancreatic microbiota and metabolome in four distinct tissue groups using 16S rRNA amplicon sequencing and untargeted metabolomics: tumor tissues from 23 PDAC patients (PDAC T), their matched para-carcinoma tissues (PCT), tumor tissues from 13 pancreatic benign tumor patients (Control T), and t
