Visuomotor Adaptation Brain Changes During a Spaceflight Analog With Elevated Carbon Dioxide (CO2): A Pilot Study.

Astronauts on board the International Space Station (ISS) must adapt to several environmental challenges including microgravity, elevated carbon dioxide (CO2), and isolation while performing highly controlled movements with complex equipment. Head down tilt bed rest (HDBR) is an analog used to study spaceflight factors including body unloading and headward fluid shifts. We recently reported how HDBR with elevated CO2 (HDBR+CO2) affects visuomotor adaptation. Here we expand upon this work and examine the effects of HDBR+CO2 on brain activity during visuomotor adaptation. Eleven participants (34

Intraocular pressure and choroidal thickness respond differently to lower body negative pressure during spaceflight.

Spaceflight-associated neuro-ocular syndrome (SANS) develops during long-duration (>1 mo) spaceflight presumably because of chronic exposure to a headward fluid shift that occurs in weightlessness. We aimed to determine whether reversing this headward fluid shift with acute application of lower body negative pressure (LBNP) can influence outcome measures at the eye. Intraocular pressure (IOP) and subfoveal choroidal thickness were therefore evaluated by tonometry and optical coherence tomography (OCT), respectively, in 14 International Space Station crewmembers before flight in the seated, sup

Mechanical loading causes site-specific anabolic effects on bone following exposure to ionizing radiation.

During spaceflight, astronauts will be exposed to a complex mixture of ionizing radiation that poses a risk to their health. Exposure of rodents to ionizing radiation on Earth causes bone loss and increases osteoclasts in cancellous tissue, but also may cause persistent damage to stem cells and osteoprogenitors. We hypothesized that ionizing radiation damages skeletal tissue despite a prolonged recovery period, and depletes the ability of cells in the osteoblast lineage to respond at a later time. The goal of the current study was to test if irradiation prevents bone accrual and bone formation

Modulation of the DNA-damage response to HZE particles by shielding.

Ions of high atomic number and energy (HZE particles) pose a significant cancer risk to astronauts on prolonged space missions. On Earth, similar ions are being used for targeted cancer therapy. The properties of these particles can be drastically altered during passage through spacecraft shielding, therapy beam modulators, or the human body. Here, we have used pertinent responses to DNA double-strand breaks (DSBs) to understand the consequences of energy loss versus nuclear fragmentation of Fe ions during passage through shielding or tissue-equivalent materials. Microneurography as a tool in clinical neurophysiology to investigate peripheral neural traffic in humans.

Microneurography is a method using metal microelectrodes to investigate directly identified neural traffic in myelinated as well as unmyelinated efferent and afferent nerves leading to and coming from muscle and skin in human peripheral nerves in situ. The present paper reviews how this technique has been used in clinical neurophysiology to elucidate the neural mechanisms of autonomic regulation, motor control and sensory functions in humans under physiological and pathological conditions. Microneurography is particularly important to investigate efferent and afferent neural traffic in unmyeli

Cleared for Launch: Pre-flight Medical Assessment in the Commercial Spaceflight Era.

Commercial human spaceflight has expanded markedly since 2021, with a growing cohort of civilian participants having undertaken suborbital, orbital free-flyer, and International Space Station visiting-crew missions. Pre-flight medical assessment of this population, however, continues to rely on regulatory guidance that predates the first civilian orbital flight by approximately fifteen years and stratifies eligibility on a single dimension of G-load exposure. The evidence accumulated since on spaceflight-associated neuro-ocular syndrome, on centrifuge-analog tolerance in participants with chro

Predicting how varying moisture conditions impact the microbiome of dust collected from the International Space Station.

BACKGROUND: The commercialization of space travel will soon lead to many more people living and working in unique built environments similar to the International Space Station, which is a specialized closed environment that contains its own indoor microbiome. Unintended microbial growth can occur in these environments as in buildings on Earth from elevated moisture, such as from a temporary ventilation system failure. This growth can drive negative health outcomes and degrade building materials. We need a predictive approach for modeling microbial growth in these critical indoor spaces. RESUL

Rumination time as an indicator of stress in the first thirty days after calving.

