Parasitic infections in humans in West Kalimantan (Borneo), Indonesia.

A survey was carried out among inhabitants of eight villages in West Kalimantan Province (Borneo), whereby blood smears were examined for malaria, stools examined for intestinal parasites and sera tested by the indirect hemagglutination test for antibodies to Entamoeba histolytica and toxoplasma gondii. The prevalence of malaria among 3017 people examined was 5.6% (Plasmodium vivax 2.8%, Plasmodium falciparum 2.8%). Brugia malayi microfilariae were found in 3.6% and Wuchereria bancrofti in 0.3%. Ninety-seven percent of 2101 stool specimens examined contained evidence of intestinal parasites. T

The expression of REG 1A and REG 1B is increased during acute amebic colitis.

Entamoeba histolytica, a protozoan parasite, is an important cause of diarrhea and colitis in the developing world. Amebic colitis is characterized by ulceration of the intestinal mucosa. We performed microarray analysis of intestinal biopsies during acute and convalescent amebiasis in order to identify genes potentially involved in tissue injury or repair. Colonic biopsy samples were obtained from 8 patients during acute E. histolytica colitis and again 60 days after recovery. Gene expression in the biopsies was evaluated using microarray, and confirmed by reverse transcriptase quantitative p

Enhanced pro-inflammatory chemokine/cytokine response triggered by pathogenic Entamoeba histolytica : basis of invasive disease.

The virulence of Entamoeba histolytica is governed by adhesion/colonization in the gut which is mediated by a galactose specific lectin. Two morphologically identical but distinct species i.e. pathogenic E. histolytica and non-pathogenic E. dispar, can be differentiated by distinct epitopes in the lectin. Both species bind to colonic epithelial cells, but only E. histolytica infection induces an inflammatory response and subsequent pathogenesis. Thus, comparing the responses of the intestinal cells to pathogenic and non-pathogenic lectins is a point of interest. The pathogenic lectin causes cy

Outbreak of amebiasis in Tbilisi, Republic of Georgia, 1998.

In 1998, we investigated a suspected outbreak of amebic liver abscesses caused by Entamoeba histolytica in the Republic of Georgia, using a case-control study. A questionnaire was administered and blood samples were obtained from cases and controls for serologic diagnosis. Medical records showed that E. histolytica infections were rarely diagnosed before 1998. However, from July through September 1998, 177 cases of suspected amebiasis were identified. Of 52 persons who had diagnosed liver abscesses, 37 (71%) were confirmed serologically to have antibodies against E. histolytica, compared with

The tumor necrosis factor alpha-stimulating region of galactose-inhibitable lectin of Entamoeba histolytica activates gamma interferon-primed macrophages for amebicidal activity mediated by nitric oxide.

Entamoeba histolytica adheres via galactose-lectin (Gal-lectin) to human colonic mucins and intestinal epithelial cells as a prerequisite to amebic invasion. Native Gal-lectin is a protective antigen in the gerbil model of amebiasis. Amino acids 596 to 1082 of Gal-lectin mediate E. histolytica adherence to target cells and stimulate tumor necrosis factor alpha (TNF-alpha) production by naive murine bone marrow macrophages (BMM). Resistance to amebiasis requires an effective cell-mediated immune response against E. histolytica trophozoites mediated by nitric oxide (NO) released from activated m

Lethal recognition between Entamoeba histolytica and the host tissues.

Lethal recognition between Entamoeba histolytica and the tissues and defence systems of the host results in a continuous interplay that determines the development of pathological lesions: (i) we have identified several of the steps and mediators utilized by the trophozoites to destroy host cells by contact-mediated cytolysis; (ii) we have established that the alternative complement system represents the main defence available to the host against the invading parasite. The amoebae recognize target cells by means of a lectin specific for N-acetylgalactosamine-containing surface glycoproteins. Th

Systemic pathology of chimpanzees.

Spontaneous lesions in chimpanzees have been described according to the organ system involved. These lesions have been reported in the literature or noted in chimpanzees at the 6571st Aeromedical Research Laboratory (ARL), Holloman AFB, N. Mex., over a 15-year period. some of the lesions seen at the ARL were the result of overt clinical disease, while others represent incidental findings, the significance and genesis of which were often obscure. Conditions involving the gastrointestinal tract, especially parasitic diseases, have been and continue to be of the highest frequency, with disorders

Household socio-economic position and individual infectious disease risk in rural Kenya.