The objectives of this study were to examine the option of being able to use rumination time (RT) as a form of stress indicator in the first thirty days after calving, and to determine the relationship between rumination time, blood cortisol levels, and lactate concentration levels in dairy cows during the first thirty days after calving. Ninety cows which produced milk (DIM) within 1-30 days were selected and categorised into the following groups: the first group (1) fell within 1-7 days after parturition (dpp) (n=30); the second group (2) fell within 8-14dpp (n=30); and the third group (3) f

Can rumination time and some blood biochemical parameters be used as biomarkers for the diagnosis of subclinical acidosis and subclinical ketosis?

According to the past reports, the utility value of monitoring rumination time (RT) around the time at which calving takes place and, in particular, during the first week of lactation, is a way of identifying in a timely fashion those cows that are at a greater level of risk when it comes to developing disease in early lactation. Recent reports have focused on the role of minerals in disease resistance in ruminants, but little is known about the concentrations blood parameters in dairy cows with subclinical acidosis and subclinical clinical ketosis. According this we hypothesised that ruminati

Effects of Command and Control Vehicle (C2V) operational environment on soldier health and performance.

The purpose of this project was to use NASA technology to assist the US Army in the assessment of motion sickness incidences and effects on soldier performance and mood states within the Command and Control Vehicle (C2V). Specific objectives were (1) to determine if there was a significant difference between three internal configurations of the C2V and/or between seats within these vehicles; (2) to determine if there was a significant difference between the park, move, or short-halt field conditions; and (3) to validate a method of converging indicators developed by NASA to assess environmenta

Low-dose simulated galactic cosmic radiation exposure after hindlimb unloading has long-term impact on skeletal muscle metabolic proteins in female mice.

Spaceflight stressors, including microgravity-induced unloading and galactic cosmic radiation (GCR), acutely disrupt mitochondrial function and contribute to skeletal muscle atrophy. The long-term remodeling of skeletal muscle following combined unloading and radiation exposure remains poorly understood. We investigated protein abundance changes 9-mo postexposure to combined unloading and radiation exposure. Female, 6-mo-old, C57Bl/6J mice underwent 5 days of hindlimb unloading (HU) or weight-bearing (WB) conditions, followed by 0, 0.5, or 1.5 Gy of simulated GCR exposure using the simplified

Knee and Hip Joint Cartilage Damage from Combined Spaceflight Hazards of Low-Dose Radiation Less than 1 Gy and Prolonged Hindlimb Unloading.

Reduced weight bearing, and to a lesser extent radiation, during spaceflight have been shown as potential hazards to astronaut joint health. These hazards combined effect to the knee and hip joints are not well defined, particularly with low-dose exposure to radiation. In this study, we examined the individual and combined effects of varying low-dose radiation (≤1 Gy) and reduced weight bearing on the cartilage of the knee and hip joints. C57BL/6J mice (n = 80) were either tail suspended via hindlimb unloading (HLU) or remained full-weight bearing (ground). On day 6, each group was divided and

Pharmaceutical countermeasures have opposite effects on the utricles and semicircular canals in man.

INTRODUCTION: Sensory conflicts in the vestibular system lead to motion sickness of which space motion sickness (SMS) is a special case. SMS affects up to 70% of the astronauts during the first 3 days in space. The search for effective countermeasures has led to several nonpharmacological and pharmacological approaches. The current study focuses on the effects of lorazepam (1 mg), meclizine (25 mg), promethazine (25 mg), and scopolamine (0.4 mg) on the vestibular system, with special focus on the canal and otolith functions separately. METHODS: The study had a placebo-controlled, single blind

Vestibular factors influencing the biomedical support of humans in space.

This paper will describe the biomedical support aspects of humans in space with respect to the vestibular system. The vestibular system is thought to be the primary sensory system involved in the short-term effects of space motion sickness although there is increasing evidence that many factors play a role in this complex set of symptoms. There is the possibility that an individual's inner sense of orientation may be strongly coupled with the susceptibility to space motion sickness. A variety of suggested countermeasures for space motion sickness will be described. Although there are no known

The effect of combined compression and tactile stimulation on ankle somatosensation in a lunar gravity lower limb load analog.

Ankle somatosensation appears to be negatively affected when in simulated hypogravity (gravity, 1 > g < 0). Developing countermeasures to reduce this negative effect is necessary for sensorimotor control as astronauts prepare to explore the Moon. Head-elevated supine lying has been found to be an effective method in simulating the physiological impact of hypogravity by reducing the weight-bearing capacity through the lower limbs. This study investigated whether wearing a combined compression sock with plantar textured sole (compression-tactile sock) is associated with enhanced somatosensory ac

Gas embolism and surfactant-based intervention: implications for long-duration space-based activity.

Intravascular gas embolism can occur with decompression in space flight, and it commonly occurs during cardiac and vascular surgery. Intravascular bubbles may be deposited into any end organ such as the heart or the brain. Surface interactions between the bubble and the endothelial cells lining the vasculature result in serious impairment of blood flow and can lead to heart attack, stroke, or even death. Surfactant-based intervention is a novel treatment for gas embolism. Intravascular surfactant can adsorb onto the gas-liquid interface and compete with blood-borne macromolecules for interfaci

Ultrasound-based Assessment of Jugular Vein Alterations in Response to Simulated Microgravity and Countermeasures: A Systematic Review.

During spaceflight, assessment of jugular vein (JV) alterations is critical as it reflects systemic responses to fluid redistribution and carries a risk of thrombosis, a potentially fatal condition if undetected. Imaging options during spaceflight are restricted, making ultrasound the only feasible modality. However, available data on JV alterations are from small astronaut cohorts (typically n <12), limiting statistical power and robust validation. As a result, ground-based microgravity simulation studies are required to characterise microgravity-induced physiological changes, including JV al

Molecular determinants of skeletal muscle force loss in response to 5 days of dry immersion in human.

BACKGROUND: Astronauts in Earth's orbit experience microgravity, resulting in a decline of skeletal muscle mass and function. On Earth, models simulating microgravity have shown that the extent of the loss in muscle force is greater than the loss in muscle mass. The reasons behind this disproportionate loss of muscle force are still poorly understood. In the present study, we hypothesize that alongside the loss in skeletal muscle mass, modifications in the expression profile of genes encoding critical determinants of resting membrane potential, excitation-contraction coupling and Ca2+ handling

Alterations in the hepatic drug-metabolizing enzyme network of mice induced by +Gz exposure and its impact on the pharmacokinetics and pharmacodynamics of modafinil.

BACKGROUND: In aviation and aerospace missions, personnel and astronauts are subjected to brief episodes of +Gz acceleration, resulting in overload stress that adversely affects the cardiovascular, respiratory, and nervous systems. This mechanical stress disrupts hemodynamic balance, tissue oxygenation, and physiological homeostasis. Increasing evidence links this stressor to hepatic injury; as the primary organ for drug metabolism, liver dysfunction may alter the expression and activity of drug-metabolizing enzymes and the pharmacokinetic profiles of circadian rhythm-regulating medications, s

Gravity Threshold and Dose Response Relationships: Health Benefits Using a Short Arm Human Centrifuge.

PURPOSE: Increasing the level of gravity passively on a centrifuge, should be equal to or even more beneficial not only to astronauts living in a microgravity environment but also to patients confined to bed. Gravity therapy (GT) may have beneficial effects on numerous conditions, such as immobility due to neuromuscular disorders, balance disorders, stroke, sports injuries. However, the appropriate configuration for administering the Gz load remains to be determined. METHODS: To address these issues, we studied graded G-loads from 0.5 to 2.0g in 24 young healthy, male and female participants,