The importance of household socio-economic position (SEP) in shaping individual infectious disease risk is increasingly recognised, particularly in low income settings. However, few studies have measured the extent to which this association is consistent for the range of pathogens that are typically endemic among the rural poor in the tropics. This cross-sectional study assessed the relationship between SEP and human infection within a single community in western Kenya using a set of pathogens with diverse transmission routes. The relationships between household SEP and individual infection wi

Detection of excretory Entamoeba histolytica DNA in the urine, and detection of E. histolytica DNA and lectin antigen in the liver abscess pus for the diagnosis of amoebic liver abscess.

BACKGROUND: Amoebic liver abscess (ALA) and pyogenic liver abscesses (PLA) appear identical by ultrasound and other imaging techniques. Collection of blood or liver abscess pus for diagnosis of liver abscesses is an invasive procedure, and the procedure requires technical expertise and disposable syringes. Collection of urine is a noninvasive procedure. Therefore, there has been much interest shown towards the use of urine as an alternative clinical specimen for the diagnosis of some parasitic infections. Here, we report for the first time the detection of E. histolytica DNA excreted in the ur

Pathogenesis of disease caused by Entamoeba histolytica: studies of adherence, secreted toxins, and contact-dependent cytolysis.

Amebiasis is due to invasion of the colonic mucosa by the enteric protozoan Entamoeba histolytica. The biology of the parasite and the pathogenic mechanisms of E. histolytica have been extensively studied. Recent studies provide new insights into the mechanism by which amebas colonize the gut, interact with bacterial flora, adhere to and lyse tissue, and combat the host's inflammatory response. Elucidation of the biochemical and cellular bases of the pathogenesis of disease caused by E. histolytica may lead to the development of an immunologic or pharmacologic means of prophylaxis.

Comparison of three commercial multiplex PCR assays for the diagnosis of intestinal protozoa.

Although microscopic examination of stool samples remains the reference method for the diagnosis of intestinal protozoal infections, these techniques are time-consuming and require operators who are experienced and well trained. Molecular biology seems to offer performances at least equivalent in terms of sensitivity and specificity for certain parasites. This study aimed to compare three multiplex PCR assays on 93 prospectively collected positive stools (prospective cohort) and a panel of 12 more Cryptosporidium-positive samples (Cryptosporidium panel). On the prospective cohort, the sensitiv

Epidemiological, clinical, and diagnostic data on intestinal infections with Entamoeba histolytica and Entamoeba dispar among returning travelers.

BACKGROUND: Among travelers returning from the tropics, Entamoeba spp. are among the most frequently detected intestinal parasites, mainly the presumable apathogenic E. dispar and the pathogenic E. histolytica. METHODS: Among 5,378 travelers seeking diagnosis and treatment for intestinal infections at the travel clinic of the University of Munich between 2005 and 2009, 103 laboratory-confirmed amebiasis cases were detected. The study compares the results of various diagnostic tests among these patients, analyzes data on co-infections and clinical symptoms, and determines the risk for acquirin

Three novel octonucleated Entamoeba lineages in pet fancy rats (Rattus norvegicus) in China: evidence from compensatory base change and phylogenetic analyses.

Entamoeba spp. are anaerobic protists primarily inhabiting the digestive tracts of vertebrates, some of which are pathogenic or zoonotic. However, genetic information on Entamoeba spp. in rodents, including rats, remains limited. This study investigated the prevalence and molecular characterization of Entamoeba in pet fancy rats in China. Fecal samples were collected from a total of 152 fancy rats from four pet shops in two provinces of China. Microscopy and PCR targeting the small subunit ribosomal RNA (SSU rRNA) gene were used for detection. Lineage differentiation was evaluated using compen

For many years, microscopic examination of stool samples has been considered to be the "gold standard" for diagnosis of intestinal parasites although the Polymerase Chain Reaction (PCR) analysis is increasingly utilized due to its high accuracy. Recently, PCR has been approved by the World Health Organization as the current method of choice for the diagnosis of Entamoeba histolytica infection. In this study we evaluated a novel immunochromatographic antigen detection rapid test, ImmunoCardSTAT CGE (Meridian Bioscence, Milan, Italy), which has been proposed for the diagnosis of infections cause
